Relationship between circulating levels of pancreatic proteolytic enzymes and pancreatic hormones.

BACKGROUND: While the close morphological relationship between the exocrine and endocrine pancreas is well established, their functional interaction remains poorly understood. The aim of this study was to investigate the associations between circulating levels of pancreatic proteolytic enzymes and insulin, as well as other pancreatic hormones. METHODS: Fasting venous blood samples were collected and analyzed for trypsin, chymotrypsin, insulin, glucagon, somatostatin, and pancreatic polypeptide. Linear regression analysis was used in unadjusted and two adjusted (accounting for prediabetes/diab

Effects of gastrointestinal hormones on pancreatic growth.

This article discusses experiments demonstrating that the gastrointestinal hormones, gastrin, secretin, and CCK (cholecystokinin), stimulate the growth of the exocrine pancreas. Exogenous gastrin, secretin, and CCK increase pancreatic weight DNA, RNA and protein content of the rat pancreas. Antrectomy, which removes most endogenous gastrin, decreases pancreatic growth. The effects of antrectomy are prevented by exogenous gastrin. Infusion of HCl into the duodenum to release secretin and infusion of amino acids into the duodenum to release CCK also stimulate pancreatic growth. These results pro

[Pancreatic hormones and hormonal regulation of insulin secretion].

Endocrine pancreas producing insulin, glucagon, somatostatin and pancreatic polypeptide is under the influence of different types of regulation; among them the regulatory role of enteropancreatic axis plays an important role. Incretin effect of glucose-dependent insulinotropic peptide (GIP) and glucagon-like peptide 1 (GLP-1) is significantly involved in the insulin secretion which is modulated by many other hormones. Diabetes mellitus, similarly to disturbances of other hormones, can cause impaired regulation of insulin and other pancreatic hormones.

Mixed endocrine pancreatic tumors producing several peptide hormones.

Twenty-four endocrine pancreatic tumors were examined immunohistochemically for insulin, glucagon, gastrin and ACTH. In seven of these tumors, more than one peptide-hormone-containing cell type was observed. These seven tumors were also examined with conventional staining methods for the presence of A1, A2, and B cells. The results showed that these staining methods do not always distinguish between the different hormone-producing cell types of endocrine pancreatic tumors. In spite of the fact that several types of hormone-secreting cells were found in the tumors, the case histories described

[Pancreatic hormones and gastrin in patients with acute dysentery].

The incretory pancreatic function has been studied in 35 patients with a moderately severe course of acute dysentery. The blood plasma content of insulin, glucagon and gastrin was determined by means of radioimmunoassay with standard kits during the acute course of the disease and prior to discharge of patients. The endocrine pancreatic function was found to be deranged with an increased insulin content during convalescence; the glucagon content was decreased during the period of the disease and the gastrin content was decreased in the acute period.

Immunoelectron study of pancreatic carcinomas using antibodies to gastrointestinal hormones.

The aim of this study was to investigate the ultrastructural appearance of pancreatic adenocarcinoma combined with glucagon and gastrin/cholecystokinin (CCK) expression. The authors investigated the ultrastructure and the immunocytochemistry of 12 human pancreatic cancer specimens and used 3 chronic pancreatitis samples and 6 adjacent histological normal pancreatic tissues (away from the tumor) as controls. The ultrastructural study revealed that chronic pancreatitis tissues were characterized by alterations of the secretory cells. The enzymic and secretory changes were confirmed by electron i

Effects of pregnancy hormones on pancreatic islets in organ culture.

Pregnancy is associated with hyperinsulinaemia and decreased glucose tolerance which may lead to gestational diabetes. Sera from pregnant women in the last trimester were found to stimulate insulin secretion of mouse pancreatic islets maintained in organ culture suggesting a direct effect of pregnancy hormones and/or metabolites on the endocrine pancreas. Previous studies have shown that progesterone and human placental lactogen (hPL) had direct effects on isolated islets and in the present study the effects of combined addition of these hormones to the cultured islets were evaluated. hPL, 1 m

[Gastro-entero-pancreatic hormones in patients with chronic Chagas' disease].

The seric levels of gastrin, pancreatic glucagon, pancreatic polypeptide, enteroglucagon, motilin and cholecistokinin were evaluated in ten patients with chronic Chagas' disease and compared with those observed in nine normal control subjects. The seric values of all the hormones were determined on basal stimulation, after continuous intravenous secretin infusion and infusion of stepwise increased concentrations of caerulein (direct stimulation), and after intravenous secretin administration followed by intraduodenal instilation of increased concentrations of phenylalanina (combined stimulatio

Leptin does not directly regulate the pancreatic hormones amylin and pancreatic polypeptide: interventional studies in humans.

OBJECTIVE: Leptin and the pancreatic hormones amylin and pancreatic polypeptide are being evaluated alone or in combination for the treatment of obesity, but their physiological regulation has not yet been fully elucidated. Thus, we examined whether amylin and pancreatic polypeptide are regulated by caloric intake and/or short- and long-term energy deprivation and whether any potential regulation is mediated by changes in leptin levels. RESEARCH DESIGN AND METHODS: We measured circulating levels of amylin and pancreatic polypeptide after 1) a 75-g glucose load in 28 healthy, normal-weight wom

Reproductive factors, exogenous hormones, and pancreatic cancer risk in the CTS.

