Chemical Proteomics Reveal the Inventory of Pyrroloquinoline Quinone Binding Proteins in Bacteria.

Pyrroloquinoline quinone (PQQ) is a bacterial redox cofactor enabling enzyme catalysis in various sugar and alcohol dehydrogenases. However, its proposed additional role as a "longevity vitamin" lacks a clear molecular basis and is thus highly debated. Here, we applied chemical proteomics to identify previously unknown classes of PQQ-binding proteins. We designed and synthesized a structurally diverse suite of five PQQ probes equipped with a diazirine photo-cross-linker and an alkyne handle for target identification. The fidelity of the probes was first evaluated for two well-characterized bac

Escherichia coli is unable to produce pyrroloquinoline quinone (PQQ).

Many bacteria can synthesize the cofactor pyrroloquinoline quinone (PQQ), a cofactor of several dehydrogenases, including glucose dehydrogenase (GCD). Among the enteric bacteria, Klebsiella pneumoniae has been shown to contain the genes required for PQQ biosynthesis. Escherichia coli and Salmonella typhimurium were thought to be unable to synthesize PQQ but it has been reported that strain EF260, a derivative of E. coli FB8, can synthesize PQQ after mutation and can oxidize glucose to gluconate via the GCD/PQQ pathway (F. Biville, E. Turlin & F. Gasser, 1991, J Gen Microbiol 137, 1775-1782). W

Protective effects of pyrroloquinoline quinone in CNS disorders.

Pyrroloquinoline quinone (PQQ) is an enzyme belonging to the family of quinone cofactors (or quinoproteins) naturally found in soil and food. PQQ was initially discovered as an essential cofactor for bacterial dehydrogenases and has since been reported to be involved in several biological processes important for mammalian growth and development. Animal studies have demonstrated that insufficient dietary intake of PQQ can lead to notable deficits, including impaired growth and compromised reproductive outcomes. In more recent years, PQQ has been recognized for its neuroprotective effects in sev

Recent progress in studies on the health benefits of pyrroloquinoline quinone.

Pyrroloquinoline quinone (PQQ), an aromatic tricyclic o-quinone, was identified initially as a redox cofactor for bacterial dehydrogenases. Although PQQ is not biosynthesized in mammals, trace amounts of PQQ have been found in human and rat tissues because of its wide distribution in dietary sources. Importantly, nutritional studies in rodents have revealed that PQQ deficiency exhibits diverse systemic responses, including growth impairment, immune dysfunction, and abnormal reproductive performance. Although PQQ is not currently classified as a vitamin, PQQ has been implicated as an important

Protection of pyrroloquinoline quinone against methylmercury-induced neurotoxicity via reducing oxidative stress.

Pyrroloquinoline quinone (PQQ) is a novel redox cofactor and also exists in various foods. In vivo as well as in vitro experimental studies have shown that PQQ functions as an essential nutrient or antioxidant. Methylmercury (MeHg), as a highly toxic environmental pollutant, could elicit central nervous system (CNS) damage. Considering the antioxidant properties of PQQ, this study was aimed to evaluate the effect of PQQ on MeHg-induced neurotoxicity in the PC12 cells. The results showed that, after pre-treatment of PC12 cells with PQQ prior to MeHg exposure, the MeHg-induced cytotoxicity was s

Endophytic Biostimulant Pyrroloquinoline Quinone Enhances Banana Growth and Primes Immunity Against Fusarium Wilt.

Pyrroloquinoline quinone (PQQ) is a redox cofactor derived from prokaryotes that participates in various biological processes involving dehydrogenase enzymes. Previous field trials identified a PQQ-producing endophyte, Burkholderia seminalis 869T2, which enhances banana growth and reduces Fusarium wilt incidence from 24.5% to 3.4%. While more recent studies have confirmed its agricultural benefits across multiple plant species, the underlying molecular mechanisms remain unclear. Here, integrated omics and imaging mass spectrometry were employed to investigate the role of PQQ in planta. Our res

PqqU (PA2289) is the primary PQQ transporter in Pseudomonas aeruginosa and links carbon metabolism to iron homeostasis.

