Unravelling Wood Extractive Resistance in Phanerochaete chrysosporium Through Random Mutagenesis.

During wood degradation, fungi have to deal with toxic and stressful compounds called wood extractives. The identification of the various detoxification strategies developed by fungi, and the molecular targets of these compounds is limited in Basidiomycetes because of the lack of genetic tools. To circumvent this problem, we have developed a direct genetic strategy in the white-rot fungus Phanerochaete chrysosporium. A library of P. chrysosporium UV mutants was generated and screened to isolate mutants resistant to Bagassa guianensis wood extractives (BWE). Wood extractives contribute to B. gu

Bioaccumulation and toxicity of CdSe/ZnS quantum dots in Phanerochaete chrysosporium.

The growing potential of quantum dots (QDs) in biomedical applications has raised considerable concerns regarding their toxicological impact. Consequently, there has been a need to understand the underlying toxicity mechanism of QDs. In this work, we comprehensively investigated the bioaccumulation and toxicity of three CdSe/ZnS QDs (COOH CdSe/ZnS 525, NH2 CdSe/ZnS 525, and NH2 CdSe/ZnS 625) in Phanerochaete chrysosporium (P. chrysosporium) using confocal laser scanning microscopy, reactive oxygen species (ROS) measurements, and cell viability assays. Isolation, characterization, and analysis of the expression of the cbhII gene of Phanerochaete chrysosporium.

Two cDNA sequences representing putative allelic variants of the Phanerochaete chrysosporium cbhII gene were isolated by hybridization to the Trichoderma reesei cbhII gene. Both of the equivalent genomic sequences were subsequently isolated by the inverse PCR technique. DNA sequencing showed that the cbhII open reading frame of 1,380 bp codes for a putative polypeptide of 460 amino acids which is interrupted by six introns. The domain structure found in T. reesei cbhII is conserved in the equivalent P. chrysosporium protein. The overall similarity between the two gene products is 54%, with the

Alteration of culture fluid proteins by cadmium induction in Phanerochaete chrysosporium.

Microorganisms need to resist the hazards posed by heavy metals during the process of metal adsorption. Phanerochaete chrysosporium is a fungus that is efficiently used for heavy-metal biosorption in wastewater treatment. Extraction and analysis of proteins induced by heavy metals can help to understand the regulatory mechanisms of P. chrysosporium in wastewater treatment. In this study, P. chrysosporium was exposed to 50 μM cadmium nitrate. A maximum cadmium adsorption capability of 77.1 mg/g dry biomass was found after 65 h, which was accompanied by a relatively higher protein concentration.

Comparative production of ligninolytic enzymes by Phanerochaete chrysosporium and Polyporus sanguineus.

The aim of the present study was to compare the effect of a wide range of culture conditions on production of ligninolytic enzymes by Polyporus sanguineus and Phanerochaete chrysosporium. Lignin peroxidase production by P. sanguineus was comparable with that of P. chrysosporium, although the culture conditions giving the highest yield varied greatly between the two fungi. Highest yield of manganese peroxidase by P. sanguineus obtained in 0.5% malt extract medium and peptone or malt extract supplemented mineral salts broth could not be surpassed by P. chrysosporium in any of the optimization ex

Exploring the Biocontrol Potential of Phanerochaete chrysosporium against Wheat Crown Rot.

The worldwide occurrence of wheat crown rot, predominantly caused by the pathogen Fusarium pseudograminearum, has a serious impact on wheat production. Numerous microorganisms have been employed as biocontrol agents, exhibiting effectiveness in addressing a wide array of plant pathogens through various pathways. The mycelium of the white-rot fungus Phanerochaete chrysosporium effectively inhibits the growth of F. pseudograminearum by tightly attaching to it and forming specialized penetrating structures. This process leads to the release of intracellular inclusions and the eventual disintegrat

Loss of Ku70 promotes mononucleate conidiation and homologous recombination in Phanerochaete chrysosporium.

Lignin is a major constituent of lignocellulose and the most abundant aromatic biopolymer on earth. It provides plants with rigidity and protection, but its recalcitrant nature also presents a significant barrier to lignocellulose valorization. The white-rot fungus Phanerochaete chrysosporium is among nature's most efficient lignin degraders, and its ligninolytic capabilities have been subjected to intensive investigations. Genome editing with precision is crucial for elucidating the in vivo mechanisms of its ligninolytic actions, but genetic manipulations of P. chrysosporium are often plagued

Phanerochaete sordida as a cause of pulmonary nodule in an immunocompromised patient: a case report.

BACKGROUND: Phanerochaete sordida is a species of wood rotting fungus, which can degrade lignin, cellulose and hemicellulose contained in wood and other hard-to-biodegrade organic substances. However, to date, there have been no other reports demonstrating that P. sordida can infect humans. CASE PRESENTATION: A 66-year-old Japanese man presented for a mass increasing in size on his left thigh. He had been suffering from rheumatoid arthritis for 18 years and chronic obstructive pulmonary disease for 20 years, for which he was being treated with 5 mg/day prednisolone and 8 mg/week methotrexate.

Lignin peroxidase of Phanerochaete chrysosporium. Evidence for an acidic ionization controlling activity.

