Kinetics of Bacterial Inactivation by Peroxynitric Acid in the Presence of Organic Contaminants.

Low-temperature atmospheric-pressure plasma has been studied for disinfection purposes. When plasma is exposed to water, reactive oxygen and nitrogen species are generated and preserved in the water fraction (plasma-treated water [PTW]), which consequently exhibits bactericidal activity. At low temperatures, one of the bactericidal components of PTW is peroxynitric acid (PNA). Importantly, PNA can also be synthesized by chemical reaction, without exposure to plasma. In this study, we evaluated the bactericidal properties of PNA based on reaction kinetics in comparison with other disinfectants.

beta-Carotene effectively scavenges toxic nitrogen oxides: nitrogen dioxide and peroxynitrous acid.

beta-Carotene absorbed 2 equimolar amounts of NO2 accompanying the complete destruction of beta-carotene. Electron spin resonance study using 2-phenyl-4,4,5,5-tetramethylimidazoline-3-oxide-1-oxyl revealed that no significant amounts of NO were released by the interaction. Nitrogen atoms derived from NO2 were tightly bound to the beta-carotene molecules. Destruction of beta-carotene was inhibited little by alpha-tocopherol and polyunsaturated fatty esters, and slightly by ascorbyl palmitate, indicating that beta-carotene was a more effective scavenger of NO2. CdTe nanocrystals-enhanced chemiluminescence from peroxynitrous acid-carbonate and its application to the direct determination of nitrite.

It was found that CdTe semiconductor nanocrystals (NCs) can induce a great sensitized effect on chemiluminescence (CL) emission from peroxynitrous acid (ONOOH)-Na(2)CO(3) system. CL spectra, fluorescence (FL) spectra, and the quenching effect of reactive oxygen species were used to investigate the CL reaction mechanism. The CL intensity was proportional to the concentration of nitrite in the range from 0.05 to 50μM. The detection limit (S/N=3) was 0.024μM and the relative standard deviation (RSD) for five repeated measurements of 0.5μM nitrite was 4.2%. This method has been successfully applie

High microbicidal effect of peroxynitric acid on biofilm-infected dentin in a root carious tooth model and verification of tissue safety.

OBJECTIVES: Root-caries, which frequently occurs in elderly people, is more difficult to treat than caries in a tooth crown, especially in filling restorations. To overcome this difficulty, it is essential to find a strategy for sufficiently sterilizing the infected dentin; however, techniques for sterilizing carious pathogens inside the biofilm, called dental plaque, have not yet been established. Recently, dental applications of plasma sterilization technology have attracted attention. The mechanism of plasma sterilization became clear, and revealed that peroxynitric acid (PNA) is an effecti

The inflammatory oxidant peroxynitrous acid modulates the structure and function of the recombinant human V3 isoform of the extracellular matrix proteoglycan versican.

Continued oxidant production during chronic inflammation generates host tissue damage, with this being associated with pathologies including atherosclerosis. Atherosclerotic plaques contain modified proteins that may contribute to disease development, including plaque rupture, the major cause of heart attacks and strokes. Versican, a large extracellular matrix (ECM) chondroitin-sulfate proteoglycan, accumulates during atherogenesis, where it interacts with other ECM proteins, receptors and hyaluronan, and promotes inflammation. As activated leukocytes produce oxidants including peroxynitrite/p

Plasma pre-treatment and peroxynitric acid soaking as a novel oxidative seed disinfection strategy against seed-borne fungal pathogens in maize.

INTRODUCTION: Seed-borne fungal contamination is an important challenge in maize production and may contribute to grain deterioration and mycotoxin contamination. This study evaluated peroxynitric acid (PNA), alone and in combination with nonequilibrium gaseous plasma pretreatment, as a rapid oxidative strategy for disinfecting maize seeds contaminated with Aspergillus flavus and Fusarium fujikuroi. METHODS: Maize seeds were artificially inoculated with approximately 10⁶ spores of either A. flavus or F. fujikuroi. For plasma pretreatment, seeds were sequentially exposed to hydrogen (0.2 s) an

Determination of nitrite and glucose in water and human urine with light-up chromogenic response based on the expeditious oxidation of 3,3',5,5'-tetramethylbenzidine by peroxynitrous acid.

