Guanidine-reactive agent phenylglyoxal induces DNA damage and cancer cell death.

BACKGROUND: DNA-damaging compounds (e.g., alkylating agents, cytotoxic antibiotics and DNA topoisomerase poisons) are the most widely used anticancer drugs. The inability of tumor cells to properly repair some types of DNA damage may explain why specific DNA-damaging drugs can selectively kill tumor cells. Phenylglyoxal is a dicarbonyl compound known to react with guanidine groups such as that of the DNA base guanine, therefore suggesting that phenylglyoxal could induce DNA damage and have anticancer activity. METHODS: Cellular DNA damage was measured by the alkaline comet assay and the γH2AX

Irreversible inactivation of red cell chloride exchange with phenylglyoxal, and arginine-specific reagent.

Chloride exchange in resealed human erythrocyte ghosts can be irreversibly inhibited with phenylglyoxal, a reagent specific for the modification of arginyl residues in proteins. Phenylglyoxal inhibits anion transport in two distinct ways. At 0 degrees C, inhibition is instantaneous and fully reversible, whereas at higher temperature in an alkaline extracellular medium, covalent binding of phenylglyoxal leads to an irreversible inhibition of the transport membranes system. Indiscriminate modification of membrane arginyl residues was prevented by reacting the with phenylglyoxal in an alkaline ex

Inhibition by trimethyl-tin, probenecid and phenylglyoxal of depolarization-induced acetylcholine release in Torpedo synaptosomes.

The effects of trimethyl-tin (anion-hydroxyde ionophore, inhibiting oxydative phosphorylation and H(+)-ATPase) probenecid (inhibitor of anion transport in neural cells) and phenylglyoxal (arginine-specific reagent, inhibiting chloride exchanges in erythrocytes) were examined in Torpedo synaptosomes prepared from electric organ. All drugs significantly reduced the stimulated release of acetylcholine triggered by depolarization of nerve endings with high-K(+) and/or gramicidin D. In contrast, trimethyl-tin, probenecid and phenylglyoxal did not affect the ionophore A23187-induced release of acety

A comparison of the effects of cyanide, hydrogen peroxide, and phenylglyoxal on eucaryotic and procaryotic Cu,Zn superoxide dismutases.

The Cu,Zn superoxide dismutases from bovine liver, yeast, Caulobacter crescentus, and Photobacter leiognathi were compared for their susceptibilities to inhibition by cyanide and to inactivation by hydrogen peroxide and phenylglyoxal. All of these enzymes were affected by these reagents, albeit with some differences in sensitivity. The yeast and the bacterial enzymes were thus more sensitive to cyanide than was the bovine enzyme, while the bovine and the yeast enzymes were inactivated more rapidly by hydrogen peroxide and less rapidly by phenylglyoxal than were their bacterial counterparts. Inactivation of liver 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase by phenylglyoxal. Evidence for essential arginine residues.

Treatment of liver 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase with the arginine-specific reagent, phenylglyoxal, irreversibly inactivated both 6-phosphofructo-2-kinase and fructose-6-bisphosphatase in a time-dependent and dose-dependent manner. Fructose 6-phosphate protected against 2,6-phosphofructo-2-kinase inactivation, whereas MgGTP protected against fructose-2,6-bisphosphatase inactivation. Semi-logarithmic plots of the time course of inactivation by different phenylglyoxal concentrations were non-linear, suggesting that more than one arginine residue was modified. The stoichiom

Essentiality of the active-site arginine residue for the normal catalytic activity of Cu,Zn superoxide dismutase.

Chemical modification of bovine and yeast Cu,Zn superoxide dismutases with phenylglyoxal diminishes the catalytic activities by greater than or equal to 98%, and treatment of these enzymes with butanedione plus borate leads to greater than or equal to 96% inactivation. The activity loss is accompanied by the modification of less than two arginine residues per subunit with no concomitant loss of Cu or Zn. The phenylglyoxal-modified enzymes require at least a 20-fold greater concentration of cyanide for 50% inhibition than do the corresponding native enzymes. Polyacrylamide-gel electrophoresis a

Identification and covalent modification of a renal brush-border anion exchanger.

