Phosphonates of Pectobacterium atrosepticum: Discovery and Role in Plant-Pathogen Interactions.

Many phytopathogens' gene products that contribute to plant-pathogen interactions remain unexplored. In one of the most harmful phytopathogenic bacterium Pectobacterium atrosepticum (Pba), phosphonate-related genes have been previously shown to be among the most upregulated following host plant colonization. However, phosphonates, compounds characterized by a carbon-phosphorus bond in their composition, have not been described in Pectobacterium species and other phytopathogenic bacteria, with the exception of Pseudomonas syringae and Pantoea ananatis. Our study aimed to determine whether Pba s

Spatiotemporal interplay between cAMP-CRP and KdgR regulates virulence in Pectobacterium carotovorum.

Phytopathogens encounter highly heterogeneous environments during infection. Carbon availability is a fundamental metabolic constraint that impacts bacterial virulence; however, the mechanism underlying the coordination of virulence traits based on metabolic cues remains unclear. We investigated the regulatory network of the soft-rot pathogen Pectobacterium carotovorum during Napa cabbage colonization, which generates tissue maceration with steep glucose and pectin gradients. Using GFP transcriptional fusions, we mapped the spatiotemporal expression of key regulatory (flhD, rsmA, and rsmB) and

Direct Binding of Salicylic Acid to Pectobacterium N-Acyl-Homoserine Lactone Synthase.

Salicylic acid (SA) is a hormone that mediates systemic acquired resistance in plants. We demonstrated that SA can interfere with group behavior and virulence of the soft-rot plant pathogen Pectobacterium spp. through quorum sensing (QS) inhibition. QS is a population density-dependent communication system that relies on the signal molecule acyl-homoserine lactone (AHL) to synchronize infection. P. parmentieri mutants, lacking the QS AHL synthase (expI-) or the response regulator (expR-), were used to determine how SA inhibits QS. ExpI was expressed in DH5α, the QS negative strain of Escherich

First Report of Pectobacterium carotovorum subsp. carotovorum on Spathiphyllum wallisii in Argentina.

Peace lily (Spathiphyllum wallisii Regel) is a popular ornamental potted plant in Argentina. During May of 2008 (austral autumn), necrotic lesions of unknown etiology were observed on S. wallisii in a nursery in Pontevedra (34°45'6″S, 58°42'42″W). Plants first showed water-soaked areas starting from the leaf tips. Infected tissue became irregular, brown, dark-to-black lesions on leaves ~12 to 14 mm in diameter surrounded by yellowish haloes. Disease incidence approached 30%. Abundant bacterial streaming was observed from lesions when examined at ×100. Bacteria isolated from lesions formed whit

Exl1 from Pectobacterium activates a jasmonate-dependent immune network in Arabidopsis thaliana.

Bacterial expansins contribute to the virulence of diverse plant pathogens, yet their biological activity and molecular roles during host interactions remain to be elucidated. Expansin-like protein, Exl1, from phytopathogens from the genus Pectobacterium, activates defense responses in Arabidopsis thaliana involving production of reactive oxygen species (ROS), and the jasmonic acid (JA), ethylene (ET), and salicylic acid (SA) pathways, ultimately reducing disease symptoms upon subsequent infections with P. brasiliense and the fungus Botrytis cinerea. However, the target of Exl1 within the plan

Biochemical and Molecular Characteristics of Pc1 Virulent Phage Isolate Infecting Pectobacterium carotovorum.

BACKGROUND AND OBJECTIVE: Pectobacterium carotovorum subsp. carotovorum is a plant-pathogenic bacterium. It is a post-harvest pathogen and causes soft rot diseases in infected plants. Different virulent bacteriophages have been isolated from different regions in the world. These bacteriophages were tolerant to high concentrations of calcium chloride and magnesium chloride. Whereas, the high concentrations of zinc chloride and aluminum chloride decreased the activity and stability of phages. Therefore, the present research aimed to study the biology of P. carotovorum phage (Pc1) by using a one-

First Report of Pectobacterium carotovorum Causing Soft Rot of Opium Poppy in Spain.

