The mechanism of action for oxyntomodulin in the regulation of obesity.

Oxyntomodulin, a product of the proglucagon gene, is released from the enteroendocrine L-cells of the gastrointestinal tract after the digestion of food, and acts via glucagon-like peptide 1 receptors in the arcuate nucleus to induce satiety. The administration of oxyntomodulin to animals and humans causes weight loss by reducing food intake in combination with increasing energy expenditure. Thus, the development of potent and long-acting analogs of oxyntomodulin is an exciting new therapeutic avenue for addressing the global obesity epidemic. This review discusses the role of oxyntomodulin in

Oxyntomodulin attenuates exendin-4-induced hypoglycemia in cattle.

Oxyntomodulin (OXM), glucagon, glucagon-like peptide-1 (GLP-1), and exendin-4 (Ex-4) are peptide hormones that regulate glucose homeostasis in monogastric and ruminant animals. Recently, we reported that the insulin-releasing effects of OXM and glucagon in cattle are mediated through both GLP-1 and glucagon receptors. The purpose of this study was to examine the mechanisms of the glucoregulatory actions induced by Ex-4, GLP-1, OXM, and glucagon and the interrelationships among these hormones in cattle. Two experiments were performed in Holstein cattle. In Experiment 1, we initially assessed th

Isolation of alligator gar (Lepisosteus spatula) glucagon, oxyntomodulin, and glucagon-like peptide: amino acid sequences of oxyntomodulin and glucagon-like peptide.

Oxyntomodulin, glucagon, and a glucagon-like peptide (GLP) have been isolated from the endocrine pancreas of the alligator gar (Lepisosteus spatula), a ganoid fish. The three peptides were isolated by gel filtration and HPLC and were identified by size, composition, and glucagon-like immunoreactivity. The amino acid sequences of the oxyntomodulin and GLP were determined. The oxyntomodulin contains 36 amino acid residues and its sequence is H S Q G T F T N D Y S K Y L D T R R A Q D F V Q W L M S T K R S G G I T. The composition of the glucagon is identical to the N-terminal 29 residues of the g

Oxyntomodulin suppresses appetite and reduces food intake in humans.

Oxyntomodulin (OXM) is released from the gut postprandially, in proportion to energy intake, and circulating levels of OXM are elevated in several conditions associated with anorexia. Central injection of OXM reduces food intake and weight gain in rodents, suggesting that OXM signals food ingestion to hypothalamic appetite-regulating circuits. We investigated the effect of iv OXM (3.0 pmol/kg.min) on appetite and food intake in 13 healthy subjects (body mass index, 22.5 +/- 0.9 kg/m(2)) in a randomized, double-blind, placebo-controlled, cross-over study. Infusion of OXM significantly reduced a

[Oxyntomodulin, a new hormonal marker of intestinal malabsorption syndromes].

Plasma oxyntomodulin-like immunoreactivity (OLI) concentrations were found to be significantly elevated in 6 patients with coeliac disease when compared with those observed in 38 healthy subjects. Furthermore, OLI hypersecretion is related to the degree of malabsorption. This marker could be used as a test for detection and follow-up of patients with malabsorptive disorders.

Diurnal profile of oxyntomodulin-like immunoreactivity in duodenal ulcer patients.

Plasma concentrations of oxyntomodulin-like immunoreactivity, a group of intestinal peptides capable of mediating an enterogastrone signal, were measured during a 24-h period in 6 duodenal ulcer patients and compared with those of 16 age-matched controls. Each subject was submitted to 18 oxyntomodulin-like immunoreactivity determinations. Four standardized meals were given during the test. Furthermore, each patient was evaluated for peak acid output after pentagastrin stimulation. The values of the duodenal ulcer subjects were predominantly within normal acid secretion limits. Fasting levels,

Metabolic clearance rates of oxyntomodulin and glucagon in the rat: contribution of the kidney.