Brush-border membrane (BBM) proteins that bind the arginine-specific reagent phenylglyoxal (PG) and interact with stilbene disulfonic derivatives were identified in canine kidney cortex. Pretreatment of BBM vesicles with PG resulted in irreversible inhibition of anion exchange as assayed by 36Cl- influx mediated via Cl-/Cl- exchange. Cl-/Cl- exchange was reversibly inhibited by the disulfonic stilbene 4,4'-dinitrostilbene-2,2'-disulfonate (DNDS). A stilbene-affinity matrix was prepared by immobilizing DNDS in polyacrylamide beads. Elution of the BBM proteins from a disulfonic stilbene (DNDS) a

Copper catalyzed photoredox synthesis of α-keto esters, quinoxaline, and naphthoquinone: controlled oxidation of terminal alkynes to glyoxals.

Herein, we report a facile visible light induced copper catalyzed controlled oxidation of terminal C[triple bond, length as m-dash]C alkynes to α-keto esters and quinoxalines via formation of phenylglyoxals as stable intermediates, under mild conditions by using molecular O2 as a sustainable oxidant. The current copper catalysed photoredox method is simple, highly functional group compatible with a broad range of electron rich and electron poor aromatic alkynes as well as aliphatic alcohols (1°, 2° and 3° alcohols), providing an efficient route for the preparation of α-keto esters (43 examples

Volatile Organic Compounds from Wolfiporia hoelen Exhibit Nematicidal Activity Against Bursaphelenchus xylophilus.

Pine wilt disease, caused by the pinewood nematode (PWN) Bursaphelenchus xylophilus, devastates pine forests worldwide. Although the medicinal fungus Wolfiporia hoelen exhibits nematicidal activity against PWN, its specific bioactive constituents remain unidentified. This study investigated the nematicidal activity of volatile organic compounds (VOCs) released from W. hoelen against PWN. Results indicated that VOCs from W. hoelen exhibit significant inhibitory efficacy against second-stage juveniles (J2s) of PWN, achieving 90% mortality after 72 h of exposure. In contrast, ovicidal activity wa

Chiral Benzoins via Asymmetric Transfer Hemihydrogenation of Benzils: The Detail that Matters.

The synthesis of enantiomerically pure benzoins by hydrogenation of readily available benzils has been long thwarted by their base-sensitivity. We show here that an iron(II) hydride complex catalyzes the asymmetric transfer hydrogenation of benzils from 2-propanol. When strictly base-free conditions are granted, excellent enantioselectivity is achieved even with o-substituted substrates, which are particularly challenging to prepare with other methods. Hence, under optimized reaction conditions, chiral benzoins were prepared in good yields (up to 83%) and excellent enantioselectivity (up to 98

The benzil rearrangement reaction: trapping of a hitherto minor product and its application to the development of a selective cyanide anion indicator.

The isolation and characterization of an intermediate from the benzil-cyanide reaction is reported. The use of this trapping chemistry to produce a chemical indicator for the cyanide anion is described. It relies on the synthesis and reaction of a pi-extended analogue of benzil. Addition of tetrabutylammonium cyanide to organic solutions of this species, referred to as compound 3 in the text, gives rise to a dramatic change in both color and fluorescence properties.

Development of a Chemoproteomic Platform to Identify Sites of (Homo)citrullination within Complex Proteomes.

Citrullination and homocitrullination of arginine and lysine can significantly impact protein structure and function. Citrullination of arginine is an enzymatic modification catalyzed by Protein Arginine Deiminases (PADs). Homocitrullination of lysine is a nonenzymatic modification that occurs in the presence of high concentrations of cyanate. Both post-translational modifications are elevated in Rheumatoid Arthritis (RA) and other inflammatory diseases. Moreover, autoantibodies targeting these PTMs are associated with the development of RA. Identifying arginine and lysine residues that are hy

A protein with anion exchange properties found in the kidney proximal tubule.

One important mechanism for reabsorption of chloride in the kidney proximal tubule involves anion exchange of chloride for a base. Anion exchange transport systems in general demonstrate sensitivity to inhibition by disulfonic stilbenes, probenecid, furosemide, and the arginyl amino group modifier phenylglyoxal. Using disulfonic stilbene affinity chromatography, we have identified and partially purified a protein with anion exchanger properties in luminal membrane vesicles isolated from rabbit kidney cortex. This protein has a molecular weight of 162 kD. The binding of the 162 kD protein to th

Structure-activity relationships of scorpion alpha-neurotoxins: contribution of arginine residues.

