Cementum and Periodontal Ligament Regeneration.

The unique anatomy and composition of the periodontium make periodontal tissue healing and regeneration a complex process. Periodontal regeneration aims to recapitulate the crucial stages of wound healing associated with periodontal development in order to restore lost tissues to their original form and function and for regeneration to occur, healing events must progress in an ordered and programmed sequence both temporally and spatially, replicating key developmental events. A number of procedures have been employed to promote true and predictable regeneration of the periodontium. Principally

Periodontal regeneration using periodontal ligament stem cell-transferred amnion.

Periodontal disease is characterized by the destruction of tooth supporting tissues. Regeneration of periodontal tissues using ex vivo expanded cells has been introduced and studied, although appropriate methodology has not yet been established. We developed a novel cell transplant method for periodontal regeneration using periodontal ligament stem cell (PDLSC)-transferred amniotic membrane (PDLSC-amnion). The aim of this study was to investigate the regenerative potential of PDLSC-amnion in a rat periodontal defect model. Cultured PDLSCs were transferred onto amniotic membranes using a glass

Periodontal wound healing in the absence of periodontal ligament cells.

This experiment was designed to study the ability of mucoperiosteal flap connective tissue to form new attachment on partially demineralized roots as well as nondemineralized roots in a wound-model where periodontal ligament (PDL) cells are absent, and ingrowth of bone granulation tissue is controlled. After reflecting mucoperiosteal flaps, bone was removed circumferentially around the distal root of mandibular second premolars and the mesial root of mandibular third premolars in five beagle dogs. The roots were completely denuded from the cementoenamel junction to the apex and curetted to rem

Conditioned Medium from Periodontal Ligament Stem Cells Enhances Periodontal Regeneration.

Periodontal disease is one of the most common infectious diseases in adults and is characterized by the destruction of tooth-supporting tissues. Mesenchymal stem cells (MSCs) comprise the mesoderm-originating stem cell population, which has been studied and used for cell therapy. However, because of the lower rate of cell survival after MSC transplantation in various disease models, paracrine functions of MSCs have been receiving increased attention as a regenerative mechanism. The aim of this study was to investigate the regenerative potential of transplanted conditioned medium (CM) obtained

Mechanoresponsive Properties of the Periodontal Ligament.

The periodontal ligament (PDL) functions as an enthesis, a connective tissue attachment that dissipates strains created by mechanical loading. Entheses are mechanoresponsive structures that rapidly adapt to changes in their mechanical loading; here we asked which features of the PDL are sensitive to such in vivo loading. We evaluated the PDL in 4 physiologically relevant mechanical environments, focusing on mitotic activity, cell density, collagen content, osteogenic protein expression, and organization of the tissue. In addition to examining PDLs that supported teeth under masticatory loading

Extracellular vesicles from induced pluripotent stem cell-derived periodontal ligament cells enhance proliferation and migration of periodontal ligament cells for periodontal regeneration.

BACKGROUND: Periodontal ligament (PDL) cells play a central role in periodontal tissue regeneration. We previously established induced pluripotent stem cell-derived PDL-like cells (iPS-PDL) as a potential alternative cell source for regenerative therapy. However, the properties and biological functions of extracellular vesicles (EVs) released by iPS-PDL remain unknown. Characterizing these EVs may facilitate the development of cell-free approaches for periodontal regeneration. OBJECTIVE: We aimed to isolate and characterize EVs derived from iPS-PDL (iPS-PDL-EVs) and compare their biological f

Compressive stress in periodontal ligament under orthodontic movements during periodontal breakdown.

INTRODUCTION: This analysis aimed to assess quantitatively and qualitatively the compressive stress (S3) in periodontal ligament in a gradual periodontal breakdown (0-8 mm) under orthodontic movements. Correlations between the applied forces, the level of bone resorption, the decrease of force magnitude, and S3 increase were also conducted. METHODS: On the basis of cone-beam computed tomography examinations (voxel size, 0.075 mm), nine 3-dimensional models of the second mandibular premolar with intact periodontium were created and then individually subjected to various levels of horizontal bo

The effects of periodontal ligament injection on pulpal and periodontal tissues.

This study was designed to investigate histologically the effects of the periodontal ligament injection on the pulp of teeth with or without subsequent amalgam restorations, and the effects of the injection on the periodontium. Eighty-eight injections were made on 55 teeth in dogs in intervals from 10-min to 30-day prefixation, while 18 teeth were not injected and evaluated as controls. After perfusion fixation, block sections were decalcified, sectioned longitudinally, stained, and evaluated microscopically by three independent examiners. The pulps were graded for degree of inflammation and t

Canine periodontal ligament stem cells as a tool for periodontal regeneration.

Canine periodontal disease, a progressive and irreversible condition, impacts dental health, posing a significant challenge in veterinary medicine. While current treatments focus on managing progression, canine periodontal ligament stem cells (cPDLSCs) offer regenerative potential through their self-renewal capacity, expression of stemness markers and multilineage differentiation. Autologous cPDLSCs have successfully regenerated alveolar bone, cementum, and Sharpey's fibres, while allogeneic cell transplants have demonstrated immunomodulatory benefits without triggering inflammatory reactions.

Periodontal Inflammation-Triggered by Periodontal Ligament Stem Cell Pyroptosis Exacerbates Periodontitis.