The half-life (t1/2) and metabolic clearance rate (MCR) of exogenous natural porcine oxyntomodulin (porcine OXM) and the synthetic analog of rat oxyntomodulin, [Nle27]-OXM (rat OXM), were compared with that of glucagon in control, sham-operated and acutely nephrectomized rats using the primed-continuous infusion technique. The half-disappearance times for porcine OXM (8.2 +/- 0.5 min) and rat OXM (6.4 +/- 0.5 min) were 3-fold slower than that of glucagon (1.9 +/- 0.1 min). Acute bilateral nephrectomy significantly prolonged the half-disappearance time of rat OXM (8.2 +/- 0.7 min) and glucagon

Peripheral oxyntomodulin reduces food intake and body weight gain in rats.

Oxyntomodulin (OXM) is a circulating gut hormone released post prandially from cells of the gastrointestinal mucosa. Given intracerebroventricularly to rats, it inhibits food intake and promotes weight loss. Here we report that peripheral (ip) administration of OXM dose-dependently inhibited both fast-induced and dark-phase food intake without delaying gastric emptying. Peripheral OXM administration also inhibited fasting plasma ghrelin. In addition, there was a significant increase in c-fos immunoreactivity, a marker of neuronal activation, in the arcuate nucleus (ARC). OXM injected directly

Preliminary evaluation of oxyntomodulin as a biomarker for metabolic risk stratification in adults with obesity.

BACKGROUND: Oxyntomodulin (OXM), secreted by enteroendocrine L-cells, regulates energy and glucose homeostasis and is a dual agonist of glucagon-like peptide-1 (GLP-1) and glucagon receptors (GCGRs), with anorexigenic, insulinotropic, and energy expenditure-enhancing effects. While synthetic analogues of OXM are under investigation as potential anti-obesity therapies, the physiological role and clinical significance of endogenous OXM in human obesity remain incompletely elucidated. This study aimed to evaluate fasting OXM levels and assess their association with anthropometric and biochemical

Oxyntomodulin and its C-terminal octapeptide inhibit liquid meal-stimulated acid secretion.

Oxyntomodulin (OXM), a 37-amino acid glucagon-containing peptide produced mainly in intestine and endocrine pancreas, is present in rat plasma and inhibits pentagastrin-stimulated gastric acid secretion in both the anesthetized and the conscious rat. In order to investigate the modifications in acid and water secretions in a physiological model, we set up a protocol which allowed us to study acid secretion in the conscious rat both under basal conditions (during an 18-hour fast) or after a physiological stimulus (a liquid meal). OXM (110 pmol X kg-1) did not modify the basal acid or water outp

Quantification of glucagon and oxyntomodulin by protein precipitation-immunoaffinity enrichment-LC-MS/MS.

INTRODUCTION: Glucagon and oxyntomodulin are peptide hormones differentially released from proglucagon that function in regulating blood glucose. Their overlapping amino acid sequences make the development of specific immunoassays difficult, but the specificity of liquid chromatography-tandem mass spectrometry can be used to distinguish the peptides. We aimed to develop a sensitive and specific mass spectrometric assay that uses non-proprietary reagents and normal-flow liquid chromatography in the simultaneous quantification of both analytes. METHODS: Bulk plasma proteins were precipitated in

A pure enteroglucagon, oxyntomodulin (glucagon 37), stimulates insulin release in perfused rat pancreas.

Oxyntomodulin, a gut peptide recently purified, has been tested in isolated perfused pancreases of normal rats. It was shown to stimulate insulin release monophasically in the presence of a low (6 mM) glucose concentration in the medium. Furthermore, oxyntomodulin potentiated glucose-induced insulin release (10 mM glucose) in a dose-dependent manner, although it was less powerful in this respect than equimolar concentrations of pancreatic glucagon. As oxyntomodulin belongs to the gut glucagon-like immunoreactants which are released during digestion, it is suggested that oxyntomodulin might be

Chemical polysialylation: design of conjugated human oxyntomodulin with a prolonged anorexic effect in vivo.

