Generic instability and cryptic diversity in Ostreinae (Ostreidae): revisiting Booneostrea Harry, 1985 and describing Ostrea sorosubucula sp. nov., a new endemic oyster from Kuwait.

The oyster traditionally referred to as Booneostrea subucula (Jousseaume in Lamy, 1925) exemplifies persistent generic instability in Ostreinae, driven by misidentified type material and debated synonymies spanning multiple genera, subfamilies and even families. We reassessed the identity of Ostrea subucula and the validity of Booneostrea Harry, 1985 using an integrative approach that combined shell morphology, anatomy and multilocus phylogenetic data. Phylogenetic analyses reveal two deeply divergent, reciprocally monophyletic lineages. The Western Indian Ocean lineage corresponds to O. subuc

Application Research on the Triplex RT-ddPCR Assay for the Simultaneous Detection of Human Astrovirus and Sapovirus in Bivalve Shellfish.

Human astrovirus (HAstV) and sapovirus (SaV) are significant pathogens associated with acute gastroenteritis in humans. This study established a triple reverse transcription-droplet digital polymerase chain reaction (PCR) (RT-ddPCR) assay incorporating MS2 phage as a process control virus for the simultaneous quantification of HAstV and SaV. The assay was validated using 240 bivalve samples, comprising five shellfish species: Ostreidae (n = 43), Ruditapes philippinarum (n = 84), Sinonovacula constricta (n = 27), Scapharca subcrenata (n = 23), and Pectinidae (n = 63). The results indicated that

Dynamics of Mud Blister Worm Infestation and Shell Repair by Oysters.

AbstractMud blister worms bore into oyster shells; and oysters respond to shell penetration by secreting new layers of shell, resulting in mud blisters on inner surfaces of oyster shells. We conducted two experiments in off-bottom oyster farms along Alabama's coast in summer 2017 to explore the dynamics of worm infestation, blister formation, and shell repair. Results support our hypothesis that only a small proportion of worms that bore into oysters cause blisters. Triploid oysters had fewer blisters than diploids, likely because of faster growth and shell repair. We treated oysters to remove

A validated DNA metabarcoding approach for routine authentication of processed bivalves.

Species mislabeling in commercial bivalves represents a growing concern for food authenticity. Given the diversity of traded bivalves, broad taxonomic coverage is essential for untargeted screening. To address this challenge, we developed and validated a DNA metabarcoding approach employing two PCR assays, a singleplex and a duplex, to amplify mitochondrial 16S rDNA fragments (160-203 bp) across seven families: Ostreidae, Pectinidae, Mytilidae, Pharidae, Veneridae, Glycymerididae, and Cardiidae. Taxa were identified at the species or genus level in 38 reference samples and four model food samp

Climate change impacts on oyster aquaculture - Part II: Impact assessment and adaptation measures.

The oyster aquaculture sector plays a major role in food security, providing a sustainable way to obtain food and livelihood for coastal and Island nations. Oysters are one of the preferred choices by aquaculturists because of their resilience to harsh climatic conditions. Nonetheless, climate change will continue to pose threats to its culture. Climate-induced hazards such as floods, storms, disease, and invasive species are some of the key factors limiting oyster production globally. A thriving aquaculture industry needs optimal conditions to maximize exploitation. Here, we continue with the

Active downward propulsion by oyster larvae in turbulence.

Oyster larvae (Crassostrea virginica) could enhance their settlement success by moving toward the seafloor in the strong turbulence associated with coastal habitats. We characterized the behavior of individual oyster larvae in grid-generated turbulence by measuring larval velocities and flow velocities simultaneously using infrared particle image velocimetry. We estimated larval behavioral velocities and propulsive forces as functions of the kinetic energy dissipation rate ε, strain rate γ, vorticity ξ and acceleration α. In calm water most larvae had near-zero vertical velocities despite prop

Changes in Larval Oyster Swimming Behavior with Salinity and Larval Age.

AbstractEastern oysters (Crassostrea virginica) are sessile, relying on a larval phase to disperse in estuaries. Oyster larval swimming behavior can alter dispersal trajectories and patterns of population connectivity. Experiments were conducted to test how both (1) acclimation time to new environmental conditions and (2) larval swimming behavior change with salinity and larval age. Acclimation time to changes in salinity was longest in lower salinity (6 ppt) and decreased with age. To test changes in behavior with salinity, larvae were placed into four salinities (6, 10, 16, and 22 ppt) where

"Nail Disease", an emerging syndrome in the Pacific oyster Magallana gigas cultivated in the Baja California Peninsula, Mexico: the challenge of determining its causal agent(s) and the importance of early warning.

In 2024, aquaculture producers reported unprecedented cases of shell malformations and mortality in Pacific oysters (Magallana gigas) in Baja California Peninsula, Mexico. Due to the shell malformations, they named this condition "nail disease." Sixty oysters from the main affected location were used for shell, bacteriological, fungal, and histopathological analysis. Shell malformations may reach 90% of the surface. Bacteriological identification based on 16S rRNA gene sequencing revealed the presence of Staphylococcus saprophyticus and Flagellimonas marinaquae. Shell decalcification exposed o

Elevated trace element concentrations in Arabian scallops, Natal rock oysters, and Intermediate clams in the Persian Gulf.

