Fasting increases 18:2-containing phosphatidylcholines to complement the decrease in 22:6-containing phosphatidylcholines in mouse skeletal muscle.

Fasting stimulates catabolic reactions in skeletal muscle to survive nutrient deprivation. Cellular phospholipids have large structural diversity due to various polar-heads and acyl-chains that affect many cellular functions. Skeletal muscle phospholipid profiles have been suggested to be associated with muscle adaptations to nutritional and environmental status. However, the effect of fasting on skeletal muscle phospholipid profiles remains unknown. Here, we analyzed phospholipids using liquid chromatography mass spectrometry. We determined that fasting resulted in a decrease in 22:6-containi

Expansion of phosphatidylcholine and phosphatidylserine/phosphatidylcholine monolayers by differently charged amphiphiles.

The degree and time-course of expansion of palmitoyloleoylphosphatidylcholine (PC) and bovine brain phosphatidylserine (PS)/PC (75:25, mol/mol) monolayers at 32 mN/m caused by differently charged amphiphiles (detergents) added to the sub-phase buffer (pH 7.4, 22 degrees C) were followed. Amphiphiles were added to the sub-phase at a concentration/monolayer area corresponding to the concentration/erythrocytes surface area where sphero-echinocytic or sphero-stomatocytic shapes are induced (0.46-14.6 microM). Nonionic, cationic and anionic amphiphiles expanded the PS/PC monolayer significantly mor

Oxidized Phosphatidylcholines Regulate Secretory Phospholipase A2 Through Membrane Nanodomain Remodeling.

Oxidative stress generates oxidized phospholipids (OxPLs) that alter membrane structure and inflammatory lipid signaling, yet the underlying biophysical mechanisms remain poorly understood. Here, we examine how two structurally distinct truncated oxidized phosphatidylcholines (OxPCs), 1-palmitoyl-2-(5'-oxo-valeroyl)-sn-glycero-3-phosphocholine (POVPC) and 1-palmitoyl-2-glutaryl-sn-glycero-3-phosphocholine (PGPC), remodel membrane lateral organization and regulate secretory phospholipase A2 (sPLA2) activity. Oxidized phosphatidylcholines trigger ferroptosis in cardiomyocytes during ischemia-reperfusion injury.

Myocardial ischemia-reperfusion (I/R) injury increases the generation of oxidized phosphatidylcholines (OxPCs), which results in cell death. However, the mechanism by which OxPCs mediate cell death and cardiac dysfunction is largely unknown. The aim of this study was to determine the mechanisms by which OxPC triggers cardiomyocyte cell death during reperfusion injury. Adult rat ventricular cardiomyocytes were treated with increasing concentrations of various purified fragmented OxPCs. Cardiomyocyte viability, bioenergetic response, and calcium transients were determined in the presence of OxPC

Mass spectrometric analyses of phosphatidylcholines in alkali-exposed corneal tissue.

PURPOSE: The aims were to determine whether exposure to sodium hydroxide results in predictable changes in phosphatidylcholine (PC) in corneal tissue and if PC profile changes correlate to exposure duration. PCs are major components of the cell membrane lipid bilayer and are often involved in biological processes such as signaling. METHODS: Enucleated porcine (n = 140) and cadaver human eyes (n = 20) were exposed to water (control) and 11 M NaOH. The corneas were excised and lipids were extracted using the Bligh and Dyer method with suitable modifications. Class-specific lipid identification

Barbiturate interaction with phosphatidylcholine.

A hydrogen-bonding interaction between phenobarbital or pentobarbital with phosphatidylcholine in chloroform is indicated by the effects of added phosphatidylcholine on the infrared and proton magnetic resonance spectra of these barbiturates. The nitrogenbound proton of the barbiturate and the orthophosphate moiety of the phosphatidylcholine molecule appear to be involved. The more pronounced effect with the two barbiturates occurs in the proton magnetic resonance spectra of phenobarbital with increased amounts of phosphatidylcholine. A plot of the chemical shift of phenobarbital N-H against t

Biosynthesis of phosphatidylcholine in bacteria.