Female steroid hormones are hypothesized to play a protective role in pancreatic cancer risk. However, results from epidemiologic studies that examined hormone-related exposures have been inconsistent. The California Teachers Study is a cohort study of female public school professionals that was established in 1995-1996. Of the 118,164 eligible study participants, 323 women were diagnosed with incident invasive pancreatic cancer through December 31, 2009. Multivariable Cox proportional hazards regression methods were used to estimate hazard ratios and 95% confidence intervals for the associati

Lighting up lizard islets: A fluorescent map of pancreatic hormones before hatching.

This study investigates the organization of pancreatic endocrine islets in lizard embryos before hatching. Immunohistochemical staining was performed on "pre-hatching" embryos from three species representing major squamate clades: Lacerta agilis (Lacertoidea), Eublepharis macularius (Gekkota), and Anolis sagrei (Iguania). Four distinct endocrine cell types were identified within the pancreas of the studied species: glucagon-producing α-cells, insulin-producing β-cells, somatostatin-producing δ cells, and pancreatic polypeptide (γ) cells. Each of these cell types exhibited a unique distribution

Action of sex-hormones on pancreatic islet function in female domestic duckling.

The aim of the present study was to examine the action of sex-hormones on the endocrine pancreas of the female domestic duckling. Estrogen alone, or in combination with progesterone, inhibited mitosis and caused degranulation only in the B-cells of the pancreatic islets. A- or D-cells of the islets were not affected. Progesterone alone had no effect on any islet cells. It is suggested that estrogen may have dual action (mitotic inhibition and beta cell stimulation) only on B-cells of the pancreatic islets of the domestic ducklings.

Spatially resolved distribution of pancreatic hormones proteoforms by MALDI-imaging mass spectrometry.

Zinc plays a crucial role both in the immune system and endocrine processes. Zinc restriction in the diet has been shown to lead to degeneration of the endocrine pancreas, resulting in hormonal imbalance within the β-cells. Proteostasismay vary depending on the stage of a pathophysiological process, which underscores the need for tools aimed at directly analyzing biological status. Among proteomics methods, MALDI-ToF-MS can serve as a rapid peptidomics tool for analyzing extracts or by histological imaging. Here we report the optimization of MALDI imaging mass spectrometry analysis of histolog

Pancreas transplantation: an immunohistochemical analysis of pancreatic hormones and HLA-DR expression.

The immunohistochemical features of pancreatic grafts in eight patients with pancreatic transplants were analyzed and compared with pancreases from five patients with chronic pancreatitis and with three pancreatic tissues without histological abnormalities. There was a significant increase in glucagon producing cells in patients with transplanted pancreases compared with those with chronic pancreatitis (P less than 0.05). A significant decline in insulin-producing cells was seen in the transplanted pancreases with rejection in comparison with normal pancreatic tissue. Immunohistochemical stain

Hormonal factors and pancreatic cancer in women.

PURPOSE: The aim of the study was to evaluate hormonal risk factors for carcinoma of the exocrine pancreas among postmenopausal women. METHODS: Mailed questionnaire data from 52 cases and 233 population-based controls in Ontario were used to assess parity, age at first birth, and other hormonal factors on pancreatic cancer risk. RESULTS: Reduced risk was seen with three or more pregnancies [adjusted odds ratio (OR) = 0.22, 95% confidence interval (CI) = 0.07-0.65] and use of oral contraceptives (adjusted OR = 0.36, 95% CI = 0.13-0.96), whereas no significant associations were found for age a

Clinical and hormonal aspects of pancreatic diabetes.

The prevalence of diabetes due to chronic pancreatitis would appear to be increasing. In western countries this is associated with the known increase in alcohol consumption and AIP. Malnutrition may be etiologic in tropical areas. The incidence of diabetes in chronic pancreatitis is dependent on a number of factors. It is more common in alcohol-induced pancreatitis, rarely occurs after the first attack but tends to increase with time and rises markedly in calcific pancreatitis. Abnormal glucose tolerance occurred in 91% of patients with calcific pancreatitis and 70% of patients with noncalific

Interdigestive cycling in chronic pancreatitis: altered coordination among pancreatic secretion, motility, and hormones.

BACKGROUND & AIMS: Secretions from the exocrine and endocrine pancreas may modulate interdigestive motility. To test this hypothesis in humans, we investigated interdigestive cycling in patients with chronic pancreatitis (CP) as a model of impaired pancreatic function. METHODS: Antroduodenal motility, pancreatic enzyme output, and pancreatic polypeptide release were monitored for two consecutive interdigestive cycles in 13 controls and 9 patients with CP. RESULTS: Interdigestive enzyme output was severely impaired in patients with CP (> 80% decrease); however, secretory cycling was still evi

Effect of duodenectomy on interdigestive pancreatic secretion, gastrointestinal motility, and hormones in dogs.

We tested the hypothesis that the duodenum is necessary to coordinate interdigestive pancreatic trypsin secretion with gastrointestinal motility and determined whether duodenectomy altered interdigestive cycles of plasma motilin and pancreatic polypeptide and their relationship to trypsin secretion and motility. Consequently, in normal and duodenectomized dogs, we measured trypsin secretion, gastrointestinal motility, and plasma concentrations of motilin and pancreatic polypeptide during the interdigestive period. After duodenectomy, peaks of trypsin secretion continued to cycle at normal inte