Recombinant gut hormone oxyntomodulin (OXM) is known to act as a satiety signal in human subjects and has therapeutic potential as an appetite controlling agent. The only form of this hormone that has a prospective use is a modified one, because native OXM has a very short half-life in vivo. Conjugation of OXM and the natural hydrophilic polymer polysialic acid (PSA) may significantly improve its half-life. Chemical polysialylation in vitro was used to create a long-acting form of OXM, the polysialic acid-oxyntomodulin (PSA-OXM) conjugate. The conjugation site was identified using mass shift c

A PEGylated analog of the gut hormone oxyntomodulin with long-lasting antihyperglycemic, insulinotropic and anorexigenic activity.

Peptide agonists of the glucagon-like peptide 1 (GLP-1) receptor (GLP1R) are rapidly gaining favor as antidiabetic agents, since in addition to increasing glucose-dependent insulin secretion, they also cause weight loss. Oxyntomodulin (OXM), a natural peptide with sequence homology to both glucagon and GLP-1, has glucose-lowering activity in rodents and anorectic activity in rodents and humans, but its clinical utility is limited by a short circulatory half-life due to rapid renal clearance and degradation by dipeptidyl peptidase IV (DPP-IV). Here, we describe the development of a novel DPP-IV

Glucagon-37 (oxyntomodulin) and glucagon-29 (pancreatic glucagon) in human bowel: analysis by HPLC and radioreceptorassay.

A method for assaying specifically the biologically active peptides Glucagon-37 (G-37/Oxyntomodulin/bioactive Enteroglucagon) and Glucagon-29 (G-29/pancreatic Glucagon) has been developed by use of high performance liquid chromatography (HPLC) of crude tissue extracts followed by radioreceptorassay in liver membranes. The peaks observed with this method in samples from human bowel have also been analysed in two other assays: stimulation of cyclic AMP accumulation in gastric glands and radioimmunoassay. Owing to the different patterns of activity of porcine G-37 and G-29 in these assays, the co

Effects of acute moderate-intensity continuous running on circulating oxyntomodulin concentrations in healthy men and women.

Oxyntomodulin (OXM) is a gut-derived peptide hormone with anorexigenic properties that reduce food intake and increase energy expenditure. This study investigated the effect of a single bout of moderate-intensity running on OXM concentrations and whether OXM response were associated with appetite perceptions and energy intake. Twenty healthy adults (10 men, 10 women; age: 25 ± 4 years; BMI: 22.2 ± 2.0 kg m-2) completed two randomised crossover trials: (1) 60-min treadmill running at 70% of peak oxygen uptake, and (2) rest control. Plasma OXM concentrations were measured at baseline (0 min), im

Central oxyntomodulin causes anorexigenic effects associated with the hypothalamus and alimentary canal in chicks (Gallus gallus).

Several peptides that are derived from proglucagon including glucagon, glucagon-like peptide-1 (GLP-1), and oxyntomodulin (OXM) cause satiety in mammals. Glucagon and GLP-1 also cause satiety in the avian, but the effects of OXM on avian appetite-related processes are not reported. Thus, this study was conducted to elucidate whether OXM induces satiety in chicks and to determine its mechanism of induction. Intracerebroventricular (ICV) OXM, in a linear-dose dependent manner, potently decreased feed and water intake. However, we found that the effect on water intake was secondary to a reduction

Glicentin and oxyntomodulin modulate both the phosphoinositide and cyclic adenosine monophosphate signaling pathways in gastric myocytes.

We have investigated the transduction pathways mediating the contractile effect of two glucagon-containing peptides, glicentin (GLIC) and oxyntomodulin (OXM), on smooth muscle cells isolated from rabbit antrum. Low concentrations of GLIC induced a biphasic and rapid (first phase at 5-8 sec) Ins(1,4,5)P3 production. By comparison, higher concentrations of OXM or OXM(19-37) were required to obtain biphasic time-courses of Ins(1,4,5)P3 production. In a Ca2+ free medium, the first phase of Ins(1,4,5)P3 production induced by GLIC or OXM was maintained, while the second phase disappeared. In saponin