Contamination of food and drinking water by dangerous levels of nitrite has always been of strong concern and toxicity of this inorganic anion calls for convenient detection methods. Although some visible approaches were developed to deal with this problem, using environmentally incompatible organic reagents or functionalized nanoparticles may greatly limit their wide applications. In this article, we report a method to visibly detect nitrite in less than 1 min at room temperature. The efficacy of the method relies on a specific reaction of HNO2 with H2O2 to produce peroxynitrous acid (HOONO),

Combined effects of long-living chemical species during microbial inactivation using atmospheric plasma-treated water.

Electrical discharges in humid air at atmospheric pressure (nonthermal quenched plasma) generate long-lived chemical species in water that are efficient for microbial decontamination. The major role of nitrites was evidenced together with a synergistic effect of nitrates and H(2)O(2) and matching acidification. Other possible active compounds are considered, e.g., peroxynitrous acid.

Investigation of the reaction of peroxynitrite with propofol at acid pH: predominant production of oxidized, nitrated, and halogenated derivatives.

We reported here the reaction, in acidic conditions, of peroxynitrite (ONOO(-)) with the anaesthetic agent propofol (2,6-diisopropylphenol, PPF). The most interesting finding is that peroxynitrite is able to nitrate and to oxidize propofol leading to 4-nitropropofol, quinone, and diphenylquinone as the major products. More surprisingly, we also found that peroxynitrite is capable of halogenating propofol in the presence of halide ions, suggesting the formation of nitrosyl chloride (NOCl) or nitryl chloride (NO(2)Cl) from the reaction of peroxynitrite with chloride ions. A significant enhanceme

Frozen Hydrogen Peroxide and Nitrite Solution: The Acceleration of Benzoic Acid Oxidation via the Decreased pH in Ice.

We investigated benzoic acid oxidation via the reaction of hydrogen peroxide (H2O2) and nitrite (NO2-). The oxidation of benzoic acid by reactive nitrous acid (HONO) was negligible, and the reactivity of the H2O2/NO2- system decreased with a decrease in temperature under aqueous conditions. However, freezing markedly accelerated the chemical reaction. Based on Raman microscope measurements, concentrated species were confirmed in certain regions of the ice. We proposed that the change in nitrite speciation (accordingly, a decrease in the pH below pKa), derived from the freezing concentration ef

Peroxynitrite oxidation of sulfhydryls. The cytotoxic potential of superoxide and nitric oxide.

Peroxynitrite anion (ONOO-) is a potent oxidant that mediates oxidation of both nonprotein and protein sulfhydryls. Endothelial cells, macrophages, and neutrophils can generate superoxide as well as nitric oxide, leading to the production of peroxynitrite anion in vivo. Apparent second order rate constants were 5,900 M-1.s-1 and 2,600-2,800 M-1.s-1 for the reaction of peroxynitrite anion with free cysteine and the single thiol of albumin, respectively, at pH 7.4 and 37 degrees C. These rate constants are 3 orders of magnitude greater than the corresponding rate constants for the reaction of hy

Separation and detection of peroxynitrite using microchip electrophoresis with amperometric detection.

Peroxynitrite (ONOO(-)) is a highly reactive species implicated in the pathology of several cardiovascular and neurodegenerative diseases. It is generated in vivo by the diffusion-limited reaction of nitric oxide (NO(*)) and superoxide anion ((*)O(2)(-)) and is known to be produced during periods of inflammation. Detection of ONOO(-) is made difficult by its short half-life under physiological conditions (approximately 1 s). Here we report a method for the separation and detection of ONOO(-) from other electroactive species utilizing a microchip electrophoresis device incorporating an amperome

Investigation of a novel sterilization method for biofilms formed on titanium surfaces.

The development of effective methods to disinfect biofilms on dental materials is medically important. This study evaluated the bactericidal effects of peroxynitric acid (HOONO2; PNA) on biofilms formed on titanium surfaces. Streptococcus gordonii was cultured on either machined or rough titanium discs that were then used to evaluate the bactericidal effects of seven reagents, i.e., normal saline, benzalkonium chloride disinfectant solution, chlorhexidine digluconate solution, three concentration types of PNA, and inactivated PNA. Using low concentration of PNA, the bacterial count based on a

Comparison of the chemical reactivity of synthetic peroxynitrite with that of the autoxidation products of nitroxyl or its anion.

