Effects of oxonic acid-induced hyperuricemia on mesenteric artery tone and cardiac load in experimental renal insufficiency.

BACKGROUND: Recent studies suggest a causal role for increased plasma uric acid in the progression of chronic renal insufficiency (CRI). However, uric acid also functions as an antioxidant with possible beneficial effects. METHODS: We investigated the influence of hyperuricemia on mesenteric arterial tone (main and second order branch) and morphology in experimental CRI. Forty-four Sprague-Dawley rats were 5/6 nephrectomized (NX) or Sham-operated and fed 2.0% oxonic acid or control diet for 9 weeks. RESULTS: Oxonic acid feeding elevated plasma uric acid levels 2.4 and 3.6-fold in the NX and

Inhibition by oxonic acid of gastrointestinal toxicity of 5-fluorouracil without loss of its antitumor activity in rats.

The possibility of decreasing the gastrointestinal (GI) toxic effects of 5-fluorouracil (5-FU) on the digestive tract such as its injury of cells and induction of diarrhea, without reducing its antitumor activity, was investigated in rats. Oxonic acid was found to inhibit the phosphorylation of 5-FU to 5-fluorouridine-5'-monophosphate catalyzed by pyrimidine phosphoribosyl-transferase in a different manner from allopurinol in cell-free extracts and intact cells in vitro. On p.o. administration of 5-FU (2 mg/kg) and a potent inhibitor of 5-FU degradation to Yoshida sarcoma-bearing rats, oxonic

Emodin, a Natural Anthraquinone, Increases Uric Acid Excretion in Rats with Potassium Oxonate-Induced Hyperuricemia.

The treatment of hyperuricemia and gout is mostly based on lowering serum uric acid levels using drugs, such as allopurinol, or increasing urinary excretion of uric acid. However, some patients still experience adverse reactions to allopurinol and turn to Chinese medicine as an alternative. Therefore, it is crucial to design a preclinical study to obtain more convincing data on the treatment of hyperuricemia and gout with Chinese medicine. This study aimed to explore the therapeutic effect of emodin, a Chinese herbal extract, in a rat model of hyperuricemia and gout. In this study, we used 36

Puerariae lobatae Radix aqueous extract ameliorates asymptomatic hyperuricemia in a potassium oxonate-induced rat model by dual modulation of uric acid production and excretion.

BACKGROUND: Puerariae lobatae Radix (PLR) is a well-known traditional Chinese medicine and edible natural nutrient, with diverse biological activities, including anti-diabetes, anti-inflammatory, anti-oxidant and liver protection. However, the effects and underlying mechanisms of PLR in hyperuricemia (HUA) are unclear. METHODS: The present study focused on the regulatory effects of aqueous extract from PLR on the asymptomatic hyperuricemia rat model, induced by potassium oxonate. Serum uric acid (SUA), serum blood urea nitrogen (BUN), creatinine (CRE), serum inflammatory factors, anthine oxid

Raising serum uric acid with a uricase inhibitor worsens PKD in rat and mouse models.

Humans are predisposed to gout because they lack uricase that converts uric acid to allantoin. Rodents have uricase, resulting in low basal serum uric acid. A uricase inhibitor raises serum uric acid in rodents. There were two aims of the study in polycystic kidney disease (PKD): 1) to determine whether increasing serum uric acid with the uricase inhibitor, oxonic acid, resulted in faster cyst growth and 2) to determine whether treatment with the xanthine oxidase inhibitor, oxypurinol, reduced the cyst growth caused by oxonic acid. Orthologous models of human PKD were used: PCK rats, a polycys

Mild Hyperuricemia Attenuates Salt-Sensitive Hypertension and Kidney Damage.

