Single-molecule photobleaching (smPB) technique is a powerful tool for characterizing molecular assemblies. It can provide a direct measure of the number of monomers constituting a given oligomeric particle and generate the oligomer size distribution in a specimen. A major current application of this technique is in understanding protein aggregation, which is linked to many incurable diseases. Quantitative measurement of the size distribution of an aggregating protein in a physiological solution remains a difficult task, since techniques such as dynamic light scattering or fluorescence correla

Photodynamic therapy (PDT) utilizing aminolevulinic acid (ALA) as a photosensitiser has been used to ablate premalignant/malignant skin conditions including superficial basal cell carcinoma (BCC) with acceptable cosmetic outcome. Clinicians continue however to face difficulties in determining the exact dose that is sufficient to achieve a complete healing from the condition where the experience of the clinician remains the only determinant factor. This inaccuracy sometimes leads to undertreating these lesions. Here, we have clinically evaluated the use of fluorescence imaging system as monitor

OBJECTIVE: Photodynamic therapy (PDT) is an alternative treatment for cancer, which involves the administration of a photosensitizing agent that is activated by light at a specific wavelength. This illumination causes after a sequence of photoreactions, the production of reactive oxygen species responsible for the death of the tumor cells but also the degradation of the photosensitizing agent, which then loose the fluorescence properties. The phenomenon is commonly known as the photobleaching process and can be considered as a therapy efficiency indicator.
METHODS: This paper presents the des

Brown carbon (BrC) aerosols are key contributors to atmospheric light absorption and photochemistry, yet their optical properties and photochemical behavior are poorly understood. To explore the influence of a microdroplet environment on the photochemical decay of a representative BrC species, single particle cavity ring-down spectroscopy was used to probe individual aerosol particles containing imidazole-2-carboxaldehyde (IC) confined inside a linear electrodynamic quadrupole balance. The aerosol particles were levitated and exposed to 405 nm wavelength laser light to drive photobleaching. Th

Semiconductor polymers are the foundation of organic electronics due to their remarkable optical features, ability to form a thin film, and low cost compared to silicon. However, some of them have intense photobleaching under UV-blue radiation, compromising several applications. In this context, we have investigated the conformational disorder effect on the real-time photobleaching kinetics of a poly[2-methoxy-5-(2-ethylhexyloxy)-1,4-phenylenevinylene] (MEH-PPV)/chloroform solution under deep-blue radiation. Our results pointed out that a 405 nm diode laser initially causes a significant confo

We describe postfabrication trimming of coupling in both laterally and vertically coupled polymer microring resonators (MRRs), using photobleaching. For both cases, a tapered directional-coupler-based simple analytical model is developed to simulate the change in coupling due to a bleaching-induced decrease in refractive index. A tightly focused laser beam spot (a few kilowatts per square centimeter) is used to precisely bleach the coupling region alone. Coupling control is achieved for (1) high-Q passive rings by bleaching the vertically coupled chromophore-doped bus waveguide, and for (2) la

Digital light processing (DLP) bioprinting holds great promise for fabricating functional organ-on-a-chip models and in vitro drug screening platforms. However, existing curing models, largely adapted from synthetic resins, overlook the dynamic photobleaching inherent in hydrogels, leading to poor prediction accuracy for enclosed microchannels. To address this challenge, a custom DLP bioprinting system tailored for high-resolution structure fabrication was developed, along with an optical penetration meter to precisely characterize the light penetration behavior of hydrogel solutions. Based on

Anisotropic diffusion governs transport in a wide range of soft and biological materials, where microstructure and molecular interactions jointly shape how matter moves. Here, we quantitatively investigate anisotropic molecular transport in lyotropic chromonic liquid crystals (LCLCs) using fluorescence recovery after photobleaching (FRAP). Disodium cromoglycate (DSCG) serves as a model LCLC system, and diffusion is measured across isotropic, nematic, and columnar phases as concentration and temperature are varied. To disentangle the roles of microstructure and molecular interactions, we employ

Extracellular vesicles (EVs) are promising biomarkers for non-invasive cancer diagnostics, but their nanoscale size and correspondingly weak fluorescence signals make single-particle detection challenging without specialized, high-cost instrumentation. Here, we present a fast Fourier transform (FFT)-based computational imaging approach for EV analysis, termed EV-FFT, which repurposes fluorophore photobleaching into a frequency-domain signature for robust single-nanoparticle detection. EV-FFT transforms time-resolved fluorescence trajectories into Fourier space to suppress high-frequency stocha

Photodynamic therapy (PDT) is recognized as a powerful method to inactivate cells. However, the photosensitizer (PS), a key component of PDT, has suffered from undesired photobleaching. Photobleaching reduces reactive oxygen species (ROS) yields, leading to the compromise of and even the loss of the photodynamic effect of the PS. Therefore, much effort has been devoted to minimizing photobleaching in order to ensure that there is no loss of photodynamic efficacy. Here, we report that a type of PS aggregate showed neither photobleaching nor photodynamic action. Upon direct contact with bacteria