Pseudomonas aeruginosa relies on the redox cofactor pyrroloquinoline quinone (PQQ) for efficient glucose and ethanol metabolism via periplasmic dehydrogenases (Gcd and ExaA). While PQQ biosynthesis is well-characterized, its uptake mechanisms remain unclear. Here, we identify PA2289 (PqqU), a TonB-dependent transporter, as the primary PQQ importer in P. aeruginosa. Growth assays with PQQ-deficient mutants (ΔpqqABCDEH) demonstrated that PqqU is the primary uptake system for exogenous PQQ, rescuing growth on glucose and ethanol. Genomic analysis across 210 P. aeruginosa and 263 Pseudomonas strai

Deciphering the assembly process of PQQ dependent methanol dehydrogenase.

Pyrroloquinoline quinone (PQQ)-dependent methanol dehydrogenases (MDHs), the periplasmic metalloenzymes in Gram-negative methylotrophic bacteria, play a pivotal role in methane and methanol bio-utilization. Although the structures of many PQQ-dependent MDHs have been resolved, including the canonical heterotetrameric enzymes composed of two MxaF and two MxaI subunits with a molecule of PQQ and a calcium ion in the active site in MxaF, the biogenesis of these enzymes remains elusive. Here, we characterize a chaperone, MxaJ, responsible for PQQ incorporation by reconstructing a PQQ-dependent MDH

Characterization of a protein-generated O₂ binding pocket in PqqC, a cofactorless oxidase catalyzing the final step in PQQ production.

PQQ is an exogenous, tricyclic, quino-cofactor for a number of bacterial dehydrogenases. The final step of PQQ formation is catalyzed by PqqC, a cofactorless oxidase. This study focuses on the activation of molecular oxygen in an enzyme active site without metal or cofactor and has identified a specific oxygen binding and activating pocket in PqqC. The active site variants H154N, Y175F,S, and R179S were studied with the goal of defining the site of O(2) binding and activation. Using apo-glucose dehydrogenase to assay for PQQ production, none of the mutants in this "O(2) core" are capable of PQ

Erroneously elevated glucose values due to maltose interference in mutant glucose dehydrogenase pyrroloquinolinequinone (mutant GDH-PQQ) based glucometer.

Currently available glucose test strip enzymes include glucose oxidase (GOD) and glucose dehydrogenase (GDH). In GDH-based glucometers, glucose oxidation can be catalysed by different cofactors: nicotinamide adenine dinucleotide (GDH-NAD), flavin adenine dinucleotide (GDH-FAD), pyrroloquinolinequinone (GDH-PQQ) and mutant GDH-PQQ. GOD-based and GDH-NAD-based glucometers are substrate-specific and do not react with sugars other than glucose. GDH-FAD reacts with xylose only in addition to glucose. GDH-PQQ is not glucose-specific; in addition to glucose, it reacts with different other sugars and

The cofactor challenge in synthetic methylotrophy: bioengineering and industrial applications.

Methanol is a promising feedstock for industrial bioproduction: it can be produced renewably and has high solubility and limited microbial toxicity. One of the key challenges for its bio-industrial application is the first enzymatic oxidation step to formaldehyde. This reaction is catalysed by methanol dehydrogenases (MDH) that can use NAD+, O2 or pyrroloquinoline quinone (PQQ) as an electron acceptor. While NAD-dependent MDH are simple to express and have the highest energetic efficiency, they exhibit mediocre kinetics and poor thermodynamics at ambient temperatures. O2-dependent methanol oxi

Effect of pyrroloquinoline quinone on neuropathic pain following chronic constriction injury of the sciatic nerve in rats.

Pyrroloquinoline quinone PQQ is a naturally occurring redox cofactor that acts as an essential nutrient, antioxidant, and redox modulator. PQQ has been demonstrated to oxidize the redox modulatory site of N-methyl-d-aspartic acid (NMDA) receptors. Such agents are known to be neuroprotective in experimental stroke models. However, there is not report about the therapeutic effect of PQQ on neuropathic pain. We tested the effects of oral administration of PQQ on neuropathic pain of rats with chronic constriction injury (CCI) of the sciatic nerve. The repeated oral administration of PQQ (20 and 40

Effects of pyrroloquinoline quinone on noise-induced and age-related hearing loss in mice.

