Native architecture and acclimation of photosynthetic membranes in a fast-growing cyanobacterium.

Efficient solar energy conversion is ensured by the organization, physical association, and physiological coordination of various protein complexes in photosynthetic membranes. Here, we visualize the native architecture and interactions of photosynthetic complexes within the thylakoid membranes from a fast-growing cyanobacterium Synechococcus elongatus UTEX 2973 (Syn2973) using high-resolution atomic force microscopy. In the Syn2973 thylakoid membranes, both photosystem I (PSI)-enriched domains and crystalline photosystem II (PSII) dimer arrays were observed, providing favorable membrane envir

Tight association of CpcL with photosystem I in Anabaena sp. PCC 7120 grown under iron-deficient conditions.

Phycobilisomes (PBSs), which are huge pigment-protein complexes displaying distinctive color variations, bind to photosystem cores for excitation-energy transfer. It is known that isolation of supercomplexes consisting of PBSs and photosystem I (PSI) or PBSs and photosystem II is challenging due to weak interactions between PBSs and the photosystem cores. In this study, we succeeded in purifying PSI-monomer-PBS and PSI-dimer-PBS supercomplexes from the cyanobacterium Anabaena sp. PCC 7120 grown under iron-deficient conditions by anion-exchange chromatography, followed by trehalose density grad

Specific light-harvesting complexes mediate grana stacking and prevent energy spillover between photosystems in plant chloroplasts.

Land plant chloroplasts are characterized by a prominent morphological feature: the stacking of thylakoid membranes into grana, ie defining a continuous membranous structure surrounded by the stroma and folded into appressed grana and non-appressed domains, enclosing the thylakoid lumen. Photosystem II and its antenna system are concentrated, whereas Photosystem I and ATPase are located in the grana end membranes and, in the stroma, membranes connecting grana. Thylakoid stacking promotes the physical separation of Photosystems I and II, thereby ensuring a balanced flow of excitation energy bet

Mg2+ limitation leads to a decrease in chlorophyll, resulting in an unbalanced photosynthetic apparatus in the cyanobacterium Synechocytis sp. PCC6803.

Mg2+, the most abundant divalent cation in living cells, plays a pivotal role in numerous enzymatic reactions and is of particular importance for organisms performing oxygenic photosynthesis. Its significance extends beyond serving as the central ion of the chlorophyll molecule, as it also acts as a counterion during the light reaction to balance the proton gradient across the thylakoid membranes. In this study, we investigated the effects of Mg2+ limitation on the physiology of the well-known model microorganism Synechocystis sp. PCC6803. Our findings reveal that Mg2+ deficiency triggers both

Photophysiological and photosynthetic complex changes during iron starvation in Synechocystis sp. PCC 6803 and Synechococcus elongatus PCC 7942.

Iron is an essential component in many protein complexes involved in photosynthesis, but environmental iron availability is often low as oxidized forms of iron are insoluble in water. To adjust to low environmental iron levels, cyanobacteria undergo numerous changes to balance their iron budget and mitigate the physiological effects of iron depletion. We investigated changes in key protein abundances and photophysiological parameters in the model cyanobacteria Synechococcus PCC 7942 and Synechocystis PCC 6803 over a 120 hour time course of iron deprivation. The iron stress induced protein (Isi

In Silico Analysis of Relationship between Proteins from Plastid Genome of Red Alga Palmaria sp. (Japan) and Angiotensin I Converting Enzyme Inhibitory Peptides.

Plastid proteins are one of the main components in red algae. In order to clarify the angiotensin I converting enzyme (ACE) inhibitory peptides from red alga Palmaria sp. (Japan), we determined the plastid genome sequence. The genome possesses 205 protein coding genes, which were classified as genetic systems, ribosomal proteins, photosystems, adenosine triphosphate (ATP) synthesis, metabolism, transport, or unknown. After comparing ACE inhibitory peptides between protein sequences and a database, photosystems (177 ACE inhibitory peptides) were found to be the major source of ACE inhibitory pe

Photoinhibition induced alterations in energy transfer process in phycobilisomes of PS II in the cyanobacterium, Spirulina platensis.