The role of arginine residues in the structure-activity relationships of alpha-scorpion neurotoxins was studied. Toxins I and II from Androctonus australis Hector (north African scorpion), containing respectively 2 and 3 arginines, were modified by phenylglyoxal or p-hydroxyphenylglyoxal. Modified derivatives were purified by reverse-phase HPLC and/or ion exchange HPLC. Subsequent bioassays showed that toxin I (AaH I) derivatives with single modifications on Arg 2 and Arg 60 had low activity (25 and 14% of residual activity, assessed in receptor binding experiments). Doubly modified (Arg 2, Ar

Structure-Guided Design of a Bioactive Covalent Small Molecule Targeting a Riboswitch.

Small molecule ligands targeting structured RNA elements hold promise for modulating RNA function, serving as chemical probes and potential therapeutics. In this study, the characterization of phenylglyoxal-based covalent probe designed to target unpaired guanine residues in structured RNAs is reported. A structure-guided design strategy was employed to modify covalently unpaired guanines critical for flavin mononucleotide (FMN) binding to the FMN riboswitch. Covalent modification occurs at the designed site and modulates riboswitch function in a cellular reporter system, highlighting the pote

Catalytic Regioselective γ-Methylenation of α,β-Unsaturated Aldehydes Using Formaldehyde via Vinylogous Aldol Condensation.

The first vinylogous aldol condensation of α,β-unsaturated aldehydes using aqueous formaldehyde is developed under mild reaction conditions to form the γ-methylenated products with excellent regioselectivity. Using this methodology, a short synthesis of α-triticene, an antifungal compound, is achieved in two steps. The practicality of this methodology is demonstrated by the gram-scale synthesis. Formation of the unusual double γ-functionalized products from crotonaldehyde and a direct asymmetric vinylogous aldol product from phenylglyoxal is also described.

Mechanism of the cyanide-catalyzed oxidation of alpha-ketoaldehydes and alpha-ketoalcohols.

Cyanide catalyzes the reduction of dioxygen or of ferricytochrome c by dihydroxyacetone phosphate. The rapid initial phase of these reactions, but not the subsequent slow phase, was augmented by incubating the triose phosphate aerobically or anaerobically at pH 9.0 prior to adding the cyanide. The aerobic incubation, which was most effective, was associated with a decline in enediol, whereas the less effective anaerobic incubation was accompanied by an increase in enediol content. This suggested that the alpha-ketoaldehyde product of autoxidation of the enediol, rather than the enediol itself,

Single-nuclei and bulk-tissue gene-expression analysis of pheochromocytoma and paraganglioma links disease subtypes with tumor microenvironment.

Pheochromocytomas (PC) and paragangliomas (PG) are rare neuroendocrine tumors associated with autonomic nerves. Here we use single-nuclei RNA-seq and bulk-tissue gene-expression data to characterize the cellular composition of PCPG and normal adrenal tissues, refine tumor gene-expression subtypes and make clinical and genotypic associations. We confirm seven PCPG gene-expression subtypes with significant genotype and clinical associations. Tumors with mutations in VHL, SDH-encoding genes (SDHx) or MAML3-fusions are characterized by hypoxia-inducible factor signaling and neoangiogenesis. PCPG h

Iodine-promoted sequential C(sp3)-H oxidation and cyclization of aryl methyl ketones with 2-(2-aminophenyl)quinazolin-4(3H)-ones.

An I2-promoted, metal-free protocol has been developed for the one-pot synthesis of 6-aroyl-5,6-dihydro-8H-quinazolino[4,3-b]quinazolin-8-ones from readily accessible substrates. This reaction involves the in situ sp3 C-H oxidation of aryl methyl ketones to phenylglyoxal, followed by imine formation and intramolecular nucleophilic addition, resulting in the formation of two new C-N bonds. Furthermore, the method is applicable to a wide range of aryl methyl ketones, including heterocycles and drug-derived substrates, yielding the desired products with yields ranging from 62% to 93%.
