Go with the Phloem: A Review of the Methods to Collect Plant Phloem Tissue.

The study of plant phloem tissue is important for understanding plant physiology, yet the anatomy and small size of the phloem tissue make direct sampling challenging. Due to the absence of a single method that allows for phloem extraction in every species, researchers have developed several strategies to address this drawback. These strategies can be categorized into three groups: classical, advanced, and mechanical extraction methods. Classical methods, including cucurbit drip, EDTA-facilitated exudation, stylectomy, and heterografting, have a long history in phloem research but are generall

Measuring Phloem Transport Velocity on a Tissue-Level Using a Phloem-Mobile Dye.

Here, we describe an in vivo dye-tracking method for measuring phloem transport velocity in seedlings, leaves, and petioles and potentially other translucent plant tissues. The method requires measurement of the fluorescent signal of a phloem-mobile dye using sensitive photo-sensors placed externally to the plant. Following dye application, velocity is determined by either following a dye pulse or using laser fluorescence bleaching. Velocity is estimated by dividing the distance traveled by the dye by the time it takes to travel. This method can be used to measure phloem transport velocity on

Quantifying Fire-Induced Phloem Damage.

Despite the phloem's crucial role in carbohydrate translocation and its high vulnerability due to its position just beneath the bark, studies directly addressing fire effects on phloem remain very limited. Thus, this chapter aims to provide a concise overview of the methodologies that have been used directly or indirectly to investigate the impacts of fire on the phloem, including  changes in phloem compounds, transport velocity, and phloem cell damage.

Transmission Electron Microscopy of the Phloem.

A universal feature of seed plants is that their phloem consists of a continuous sieve-tube system extending throughout the plant, which is highly pressurized due to its sugar content. This cellular continuity, combined with the pressure-driven, osmotically generated flow in the source leaves, enables assimilates to reach all sink organs. However, these two key features of the phloem, the cellular continuity and high pressure, pose significant challenges when attempting to fix the phloem for transmission electron microscopy. In most cases, the tissue preparation required for fixation triggers

Manipulating phloem iron transport using the phloem-specific promoter OsSUT1 enhances grain iron accumulation in rice.

Human diets are commonly deficient in iron. Hence, increasing the iron content in rice, a major staple crop globally, will help overcome this iron deficiency. The phloem is an important route for iron distribution. However, little is known about the effects of manipulating iron transport in the phloem on iron content. The phloem-specific OsSUT1 promoter is used to drive the expression of iron transport genes OsYSL9 and OsIRT1. The phloem exudate iron content, iron harvest index and remobilization contribution rate are enhanced in pOsSUT1::OsYSL9 (SY) and pOsSUT1::OsIRT1 (SI) transgenic lines.

Collection and Analysis of Phloem Lipids.

The plant phloem is a long-distance conduit for the transport of assimilates but also of mobile developmental and stress signals. These signals can be sugars, metabolites, amino acids, peptides, proteins, microRNA, or mRNA. Yet small lipophilic molecules such as oxylipins and, more recently, phospholipids have emerged as possible long-distance signals as well. Analysis of phloem (phospho)lipids, however, requires enrichment, purification, and sensitive analysis. This chapter describes the EDTA-facilitated approach of phloem exudate collection, phase partitioning against chloroform-methanol for

Using Aphids to Sample Phloem Sap.

Phloem transport is crucial for plant growth and development because it mediates the transport of metabolites, mineral nutrients, proteins, and secondary compounds between plant tissues. The type of phloem loading with such compounds depends on their concentration in the phloem and the surrounding cells. To investigate these processes, it is very important to collect phloem sap from intact plants and determine the concentrations of the various compounds. This chapter describes aphid stylectomy, in which severed aphid stylets are used to collect phloem sap. Compared to other methods for collect

Predicting Phloem Transport Rate in Crops.

Accessing phloem sap movement properties, such as transport rate and front velocity, is difficult and involves technologies that are not routinely implementable at high throughput. This can be problematic if quantitative information on phloem transport is to be used for comparing varieties for breeding. We present here how to calculate an average phloem transport rate from agronomic data, using two crops, wheat and oil palm, as examples. When physiological variables are also known for the same agronomic trial, phloem transport rate can be related to other processes such as water movement in xy

Mechanisms of phloem unloading and post-phloem transport of photoassimilates during the rapid elongation of young Dendrocalamus brandisii shoots.

Phloem unloading plays a key role in the transport and allocation of photoassimilates. Elucidating the unloading process and metabolic dynamics of photoassimilates in rapidly elongating internodes is crucial for understanding the rapid growth mechanism of bamboo. We systematically investigated the phloem unloading and the post-unloading transport mechanism in internodes of young Dendrocalamus brandisii culms through integrated anatomical observations, transcriptomic analysis, and gene functional verification. Phloem unloading shifts from coexisting apoplasmic and symplasmic pathways in immatur

Predicting Phloem Loading Types with Electron Microscopy.

Phloem loading is a critical step in the transport of photosynthetically produced sugars from leaves to sink tissues. The type of phloem loading-active apoplasmic, active symplasmic, or passive symplasmic-can be inferred from the density and structure of plasmodesmata (PD) along the phloem-loading pathway. This protocol describes a method to predict phloem loading types using transmission electron microscopy (TEM) to quantify PD density. The method follows established standards for sample preparation, imaging, and data analysis, enabling comparative studies across species.