Exposure of algae or plants to irradiance from above the light saturation point of photosynthesis is known as high light stress. This high light stress induces various responses including photoinhibition of the photosynthetic apparatus. The degree of photoinhibition could be clearly determined by measuring the parameters such as absorption and fluorescence of chromoproteins. In cyanobacteria and red algae, most of the photosystem (PS) II associated light harvesting is performed by a membrane attached complex called the phycobilisome (PBS). The effects of high intensity light (1000-4000 micromo

Cyclic Electron Transport via the NDH Complex Sustains Photosynthesis and Productivity Under Fluctuating and Sub-Optimal Environments.

The chloroplast NADH dehydrogenase-like (NDH) complex mediates cyclic electron transport (CET) around photosystem I (PSI) and contributes to photosynthetic regulation and photoprotection under various environmental stresses. Although NDH function has been extensively characterized under controlled conditions, NDH-deficient mutants often show only subtle phenotypes in such environments, leaving its physiological importance to photosynthesis in plants grown under naturally fluctuating field conditions poorly understood. Here, we evaluated growth, yield, and photosynthetic performance of NDH-defi

Interplay between CO2 and light governs carbon partitioning in Chlamydomonas reinhardtii.

Increasing CO2 availability is a common practice at the industrial level to trigger biomass productivity in microalgae cultures. Still, the consequences of high CO2 availability in microalgal cells exposed to relatively high light require further investigation. Here, the photosynthetic, physiologic, and metabolic responses of the green microalga model Chlamydomonas reinhardtii were investigated in high or low CO2 availability conditions: high CO2 enabled higher biomass yields only if sufficient light energy was provided. Moreover, cells grown in high light and high CO2 availability were charac

Photoprotection of PSI by Far-Red Light Against the Fluctuating Light-Induced Photoinhibition in Arabidopsis thaliana and Field-Grown Plants.

It has been reported that PSI photoinhibition is induced even in wild-type plants of Arabidopsis thaliana, rice and other species by exposure of leaves to fluctuating light (FL) for a few hours. Because plants are exposed to FL in nature, they must possess protective mechanisms against the FL-induced photodamage. Here, using A. thaliana grown at various irradiances, we examined PSI photoprotection by far-red (FR) light at intensities comparable with those observed in nature. Dark-treated leaves were illuminated by red FL alternating high/low light at 1,200/30 µmol m-2 s-1 for 800 ms/10 s. By t

Lumen alkalization under elevated bicarbonate concentration outside isolated thylakoids of higher plants.

Illumination causes alkalization of chloroplast stroma, which is accompanied with a substantial increase in stromal bicarbonate concentration there. Whether bicarbonate itself contributes to regulation of thylakoid lumen pH has remained unclear. Here we show that, increasing bicarbonate concentration in the suspension of the isolated pea thylakoids increases luminal pH and stimulates electron transport through the photosynthetic electron transport chain. Addition of bicarbonate stimulated the electron transport in the functionally isolated photosystem I in the case when a pH sensitive electron

Evidence for Protein Dynamics-Induced Energy Transfer Pathway Changes in Photosynthetic Complexes.

Photosystem I (PSI) is a chlorophyll-binding protein complex that plays a central role in photosynthesis. PSI isolated from cyanobacteria, Thermosynechococcus vestitus, contains a light-harvesting system that consists of approximately 100 chlorophyll a molecules. The effect of the protein fluctuation dynamics on the excitation energy migration within the pigment array is fundamentally important. In 2009, Brecht and co-workers proposed that protein dynamics can induce variations in the excitation energy transfer (EET) pathways within PSI ( Brecht et al. Proc. Natl. Acad. Sci. U. S. In situ structures of plant photosystem supercomplexes.

Photosynthesis sustains life on Earth by converting light to chemical energy through the coordinated action of photosystem I (PSI) and photosystem II (PSII) within thylakoid membranes1-4. Although structures of isolated photosystems are available, their native organization in chloroplasts remains unknown. Here, using in situ cryo-electron microscopy, we directly imaged Oryza sativa (rice) chloroplasts and determined structures of photosystem supercomplexes in their native membrane environment. We resolved a C2S2M2L4-type PSII-light harvesting complex II (LHCII) supercomplex, including four LHC

Multiple sources of carbonic anhydrase activity in pea thylakoids: soluble and membrane-bound forms.

