Immunohistochemical distribution of tachykinins in the CNS of the lamprey Petromyzon marinus.

The presence of tachykinins in the CNS of vertebrates has been known for many decades, and numerous studies have described their distribution in mammals. Tachykinins were also reported in the CNS of lampreys using immunohistochemistry, chromatography, and radioimmunoassay methods, but the use of substance P (SP)-specific antibodies to reveal those tachykinins could have led to an underestimation of their number in this genus. Therefore, we carried out a new immunohistochemical study on Petromyzon marinus using a commercial polyclonal antibody that binds not only to mammalian SP, but also to ot

Sea lamprey Petromyzon marinus: an exception to the rule of homing in anadromous fishes.

Anadromous fishes are believed to make regular circuits of migration in the sea before homing to their natal rivers. Sea lamprey Petromyzon marinus is an anadromous fish that is an exception to this life-history pattern. It also differs from other anadromous fishes in that its adult phase is parasitic, a feeding strategy that should make homing problematic for lamprey cohorts that become widely dispersed through transport by the diverse hosts they parasitize. We sequenced a portion of the mitochondrial DNA control region from sea lampreys collected from 11 North American east coast rivers to t

Melatonin secretion in vitro from the pineal complex of the lamprey Petromyzon marinus.

Melatonin secretion from cultured pineal complexes of the lamprey, Petromyzon marinus, was measured by radioimmunoassay (RIA) for 6 days under either a 12:12 light:dark cycle (L:D) or continuous darkness (D:D) at either 10 degrees or 20 degrees. Under the L:D cycle, melatonin release was completely suppressed during daytime and high during night-time, with larger amplitude at 20 degrees than at 10 degrees. Under D:D conditions, no significant circadian rhythmicity in secretion could be revealed by periodogram analysis at either 10 degrees or 20 degrees. This is the first direct evidence of mel

Convoluted bile ducts in the liver of the larval lamprey, Petromyzon marinus L.

The three-dimensional structure of the bile ducts and their relationship to the blood vessels were studied in the larval lamprey by scanning electron microscopy of the intact tissue and of biliary and vascular casts. The intrahepatic gall bladder is situated in the cephalic portion of the liver and a cystic duct is connected to a straight intrahepatic common bile duct, which extends to the extrahepatic bile duct at the caudal end of the liver. Several smaller intrahepatic common bile ducts are connected directly to the intrahepatic common bile duct, are convoluted or serpiginous and are surrou

Dopaminergic modulation of olfactory-evoked motor output in sea lampreys (Petromyzon marinus L.).

Detection of chemical cues is important to guide locomotion in association with feeding and sexual behavior. Two neural pathways responsible for odor-evoked locomotion have been characterized in the sea lamprey (Petromyzon marinus L.), a basal vertebrate. There is a medial pathway originating in the medial olfactory bulb (OB) and a lateral pathway originating from the rest of the OB. These olfactomotor pathways are present throughout the life cycle of lampreys, but olfactory-driven behaviors differ according to the developmental stage. Among possible mechanisms, dopaminergic (DA) modulation in

GABA immunoreactivity in the olfactory bulbs of the adult sea lamprey Petromyzon marinus L.

The distribution of gamma-aminobutyric acid (GABA) immunoreactivity in the olfactory bulbs of the adult sea lamprey was studied using an antibody against this transmitter. Five types of GABA-immunoreactive (GABAir) cells were observed. Medium-sized GABAir cells (periglomerular cells) were located around the olfactory glomeruli and occasionally within them. In the inner cellular layer of the bulbs and around the olfactory ventricles, two types of GABAir perikarya were present: some medium-sized GABAir cells and numerous small GABAir cells (granules). In the walls of the olfactory ventricle, som

Liver metabolomic profiles of sea lamprey (Petromyzon marinus) are influenced by sex and maturation stages.

INTRODUCTION: Sea lamprey (Petromyzon marinus) is a unique vertebrate model to examine how liver metabolomes support different reproductive functions. Juvenile sea lamprey prey on other fish species by attaching to their body and feeding on their blood and body fluids. Once reaching adulthood, they cease feeding, migrate to spawning streams and begin their final sexual maturation. During these processes, the male livers produce large quantities of bile acid pheromone precursors to be modified and released via gills, whereas the female livers synthesize vast amounts of vitellogenin (yolk lipoph

Molecular cloning and pharmacological characterization of two novel GnRH receptors in the lamprey (Petromyzon marinus).

This paper reports the identification, expression, binding kinetics, and functional studies of two novel type III lamprey GnRH receptors (lGnRH-R-2 and lGnRH-R-3) in the sea lamprey, a basal vertebrate. These novel GnRH receptors share the structural features and amino acid motifs common to other known gnathostome GnRH receptors. The ligand specificity and activation of intracellular signaling studies showed ligands lGnRH-II and -III induced an inositol phosphate (IP) response at lGnRH-R-2 and lGnRH-R-3, whereas the ligand lGnRH-I did not stimulate an IP response. lGnRH-II was a more potent ac

The origin of trochlear motoneurons in the larval sea lamprey, Petromyzon marinus L. An HRP study.

The origin of trochlear motoneurons in larval lampreys was studied by injection of HRP into the orbit. Immature motoneurons had ventricular attachments, the position of which with respect to the ventricular sulci was used to study their regions of origin. Motoneurons originate from the region between the sulcus intermedius dorsalis and the sulcus intermedius ventralis, which in other parts of the brain were identified as the visceromotor and viscerosensory columns. Both ipsilateral and contralateral immature motoneurons were found. The significance of these findings is discussed in relation to

Serotonergic nerve fibers in the primary olfactory pathway of the larval sea lamprey, Petromyzon marinus.

