Permafrost Immunity.

Thawing permafrost is a serious and worrisome threat to the environment, because it releases trapped heavy metals and greenhouse gasses. Thawing permafrost is also a health threat because, in addition to releasing these noxious gasses, thawing permafrost may free novel and undiscovered antibiotic-resistant bacteria, viruses, fungi and parasites among a plethora of dormant pathogens. Our immune system is ill-prepared to counter these challenges, and will require significant adaptation, or allostasis, which can be subsumed under the generic term of permafrost immunity. Since most of the most gra

Approaches for the complex assessment of polychemical pollution of permafrost-affected soils and the upper layer of permafrost.

The analysis of about 200 samples taken from 42 permafrost-affected soil profiles was carried out on four key sites in different regions of cryolithozone (West Siberia, Central, North, and North-East Yakutia) characterized by different active layer depths and soil lithology. The aim of the study was to determine the influence of different processes of cryogenic mass-exchange on the redistribution and accumulation of major pollutants such as petroleum products, acid-soluble forms of trace elements, polycyclic hydrocarbons, and technogenic radionuclides transferred via atmospheric transport or a

CD71: Role in permafrost immunity.

Iron, an essential constituent of cell metabolism, is transported intra-cellularly bound to the ubiquitous 76 kDa blood glycoprotein transferrin via the transferrin receptor, CD71. Because of its structure, CD71 facilitates the binding and penetration of a large variety of viruses into the host. Among which the hemorrhagic fever-causing New World mammarena viruses (family of single stranded ambisense segmented RNA Arenaviridae), the single stranded positive sense RNA hepatitis C virus, the single stranded negative sense segmented influenza A virus, the single stranded negative sense RNA rabies

Permafrost viremia and immune tweening.

The immune system, an exquisitely regulated physiological system, utilizes a wide spectrum of soluble factors and multiple cell populations and subpopulations at diverse states of maturation to monitor and protect the organism against foreign organisms. Immune surveillance is ensured by distinguishing self-antigens from self-associated with non-self (e.g., viral) peptides presented by major histocompatibility complexes (MHC). Pathology is often identified as unregulated inflammatory responses (e.g., cytokine storm), or recognizing self as a non-self entity (i.e., auto-immunity). Artificial int

Movement of trichloroethene in a discontinuous permafrost zone.

At a site with discontinuous permafrost in Fairbanks, Alaska, releases of trichloroethene (TCE), an industrial solvent, have caused contamination of the groundwater. The objective of this study was to investigate the relationship between the migration pathway of the TCE groundwater plume and the distribution of the discontinuous permafrost at the site. The TCE plume configuration is substantially different than what regional hydrology trends would predict. Using GIS, we conducted a geostatistical analysis of field data collected during soil-boring installations and groundwater monitoring well

Permafrost Thaw Increases Methylmercury Formation in Subarctic Fennoscandia.

Methylmercury (MeHg) forms in anoxic environments and can bioaccumulate and biomagnify in aquatic food webs to concentrations of concern for human and wildlife health. Mercury (Hg) pollution in the Arctic environment may worsen as these areas warm and Hg, currently locked in permafrost soils, is remobilized. One of the main concerns is the development of Hg methylation hotspots in the terrestrial environment due to thermokarst formation. The extent to which net methylation of Hg is enhanced upon thaw is, however, largely unknown. Here, we have studied the formation of Hg methylation hotspots u

Abrupt stream acidification and metal mobilization from permafrost degradation.

Stream chemistry and ecosystem function are being transformed by abrupt acceleration of sulfide-mineral oxidation in permafrost-underlain headwater catchments of the Yukon and Mackenzie river basins-the two largest (sub)Arctic rivers in North America. Over the past decade, dozens of acidic (pH ~3) seepages have emerged in these headwaters, causing vegetation dieback and mobilizing metals at acutely toxic concentrations in receiving streams. Acid generated during sulfide-mineral oxidation also accelerates carbon dioxide emissions by driving carbonate-mineral dissolution. Downstream (sub)Arctic

Positive rhizosphere priming accelerates carbon release from permafrost soils.