The active site amino acid residues of lignin peroxidase are homologous to those of other peroxidases; however, in contrast to other peroxidases, no pH dependence is observed for the reaction of ferric lignin peroxidase with H2O2 to form compound I (Andrawis, A., Johnson, K.A., and Tien, M. (1988) J. Biol. Chem. 263, 1195-1198). Chloride binding is used in the present study to investigate this reaction further. Chloride binds to lignin peroxidase at the same site as cyanide and hydrogen peroxide. This is indicated by the following. 1) Chloride competes with cyanide in binding to lignin peroxid

Engineered CRISPR-Cas9 Vector for Efficient Agrobacterium-Mediated Transformation of Phanerochaete chrysosporium.

Phanerochaete chrysosporium is a model white-rot-fungus with exceptional lignolytic activity, but its genetic manipulation has remained a major challenge due to low transformation efficiencies. Here, we report the development of an optimized Agrobacterium-mediated transformation (AMT) protocol optimized for this species. A CRISPR-Cas9 compatible binary vector was modified by replacing the mCherry cassette with a strong sGFP reporter under the constitutive 35S promoter, enabling reliable identification of transformants. Using mycelial disks as starting material and applying a sandwich selection

Alleviating the toxicity of quantum dots to Phanerochaete chrysosporium by sodium hydrosulfide and cysteine.

Quantum dots (QDs) have caused large challenges in clinical tests and biomedical applications due to their potential toxicity from nanosize effects and heavy metal components. In this study, the physiological responses of Phanerochaete chrysosporium (P. chrysosporium) to CdSe/ZnS QDs with either an inorganic sulfide NaHS or an organic sulfide cysteine as antidote have been investigated. Scanning electron microscope analysis showed that the hyphal structure and morphology of P. chrysosporium have obviously changed after exposure to 100 nM of COOH CdSe/ZnS 505, NH2 CdSe/ZnS 505, NH2 CdSe/ZnS 565

Reduction of quinones and radicals by a plasma membrane redox system of Phanerochaete chrysosporium.

Quinones which are produced during the mineralization of lignin and xenobiotics by the white rot fungus Phanerochaete chrysosporium were reduced by a plasma membrane redox system of the fungus. Both intracellular enzymes and the plasma membrane redox system were able to reduce 1,4-benzoquinone. However, no quinone reductase activity was observed with the extracellular culture fluid. The intracellular reductase activity had a pH optimum between 6.0 and 7.0 and a Km of 150 microM. Reduction of 1,4-benzoquinone by the plasma membrane redox system had a pH optimum between 7.5 and 8.5 and exhibited

Flavin-containing monooxygenases from Phanerochaete chrysosporium responsible for fungal metabolism of phenolic compounds.

We investigated the cellular responses of the white-rot basidiomycete Phanerochaete chrysosporium against vanillin. Based upon a proteomic survey, it was demonstrated that two flavin-containing monooxygenases (PcFMO1 and PcFMO2) are translationally up-regulated in response to exogenous addition of vanillin. To elucidate their catalytic functions, we cloned cDNAs and heterologously expressed them in Escherichia coli. The recombinant PcFMO1 showed catalytic activities against monocyclic phenols such as phenol, hydroquinone, and 4-chlorophenol. In addition, the product from hydroquinone was ident

H2O2-dependent decolorization of Poly R-481 by particulate fractions from Phanerochaete chrysosporium.

A cell-free preparation from Phanerochaete chrysosporium culture medium decolorized the polymeric dye Poly R-481. The majority of this decolorization activity sedimented when centrifuged at 150,000 X g, indicating that it was associated with a particulate body. The activity was sensitive to heat, azide and cyanide, was stimulated by exogenously added H2O2, and was optimal around pH 4. Electron micrographs of the sedimented culture medium fraction showed the presence of numerous particulate structures. A similar dye decolorization activity from sonicated mycelium also sedimented at 150,000 X g.

Facile green extracellular biosynthesis of CdS quantum dots by white rot fungus Phanerochaete chrysosporium.

This study details a novel method for the extracellular microbial synthesis of cadmium sulfide (CdS) quantum dots (QDs) by the white rot fungus Phanerochaete chrysosporium. P. chrysosporium was incubated in a solution containing cadmium nitrate tetrahydrate, which became yellow from 12h onwards, indicating the formation of CdS nanocrystals. The purified solution showed a maximum absorbance peak between 296 and 298 nm due to CdS particles in the quantum size regime. The fluorescence emission at 458 nm showed the blue fluorescence of the nanoparticles. X-ray analysis of the nanoparticles confirm

1H NMR investigation of manganese peroxidase from Phanerochaete chrysosporium. A comparison with other peroxidases.

1H NMR spectra at 200- and 600-MHz of manganese peroxidase from Phanerochaete chrysosporium and of its cyanide derivative are reported. The spectrum of the native protein is very similar to that of other peroxidases. The assignment of the spectrum of the cyanide derivative has been performed through 1D NOE, 2D NOESY, and COSY experiments. This protein is very similar to lignin peroxidase, the only meaningful difference being the shift of H delta 2 of the proximal histidine. The spectra of the cyanide derivative of these two proteins are compared with those of horseradish peroxidase and cytochr

Biodegradation of 2,2',4,4'-tetrabromodiphenyl ether (BDE-47) by Phanerochaete chrysosporium in the presence of Cd2.

Aerobic biodegradation of 2,2',4,4'-tetrabrominated diphenyl ether (BDE-47) by Phanerochaete chrysosporium in the presence of Cd2+ was investigated in this study. The results showed that P. chrysosporium could effectively degrade BDE-47, and its extracellular enzyme played an important role in the process of decomposition. BDE-47 biodegradation by fungi was more tolerant than extracellular enzyme in the presence of Cd2+.