Donors of nitroxyl (HNO) exhibit pharmacological properties that are potentially favorable for treatment of a variety of diseases. To fully evaluate the pharmacological utility of HNO, it is therefore important to understand its chemistry, particularly involvement in deleterious biological reactions. Of particular note is the cytotoxic species formed from HNO autoxidation that is capable of inducing double strand DNA breaks. The identity of this species remains elusive, but a conceivable product is peroxynitrous acid. However, chemical comparison studies have demonstrated that HNO autoxidation

A Two-Photon Fluorescent Probe for the Visual Detection of Peroxynitrite in Living Cells and Zebrafish.

Peroxynitrite (ONOO-), as an important reactive oxygen species (ROS), holds great potential to react with a variety of biologically active substances, leading to the occurrence of various diseases such as cancer and neurodegenerative diseases. In this work, we developed a novel mitochondria-localized fluorescent probe, HDBT-ONOO-, which was designed as a mitochondria-targeting two-photon fluorescence probe based on 1,8-naphthylimide fluorophore and the reactive group of 4-(bromomethyl)-benzene boronic acid pinacol ester. More importantly, the probe exhibited good biocompatibility, sensitivity,

Engineering Coumarin-Based Afterglow Luminescence Probes for Activatable Imaging Peroxynitrite In Vivo.

Afterglow luminescence imaging materials have evolved from inorganic to organic systems, and further from macromolecular polymers to small molecules, highlighting the imperative of developing safe and efficient organic afterglow molecules for bioimaging. However, flexible organic afterglow molecules with well-characterized structure-luminescence intensity relationships for high-contrast activatable imaging remain scarce. Here, we report for the first time that coumarin derivatives exhibit bright afterglow luminescence and reveal that rigidified coumarin structures display stronger emission tha

Self-Targeting Carbon Quantum Dots for Peroxynitrite Detection and Imaging in Live Cells.

Peroxynitrite (ONOO-), a highly reactive nitrogen species (RNS) generated mainly in mitochondria, has been identified to be associated with numerous pathophysiological processes, and thus accurate ONOO- imaging with superior sensitivity and selectivity is highly desirable. Herein, we prepared a new type of carbon quantum dots (CQDs) with mitochondria-targeting function without the aid of any targeting molecules via a simple one-step hydrothermal route. The as-prepared CQDs not only displayed relatively uniform size distribution, few surface defects, high photostability, and excellent biocompat

Formation of methane sulfinic acid in the gas-phase OH-radical initiated oxidation of dimethyl sulfoxide.

Dimethyl sulfoxide (CH3S(O)CH3: DMSO) is an important product of dimethyl sulfide (CH3SCH3: DMS) photooxidation. The mechanism of the OH-radical initiated oxidation of DMSO is still highly uncertain and a major aim of recent studies has been to establish if methane sulfinic acid (CH3S(O)OH: MSIA) is a major reaction product In the present work the products of the OH-radical gas-phase oxidation of dimethyl sulfoxide have been investigated in the absence and presence of NOx All experiments were performed in a 1,080 L reaction chamber in 1,000 mbar synthetic air at 284 +/- 2 K using long-path FT-

Kinetics-Based Quantification of Peroxynitrite Using the Oxidative Decarbonylation of Isatin.

Peroxynitrite and its radical decomposition products are highly reactive nitrogen and oxygen species that can influence the balance between health and disease in multiple organ systems. Despite vigorous research activity, real-time quantitative monitoring of peroxynitrite generated by donor compounds remains challenging. Here, we report a kinetics-based fluorescence method for quantitative tracking of peroxynitrite generation using the oxidative decarbonylation of isatin to form anthranilic acid as a fluorescent probe. This method relies on knowledge of the rate of the reaction of peroxynitrit

Protection against peroxynitrite reactions by flavonoids.

Peroxynitrite is believed to contribute to pathogenesis of various diseases. Search for drugs and nutraceuticals able to limit peroxynitrite reactions is thus of interest. This study aimed to compare the capacity of 16 flavonoids and several other compounds present in food to prevent peroxynitrite reactions in three different test systems, based on fluorescein bleaching, tyrosine nitration and serum albumin thiol oxidation. The sequence of protective capacity of the antioxidants was different in various systems. Correlation analysis revealed that the hydroxyl group at the R'4 position of the f