Thawing permafrost soils are predicted to release substantial amounts of carbon by 2100. In addition to this, warming-induced active-layer deepening and increased rooting depth may result in further carbon losses from previously-frozen soil by stimulating microbial communities through fresh carbon inputs inducing positive rhizosphere priming. While models based on temperate data predict significant permafrost carbon loss through rhizosphere priming, data from permafrost soils are lacking. Here, we provide direct evidence of live plant-induced positive rhizosphere priming in permafrost and acti

Seasonal Fluctuations in Iron Cycling in Thawing Permafrost Peatlands.

In permafrost peatlands, up to 20% of total organic carbon (OC) is bound to reactive iron (Fe) minerals in the active layer overlying intact permafrost, potentially protecting OC from microbial degradation and transformation into greenhouse gases (GHG) such as CO2 and CH4. During the summer, shifts in runoff and soil moisture influence redox conditions and therefore the balance of Fe oxidation and reduction. Whether reactive iron minerals could act as a stable sink for carbon or whether they are continuously dissolved and reprecipitated during redox shifts remains unknown. We deployed bags of

Diversity and potential ecological roles of viruses in Pleistocene permafrost.

BACKGROUND: Ancient permafrost, formed during past glacial periods, is widespread in Siberia and other Arctic regions. Even though these soils have remained below 0 °C over geological time periods, the widespread presence of bacteria and archaea in permafrost is well documented. However, the diversity of viruses in ancient permafrost of different geological ages and their potential ecological roles are still poorly understood. RESULTS: We applied metagenomics to characterize viruses from Middle to Late Pleistocene permafrost sediments from Siberia. A total of 2697 viral operational taxonomic

Permafrost dynamics and the risk of anthrax transmission: a modelling study.

A recent outbreak of anthrax disease, severely affecting reindeer herds in Siberia, has been reportedly associated to the presence of infected carcasses or spores released from the active layer over permafrost, which is thawing and thickening at increasing rates, thus underlying the re-emerging nature of this pathogen in the Arctic region because of warming temperatures. Anthrax is a global zoonotic and epizootic disease, with a high case-fatality ratio in infected animals. Its transmission is mediated by environmental contamination through highly resistant spores which can persist in the soil

Does maritime Antarctic permafrost harbor environmental fungi with pathogenic potential?

We assessed the potentially pathogenic fungi present in Antarctic permafrost and the overlying active layer on King George, Robert, Livingston and Deception Islands in the South Shetland Islands archipelago, maritime Antarctica. Permafrost and active layer sub-samples were incubated at 37 °C to select fungi able to grow inside the human body. A total of 67 fungal isolates were obtained, 27 from the permafrost and 40 from the active layer. These represented 18 taxa of the genera Alternaria, Aspergillus, Curvularia, Penicillium, Rhodotorula and Talaromyces. The majority of fungi detected occurre

Snowball Earth termination by destabilization of equatorial permafrost methane clathrate.

The start of the Ediacaran period is defined by one of the most severe climate change events recorded in Earth history--the recovery from the Marinoan 'snowball' ice age, approximately 635 Myr ago (ref. 1). Marinoan glacial-marine deposits occur at equatorial palaeolatitudes, and are sharply overlain by a thin interval of carbonate that preserves marine carbon and sulphur isotopic excursions of about -5 and +15 parts per thousand, respectively; these deposits are thought to record widespread oceanic carbonate precipitation during postglacial sea level rise. This abrupt transition records a cli

Pleistocene mitogenomes reconstructed from the environmental DNA of permafrost sediments.

