Identification of patulin-producing Penicillium isolates from decayed apples, their patulin production profiles, and the feasibility of high-purity patulin preparation.

This study presents a combined evaluation of patulin contamination by integrating fungal identification, assessment of toxin production, and the feasibility of obtaining high-purity patulin from biological sources. Fungal isolates recovered from naturally decayed apples collected from different agricultural environments in Türkiye were identified using MALDI-TOF MS and morphological characteristics, and were assigned to Penicillium expansum at a probable species level. The patulin production potential of the isolates was evaluated by inoculating them onto red, yellow, and green apple matrices

In Vitro and In Vivo Evaluation of Chlorogenic Acid-Encapsulated Lignin on Patulin Adsorption and Alleviation of Patulin-Induced Colonic Damage.

Patulin (PAT) is a water-soluble mycotoxin that causes digestive tract damage and liver and kidney function abnormalities. The current control approaches only reduce the amount of PAT in raw materials and food, which is difficult to remove once ingested in the body. In this study, lignin-based cross-linked particles loaded with chlorogenic acid were prepared, which intervened the liver and colon damage caused by PAT in mice. In the simulated digestion process in vitro, the accumulated adsorption capacity of the LB/CA-SH for PAT was 0.934 mg/g. LB/CA-SH intervention reversed the shortening of t

Reduction of patulin during apple juice clarification.

Patulin is a mycotoxin produced by a number of molds involved in fruit spoilage. This compound is carcinogenic and teratogenic. Various methods are currently used to reduce the levels of patulin in apple juice, namely, charcoal treatment, chemical preservation (sulfur dioxide), gamma irradiation, fermentation, and trimming of fungus-infected apples. Many of these processes are expensive and time-consuming. Therefore, there is a need to find a convenient and economical process to control patulin levels. This study was undertaken to evaluate the effectiveness of several clarification processes f

Immunosuppressive effects of the mycotoxin patulin in macrophages.

Patulin (PAT) is a fungi-derived secondary metabolite produced by numerous fungal species, especially within Aspergillus, Byssochlamys, and Penicillium genera, amongst which P. expansum is the foremost producer. Similar to other fungi-derived metabolites, PAT has been shown to have diverse biological features. Initially, PAT was used as an effective antimicrobial agent against Gram-negative and Gram-positive bacteria. Then, PAT has been shown to possess immunosuppressive properties encompassing humoral and cellular immune response, immune cell function and activation, phagocytosis, nitric oxid

Effect of preservatives on patulin production by Penicillium expansum.

Penicillium expansum has been grown on Capek-Dox medium using glucose and fructose as carbon source. Preservatives used in fruit processing and introduced in the medium were sorbic acid, formic acid, benzoic acid, SO2 and saccharose. Sulphur dioxide had a most inhibitory effect on mycelium growth and patulin production, formic acid concentration of 0.025% increased the amount of patulin by about 30% as compared to the culture with no preservatives. However its higher concentrations inhibited synthesis of this mycotoxin. Sorbic acid concentration of 0.1% stimulated the fungus strains examined i

Stability of patulin to sulfur dioxide and to yeast fermentation.

The affinity of patulin for sulfur dioxide (SO2) is much less than was previously reported and is of little significance at the SO2 concentrations (below 200 ppm) used in the processing of apple juice and cider. However, at concentrations of 2000 ppm SO2 and 15 ppm patulin, combination was 90% complete in 2 days. Removal of SO2 liberated only part of the patulin, which suggests that 2 mechanisms are involved: one reversible (opening the hemiacetal ring) and one irreversible (SO2 addition at the double bond). Test with 2 yeasts used in English commercial cider making confirmed that patulin is e

Evaluation of the reproductive toxicity of patulin in growing male rats.

Patulin is a mycotoxin produced by several Penicillium, Aspergillus and Byssachlamys species. Patulin can be produced on different food products including fruits, grains, cheese, cured meats, but in natural situations patulin is exclusively found in apple and apple products. Patulin, at dose of 0.1mg/kg bw/day, was administered by gavage to the growing male rats aged 5-6 week for 60 or 90 days. At the end of the experiment, sperm counts and morphology were investigated. Also, effects of patulin on the epididymis, seminal vesicle and prostate tissues were examined histopathologically and morpho

Quantitation of patulin pathway metabolites using gas-liquid chromatography.