Opium poppy (Papaver somniferum L.) is an economically important pharmaceutical crop in Spain. Approximately 8,000 ha are cultivated annually in southern and central Spain. To improve yields, opium poppy cultivation is expanding to more humid or irrigated areas of Spain. In the springs of 2005 and 2007, we observed poppy plants with wilt and stem rot symptoms in irrigated, commercial opium poppy (cv. Nigrum) at Carmona and Écija, which are in Seville Province in southern Spain. Closer observations of affected plants revealed darkening and water soaking of the leaves and stem at the soil level,

Impact of Homologous Recombination on Core Genome Evolution and Host Adaptation of Pectobacterium parmentieri.

Homologous recombination is a major force mechanism driving bacterial evolution, host adaptability, and acquisition of novel virulence traits. Pectobacterium parmentieri is a plant bacterial pathogen distributed worldwide, primarily affecting potatoes, by causing soft rot and blackleg diseases. The goal of this investigation was to understand the impact of homologous recombination on the genomic evolution of P. parmentieri. Analysis of P. parmentieri genomes using Roary revealed a dynamic pan-genome with 3,742 core genes and over 55% accessory genome variability. Bayesian population structure

Host range and molecular phylogenies of the soft rot enterobacterial genera pectobacterium and dickeya.

ABSTRACT Pectobacterium and Dickeya spp. are related broad-host-range entero-bacterial pathogens of angiosperms. A review of the literature shows that these genera each cause disease in species from at least 35% of angiosperm plant orders. The known host ranges of these pathogens partially overlap and, together, these two genera are pathogens of species from 50% of angiosperm plant orders. Notably, there are no reported hosts for either genus in the eudicots clade and no reported Dickeya hosts in the magnoliids or eurosids II clades, although Pectobacterium spp. are pathogens of at least one p

Phylogeny and virulence of naturally occurring type III secretion system-deficient Pectobacterium strains.

Pectobacterium species are enterobacterial plant-pathogenic bacteria that cause soft rot disease in diverse plant species. Previous epidemiological studies of Pectobacterium species have suffered from an inability to identify most isolates to the species or subspecies level. We used three previously described DNA-based methods, 16S-23S intergenic transcribed spacer PCR-restriction fragment length polymorphism analysis, multilocus sequence analysis (MLSA), and pulsed-field gel electrophoresis, to examine isolates from diseased stems and tubers and found that MLSA provided the most reliable clas

Kojic acid attenuates tomato soft rot caused by Pectobacterium carotovorum subsp. carotovorum through quorum quenching.

Pectobacterium carotovorum subsp. carotovorum (Pcc) is the causal agent of bacterial soft rot disease, which triggers rapid decay of infected tomato fruit. Kojic acid, primarily a fermentation metabolite of Aspergillus oryzae, exhibits biocontrol activity against postharvest soft rot of tomato. In this work, we investigated the interference of kojic acid with quorum sensing (QS) in Pcc and elucidated the relevant mechanisms to provide novel theoretical insights into the management of tomato postharvest soft rot. The results demonstrated that kojic acid at a sub-inhibitory concentration (1/4 MI

Priming of the Arabidopsis pattern-triggered immunity response upon infection by necrotrophic Pectobacterium carotovorum bacteria.

Boosted responsiveness of plant cells to stress at the onset of pathogen- or chemically induced resistance is called priming. The chemical β-aminobutyric acid (BABA) enhances Arabidopsis thaliana resistance to hemibiotrophic bacteria through the priming of the salicylic acid (SA) defence response. Whether BABA increases Arabidopsis resistance to the necrotrophic bacterium Pectobacterium carotovorum ssp. carotovorum (Pcc) is not clear. In this work, we show that treatment with BABA protects Arabidopsis against the soft-rot pathogen Pcc. BABA did not prime the expression of the jasmonate/ethylen

The chemical structure of the O-antigen and enterobacterial common antigen of Pectobacterium brasiliense NCPPB 4609TS.

