Antineurofilament antibodies in postencephalitic and idiopathic Parkinson's disease.

Autoantibodies to neurofilaments were found by the immunofluorescence technique in serum of patients with postencephalitic (von Economo's) and idiopathic Parkinson's disease in the same proportion as in age-matched neurological and non-neurological controls. In addition, similar neurofibrillary staining was detected in age groups of 29 years and younger, but rarely in the first year of life. Persons over 70, with or without disease, showed a prevalence of antibodies significantly higher than in persons under 70. Serum from 1 case of Alzheimer's disease out of 4 tested, was positive for neurofi

Von Economo's disease and postencephalitic parkinsonism responsive to carbidopa and levodopa.

Postencephalitic parkinsonism (PEP) is currently perceived as having a very close etiologic relationship with encephalitis lethargica (von Economo's disease [EL]), with PEP developing immediately after the acute phase of EL or at some time later. EL was classically described by von Economo and has somnolent-ophthalmoplegic, hyperkinetic, and amyostatic-akinetic forms. Previous cases have reported success with levodopa in PEP. We report a case with PEP showing success after administration of levodopa and carbidopa. This case would support the hypothesis that this syndrome effects dopamine neuro

Progressive supranuclear palsy: new disease or variant of postencephalitic parkinsonism?

We review the etiological importance of the epidemic encephalitis for progressive supranuclear palsy (PSP) and addresses the question of whether the explosion of PSP literature in the mid-20th century reflects the appearance of a new disease. We examined 2,000 studies on Parkinson's disease from 1861 to 1963 and found PSP-like cases in the past, before the epidemic encephalitis era. It can be assumed that PSP is neither a new disease nor a variant of postencephalitic parkinsonism.

Astrogliosis in von Economo's and postencephalitic Parkinson's diseases supports probable viral etiology.

A marked generalized astrogliosis was observed in the frontal and temporal white matter from a case of von Economo's disease and another of postencephalitic Parkinson's disease, which areas were otherwise devoid of any other demonstrable microscopic lesions. No similar astrocytic reaction of any severity was observed in the same areas in a number of other brain diseases or controls, except when other kinds of lesions were present in the same section, with reactive astrocytes being present within the primary or defining lesion or immediately close by. The marked astrogliosis in von Economo's an

Pathological tau proteins in postencephalitic parkinsonism: comparison with Alzheimer's disease and other neurodegenerative disorders.

Immunohistochemical and biochemical analyses of hyperphosphorylated tau proteins, the major component of neurofibrillary tangles, were performed in different brain regions from patients presenting with postencephalitic parkinsonism. Neurofibrillary tangles were found in hippocampus, neocortical areas (mostly in supragranular layers), and several subcortical structures. By immunoblotting, a tau protein triplet similar to the one seen in Alzheimer's disease was observed. This biochemical approach allows for the definition of postencephalitic parkinsonism from certain neurodegenerative disorders

Frequency of Alzheimer's neurofibrillary tangle in the brains of progressive supranuclear palsy, postencephalitic parkinsonism, Alzheimer's disease, senile dementia and non-demented elderly person.

The frequency and the distribution of Alzheimer's neurofibrillary tangles were examined in those cases of progressive supranuclear palsy (one case), postencephalitic parkinsonism (one case), Alzheimer's disease (two cases), senile dementia (two cases) and non-demented elderly person (three cases). The light microscopic pattern of Alzheimer's neurofibrillary tangles seen in progressive supranuclear palsy was similar to those in postencephalitic parkinsonism, Alzheimer's disease, senile dementia and non-demented elderly person. Pathological overlap in cases of parkinsonism associated with neurofibrillary tangles. A study of recent cases of postencephalitic parkinsonism and comparison with progressive supranuclear palsy and Guamanian parkinsonism-dementia complex.

In recent years a number of patients suffering from long-standing postencephalitic parkinsonism have donated their brains to the United Kingdom Parkinson's Disease Society Brain Bank, in London. In view of the paucity of detailed neuropathological reports of the disease since the 1940s, we have carried out a clinicopathological study of eight recent cases. A spectrum of pathological change was seen, with highly variable involvement of cortical, subcortical and brainstem structures. There was no correlation between severity of disease and severity of pathology. The anatomical distribution of le

Distribution of neurofibrillary tangles and senile plaques in the cerebral cortex in postencephalitic parkinsonism.

Postencephalitic parkinsonism is characterized neuropathologically by severe loss of pigmented neurons in the substantia nigra and the presence of high densities of neurofibrillary tangles in several brainstem structures. In 5 cases of postencephalitic parkinsonism, we observed that the neurofibrillary tangle distribution in the cerebral cortex predominated in the hippocampus and entorhinal cortex. In the prefrontal and inferior temporal cortex, neurofibrillary tangles were preferentially localized in layers II and III. This pattern contrasts with the neurofibrillary tangle distribution observ

[Changes in lipid metabolism in patients with Parkinson disease after combined treatment with L-deprenil (iumex) and antiparkinson drugs].

Overall 85 patients with different forms of parkinsonism placed under observation were examined for lipid fractions in the blood serum and cerebrospinal fluid. The greatest increase of blood serum phospholipids was discovered in patients with postencephalitic parkinsonism; patients with vascular and posttraumatic parkinsonism, tended towards their decrease as compared to the control group. In patients with Parkinson's disease, the content of phospholipids in the blood serum was within normal. In patients with postencephalitic parkinsonism and Parkinson's disease, there was a rise of lysolecith

Oculogyric crises and parkinsonism. A case of recent onset.

Oculogyric crisis in association with postencephalitic parkinsonism has been reported only following encephalitis lethargica (Economo's disease). Generally, it has been assumed that this phenomenon would fade away with the demise of these postencephalitic patients. Therefore, we report a case of recent onset of persistent oculogyric crisis with parkinsonism in a 35-year-old man following an apparent attack of encephalitis in 1972.

Parkinson's disease: distribution of Lewy bodies and monoamine neuron system.

A systematic study of the central and peripheral nervous systems in 3 cases of Parkinson's disease has demonstrated that Lewy bodies are present in 27 nuclei. Of these 20 nuclei (12 pigmented and 8 unpigmented) are involved in 2 or all 3 cases. It is noticed that the distribution of Lewy bodies in Parkinson's disease described here corresponds surprisingly well to that of monoamine (dopamine, noradrenaline and serotonin) cell bodies demonstrated in rats by the histochemical fluorescence method. This correlation is similar to that of Alzheimer's neurofibillary changes in postencephalitic Parkin

Response of patients with postencephalitic Parkinsonism to levodopa.

Thirty patients with postencephalitic Parkinsonism were treated with levodopa. In seven patients, treatment was withdrawn because of adverse reactions or insufficient benefit. Improvements observed in 26 patients who remained on treatment for a sufficient period to permit adequate assessment were comparable with those previously reported in Parkinson's disease. Some sequelae of encephalitis lethargica-notably, oculogyria and drooling-were appreciably improved in most patients affected, whereas choreoathetoid dyskinesias were aggravated by levodopa. Adventitious choreiform involuntary movements

Excretion of dopamine in diseases of basal ganglia.

The urinary excretion of catecholamines has been measured in 32 patients with disorders of the basal ganglia. Sixteen patients with Parkinsonism (idiopathic, postencephalitic, and arteriosclerotic types) had a significantly lower amount of dopamine in the urine during a 24-hour period than a group of 24 normal control subjects. In a group of 16 patients with various striatal syndromes the excretion of dopamine and epinephrine was significantly higher than normal. Norepinephrine excretion was similar in the three groups. The lowest mean value of urinary dopamine was found in postencephalitic Pa