A gas-liquid chromatographic system which allows most of the metabolites of the patulin pathway to be separated and quantitated has been developed. The metabolites are mostly phenols, and after conversion to their trimethylsilyl derivatives they are efficiently separated on a 3.18 mm O.D. stainless-stell column of 10% QF-1 on Gas-Chrom Q using a temperature programme. The detection limits for the three phenolic acids, 6-methylsalicylic acid, m-hydroxybenzoic acid and gentisic acid are all markedly lower than those obtained in previous systems. The usefulness of the system has been assessed by

Involvement of caspase in patulin-induced hepatotoxicity in vitro and in vivo.

Patulin (PAT) a kind of mycotoxin, is a widely disseminated mycotoxin found in agricultural products and could cause liver damage. However, evidence on the underlying mechanisms of patulin is still lacking. In the present study, Human liver cancer cells (HepG2) together with a mouse model were used to explore the possible effect and mechanism. The results demonstrated that PAT treatment inhibited cell proliferation and caused liver toxicity in mice. In vitro, PAT inhibited the growth of HepG2 cells in a dose-dependent manner and a time-dependent manner; lipid peroxidation, malondialdehyde (MDA

Intrinsic limitations on the continued production of the antibiotic patulin by Penicillium urticae.

The initial cause of the cessation of patulin biosynthesis in submerged cultures of Penicillium urticae was sought. The extrinsic limitations to patulin biosynthesis in fermentor cultures were successfully removed and kinetic profiles of the metabolites of the pathway and of three pathway enzymes were determined over a 60-h period. These profiles indicated that the first intrinsic limitation involved a decrease in the cellular content of the first enzyme of the pathway, 6-methylsalicylic acid synthetase. Other enzyme contents also decreased but on a significantly longer time scale. Thus the in

Elimination of patulin in apple juice using cysteine capped Fe3O4 nanoadsorbent.

The present study evaluated the preparation of L-cysteine capped Fe3O4 (magnetite) nanoparticles (LCys-Fe3O4 NPs), with a particular focus on their potential as an adsorbent and also the efficiency of these NPs in the removal of patulin from apple juice. The methods employed, including Scanning Electron Microscopy (SEM), Transmission Electron Microscopy (TEM), X-ray powder diffraction analysis (XRD), X-ray Photoelectron Spectroscopy (XPS), and Fourier Transform Infrared Spectroscopy (FT-IR), effectively showed the synthesis of LCys-Fe3O4 NPs. The proposed method was not only used for patulin r

Patulin Induces Acute Kidney Injury in Mice through Autophagy-Ferroptosis Pathway.

Patulin (PAT) is a common mycotoxin, widely found in cereals, seafood, nuts, and especially in fruits and their products. Exposure to this mycotoxin has been reported to induce kidney injury. However, the possible mechanism remains unclear. In our study, short-term high-dose intake of PAT caused acute kidney injury (AKI) in mice. We performed high-throughput transcriptional sequencing to identify differentially expressed genes (DEGs) between the treatment and control groups. The ferroptosis signaling pathway had the highest enrichment, suggesting ferroptosis is involved in PAT-induced AKI. Fur

Patulin Alters Insulin Signaling and Metabolic Flexibility in HepG2 and HEK293 Cells.

Non-communicable diseases (NCDs) have risen rapidly worldwide, sparking interest in causative agents and pathways. Patulin (PAT), a xenobiotic found in fruit products contaminated by molds, is postulated to be diabetogenic in animals, but little is known about these effects in humans. This study examined the effects of PAT on the insulin signaling pathway and the pyruvate dehydrogenase complex (PDH). HEK293 and HepG2 cells were exposed to normal (5 mM) or high (25 mM) glucose levels, insulin (1.7 nM) and PAT (0.2 μM; 2.0 μM) for 24 h. The qPCR determined gene expression of key enzymes involved

Combining Patulin with Cadmium Induces Enhanced Hepatotoxicity and Nephrotoxicity In Vitro and In Vivo.

Food can be contaminated by various types of contaminants such as mycotoxins and toxic heavy metals. Therefore, it is very likely that simultaneous intake of more than one type of food contaminant by consumers may take place, which provides a strong rationale for investigating the combined toxicities of these food contaminants. Patulin is one of the most common food-borne mycotoxins, whereas cadmium is a representative of toxic heavy metals found in food. The liver and kidneys are the main target organ sites for both patulin and cadmium. We hypothesized that simultaneous exposure to patulin an