BACKGROUNDS: This study aimed to quantify trace element concentrations in Arabian scallops (Mimachlamys sanguinea), Natal rock oysters (Saccostrea cucullata), and Intermediate clams (Tivela damaoides). These elements accumulate over time from water, food, and sediment, potentially leading to toxicity. MATERIALS AND METHODS: Specimens were collected from coastal waters around Larak Island, processed, and analyzed for trace elements including copper (Cu), iron (Fe), cadmium (Cd), lead (Pb), arsenic (As), mercury (Hg), zinc (Zn), vanadium (V), magnesium (Mg), manganese (Mn), selenium (Se), nicke

Spatial and temporal consistency in green algae-induced oyster microbiota dysbiosis and associated increased disease risk.

Growing interest in microorganisms has extended to the microbiota of hosts and how they interact, as many diseases result from a microbiota imbalance. Recent studies on Pacific oysters Magallana gigas have shown that individuals exposed to particular communities, like green algae holobionts from the genus Ulva, were more susceptible to the infection by the Ostreid herpesvirus type 1 (OsHV-1), coinciding with dysbiosis of the bacterial microbiota and modification of host immune response. Considering that the microbiota of macroalgae and oysters vary with time and geographical location, dysbiosi

Direct and indirect ecosystem responses to vehicle compaction of soft sediments.

Change in an ecological community after a disturbance may be a direct impact or may be indirectly mediated through the response of a highly influential species. In northeastern Pacific tidal flats, a native bioturbator, ghost shrimp Neotrypaea californiensis, engineers soft-sediment habitat and interacts antagonistically with bivalve shellfish. Vehicle compaction has been used in pest control of ghost shrimp, but this disturbance lacks quantitative evidence of its efficacy and environmental impacts. Through three large (∼10 ha) experiments in Grays Harbor, Washington, USA, we tested the direct

Genetic detection and discrimination of closely related oyster pathogens for biosecurity and shellfish stock management.

Disease outbreaks among shellfish cause significant declines in both wild fisheries and aquaculture production globally. In Aotearoa New Zealand, a long history of flat oyster infections by the haplosporidian parasite Bonamia exitiosa has influenced fishery dynamics in the Foveaux Strait. B. ostreae, which causes high host mortality, was detected in both wild and farmed oysters in the Marlborough Sounds in 2015 and at Stewart Island in 2017, prompting the development of rapid diagnostic tests with high sensitivity and specificity that can be used to simultaneously monitor both Bonamia species.

Current decontamination challenges and potentially complementary solutions to safeguard the vulnerable seafood industry from recalcitrant human norovirus in live shellfish: Quo Vadis?

Safeguarding the seafood industry is important given its contribution to supporting our growing global population. However, shellfish are filter feeders that bioaccumulate microbial contaminants in their tissue from wastewater discharged into the same coastal growing environments leading to significant human disease outbreaks unless appropriately mitigated. Removal or inactivation of enteric viruses is very challenging particularly as human norovirus (hNoV) binds to specific histo-blood ligands in live oyster tissue that are consumed raw or lightly cooked. The regulatory framework that sets ou

Exploring PAH kinetics in wild vs. transplanted triploid and diploid oysters at a contaminated field site using immunological techniques.

Crassostrea virginica is a well-established bivalve species for biomonitoring persistent organic pollutants such as polycyclic aromatic hydrocarbons (PAH) in aquatic environments. Differing biomonitoring methods employing either wild oysters inhabiting sites of interest or naïve cultured oysters deployed to sites for extended periods can be used for site evaluations. However, important differences in total contaminant concentrations accumulated have been observed between the wild and transplanted groups. Furthermore, although rearing cultured triploid oysters is widely popular in commercial fa

Zinc from oyster tissue as causative factor in mouse deaths in official bioassay for paralytic shellfish poison.

Toxicity (extreme weakness, body temperature drop, cyanosis, some slow deaths) in test mice, upon intraperitoneal injection of standard-method paralytic shellfish poison (PSP) extracts of some PSP-free oysters, is consistent with the relatively high levels of zinc in these extracts. As a rough guideline, the threshold for a toxic response corresponds to a drained tissue zinc level of over 900 micrograms/g. The identification of zinc as the substance responsible has been supported by inducing toxicity in control extracts by spiking with nontoxic levels of zinc, and by eliminating toxicity from

Examination of the potential relationship between boring sponges and pea crabs and their effects on eastern oyster condition.

The eastern oyster Crassostrea virginica provides a number of ecosystem services and is an important commercial fishery species along the US East and Gulf Coasts. Oyster populations have declined dramatically due to overharvesting, habitat loss, and disease. As restoration efforts and aquaculture of oysters continue to increase throughout their range, it is important to consider the impacts of a number of potential oyster pests, including the boring sponge Cliona spp. and the pea crab Zaops (Pinnotheres) ostreum, on oyster populations. Both of these have been demonstrated to reduce oyster grow

Effectiveness of standard UV depuration at inactivating Cryptosporidium parvum recovered from spiked Pacific oysters (Crassostrea gigas).

When filter-feeding shellfish are consumed raw, because of their ability to concentrate and store waterborne pathogens, they are being increasingly associated with human gastroenteritis and have become recognized as important pathogen vectors. In the shellfish industry, UV depuration procedures are mandatory to reduce pathogen levels prior to human consumption. However, these guidelines are based around more susceptible fecal coliforms and Salmonella spp. and do not consider Cryptosporidium spp., which have significant resistance to environmental stresses. Thus, there is an urgent need to eval