Pectobacterium brasiliense is a widely distributed phytopathogenic bacterium that causes diseases such as soft rot and blackleg, leading to significant yield losses in potatoes as well as other vegetables and ornamental plants. Lipopolysaccharide (LPS) is an important virulence factor that plays an essential role in colonisation of plant tissues and overcoming the host defence mechanisms. The O-polysaccharide from the LPS of P. brasiliense strain NCPPB 4609TS (=CFBP 6617TS = LMG 21371TS = IFB5390) was structurally characterised using spectroscopic techniques and chemical methods. A Novel Plant Secondary Metabolite-Binding Gene Family Required for Full Virulence in Pectobacterium parvum.

Pectobacterium parvum is an emerging phytopathogen causing aerial stem rot of potato. P. parvum has a broad host range, with Chinese cabbage as one of its important hosts. Through multi-omics analysis, we discovered a previously uncharacterized gene family highly expressed during in planta infection, designated the plant secondary metabolite-binding (PSMB) family. This family likely originated via horizontal gene transfer and underwent lineage-specific expansion in P. parvum, forming a four-paralog pathogenicity island. Structural modelling revealed that PSMB proteins resemble the rhizobial Rh

Control of the bacterial soft rot pathogen, Pectobacterium carotovorum by Bacillus velezensis CE 100 in cucumber.

Pectobacterium carotovorum is a problematic bacterial pathogen causing soft rot in different vegetable crops, resulting in yield losses during pre- and post-harvest periods. In this study, Bacillus velezensis CE 100 showed antibacterial activity against P. carotovorum. Co-inoculation experiment indicated that B. velezensis CE 100 reduced the proliferation rate of P. carotovorum at the early incubation period and that a long incubation time induced a loss of viability of the bacterial pathogen. Agar well diffusion assay revealed that the culture filtrate of strain CE 100 affected the growth of

Quorum sensing coordinates brute force and stealth modes of infection in the plant pathogen Pectobacterium atrosepticum.

Quorum sensing (QS) in vitro controls production of plant cell wall degrading enzymes (PCWDEs) and other virulence factors in the soft rotting enterobacterial plant pathogen Pectobacterium atrosepticum (Pba). Here, we demonstrate the genome-wide regulatory role of QS in vivo during the Pba-potato interaction, using a Pba-specific microarray. We show that 26% of the Pba genome exhibited differential transcription in a QS (expI-) mutant, compared to the wild-type, suggesting that QS may make a greater contribution to pathogenesis than previously thought. We identify novel components of the QS re

Trichokonins from Trichoderma pseudokoningii SMF2 induce resistance against Gram-negative Pectobacterium carotovorum subsp. carotovorum in Chinese cabbage.

Peptaibols, mainly produced by Trichoderma, play a pivotal role in controlling plant disease caused by fungi, virus, and Gram-positive bacteria. In the current study, we evaluated the control effect of Trichokonins, antimicrobial peptaibols from Trichoderma pseudokoningii SMF2, on soft rot disease of Chinese cabbage caused by a Gram-negative bacterium Pectobacterium carotovorum subsp. carotovorum and analyzed the mechanism involved. Trichokonins treatment (0.3 mg L(-1) ) enhanced the resistance of Chinese cabbage against Pcc infection. However, Trichokonins could hardly inhibit the growth of P

Plant growth-promoting archaea trigger induced systemic resistance in Arabidopsis thaliana against Pectobacterium carotovorum and Pseudomonas syringae.

Archaea have inhabited the earth for a long period of time and are ubiquitously distributed in diverse environments. However, few studies have focused on the interactions of archaea with other organisms, including eukaryotes such as plants, since it is difficult to cultivate sufficient numbers of archaeal cells for analysis. In this study, we investigated the interaction between soil archaea and Arabidopsis thaliana. We demonstrate for the first time that soil archaea promote plant growth and trigger induced systemic resistance (ISR) against the necrotrophic bacterium Pectobacterium carotovoru