BACKGROUND: Asymptomatic hyperuricemia is associated with poor outcomes in kidney and cardiovascular diseases, but its causative role remains controversial. Clinical studies have shown a positive correlation between increased dietary sodium intake and urinary uric acid excretion, suggesting that hyperuricemia may influence salt-sensitive hypertension and associated kidney damage. METHODS: To study the effects of mild hyperuricemia on salt-sensitive hypertension, male and female Dahl SS rats were fed a diet containing a uricase inhibitor (2% oxonic acid) and 4% NaCl (high salt) or a high salt-

Experimental meningitis in the rat: protection by uric acid at human physiological blood concentrations.

The natural peroxynitrite scavenger uric acid was previously shown to be protective in a rat model of pneumococcal meningitis; however, rats have much lower blood uric acid levels than humans. Therefore, we evaluated its therapeutic effect at human physiological blood concentrations. Intraperitoneal pretreatment with uric acid increased its blood concentrations from 44.9+/-10.0 microM in untreated rats to 169.8+/-122.6 microM and reduced the cerebrospinal fluid (CSF) pleocytosis from 12767+/-2520 to 8376+/-2450 cells/microl (P<0.05) and the intracranial pressure from 11.6+/-3.0 to 4.3+/-1.2 mm

The influence of oxonate-induced hyperuricemia and allopurinol on behavioral reactions of random-bred mice.

BACKGROUND: Purine metabolism specificity, namely the high level of uric acid in the blood, is considered to be significant in the appearance of intellectually-developed primates, as well as in later periods of human development, for personal activity and achievements and even for the formation of genius. Nowadays hyperuricemia is mostly associated with the pathogenesis of hypertension and metabolic syndrome. Its influence on the central nervous system is predominantly set aside. METHODS: The behavioral reactions of random-bred mice with uricase inhibition due to potassium oxonate or xanthine

Potassium oxonate induces acute hyperuricemia in the tree shrew (tupaia belangeri chinensis).

Potassium oxonate, a selectively competitive uricase inhibitor, produced hyperuricemia (HUA) in rodents in a previous study. In this study, we employed the tree shrew as an animal model to study potassium oxonate-induced HUA. The effect of allopurinol (ALLO), a uric acid reducer, was also examined in this model. Potassium oxonate at doses of 5, 20, 40, 60, 80, 100, and 1,000 mg/kg was given intraperitoneally to tree shrews. The results showed that potassium oxonate can effectively increase the levels of uric acid in tree shrews at doses ranging from 40 to 100 mg/kg. Semiquantitative RT-PCR sho

Use of the uricase-inhibited rat as an animal model in toxicology.

An accessible, reproducible, and inexpensive animal model for toxicologic evaluation of hyperuricemic conditions has been required for some time. A number of authors have tried to develop such a model by administering high doses of uric acid to various animal species (dog, rabbit, rat) but the potent liver uricase in these species prevented development of sustained hyperuricemia. Johnson et al. [4], Stavric et al. [5], and a number of other investigators [72, 75] successfully used potassium oxonate [63] to block the effect of hepatic uricase and to produce hyperuricemia in rats [4, 5, 68, 69,

β-Carboline alkaloid harmaline alleviates hyperuricemia-mediated renal inflammation by suppressing oxidative stress.

Hyperuricemia is a metabolic disorder marked by elevated serum uric acid levels and is closely linked to the development of gout and progressive renal dysfunction. It is increasingly recognized as a major public health concern due to its association with chronic kidney disease and cardiovascular complications. Hyperuricemia induces kidney impairment through a complex interplay of oxidative and inflammatory stress. Harmaline, a β-carboline alkaloid primarily found in Peganum harmala, exhibits antioxidant and anti-inflammatory activities. Considering the roles of oxidative and inflammatory stres

Podocyte Injury and Albuminuria in Experimental Hyperuricemic Model Rats.

Although hyperuricemia is shown to accelerate chronic kidney disease, the mechanisms remain unclear. Accumulating studies also indicate that uric acid has both pro- and antioxidant properties. We postulated that hyperuricemia impairs the function of glomerular podocytes, resulting in albuminuria. Hyperuricemic model was induced by oral administration of 2% oxonic acid, a uricase inhibitor. Oxonic acid caused a twofold increase in serum uric acid levels at 8 weeks when compared to control animals. Hyperuricemia in this model was associated with the increase in blood pressure and the wall-thicke

Efficient elimination of caffeine from water using Oxone activated by a magnetic and recyclable cobalt/carbon nanocomposite derived from ZIF-67.