Traditionally, paleontologists have relied on the morphological features of bones and teeth to reconstruct the evolutionary relationships of extinct animals.1 In recent decades, the analysis of ancient DNA recovered from macrofossils has provided a powerful means to evaluate these hypotheses and develop novel phylogenetic models.2 Although a great deal of life history data can be extracted from bones, their scarcity and associated biases limit their information potential. The paleontological record of Beringia3-the unglaciated areas and former land bridge between northeast Eurasia and northwes

The Degree of Permafrost Degradation Determines the Stability of Soil Organic Matter.

Global warming profoundly alters the stability of soil organic matter (SOM) in permafrost zones, yet the underlying mechanisms governing SOM stability in permafrost landscapes at different stages of degradation remain unclear. This study utilised soil samples from the 0-50 cm layer collected from both continuous and isolated patches permafrost zones in the Daxing'an Mountains for laboratory incubation experiments at constant temperatures of 5 and 15 °C. The results for SOC, Fe and Al oxide content, δ 13C and MBC indicate that SOM in isolated patchy permafrost possesses greater bioavailability

Cooling perspectives on the risk of pathogenic viruses from thawing permafrost.

Climate change is inducing wide-scale permafrost thaw in the Arctic and subarctic, triggering concerns that long-dormant pathogens could reemerge from the thawing ground and initiate epidemics or pandemics. Viruses, as opposed to bacterial pathogens, garner particular interest because outbreaks cannot be controlled with antibiotics, though the effects can be mitigated by vaccines and newer antiviral drugs. To evaluate the potential hazards posed by viral pathogens emerging from thawing permafrost, we review information from a diverse range of disciplines. This includes efforts to recover infec

Recent advances in the study of mercury biogeochemistry in Arctic permafrost ecosystems.

Permafrost predominates in polar and high mountain regions, encompassing nearly 15 % of the exposed land in the Northern Hemisphere. It denotes soil or rock that remains at or below 0 °C for the duration of at least two consecutive years. These frozen soils serve as a barrier to contaminants that are stored and accumulated in permafrost over extended periods of time. One of these chemical compounds is mercury (Hg), a heavy metal well recognized for its severe toxic effects. Mercury presents a major risk worldwide to ecosystems, biota and human health and is strengthened by the Minamata Convent

Seasonal Regime Shift in the Viral Communities of a Permafrost Thaw Lake.

Permafrost thaw lakes including thermokarst lakes and ponds are ubiquitous features of Subarctic and Arctic landscapes and are hotspots of microbial activity. Input of terrestrial organic matter into the planktonic microbial loop of these lakes may greatly amplify global greenhouse gas emissions. This microbial loop, dominated in the summer by aerobic microorganisms including phototrophs, is radically different in the winter, when metabolic processes shift to the anaerobic degradation of organic matter. Little is known about the viruses that infect these microbes, despite evidence that viruses

Rock weathering can counteract river CO2 emissions induced by permafrost thaw.

Climate-induced permafrost thaw unlocks large stores of organic carbon that are mineralized and emitted as carbon dioxide (CO2) from rivers to the atmosphere1. Concurrently, warming and permafrost thaw can increase mineral weathering rates, thus affecting the release and sequestration of inorganic carbon2-4. Yet how these biological and geological carbon cycles interact and jointly affect CO2 dynamics (emission compared with drawdown) in permafrost rivers remains unknown5. Here we combine CO2 emissions, organic and inorganic solute concentrations, dual carbon isotopes (δ13C-Δ14C) and geochemic

Climate-carbon feedback tradeoff between Arctic and alpine permafrost under warming.

Whether greenhouse gas (GHG) emissions from permafrost will trigger positive climate feedbacks under warming remains unknown. Here, we synthesized the response of growing season carbon dioxide (CO2), methane (CH4), and nitrous oxide (N2O) emissions to experimentally manipulated warming of ~2°C for permafrost in alpine and Arctic regions. Warming weakened the GHG sink of alpine permafrost, thereby increasing (13%) its global warming potential, but strengthened the GHG sink of Arctic permafrost and decreased (-10%) its global warming potential. When warming caused drying of alpine permafrost soi