Although the primary biological function of retinal photoreceptors is to absorb light and provide visual information, extensive exposure to intense light could increase the risk of phototoxic reactions mediated by products of rhodopsin bleaching that might accumulate in photoreceptor outer segments (POS). The phototoxicity of POS, isolated from bovine retinas, was examined in cultured retinal pigment epithelium cells (ARPE-19) containing phagocytised POS and in selected model systems by determining POS ability to photogenerate singlet oxygen, and photoinduce oxidation of cholesterol and serum

Phototoxicity and photobleaching are major limitations in live-cell fluorescence microscopy. They are caused by fluorophores in an excited singlet or triplet state that generate singlet oxygen and other reactive oxygen species. The principle of controlled light exposure microscopy (CLEM) is based on non-uniform illumination of the field of view to reduce the number of excited fluorophore molecules. This approach reduces phototoxicity and photobleaching 2- to 10-fold without deteriorating image quality. Reduction of phototoxicity and photobleaching depends on the fluorophore distribution in the

This chapter details quantitative imaging of the Mero-CBD biosensor, which reports activation of endogenous Cdc42 in living cells. The procedures described are appropriate for imaging any biosensor that uses two different fluorophores on a single molecule, including FRET biosensors. Of particular interest is an algorithm to correct for fluorophore photobleaching, useful when quantitating activity changes over time. Specific topics include procedures and caveats in production of the Mero-CBD sensor, image acquisition, motion artifacts, shading correction, background subtraction, registration, a

BACKGROUND: Human nasal epithelial (HNE) cells can be sampled noninvasively and cultured to provide a model of the airway epithelium that reflects cystic fibrosis (CF) pathophysiology. We hypothesised that in vitro measures of HNE cell physiology would correlate directly with in vivo measures of lung physiology and therapeutic response, providing a framework for using HNE cells for therapeutic development and precision medicine.
METHODS: We sampled nasal cells from participants with CF (CF group, n=26), healthy controls (HC group, n=14) and single CF transmembrane conductance regulator (CFTR)

PURPOSE: The R120G mutation of alphaB-crystallin is known to cause desmin-related myopathy, but the mechanisms underlying the formation of cataract are not clearly established. We hypothesize that alteration of protein-protein interaction between R120G alphaB-crystallin and lens intermediate filament proteins is one of the mechanisms of congenital cataract.
METHODS: Protein-protein interactions were determined by confocal fluorescence resonance energy transfer (FRET) microscopy using green fluorescence protein (GFP) as the donor and red fluorescence protein (RFP) as the acceptor. The lens vim

Connective tissues are complex structures which contain collagen and elastin fibers. These fiber-based structures have a great influence on material mechanical properties and need to be studied at the microscopic scale. Several microscopy techniques have been developed in order to image such microstructures; among them are two-photon excited fluorescence microscopy and second harmonic generation. These observations have been coupled with mechanical characterization to link microstructural kinematics to macroscopic material parameter evolution. In this study, we present a new approach to measur

Dye-encoded bead-based assays are widely used for diagnostics. Multiple bead populations are required for multiplexing and can be produced using different dye colors, labeling levels, or combinations of dye ratios. Ready-to-use multiplex bead populations restrict users to specific targets, are costly, or require specialized instrumentation. In-house methods produce few bead plexes or require many fine-tuning steps. To expand bead encoding strategies, we present a simple, safe, and cost-effective bench-top system for generating bead populations using photobleaching. By photobleaching commercial

Fluorescence photobleaching recovery (FPR) measurements of virus glycoproteins on the surfaces of cells infected with vesicular stomatitis virus (VSV) and Sindbis virus showed that the VSV glycoprotein (G) remained mobile throughout the infectious cycle, whereas Sindbis virus glycoproteins (E1, E2) were partially mobile early after infection and immobile at later times when greater amounts of these proteins were on the cell surface. A highly mobile fraction of Sindbis virus glycoproteins was detected throughout the replication cycle of a temperature-sensitive mutant unable to form virus partic

Melanin pigments find a diverse field of practical application including art. One of the authors has developed a method of printing images with genetically modified Escherichia coli cells that produce melanin onto paper using the screen-printing process. The method prints the cell culture and then incubates the paper over 3 days, allowing the cells to deposit the melanin as they reproduce. However, using melanin comes with challenges related to color fading of "melanin-on-paper." The aim of this study was to evaluate how melanin-on-paper was photobleached. First, we characterized E. coli (EC)-

Cells respond to their environment via an intricate cellular signaling network, directing cell fate. Changes in cell fate are characterized by changes in gene transcription, dictated by (master) transcription factor activity. SOX9 is the master transcription factor for chondrocyte development. Its impaired function is implicated in osteoarthritis and growth disorders, such as dwarfism. However, the factors regulating SOX9 transcriptional activity are not yet fully mapped. Current methods to study transcription factor activity are indirect and largely limited to quantification of SOX9 target ge
