Five novel Paenibacillus species from the plant rhizosphere: Paenibacillus corni sp. nov., Paenibacillus vaccinii sp. nov., Paenibacillus ericacearum sp. nov., Paenibacillus polytrichii sp. nov., and Paenibacillus artemisiae sp. nov.

Bacterial strains were isolated from different locations and screened for plant growth promoting (PGP) features. Comparative analyses of the 16S gene sequences of 5 of the strains indicated taxonomic relatedness within the genus Paenibacillus. A polyphasic taxonomic approach was employed to study the strains in detail to clarify their phylogenetic position. Genome-based analyses, including digital DNA-DNA hybridization (dDDH) and average nucleotide identity (ANI), revealed values consistently below the accepted species delineation thresholds when compared with closest relatives of each strain.

Paenibacillus encodes a membrane-localized Spo0B.

Sporulation is a strategy employed by many bacteria to survive harsh environmental conditions. The genus Paenibacillus includes spore-forming species notorious for spoiling pasteurized dairy products and for causing American foulbrood in honeybee larvae, leading to colony collapse. Human pathogens within Paenibacillus are also a growing threat, causing fatal opportunistic infections. Here, we present a comprehensive survey of sporulation genes across 1,460 high-quality Paenibacillus genomes. We find that all members of the sporulation-initiating phosphorelay are well conserved, but that the Sp

Brain abscess caused by Paenibacillus lactis.

We present a case of a 30-year-old male patient who was admitted to our institution with a 4-month history of persistent headache, pain in the left half of the face and the left ear. The initial magnetic resonance imaging detected an inflammatory process in the left pyramid interpreted as petrous apicitis. Subsequently, he developed generalized seizures. Follow-up computed tomography scanning with contrast enhancement demonstrated newly-formed brain abscess in the basal region of the left temporal lobe. The patient underwent microsurgical evacuation and resection of the abscess. Microbiologica

Comparative Genomics of Paenibacillus Secondary Metabolism: Unveiling the Putative Biosynthetic Gene Cluster for Paenialvins in Paenibacillus Alvei Strain 32.

In this study, we used comparative genomics and culture-based methods to investigate Biosynthetic Gene Clusters (BGCs) responsible for the production of antimicrobial peptides. Paenibacillus alvei strain 32 was isolated from a cystic fibrosis sputum. Its genome was sequenced using Illumina, showing a size of 6,584,590 bp with 239 contigs assembled in 26 scaffolds, an average coverage of 243X, and 6,832 coding sequences. ANI analysis and in silico DNA-DNA hybridization showed its affiliation inside Paenibacillus alvei, with a clear separation from other related strains, leading us to propose a

Enhancement of reduction of biodesulfurization end products by Paenibacillus strains.

The work reported here aimed to enhance the reduction of the downstream intermediates 2-hydroxybiphenyl (2-HBP) and 2-(2'hydroxyphenyl ethan-1-al) produced by biodesulfurization of DBT (dibenzothiophene) and BT (benzothiophene) using Paenibacillus strains (32O-W and 32O-Y). Salicylaldehyde was used as a surrogate for 2-(2'hydroxybiphenyl ethan-1-al), as the two compounds are structurally very similar while the latter is not commercially available. Five strategies were tested using growth in media containing either 2-HBP or salicylaldehyde: use of single strain cultures, co-culturing, genetic e

Paenibacillus Sepsis and Meningitis in a Premature Infant: A Case Report.

Infections due to bacteria of the genus Paenibacillus are exceedingly rare and therefore predominately described on a case-by-case basis. Here, we present a case of a 25-day-old premature neonate who died from presumed Paenibacillus sepsis and meningitis. Most prior reported cases of Paenibacillus bacteremia were among patients who had prosthetic medical devices, were immunocompromised, or were injection drug users. However, to our knowledge, this is the first reported case of infant death from presumed Paenibacillus thiaminolyticus. This case suggests the potential for severe human infection

Genetic diversity among isolates of Paenibacillus larvae from Austria.