Phosphatidylcholine (PC) is the major membrane-forming phospholipid in eukaryotes and can be synthesized by either of two pathways, the methylation pathway or the CDP-choline pathway. Many prokaryotes lack PC, but it can be found in significant amounts in membranes of rather diverse bacteria and based on genomic data, we estimate that more than 10% of all bacteria possess PC. Enzymatic methylation of phosphatidylethanolamine via the methylation pathway was thought to be the only biosynthetic pathway to yield PC in bacteria. However, a choline-dependent pathway for PC biosynthesis has been disc

Interaction of trichloroethylene with phosphatidylcholine and its localization in the phosphatidylcholine vesicles: a 1H-NMR study.

The interaction of trichloroethylene, an inhalation general anesthetic, with egg yolk phosphatidylcholine (EYPC) and its localization in small unilamellar vesicles (SUV) has been studied by Nuclear Magnetic Resonance. These studies demonstrate that inhalation anesthetic can interact nonspecifically with lipid molecules. Furthermore, it suggests that the action of inhalation anesthetics is directed primarily to the interfacial region of the lipid membrane.

Increased storage and secretion of phosphatidylcholines by senescent human peritoneal mesothelial cells.

BACKGROUND/AIMS: Human peritoneal mesothelial cells (HPMC) secrete phosphatidylcholines (PC) which form a lipid bilayer lining the peritoneum. They prevent frictions and adhesions and act as a barrier to the transport of water-soluble solutes while permitting water flux. PC may play an essential role in peritoneal integrity and function, the role of PD induced HPMC senescence on PC homeostasis, however, is unknown. METHODS: HPMC cell lines were isolated from four non-uremic patients. Expression of the three PC synthesis genes (rt-PCR), and cellular storage and secretion of PC (ESI-mass-spectr

Lubricating recovery of damaged pleural mesothelium: effect of time and of phosphatidylcholines.

Effect of time and phosphatidylcholines (PCs) on lubrication of damaged mesothelium has been investigated. Marked increase in coefficient of kinetic friction (μ) of pleural specimens after mesothelial blotting and rewetting decreased by 23.4±3.5%, 41.8±3.8%, and 40.5±2.7% after 30min, 1h, and 2h. Hence, damaged mesothelium is able to partially reset lubricating molecules on its surface. Increase in μ of post-blotting Ringer 2h after addition of unsaturated PCs (3mg/ml) decreased a little more than after 2h Ringer. Effects of unsaturated and saturated PCs were similar, contrary to expectation r

Heterogeneous catalytic transesterification of phosphatidylcholine.

The transesterification of phosphatidylcholine (PC) via homogeneous and heterogeneous catalysis was investigated for the production of fatty acid methyl esters (FAME) i.e. biodiesel. Calcium methoxide and calcium oxide were used as heterogeneous catalysts, and KOH as a homogeneous catalyst for the transesterification of phosphatidylcholine (PC)--a polar phospholipid prevalent in eukaryotic organisms. The initial reaction rate was higher for KOH (24.23 g of FAME/g of catalyst.min) than for calcium methoxide (17.06 g of FAME/g of catalyst.min) and calcium oxide (1.06 g of FAME/g of catalyst.min)

Pathways for phosphatidylcholine biosynthesis in bacteria.

Phosphatidylcholine (PC) is the major membrane-forming phospholipid in eukaryotes with important structural and signalling functions. Although many prokaryotes lack PC, it can be found in significant amounts in membranes of rather diverse bacteria. Two pathways for PC biosynthesis are known in bacteria, the methylation pathway and the phosphatidylcholine synthase (PCS) pathway. In the methylation pathway, phosphatidylethanolamine is methylated three times to yield PC, in reactions catalysed by one or several phospholipid N-methyltransferases (PMTs). In the PCS pathway, choline is condensed dir

Preparation and characterization of 8a-(phosphatidylcholine-dioxy)-alpha-tocopherones and their formation during the peroxidation of phosphatidylcholine in liposomes.