Neutron Imaging to Reveal Phloem Function in Trees.

Understanding phloem function is key to understanding the regulation of plant function. The phloem distributes the main building blocks that support plant function and growth from the production sites to sites of consumption and storage. It also distributes important signaling molecules and interacts with the adjacent water-transport tissue, the xylem, to support hydraulic integrity of plants. Yet, studying phloem function is challenging because of the sensitivity of this tissue to mechanical stimulation that can lead to blockage of flow. Furthermore, its small size, adjacent location, and int

Measurement of Phloem-Based Electrical Signals in Plants.

Electrical signals are vital components of long-distance signaling in plants, enabling rapid systemic responses to environmental stimuli. These signals, which include action potentials (APs), variation potentials (VPs), and system potentials (SPs), that can also combine into electropotential waves (EPWs), are primarily propagated through the vascular system, with the phloem playing a central role. EPWs, encompassing APs, VPs, and SPs, are integral to systemic signaling and inter-organ communication via the phloem, and are closely associated with plant responses to pests and pathogens. The comp

Cadmium transport characteristics in the phloem of sweet potato.

Cadmium (Cd) pollution in farmland soil poses serious threats to human health. The Cd transport and accumulation in the aboveground tissues of sweet potato (Ipomoea batatas L.) variety "Fu Cai 18″ and the characteristics of phloem-mediated Cd transport were elucidated in this study. Sweet potato seedlings grown in hydroponics were analyzed. The scanning through μ-XRF revealed that Cd was mainly distributed in cortex cells and vascular tissues of the stem; in leaves, it was mainly distributed in veins. The root regeneration and leaf smearing tests revealed that Cd accumulated in the stem can be

Visual Detection of Inorganic Orthophosphate in Phloem Cells.

The phloem plays a vital role in actively transporting soluble organic compounds-such as sugars, minerals, and proteins-from source to sink tissues, a process driven by turgor pressure and adenosine triphosphate (ATP)-dependent energy. Since ATP levels in phloem cells are closely linked to inorganic orthophosphate (Pi) availability, precise and efficient detection of Pi in these cells is essential for understanding phloem function. To address this, we developed the inorganic orthophosphate staining assay (IOSA), a method for visually detecting Pi in phloem cells. As a demonstration, we have su

Immunohistochemical Localization of Proteins in the Phloem: Problems and Solutions.

The technique of immunolocalization in differentiated phloem cells is a challenging task given their specific functions, anatomy, and cell organellar infrastructure. Incorporation of specific cell wall components in thickened cell walls of phloem cells is often the source of unspecific labeling and erroneous localization data. Furthermore, tissue preservation of cells that are exposed to high osmotic pressure is a very challenging task, encountering many pitfalls. Therefore, special care is required regarding the generation and purification of specific antibodies and in the protocols used for

Phloem mobility of xenobiotics VIII. A short review.

Great strides have been made in the last 15 years in our understanding of phloem mobility of xenobiotics. The subject has been transformed from a poorly understood phenomenon to a process that can be accurately described by the physicochemical properties of the xenobiotic and the nature of the vascular system through which it moves. The basic tenet of the unified mathematical model is that the combination of the permeability and the acid dissociation constant (pK(a)) determines phloem mobility, and this has been largely validated for many compounds in many plant systems. More precise testing o

Effect of petiole anoxia on Phloem transport in squash.

Translocation of (14)C-labeled assimilates in Early Prolific Straightneck squash (Cucurbita melopepo torticollis Bailey) through a 15-centimeter oxygen-deficient zone of the petiole was studied as a function of varying periods of anaerobiosis (N(2) atmosphere). Initiation of anaerobic conditions caused an immediate and rapid decline in translocation to about 35 to 45% of the pretreatment rate within 30 to 40 minutes. This inhibition response (first inhibition response) was transient, however, and full recovery to the pretreatment rate occurred during the ensuing 60 to 90 minutes. Following thi

Phloem Catastrophe: Identifying Plant Tipping Points Through Bifurcation Analysis.

This chapter presents a mathematical framework to identify conditions leading to phloem failure using bifurcation analysis and catastrophe theory. The approach assumes that at equilibrium, sucrose production through leaf photosynthesis is balanced by sucrose transport within the phloem. Leaf photosynthesis is modeled according to stomatal optimality theories, which propose that stomatal opening adjusts to maximize carbon uptake while being constrained by resource availability (usually water per unit leaf area in the rooting zone). Sucrose transport in the phloem vasculature is described by the

Phloem small RNAs, nutrient stress responses, and systemic mobility.

BACKGROUND: Nutrient availabilities and needs have to be tightly coordinated between organs to ensure a balance between uptake and consumption for metabolism, growth, and defense reactions. Since plants often have to grow in environments with sub-optimal nutrient availability, a fine tuning is vital. To achieve this, information has to flow cell-to-cell and over long-distance via xylem and phloem. Recently, specific miRNAs emerged as a new type of regulating molecules during stress and nutrient deficiency responses, and miR399 was suggested to be a phloem-mobile long-distance signal involved i