In this study, serotonin (5-hydroxytryptamine; 5HT)-immunoreactive (5HT-IR) neuronal fibers were identified in the primary olfactory pathway of the sea lamprey. These neurons are likely part of a nonolfactory neural system that innervates the olfactory sac. Cell bodies with 5HT immunoreactivity predominated in the lamina propria of the rostral portion of the nasal cavity and were less prevalent adjacent to the olfactory epithelium. The 5HT-IR fibers were parallel to axons of the olfactory receptor neurons in the lamina propria of the olfactory mucosa and in the olfactory nerve. Serotonergic fi

Cloning and characterization of the pineal gland-specific opsin gene of marine lamprey (Petromyzon marinus).

We have cloned and sequenced the pineal gland-specific opsin (P opsin) gene, p(Pm), of marine lamprey, P. marinus. The deduced P opsin, P(Pm), consisting of 444 aa, is about 90 aa longer than the orthologous opsins of chicken, pigeon, and American chameleon. The evolution of P(Pm) is characterized not only by a significantly higher rate of aa replacement than the other P opsins but also by a larger number of unique aa replacements at highly conserved sites in the nonTM region. These changes in P(Pm) might have been important in achieving marine lamprey-specific interaction between P(Pm) and ot

Thyroid hormone and retinoid X receptor function and expression during sea lamprey (Petromyzon marinus) metamorphosis.

Sea lampreys (Petromyzon marinus) are members of the ancient class Agnatha and undergo a metamorphosis that transforms blind, sedentary, filter-feeding larvae into free-swimming, parasitic juveniles. Thyroid hormones (THs) appear to be important for lamprey metamorphosis, however, serum TH concentrations are elevated in the larval phase, decline rapidly during early metamorphosis and remain low until metamorphosis is complete; these TH fluctuations are contrary to those of other metamorphosing vertebrates. Moreover, thyroid hormone synthesis inhibitors (goitrogens) induce precocious metamorpho

Fine structure of the liver in the larval lamprey, Petromyzon marinus L.; bile ducts and gall bladder.

The purpose of the present investigation was to describe ultrastructural characteristics of hepatic bile ducts and the gall bladder in larvae of the sea lamprey, Petromyzon marinus, using freeze--fracture replicas as well as ultrathin sections. Comparison of these structures with those of other vertebrates was necessary to provide a basis in future studies for characterization of biliary degeneration during metamorphosis. The bile ducts were composed of a simple cuboidal to columnar epithelium with the cells separated by wide lateral intercellular spaces and containing a prominent brush border

A new clarification method to visualize biliary degeneration during liver metamorphosis in Sea Lamprey (Petromyzon marinus).

Biliary atresia is a rare disease of infancy, with an estimated 1 in 15,000 frequency in the southeast United States, but more common in East Asian countries, with a reported frequency of 1 in 5,000 in Taiwan. Although much is known about the management of biliary atresia, its pathogenesis is still elusive. The sea lamprey (Petromyzon marinus) provides a unique opportunity to examine the mechanism and progression of biliary degeneration. Sea lamprey develop through three distinct life stages: larval, parasitic, and adult. During the transition from larvae to parasitic juvenile, sea lamprey und

Plasma estradiol and progesterone after hypophysectomy and substitution with pituitary in female sea lampreys (Petromyzon marinus).

This study investigated plasma steroid concentrations 1 week after total hypophysectomy in adult female sea lampreys. Plasma estradiol significantly decreased (to 1.19 +/- 0.17 SE ng/ml) following hypophysectomy and was elevated (to 1.91 +/- 0.11 ng/ml) to a value similar to controls by a single injection of two pituitary equivalents in adult female sea lampreys in the stage of final maturation. Plasma progesterone also significantly decreased (to 69 +/- 10 pg/ml) following hypophysectomy and was elevated (to 183 +/- 71 pg/ml) to a value not different from uninjected controls by a single injec

Analysis of DNA damage in sea lamprey (Petromyzon marinus) spermatozoa by UV, hydrogen peroxide, and the toxicant bisazir.

In this study we sought to demonstrate that Comet assay can be applied to sea lamprey sperm DNA fragmentation and used to describe the relationship between sperm DNA damage and sperm fertilizing ability. We show that the assay can be used reliably and accurately, and unlike in the case of mammals, there is no need for additional steps related to improvement of efficacy of lysis and DNA decondensation. This agrees with the presence of histone proteins in lamprey sperm. An increase in DNA fragmentation was noted during short-term storage of milt on ice (0-4 days). We demonstrated genotoxic effec

Neural projections of the pineal organ in the larval sea lamprey (Petromyzon marinus L.) revealed by indocarbocyanine dye tracing.

The distribution of the central neural connections of the pineal organ of the larval sea lamprey was investigated by means of anterograde and retrograde tracing with the fluorescent lipophilic dye, DiI (1,1'-dioctadecyl 3,3,3',3'-tetramethylindocarbocyanine perchlorate). Pinealofugal projections are well developed in larvae, extending from the posterior commissure into the diencephalon and mesencephalon. Small numbers of neurons were retrogradely labelled in the transition zone between the diencephalon and the mesencephalic tegmentum. These cells may constitute the first pinealopetal system de

Fine structure and immunocytochemistry of cells within the endocrine pancreas of larval and adult sea lampreys, Petromyzon marinus L.

Immunocytochemistry with protein A-gold and routine electron microscopy were used to identify cell types within the endocrine pancreas of larvae, juvenile adults, and upstream-migrant adults of the sea lamprey, Petromyzon marinus. The larval pancreatic islets are composed only of insulin-immunoreactive B-cells, which are uniform in their fine structure. The cranial and caudal pancreatic tissue in both adult periods contains three cell types: B-cells, somatostatin-immunoreactive D-cells, and a third cell type of unknown content.
