For more than a decade now, a search for answers to the following two questions has taken us on a new and exciting journey into the world of beta- and gamma-peptides: What happens if the oxygen atoms in a 3i-helix of a polymeric chain composed of (R)-3-hydroxybutanoic acid are replaced by NH units? What happens if one or two CH2 groups are introduced into each amino acid building block in the chain of a peptide or protein, thereby providing homologues of the proteinogenic alpha-amino acids? Our journey has repeatedly thrown up surprises, continually expanding the potential of these classes of

Antimicrobial resistance is a critical global healthcare issue that urgently needs the development of new antimicrobial agents. Herein, we developed new and discrete cationic amphipathic cyclic peptidomimetics as potential antimicrobial agents to combat multidrug-resistant bacteria. These cyclic oligomers of different alkyl chain lengths are easy to obtain via carbodiimide-mediated condensation reaction and using aza-crown ether as the core reactant. The antimicrobial activity, membrane perturbation effects, and toxicity of the compounds are strongly dependent on the hydrophobic alkyl chain le

Mediation of protein-protein interactions with molecules that bind strongly and selectively to one of the partners at the protein interface is a promising therapeutic strategy for myriad diseases. One such approach is the rational design of non-peptidic scaffolds that reproduce the display of amino acid side chains from one face of a secondary structural element. We have previously disclosed proof-of principle syntheses of β-strand mimetics composed of alternating (hetero)aromatic and cyclic urea units, conformationally preorganised through dipolar repulsion in organic solvents, that are in go

Biologics such as monoclonal antibodies (mAb) and soluble receptors represent new classes of therapeutic agents for treatment of several diseases. High affinity and high specificity biologics can be utilized for variety of clinical purposes. Monoclonal antibodies have been used as diagnostic agents when coupled with radionuclide, immune modulatory agents or in the treatment of cancers. Among other limitations of using large molecules for therapy the actual cost of biologics has become an issue. There is an effort among chemists and biologists to reduce the size of biologics which includes mono

The central position of thrombin in the coagulation cascade has made it a popular target for discovery of novel antithrombotic agents. Starting with hirudin, a natural peptide isolated from the medicinal leech, its shorter synthetic analogue hirulog, and argatroban, the first therapeutically used synthetic small-molecule thrombin active site inhibitor, hundreds of direct thrombin inhibitors have been discovered over the last 20 years. Most of them are peptidomimetic compounds, based on the amino acid sequence of fibrinogen which binds into the thrombin active site. Since elucidation of the cryst

The (re)emergence of multidrug-resistant viruses and the emergence of new viruses highlight the urgent and ongoing need for new antiviral agents. The use of peptidomimetics as therapeutic drugs has often been associated with advantages, such as enhanced binding affinity, improved metabolic stability, and good bioavailability profiles. The development of novel antivirals is currently driven by strategies of converting peptides into peptidomimetic derivatives. In this review, we outline different structural modification design strategies for developing novel peptidomimetics as antivirals, involv

Herein we report the synthesis of a series of novel constrained peptidomimetics 2-10 endowed with a dipeptide backbone (D-Ser-Gly) and a vinyl ester warhead, structurally related to a previously identified lead compound 1, an irreversible inhibitor of falcipain-2, the main haemoglobinase of lethal malaria parasite Plasmodium falciparum. The new compounds were evaluated for their inhibition against falcipain-2, as well as against cultured P. falciparum. The inhibitory activity of the synthesized compounds was also evaluated against another protozoal cysteine protease, namely rhodesain of Trypan

Peptidomimetics have emerged as indispensable tools in medicinal chemistry, renowned for their therapeutic uses and ability to mimic protein secondary structures. Despite their biological and structural relevance, their use in the design of higher order supramolecular structures has remained largely unexplored. Herein, we report supramolecular hydrogelation from a minimalist-designed peptidomimetic composed of a non-canonical β-hydroxy-γ-amino acid (statine) and canonical phenylalanine (Phe). The resultant hydrogel comprises entangled fibers, exhibits viscoelastic and thixotropic properties, i

Human immunodeficiency virus 1 (HIV-1) infection remains a public health concern globally. Although great strides in the management of HIV-1 have been achieved, current highly active antiretroviral therapy is limited by multidrug resistance, prolonged use-related effects, and inability to purge the HIV-1 latent pool. Even though novel therapeutic options with HIV-1 broadly neutralizing antibodies (bNAbs) are being explored, the scalability of bNAbs is limited by economic cost of production and obligatory requirement for parenteral administration. However, these limitations can be addressed by

Misfolding of the human protein α-synuclein results in toxic fibrils and the aggregation of Lewy bodies, which are a hallmark of Parkinson's disease in brain tissue. Here we disclose a supramolecular approach where peptidomimetics are rationally designed and pre-organised to recognize the surface of native helical α-Syn by forming complementary contacts with key patches of protein surface composed of charged and hydrophobic residues. Under lipid-catalyzed conditions the mimetics slow the rate of aggregation (thioflavin-T assay) and disrupt the misfolding pathway (electron microscopy of aggrega