alpha-Tocopherol was reacted with the phosphatidylcholines (PCs), 1-palmitoyl-2-linoleoyl-3-sn-PC (PLPC), 1-palmitoyl-2-linolenoyl-3-sn-PC, 1-palmitoyl-2-arachidonoyl-3-sn-PC (PAPC) and 1-stearoyl-2-arachidonoyl-3-sn-PC, in the presence of the free radical initiator, 2,2'-azobis (2,4-dimethylvaleronitrile), at 37 degrees C. The addition products of alpha-tocopherol with the PC peroxyl radicals were isolated and identified as 8a-(PC-dioxy)-alpha-tocopherones, in which the peroxyl radicals derived from each PC molecule attacked the 8a-position of the alpha-tocopheroxyl radical. Oxidized phosphatidylcholines inhibit airway smooth muscle β2-adrenergic receptors via protein kinase C.

Understanding mechanisms for β2 adrenergic receptor (β2AR) insensitivity in asthma is incomplete. We discovered that the accumulation of oxidized phosphatidylcholines (OxPCs) in the lung correlates with airway hyperresponsiveness and that OxPCs induce contraction and cytokine synthesis in human airway smooth muscle (HASM) cells. Here, we test whether OxPCs impair β2AR agonist bronchodilator responses and related mechanism(s) for β2AR insensitivity. Using tracheal rings from BALB/c mice, we assayed the effects of oxidized 1-palmitoyl-2-arachidonoyl-sn-glycero-3-phosphocholine (OxPAPC) on β2AR a

High level of phosphatidylcholines/lysophosphatidylcholine ratio in urine is associated with prostate cancer.

The altered levels of phospholipids (PLs) and lysophospholipids (LPLs) in prostate cancer (CaP) and benign tissues in our previous findings prompted us to explore PLs and LPLs as potential biomarkers for CaP. Urinary lipidomics has attracted increasing attention in clinical diagnostics and prognostics for CaP. In this study, 31 prostate tissues obtained from radical prostatectomy were assessed using high-resolution matrix-assisted laser desorption/ionization imaging mass spectrometry (HR-MALDI-IMS). Urine samples were collected after digital rectal examination (DRE), and urinary lipids were ex

Alterations in rat pulmonary phosphatidylcholines after chronic exposure to ambient fine particulate matter.

This study elucidated the underlying pathophysiological changes that occur after chronic ambient fine particulate matter (PM2.5) exposure via a lipidomic approach. Five male Sprague-Dawley rats were continually whole-body exposed to ambient air containing PM2.5 at 16.7 ± 10.1 μg m(-3) from the outside of the building for 8 months, whereas a control group (n = 5) inhaled filtered air. Phosphorylcholine-containing lipids were extracted from lung tissue and profiled using ultraperformance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). Phosphatidylcholine: Greasing the Cholesterol Transport Machinery.

Negative feedback regulation of cholesterol metabolism in mammalian cells ensures a proper balance of cholesterol with other membrane lipids, principal among these being the major phospholipid phosphatidylcholine (PC). Processes such as cholesterol biosynthesis and efflux, cholesteryl ester storage in lipid droplets, and uptake of plasma lipoproteins are tuned to the cholesterol/PC ratio. Cholesterol-loaded macrophages in atherosclerotic lesions display increased PC biosynthesis that buffers against elevated cholesterol levels and may also facilitate cholesterol trafficking to enhance choleste

Mass spectrometric analysis of the amiodarone-induced alteration of phosphatidylcholines in living single cells.

Amiodarone is one of the cationic amphiphilic drugs which cause the drug-induced phospholipidosis (DIPL). With the accumulation of phospholipids, the multiple-membrane structures called lamellar bodies emerge in the cell. Since the pathogenesis mechanism has not been clarified yet, DIPL is still concerned in the clinical use of amiodarone and the new drug development processes. In this study, we have focused on phosphatidylcholines (PCs) which are the representative phospholipids and have developed a single-cell analytical method for the determination of PCs accumulated in the lamellar bodies.
