Phellinus linteus polysaccharide extract improves insulin resistance by regulating gut microbiota composition.

The hypoglycemic effect of Phellinus linteus polysaccharide extract (PLPE) has been documented in several previous studies, but the functional interactions among PLPE, gut microbiota, and the hypoglycemic effect remain unclear. We examined the regulatory effect of PLPE on gut microbiota, and the molecular mechanism underlying improvement of insulin resistance, using a type 2 diabetic rat model. Here, 24 male Sprague-Dawley rats were randomly divided into four groups that were subjected to intervention of saline (normal and model control group), metformin (120 mg/kg.bw), and PLPE (600 mg/kg.bw)

Phellinus linteus and igniarius (L.) Quel. have been used in traditional Asian medicine for over two centuries against a variety of diseases. Polysaccharides from their fruiting bodies show strong immunomodulatory activity. In this study we characterized the structure and composition of polysaccharides from Phellinus linteus and Phellinus igniarius by HPLC, GC-MS and NMR (1-H, 13-C, COSY, NOESY and TOCSY). The polysaccharides from P. linteus and P. igniarius mainly contained glucose with minor proportions of mannose, galactose, xylose, arabinose and rhamnose. Methylation analyses showed that t

Anti-breast cancer effect of Phellinus pini and its chemical composition characterization.

Phellinus pini (P. pini) has been widely utilized as a traditional medicinal macrofungus in China and various East Asian countries. Modern pharmacological studies have shown that P. pini has important application value in anti-tumor therapy. However, its main anti-breast cancer active substances and mechanism are still unclear. In this study, we demonstrated that the methanol extract of Phellinus pini (PPE) could significantly inhibit the viability of 4 T1 cells, induce apoptosis, decrease mitochondrial membrane potential and S phase arrest of 4 T1 cells. PPE can restore the imbalance of intes

Phellinus linteus polysaccharides mediates acetaminophen-induced hepatotoxicity via activating AMPK/Nrf2 signaling pathways.

Overdose of acetaminophen (APAP) is currently one of the main causes of hepatoxicity and acute liver injury, which is often linked to oxidative stress. Phellinus linteus polysaccharides (Phps) have shown many hepatoprotective effects, however, the mechanism of Phps on APAP-induced acute liver injury has not been further elucidated. The aim of this study is to investigate the underlying mechanism of Phps to acute liver injury. The expression of AMPK/Nrf2 and autophagy were detected using western blot. The results indicated that Phps treatment effectively alleviated APAP-induced acute liver inju

Inhibitory Effect of Styrylpyrone Extract of Phellinus linteus on Hepatic Steatosis in HepG2 Cells.

The prevalence of nonalcoholic fatty liver disease (NAFLD) is estimated to be approximately about 25.24% of the population worldwide. NAFLD is a complex syndrome and is characterized by a simple benign hepatocyte steatosis to more severe steatohepatitis in the liver pathology. Phellinus linteus (PL) is traditionally used as a hepatoprotective supplement. Styrylpyrone-enriched extract (SPEE) obtained from the PL mycelia has been shown to have potential inhibition effects on high-fat- and high-fructose-diet-induced NAFLD. In the continuous study, we aimed to explore the inhibitory effects of SPE

Effects of mycelial culture of Phellinus linteus on ethanol-induced gastric ulcer in rats.

The gastroprotective effects of a mycelial culture of Phellinus linteus (MCPL) were evaluated by determining the ulcer index, gastric mucus content, histopathological observation and histochemical properties of mucin in an ethanol-induced ulcer model of rats. Preadministration with MCPL at doses of 20 and 60 mg/kg, showed a significant decrease of bleeding and ulcer index and alleviated the histopathological changes induced by ethanol such as hemorrhage and necrosis. Ethanol treatment decreased the gastric adhesion mucus content, but a higher level of gastric mucus persisted after preadministr

Phellinus pini Polysaccharides Regulate Gut Microbiota-Host Metabolism to Attenuate Influenza-Driven Lung Injury.

Phellinus pini is a large fungus with significant medicinal value. Building on the previously discovered inhibitory activity of Phellinus pini polysaccharides (PPP) against influenza viruses, this study established an influenza A mouse model through the intranasal instillation of an H1N1/PR8 virus solution. Using a combination of metabolomics and gut microbiota analysis techniques, the study further elucidated a novel mechanism by which PPP regulates influenza-induced lung damage via the "gut-lung axis". Results showed PPP reversed lung lesions, reduced the lung index and viral load in lung ti

Phellinus igniarius-derived carbon dots suppress liver cancer through the ROS/MAPK signaling axis.

Liver cancer is one of the most common and highly lethal malignancies worldwide, and current therapeutic strategies remain limited, posing significant clinical challenges. Thermal processing of natural products is not only used for the extraction of active constituents, but may also induce structural reconstruction and bioactivity transformation of endogenous components. Recent studies have demonstrated that certain natural products can generate carbon dots (CDs) with specific biological activities under high-temperature conditions, suggesting that these nanoparticles may serve as novel active