Periodontitis is an immune inflammatory disease that leads to progressive destruction of bone and connective tissue, accompanied by the dysfunction and even loss of periodontal ligament stem cells (PDLSCs). Pyroptosis mediated by gasdermin-D (GSDMD) participates in the pathogenesis of inflammatory diseases. However, whether pyroptosis mediates PDLSC loss, and inflammation triggered by pyroptosis is involved in the pathological progression of periodontitis remain unclear. Here, we found that PDLSCs suffered GSDMD-dependent pyroptosis to release interleukin-1β (IL-1β) during human periodontitis.

Periodontal healing by periodontal ligament fiber with or without cells: A preclinical study of the decellularized periodontal ligament in a tooth replantation model.

BACKGROUND: The periodontal ligament (PDL) comprises cells embedded in the extracellular matrix (ECM) and is required for periodontal healing upon tooth transplantation. However, the extent to which the ECM contributes to periodontal regeneration remains unknown. This study aimed to evaluate effects of separate PDL matrix without cells on periodontal healing. METHODS: Mandibular premolars extracted from six beagle dogs, were 1) left untreated, 2) decellularized, or 3) surface planed. Cytologic, histologic, scanning electron microscopy, and transmission electron microscopy analyses confirmed t

Biomimetic Tubular Matrix Induces Periodontal Ligament Principal Fiber Formation and Inhibits Osteogenic Differentiation of Periodontal Ligament Stem Cells.

Periodontal ligament (PDL) is assembled from highly organized collagen fiber bundles (PDL principal fibers) that are crucial in supporting teeth and buffering mechanical force. Therefore, regeneration of PDL needs to reconstruct these well-ordered fiber bundles to restore PDL functions. However, the formation of PDL principal fibers has long been a challenge due to the absence of an effective three-dimensional (3D) matrix to guide the growth of periodontal ligament stem cells (PDLSCs) and to inhibit the osteogenic differentiation of PDLSCs during the PDL principal fibers deposition. In this wo

The regenerative potential of the periodontal ligament.

The current decline in the incidence of dental caries indicates that patients will retain most of their dentition. Reconstruction or regeneration of the entire attachment apparatus, including the periodontal ligament, cementum, and bone, is an attainable therapeutic goal. Clinical treatments designed to reattach connective tissue to exposed root surfaces commonly result in the formation of a long junctional epithelial attachment. This attachment is probably not an effective barrier to bacterial toxins and may allow recurrent pocket formation. To enhance reattachment of connective tissue, root-

Material properties of periodontal ligaments.

The mechanism of orthodontic teeth movement is not entirely explained. The principal reaction on tissues at the cellular and molecular level is initiated by the force applied to the tooth crown and transferred in turn upon the periodontal ligament (PDL). It seems, therefore, that the PDL and particularly its properties play a key role in bone remodelling. One of the more commonly used methods, which is capable of analysis of a wide range of orthodontic movements or distribution of stress and strain within teeth and periodontium, is the finite element method (FEM). Aiming to achieve the FEM mod

Effects of periodontal disease on the proteomic profile of the periodontal ligament.

Periodontal disease affects over 1 billion people globally. This study investigated how periodontitis affects the protein profile of the periodontal ligament (PDL) in rats. Eight Holtzman rats were divided into control and experimental periodontitis groups. The PDL was isolated using laser capture microdissection and protein extracts were analyzed by mass spectrometry. Data analysis utilized specialized software, and Gene Ontology enrichment analysis identified significant protein functions. The data are available via ProteomeXchange with identifier PXD055817. Proteins such as SerpinB1, C5, an

Therapeutic potential of periodontal ligament stem cells.

Inflammatory periodontal disease known as periodontitis is one of the most common conditions that affect human teeth and often leads to tooth loss. Due to the complexity of the periodontium, which is composed of several tissues, its regeneration and subsequent return to a homeostatic state is challenging with the therapies currently available. Cellular therapy is increasingly becoming an alternative in regenerative medicine/dentistry, especially therapies using mesenchymal stem cells, as they can be isolated from a myriad of tissues. Periodontal ligament stem cells (PDLSCs) are probably the mo

Unipolarity of fibroblasts in rodent periodontal ligament.

In order to investigate whether fibroblasts in rodent periodontal ligament have a structural polarity, the position of the Golgi apparatus or the centriolar region in the cells was studied using light and electron microscopy. It appeared that, in the periodontal ligament of continuously erupting mouse and rat incisors, centrioles in fibroblasts on the tooth side of the ligament are preferably located in the anterior (occlusally directed) part of the cytoplasm. Polarity of fibroblasts in a single direction was less pronounced or absent on the bone side of the tissue. In the mouse, fibroblasts i

Programmed cell death of periodontal ligament cells.

The periodontal ligament is a crucial tissue that provides support to the periodontium. Situated between the alveolar bone and the tooth root, it consists primarily of fibroblasts, cementoblasts, osteoblasts, osteoclasts, periodontal ligament stem cells (PDLSCs), and epithelial cell rests of Malassez. Fibroblasts, cementoblasts, osteoblasts, and osteoclasts are functionally differentiated cells, whereas PDLSCs are undifferentiated mesenchymal stem cells. The dynamic development of these cells is intricately linked to periodontal changes and homeostasis. Notably, the regulation of programmed ce
