Despite being common worldwide, parapoxvirus infections are regarded as neglected zoonoses because their incidence is either unknown or grossly overestimated. In ruminants all throughout the world, parapoxvirus produces oral lesions and infectious pustular dermatitis. The pathogen is typically spread directly via items contaminated with parapoxvirus and indirectly via a near contact with dermatological lesions that contain the virus on affected animals. Animals infected with the parapoxvirus typically exhibit no clinical symptoms, and the mode of parapoxvirus transmission is occasionally uncle

This report describes the clinical and laboratory findings for 5 sheep from 3 different flocks with extensive proliferative skin lesions grossly resembling warts on the distal limbs. The lesions affected the front and rear extremities in all sheep, and 2 sheep also had lesions around the head. The sheep exhibited signs of pain when the lesions were touched, and most sheep were reluctant to move. Various empirical treatments, including systemic antibiotics, topical antibiotics, and antifungal ointments, were administered without clinical improvement. Diagnostic tests including skin biopsy and h

In 2026, a widespread outbreak of ulcerative dermatitis of unknown aetiology occurred in breeding horses in Ireland. The lesions resembled those associated with equine herpesvirus 3 (EHV3) infection, i.e., papules, vesicles, and ulcers on the penis of stallions and on the vulval and anal areas of mares. This study aimed to identify the probable causative agent. The methods applied included metagenomic sequencing of DNA, phylogenetic analysis, and real-time PCR. Metagenomic analysis directly from a penile sample confirmed the presence of equine parapoxvirus (EqPPV) DNA. Phylogenetic analysis in

A 53-year-old woman presented with an enlarging, tender, ulcerated nodule on her dorsal hand after being scratched by a stray kitten. Histology was consistent with orf parapoxvirus infection. The lesion resolved without scarring after several weeks of treatment with imiquimod 5 percent cream. Orf is a zoonosis transmitted to humans from sheep and goats by direct contact or by fomites. Transmission by cat scratch has not been previously described in the literature.

BACKGROUND: Detection of parapoxviruses is important in various animals as well as in humans as zoonotic infections. Reliable detection of parapoxviruses is fundamental for the exclusion of other rash-causing illnesses, for both veterinarians and medical practitioners. To date, however, no real-time PCR assay for the detection of parapoxviruses has been reported.
METHODS: A minor groove binder-based quantitative real-time PCR assay targeting the B2L gene of parapoxviruses was developed on the ABI Prism and the LightCycler platforms.
RESULTS: The real-time PCR assay successfully amplified DNA

The aim of this study was to evaluate the persistence and infectivity of a Parapoxvirus orf sample that was kept under refrigeration for 35 years, originating from an animal with a presumptive diagnosis of contagious ecthyma. The sample, identified as CIZIP/1989, was collected from a goat with clinical suspicion of contagious ecthyma in 1989, originating from a flock of sheep and goats in São Paulo, Brazil. After 35 years of refrigeration under the stress of temperature variations, viral isolation was performed in cell culture using MDBK cells. In addition, a Polymerase Chain Reaction (PCR) wa

A horse in Finland exhibited generalized granulomatous inflammation and severe proliferative dermatitis. After euthanization, we detected poxvirus DNA from a skin lesion sample. The virus sequence grouped with parapoxviruses, closely resembling a novel poxvirus detected in humans in the United States after horse contact. Our findings indicate horses may be a reservoir for zoonotic parapoxvirus.

BACKGROUND: Parapoxviruses (PPV) cause skin and mucous membrane signs to several animal species and humans worldwide. Equine parapoxvirus (EqPPV) was first detected in a sick horse in Finland in 2013. It is potentially zoonotic, and a similar virus has been detected in humans in the USA. In winter 2021–2022, EqPPV caused a large-scale pastern dermatitis epidemic in racehorses all over Finland. Field reports suggest that similar epidemics of unverified cause have also occurred in 2015 and 2019. The aim of this study was to develop a serological test and study the immune response, seroprevalence

A Mediterranean monk seal was shown by immunohistochemical and polymerase chain reaction techniques to be dually infected with a Leishmania sp. and parapoxvirus. The pathological findings included a deep ulcer on the side of the head, ulcers on the gingival and inner aspect of the lower lip, enlarged lymph nodes and tonsils, and respiratory lesions (pulmonary consolidation, oedema, haemorrhages and emphysema; tracheal and bronchial congestion, exudates and haemorrhage). Amastigotes were demonstrated in macrophages in the lymph nodes and spleen, and intracytoplasmic inclusion bodies were observ