We investigated whether the oxidoreductase cofactor pyrroloquinoline quinone (PQQ) prevents noise-induced and age-related hearing loss (NIHL and ARHL) in mice. To assess NIHL, 8 week-old mice with and without PQQ administration were exposed to noise for 4 h. PQQ was orally administered for one week before and after noise exposure and subcutaneously once before noise exposure. For ARHL evaluation, mice were given drinking water with or without PQQ starting at 2 months of age. In the NIHL model, PQQ-treated mice had auditory brainstem response (ABR) thresholds of significantly reduced elevation

Pyrroloquinoline quinone-conferred neuroprotection in rotenone models of Parkinson's disease.

Pyrroloquinoline quinone (PQQ), a redox cofactor in the mitochondrial respiratory chain, has proven to protect neurons against glutamate-induced damage both in vitro and in vivo. This study was aimed to investigate the possible neuroprotective effects of PQQ in rotenone-induced Parkinson's disease (PD) model. Pre-treatment with PQQ prevented cultured SH-SY5Y cells from rotenone-induced apoptosis, accompanied by modulation of apoptosis-related proteins (Bcl-2, Bax and Smac), restoration of the mitochondrial membrane potential, inhibition of intracellular reactive oxygen species (ROS) production

Pyrroloquinoline quinone protects heat-stressed porcine oocytes and improves early embryonic development via maintaining mitochondrial integrity and redox homeostasis.

Heat stress (HS) is a major environmental factor that impairs female fertility by disrupting oocyte quality and developmental competence. Pyrroloquinoline quinone (PQQ), a redox cofactor with strong antioxidant and mitochondrial-protective properties, has recently attracted attention for its cytoprotective roles in mammalian cells. Here, we investigated whether PQQ supplementation could alleviate HS-induced porcine oocyte damage and improve subsequent embryonic development. Our results showed that PQQ treatment significantly restored the rates of germinal vesicle breakdown (GVBD) and first pol

Pyrroloquinoline quinone targets senescent osteoclasts and reduces spinal pain and endplate degeneration in a lumbar spine instability mouse model.

BACKGROUND: Pyrroloquinoline Quinone (PQQ) is a redox cofactor with potent antioxidant and anti-inflammatory properties. This study investigated the therapeutic potential of PQQ in alleviating spinal pain and degeneration in a lumbar spine instability (LSI) mouse model. LSI is commonly associated with spinal pain and structural degeneration, with senescent osteoclasts (SnOCs) involved in spinal hypersensitivity and degeneration. METHODS: LSI was induced in 8-week-old C57/BL6 mice by surgically resecting the L3–L5 spinous processes and associated ligaments. The mice were treated with PQQ, and b

Pyrroloquinoline quinone drives ATP synthesis in vitro and in vivo and provides retinal ganglion cell neuroprotection.

Retinal ganglion cells are highly metabolically active requiring strictly regulated metabolism and functional mitochondria to keep ATP levels in physiological range. Imbalances in metabolism and mitochondrial mechanisms can be sufficient to induce a depletion of ATP, thus altering retinal ganglion cell viability and increasing cell susceptibility to death under stress. Altered metabolism and mitochondrial abnormalities have been demonstrated early in many optic neuropathies, including glaucoma, autosomal dominant optic atrophy, and Leber hereditary optic neuropathy. Pyrroloquinoline Quinone (PQQ) Attenuates Hydrogen Peroxide-Induced Injury Through the Enhancement of Mitochondrial Function in Human Trabecular Meshwork Cells.

Mitochondrial metabolism in the trabecular meshwork (TM) plays a critical role in maintaining intraocular pressure homeostasis by supporting the energy-demanding processes involved in aqueous humour outflow. In primary open-angle glaucoma, oxidative stress impairs mitochondrial function, leading to TM dysfunction. Therefore, understanding and targeting mitochondrial health in TM cells could offer a novel therapeutic strategy. Pyrroloquinoline quinone (PQQ) is a redox cofactor with antioxidant and mitochondrial-enhancing properties. However, its effects on human TM (HTM) cells remain largely un