Carbonic anhydrase (CA) activity of pea thylakoids, thylakoid membranes enriched with photosystem I (PSI-membranes), or photosystem II (PSII-membranes) as well as both supernatant and pellet after precipitation of thylakoids treated with detergent Triton X-100 were studied. CA activity of thylakoids in the presence of varying concentrations of Triton X-100 had two maxima, at Triton/chlorophyll (triton/Chl) ratios of 0.3 and 1.0. CA activities of PSI-membranes and PSII-membranes had only one maximum each, at Triton/Chl ratio 0.3 or 1.0, respectively. Two CAs with characteristics of the membrane

Application of Simultaneous Active and Passive Fluorescence Observations: Extending a Fluorescence-Based qL Estimation Model.

The fraction of open Photosystem II (PSII) reaction centers (qL) is critical for connecting broadband PSII fluorescence (ChlFPSII) with the actual electron transport from PSII to Photosystem I. Accurately estimating qL is fundamental for determining ChlFPSII, which, in turn, is vital for mechanistically estimating the actual electron transport rate and photosynthetic CO2 assimilation. Chlorophyll fluorescence provides direct physiological insights, offering a robust foundation for qL estimation. However, uncertainties in the ChlFPSII-qL relationship across different plant functional types (PFT

Detergent Choice Shapes the Solution Structures of Photosystems I and II: Implications for Crystallization and High-Resolution Studies.

Photosystems I (PSI) and II (PSII) are pigment-protein complexes that perform the light-driven charge separation necessary to convert solar energy into a biochemically usable form in a fundamental process called photosynthesis. Small-angle X-ray scattering provides unique structural insights into PSI and PSII in solution under near-physiological conditions. Here, we study the solubilization of PSI and PSII with different detergents, the octaethylene glycol monododecyl ether (C12E8) and the most commonly used n-dodecyl-β-D-maltoside (DDM). It is noteworthy that the volume of the C12E8 detergent

Daily Heat Stress Induces Donor-Side Limitation of PSI Via Downregulation of the Cyt b6f Complex.

The rapidly warming climate is driving increasingly frequent and intense heat waves worldwide, posing major challenges to plant metabolism and growth. Despite the central role of photosynthesis in plant growth, the effects of prolonged heat stress on the primary reactions of photosynthesis remain poorly understood. Here, we studied how long-term heat exposure affects growth and photosynthetic performance in the model plant Arabidopsis thaliana. Plants were exposed daily to high temperature (38°C) for 4 h throughout the growth period. Conflicting to earlier studies, we show that PSII-LHCII rema

Differences between barley and maize revealed in limitations of photosystems I and II under high temperature and low air humidity.

Non-photochemical quenching and limitations of the photosystem I and photosystem II activities were studied in C3 -plant barley and C4 -plant maize. Plants were exposed to prolonged heat stress under high and low air humidity. Both species decreased non-photochemical quenching at 37-42°C, which increased at 46°C. A decrease of photosystem II activity at 46°C in lower air humidity was achieved through different mechanisms. In barley, photosystem II was downregulated by the increase of non-photochemical quenching. In maize, photosystem II was downregulated by the increase of acceptor-side limita

Photochemistry of Photosystems II and I in Rice Plants Grown under Different N Levels at Normal and High Temperature.

Although N levels affect leaf photosynthetic capacity, the effects of N levels on the photochemistry of photosystems II and I (PSII and PSI, respectively) are not well-understood. In the present study, we examined this aspect in rice (Oryza sativa L. 'Hitomebore') plants grown under three different N levels at normal or high temperatures that can occur during rice culture and do not severely suppress photosynthesis. At both growth temperatures, the quantum efficiency of PSII [Y(II)] and the fraction of the primary quinone electron acceptor in its oxidized state were positively correlated with