The Poxviridae family consists of large, double-stranded DNA viruses, with significant implications for human and animal health. Among these, the genus Parapoxvirus, which includes ORF virus (ORFV), bovine papular stomatitis virus (BPSV), and pseudocowpox virus (PCPV), demonstrates unique evolutionary characteristics. This study investigated the variability, evolutionary drivers, and structural characteristics of A33 and A34 proteins, key glycoproteins involved in viral dissemination and immune evasion. Our findings revealed that A33 and A34 exhibit higher variability in Parapoxvirus compared

BACKGROUND: Several viruses belonging to the family Poxviridae can cause infections in humans and animals. In Corsica, livestock farming (sheep, goats, pigs, and cattle) is mainly mixed, leading to important interactions between livestock, wildlife, and human populations. This could facilitate the circulation of zoonotic diseases, and makes Corsica a good example for studies of tick-borne diseases.
OBJECTIVES: To gain understanding on the circulation of poxviruses in Corsica, we investigated their presence in tick species collected from cattle, sheep, horses, and wild boar, and characterized

AIM: Inactivated Orf virus (ORFV, Parapoxvirus ovis) demonstrates strong antiviral activity in animal models including a human hepatitis B virus (HBV)-transgenic mouse. In addition, expression of interferon (IFN)-γ and interleukin-10 (IL-10) was induced after administration of inactivated ORFV in these mice. IFN-γ and IL-10 are known to elicit antifibrotic activity. We therefore aimed to study antifibrotic activity of inactivated ORFV in models of liver fibrosis.
METHODS: We characterized ORFV-induced hepatic cytokine expression in rats. We then studied ORFV in two models of liver fibrosis in

BACKGROUND: Equine parapoxvirus (EqPPV) has emerged as a cause of recurring pastern dermatitis outbreaks in horses in Finland. First detected in 2013, EqPPV has since been associated with potentially hundreds of clinical cases and suspected zoonotic transmission. Based on serological data and field reports, EqPPV causes pastern dermatitis outbreaks in Finland every few years. This study aimed to investigate the environmental presence and plausible transmission dynamics of the virus to prepare for future outbreaks and give targeted guidelines to stable staff and veterinarians.

Parapoxvirus (PPV) causes papular stomatitis and contagious pustular dermatitis in ruminants worldwide. The virus is generally transmitted through close contact with skin lesions containing PPV in infected animals and indirectly through PPV-contaminated materials. PPV-infected animals frequently do not show clinical signs and the route of PPV transmission is sometimes unclear. In this study, the possibility of mechanical transmission of PPV by houseflies (Musca domestica) was investigated using polymerase chain reaction (PCR) gene surveillance. Samples were collected from cattle, sheep, barn e

Parapoxvirus (PPV) infections are of worldwide importance, particularly in sheep and goat herds. Owing to the zoonotic potential of all PPV species, they are a permanent threat to human health as well. The virus is also known to affect wildlife, as reported for pinnipeds, red deer and several other wild ruminants. PPVs found in red deer have been claimed as a unique species according to certain genomic features. So far infection of wildlife has been recognized because of clinical manifestation such as inflammation, stomatitis or typical pox-like lesions in the skin or mucous membranes. Here we

The parapoxvirus Orf virus (ORFV), has long been recognized for its immunomodulatory properties in permissive and non-permissive animal species. Here, a new recombinant ORFV expressing the full-length spike (S) protein of Porcine epidemic diarrhea virus (PEDV) was generated and its immunogenicity and protective efficacy were evaluated in pigs. The PEDV S was inserted into the ORFV121 gene locus, an immunomodulatory gene that inhibits activation of the NF-κB signalling pathway and contributes to ORFV virulence in the natural host. The recombinant ORFV-PEDV-S virus efficiently and stably express

Human parapoxvirus infections are rare, self-limiting, zoonotic diseases. A 35-year-old veterinarian presented with a generalized rash of large umbilicated vesicles that appeared after antibiotic treatment for erysipelas on the forearm. The erysipelas arose from an erupted pustular thumb lesion that appeared after examining a sheep. An outbreak of chickenpox in the village suggested parapoxvirus or varicella zoster virus (VZV) was the most likely agent. No poxvirus was detected by electron microscopy or in cell cultures from lesion material. PCR revealed parapoxvirus DNA with a sequence simila