Alzheimer's disease is characterized by the oligomerization and amyloid fibril formation of amyloid β-peptide (Aβ). We describe a novel class of small water-soluble Aβ binding peptidomimetics based on two hydrophobic Ala-Val and Val-Leu dipeptides linked to a D-glucopyranosyl scaffold through aminoalkyl and carboxyethyl links in C1 and C6 positions. These compounds combine the targeting of hydrophobic recognition interfaces with an original hydrophilic sugar β-breakage strategy. These molecules were shown, by fluorescence thioflavin-T assays, to dramatically slow down the kinetics of amyloid f

In the search for new and effective prodrugs against the herpes simplex virus, a series of acyclovir analogues with a thiazole ring containing amino acids (glycine, alanine, valine, leucine) has been investigated. The chemical stability of some of the compounds containing different residues was studied at pH 1 and pH 7.4 at a temperature of 37°C. An HPLC method was developed for quantification of the unchanged ester concentration. Some of the esters (Gly-thiazole, Ala-thiazole-acyclovir, Leu-thiazole-acyclovir) were rather unstable, especially under acidic conditions, and underwent rapid hydrol

Cationic amphipathic poly(ethylene glycol)-based polymers are generated with synthetic efficiencies allowing their evaluation as antimicrobial peptide (AMP) mimetics. Accordingly, statistical copolymers with cationic units consistently functionalized with guanyl groups, but different side-chain lengths, and hydrophobic units displaying long aliphatic, branched, and/or aromatic side chains are produced and tested for their antimicrobial and hemolytic properties. The results obtained indicate that candidates with activities and selectivity commensurate to some AMPs can be obtained and that furth

In response to the growing threat of antimicrobial resistance, scaffold-based peptidomimetics were explored as potential antibacterial agents. This innovative design approach involved fragment display on small-molecule scaffolds of ultrashort peptides and peptidomimetics with alternating cationic/hydrophobic motifs. Compounds, based on ultrashort peptides or peptide/β-peptoid hybrids attached to small-molecule scaffolds comprising bis-, tris-, and tetrakis(bromomethyl)benzene derivatives as well as triamines, were tested for antibacterial activity against a panel of Gram-negative and Gram-posi

Worldwide efforts are underway to develop new antimicrobial agents against bacterial resistance. To identify new compounds with a good antimicrobial profile, we designed and synthesized two series of small cationic antimicrobial peptidomimetics (1-8) containing unusual arginine mimetics (to introduce cationic charges) and several aromatic amino acids (bulky moieties to improve lipophilicity). Both series were screened for in vitro antibacterial activity against a representative panel of Gram-positive (Staphylococcus aureus and Staphylococcus epidermidis) and Gram-negative (Escherichia coli and

The development of age-related neurodegenerative diseases is associated with the accumulation of damaged and misfolded proteins. Such proteins are eliminated from cells by proteolytic systems, mainly by 20S proteasomes, whose activity declines with age. Its stimulation has been recognized as a promising approach to delay the onset or ameliorate the symptoms of neurodegenerative disorders. Here we present peptidomimetics that are very effective in stimulating the proteasome in biochemical assays and in cell culture. They are stable in human plasma and capable of penetrating the cell membranes.

Thirteen cationic peptidomimetics derived from amino acids bearing an alkyl or ethynylphenyl moiety that mimic the structure of cationic antibacterial peptides were designed and synthesized using a simple coupling reaction of an amino acid with a substituted amine. Antibacterial activities of the resulting peptidomimetics against drug-sensitive bacteria, such as Gram-positive Staphylococcus aureus (S. aureus) and Bacillus subtilis, Gram-negative Escherichia coli (E. coli) and Salmonella enterica, and a drug-resistant bacterium, methicillin-resistant S. aureus (MRSA), were systematically evalua

The human immune system is a highly integrated and dynamic network designed to detect, respond to, and eliminate a wide range of infectious agents while preserving tissue homeostasis. This chapter provides an overview of the fundamental organization and function of innate and adaptive immunity, followed by a focused description of the human infection cycles of major pathogens of global health relevance, including Trypanosoma brucei (human African trypanosomiasis), T. cruzi (Chagas disease), Leishmania spp., Schistosoma mansoni, Plasmodium spp., and arboviruses such as Zika and dengue viruses.

The library of selected peptidomimetics was synthesised via Passerini 3-component reaction. The SAR study of obtained α-acyloxy carboxamides for the catalytic activity of lipase from Candida rugosa was undertaken. The compounds were tested as potential CRL modulators on model 4-nitrophenol laurate hydrolysis reaction. Subsequently, three activators (EC50: 33-41 nM) and one inhibitor (IC50: 3.35 μM) were identified. The highest activity was observed in the presence of 1-((4-methoxybenzyl)amino)-1-oxo-3-phenylpropan-2-yl 2-phenylacetate, then the CRL was 2.3-fold more active than native enzyme,

Peptidomimetics have been extensively explored in many area due to their ability to improve pharmacological qualities and interesting biological activities. Cycles could be incorporated in peptides to reduce their flexibility, often enhancing the affinity for a certain receptor. Many efforts have been made to synthesize various peptidomimetics. Among them, the Ugi reaction is a popular way for the synthesis of peptidomimetics because it provides peptide-like products. The Ugi reaction consists of the condensation of an aldehyde or ketone, a carboxylic acid, an amine, and an isocyanide usually