To eliminate caffeine, one of the most common pharmaceuticals and personal care products, from water, Oxone (peroxymonosulfate salt) was proposed to degrade it. To accelerate the generation of sulfate radicals from Oxone, a magnetic cobalt/carbon nanocomposite (CCN) was prepared from a one-step carbonization of a cobalt-based Zeolitic Imidazolate Framework (ZIF-67). The resultant CCN exhibits immobilized cobalt and increased porosity, and can be magnetically manipulated. These characteristics make CCN a promising heterogeneous catalyst to activate Oxone for caffeine degradation. Factors affect

Hyperuricemic effects of cholinergic agents in rats.

The hyperuricemic effect of cholinergic agents was studied in rats given potassium oxonate which inhibits urate oxidase and in functionally hepatectomized rats. Acetylcholine slightly increased plasma uric acid in the oxonate-treated animals, and the effect was markedly enhanced by pretreatment with physostigmine. Carbachol alone showed the hyperuricemic effect at a smaller dose than acetylcholine. The hyperuricemic effects of physostigmine plus acetylcholine and carbachol alone were also observed in eviscerated rats with nonfunctioning livers. Nephrectomy markedly potentiated cholinergic agen

Establishment and optimization of a novel mouse model of hyperuricemic nephropathy.

Hyperuricemia is a metabolic disorder characterized by elevated serum uric acid levels. Soluble urate can activate immune responses, and the excessive accumulation of urate in the kidneys results in hyperuricemic nephropathy (HN). However, the lack of an established HN model is a major obstacle to advancing research into the pathogenesis of HN and the development of novel drugs. In this study, we generated and evaluated an optimized mouse model of HN by the combined administration of potassium oxonate and hypoxanthine at various dosages. Our results demonstrated that intraperitoneal injection

Metabolic and cardiovascular effects of chronic mild hyperuricemia in rodents.

Mildly elevated serum uric acid levels are common in people with metabolic syndrome and type 2 diabetes mellitus (T2DM), but whether elevated uric acid has a causal role in the pathogenesis of diabetes remains uncertain. We tested whether chronic mild hyperuricemia in rodents under controlled laboratory conditions can cause glucose intolerance in otherwise healthy animals, or whether it can worsen glucometabolic control in animals that are genetically predisposed to T2DM. We used an established model of experimental hyperuricemia in rodents with potassium oxonate dietary supplementation, which

Occurrence and removal of antibiotics from aquaculture wastewater by solar-driven Fe(VI)/oxone process.

In this study, the occurrence and removal of ten selected antibiotics from aquaculture wastewater by the process solar + Fe(VI)+oxone were investigated. The detection levels of the antibiotics in the aquaculture wastewater samples were at ng/L. The degradation of the selected antibiotics under the process solar + Fe(VI)+oxone followed pseudo-first-order kinetics. As the most abundant antibiotic in the studied aquaculture wastewater, norfloxacin (NFX) was used as the model compound to study the reaction mechanism and detoxification ability of the treatment system, as well as the effects of reac

[The Xanthine Oxidase Inhibitory Activity and Hypouricemic Effects of Crude Drugs Obtained from the Silkworm in Mice].

This study evaluated the effects of crude drugs obtained from the silkworm in mice with oxonic acid-induced hyperuricemia using xanthine oxidase inhibitory activity and plasma uric acid levels. The plasma uric acid level was analyzed using an improved HPLC with UV detection (HPLC-UV) method, which enabled high-sensitivity analysis of a microliter of plasma. Using this method, we evaluated natural products administered orally to the hypouricemic mice. The plasma uric acid level of mice administered a water-soluble extract from silkworm larvae with botrytis (used in traditional Chinese medicine