Genetic diversity of 214 Paenibacillus larvae strains from Austria was studied. Genotyping of isolates was performed by polymerase chain reaction (PCR) with primers corresponding to enterobacterial repetitive intergenic consensus (ERIC), BOX repetitive and extragenic palindromic (REP) elements (collectively known as rep-PCR) using ERIC primers, BOX A1R and MBO REP1 primers. Using ERIC-PCR technique two genotypes could be differentiated (ERIC I and II), whereas using combined typing by BOX- and REP-PCR, five different genotypes were detected (ab, aB, Ab, AB and alphab). Genotypes aB and alphab

Paenibacillus tabacirhizosphaerae sp. nov., isolated from rhizosphere soil of tobacco.

Two Gram-staining positive, aerobic, non-motile, endospore-forming and rod-shaped bacteria, designated strains SI92T and SI8, respectively, were isolated from the rhizosphere soil of tobacco collected from south-western China. Strain SI92T shared the highest 16S rRNA gene similarity to Paenibacillus silvestris (97.9%) and Paenibacillus phytorum (97.9%), and strain SI8 showed the highest 16S rRNA gene similarity with P. phytorum (98.0%). Phylogenetic analysis based on 16S rRNA gene sequences showed that strains SI92T and SI8 clustered with the genus Paenibacillus and formed a distinct subclade.

Recombinant production of Paenibacillus wynnii β-galactosidase with Komagataella phaffii.

BACKGROUND: The β-galactosidase from Paenibacillus wynnii (β-gal-Pw) is a promising candidate for lactose hydrolysis in milk and dairy products, as it has a higher affinity for the substrate lactose (low KM value) compared to industrially used β-galactosidases and is not inhibited by the hydrolysis-generated product D-galactose. However, β-gal-Pw must firstly be produced cost-effectively for any potential industrial application. Accordingly, the yeast Komagataella phaffii was chosen to investigate its feasibility to recombinantly produce β-gal-Pw since it is approved for the regulated producti

Molecular diversity of Paenibacillus larvae strains isolated from Lithuanian apiaries.

Paenibacillus larvae bacterium is known to be the causative agent of American foulbrood (AFB), a widespread, highly contagious and fatal disease in honey bees (Apis mellifera). There are four genotypes of Paenibacillus larvae that are named after their enterobacterial repetitive consensus (ERIC), and a fifth ERIC genotype has recently been found. In this study, a total of 108 independent P. larvae isolates from different geographical regions in Lithuania collected between 2011 and 2021 were investigated by molecular methods. The aims of this study were to detect which enterobacterial repetitiv

The N-terminus of Paenibacillus larvae C3larvinA modulates catalytic efficiency.

C3larvinA was recently described as a mono-ADP-ribosyltransferase (mART) toxin from the enterobacterial repetitive intergenic consensus (ERIC) III genotype of the agricultural pathogen, Paenibacillus larvae. It was shown to be the full-length, functional version of the previously described C3larvintrunc toxin, due to a 33-residue extension of the N-terminus of the protein. In the present study, a series of deletions and substitutions were made to the N-terminus of C3larvinA to assess the contribution of the α1-helix to toxin structure and function. Catalytic characterization of these variants

Immobilization of Paenibacillus polymyxa with biopolymers to enhance the production of 2,3-butanediol.

BACKGROUND: Paenibacillus polymyxa, is a Gram-positive, plant growth promoting bacterium, known for producing 98% optically pure 2,3-butanediol, an industrially valuable chemical for solvents, plasticizers and resins. Immobilization of Paenibacillus polymyxa has been proposed to improve the cell stability and efficiency of the fermentation process, reduce contamination and provide easy separation of butanediol in the culture broth as compared to conventional bioprocesses. This research aimed to explore the potential of Paenibacillus polymyxa with immobilization technique to produce 2,3-butaned

α-L-fucosidase from Paenibacillus thiaminolyticus: its hydrolytic and transglycosylation abilities.

In this work, focused on possible application of α-L-fucosidases from bacterial sources in the synthesis of α-L-fucosylated glycoconjugates, several nonpathogenic aerobic bacterial strains were screened for α-L-fucosidase activity. Among them Paenibacillus thiaminolyticus was confirmed as a potent producer of enzyme with the ability to cleave the chromogenic substrate p-nitrophenyl α-L-fucopyranoside. The gene encoding α-L-fucosidase was found using the genomic library of P. thiaminolyticus constructed in the cells of Escherichia coli DH5α and sequenced (EMBL database: FN869117, carbohydrate-a
