The avoidance response in Phycomyces.

Phycomyces sporangiophores grow away from stationary objects, a phenomenon known as the avoidance response. Evidence is presented suggesting that a growth-stimulating gas is emitted from the sporangiophore and is then swept to the leeward side by air currents resulting in higher gas concentration on that side. The presence of a stationary barrier decreases the passive movement of the gas away from the leeward side. It is proposed that an increase of this gas on one side causes that side to grow faster. Indirect evidence suggests that the gas is water vapor.

Alternative respiration of fungus Phycomyces blakesleeanus.

Respiratory characteristics of germinating spores, developing mycelium and mitochondria of the fungus Phycomyces blakesleeanus were investigated by means of oxygen Clark-type electrode. The effects of respiratory inhibitors and metabolic compounds on oxygen consumption were tested. It was demonstrated that P. blakesleeanus apart of cyanide-sensitive respiration, CSR, possess alternative respiration, (cyanide-resistant respiration, CRR) which is constitutive and whose capacity decreases during development. Maximum is observed for activated spores where CRR capacity is significantly greater than

A macroconidial synanamorph of Kendrickiella phycomyces (Pezizomycotina incertae sedis).

A macroconidial synanamorph, namely a phragmosporous phialidic morph of Kendrickiella phycomyces (Pezizomycotina incertae sedis), was newly observed in authentic cultures isolated from the Kitora and Takamatsuzuka Tumuli in Japan. The synanamorph was sparsely produced throughout of the colonies on PDA after a few days at room temperature in cultures revived from lyophiles or in streak cultures, both derived from germinated Kendrickiella-type aseptate microconidia. After 1-2 wk, the microconidial synanamorph (the Kendrickiella-type synanamorph) and creeping hyphae completely overgrew the macroc

A cytoplasmically inherited mutation in the fungus Phycomyces blakesleeanus.

Fourteen mutants of the fungus Phycomyces blakesleeanus, showing high levels of resistance to copper, were isolated. In all the mutants, copper resistance behaved as a very variable and unstable trait. In the mutant strain MU102, the mutation was demonstrated to be cytoplasmically inherited. In addition, this mutant strain differed from the wild-type in growth, respiration rate, and shape and viability of spores.

Identification of the ergC gene involved in polyene drug sensitivity in the Mucorales species Phycomyces blakesleeanus.

BACKGROUND: A strain of Phycomyces blakesleeanus (Mucorales, Mucoromycota) that was previously isolated after ultraviolet mutagenesis has altered responses to polyene antifungal drugs, sterol profiles, and phototropism of its sporangia. In this study, the genetic basis for these changes was sought. METHODS AND RESULTS: Two base pair substitutions were identified in the mutant within a P. blakelesleeanus gene that is homologous to others characterized from fungi, such as the Saccharomyces cerevisiae ERG3 gene, encoding sterol Δ5,6-desaturase. The polyene resistance and growth reduction phenoty

The effect of anoxia on PolyP content of Phycomyces blakesleeanus mycelium studied by 31P NMR spectroscopy.

The effect of anoxia and the respiratory chain inhibitors azide and cyanide on the polyphosphate content of Phycomyces was studied by in vivo (31)P NMR spectroscopy. Anoxia was manifested by a decrease of core polyphosphates (PP(i)) and increase of intracellular inorganic phosphate (P(i)) signal. Normalized changes in PP(i)/P(i) ratio between control and nitrogen-purged mycelia suggest that the sensitivity to anoxia differs with growth phases. Azide acts in the same way as anoxia, by decreasing the PP(i)/P(i) ratio, while cyanide causes an increase of the PP(i)/P(i) ratio.

Internal ophthalmoplegia resulting from acute orbital phycomycosis.

Phycomycosis is an opportunistic infection occurring in immunocompromised hosts. It is the most acutely fatal fungal disease known. Therefore, early recognition and treatment are essential. Ocular and orbital involvement is part of a rhino-orbital-cerebral form of the disease. The orbital apex syndrome is the hallmark orbital involvement. A case of phycomycosis with internal ophthalmoplegia alone is reported. The internal ophthalmoplegia is probably secondary to involvement at the apex of the orbit, although not sufficiently diffuse to result in the complete orbital apex syndrome.

[Necrotizing fasciitis in an immunocompetent patient caused by Apophysomyces elegans].

A case study is presented of a 7-year-old boy, seriously injured in a car accident, who developed a fatal infection due to Aphophysomyces elegans--a mold of the Mucoracea family. Fungal invasion was initially manifested by a spotted wound in the left lumbar region which developed into a necrotizing fasciitis. Later this progressed to the right lumbar area, including the gluteus and the corresponding flank. Antimycotic treatment proved ineffective, and the child died 8 weeks after the accident. Other cases due to this fungus are reviewed.

Survival of a recipient of renal transplantation after pulmonary phycomycosis.

A 48-year-old nondiabetic man maintained on prednisone, azathioprine, and cyclosporine after cadaveric renal transplantation developed multiple unilateral phycomycotic pulmonary abscesses. Despite treatment with amphotericin B the patient remained febrile. Surgical resection of these pulmonary abscesses was performed and the patient remains free of disease 1 year after operation. To our knowledge, this patient represents the first reported case of pulmonary phycomycosis in a recipient of renal transplantation without diabetes mellitus.

Biocontrol Potential of Native Trichoderma Strains Toward Soil-Borne Phytopathogenic and Saprotrophic Fungi.

The potential of Trichoderma fungi as biocontrol agents has not yet been fully explored, as there is a large repertoire of inter- and intra-species variation in their phytopathogenic antagonistic effects due to different adaptations of individual Trichoderma strains. In the present study, we investigated the biocontrol efficacy of eight native isolates of Trichoderma spp. against the soilborne phytopathogens Sclerotinia sclerotiorum and Rhizoctonia solani and a representative of the Mucoromycota, Phycomyces blakesleeanus. An in vitro dual culture test showed a complete (100%) inhibition of S.

Transcription Factors Tec1 and Tec2 Play Key Roles in the Hyphal Growth and Virulence of Mucor lusitanicus Through Increased Mitochondrial Oxidative Metabolism.

Mucormycosis is a lethal and difficult-to-treat fungal infection caused by fungi of the order Mucorales. Mucor lusitanicus, a member of Mucorales, is commonly used as a model to understand disease pathogenesis. However, transcriptional control of hyphal growth and virulence in Mucorales is poorly understood. This study aimed to investigate the role of Tec proteins, which belong to the TEA/ATTS transcription factor family, in the hyphal development and virulence of M. lusitanicus. Unlike in the genome of Ascomycetes and Basidiomycetes, which have a single Tec homologue, in the genome of Mucoral

Real-time application of ITS and D1-D3 nanopore amplicon metagenomic sequencing in fungal infections: Enhancing fungal infection diagnostics.

While fungal infections cause considerable morbidity and mortality, the performance of the current diagnostic tests for fungal infection is low. Even though fungal metagenomics or targeted next-generation sequencing have been investigated for various clinical samples, the real-time clinical utility of these methods still needs to be elucidated. In this study, we used internal transcribed spacer (ITS) and D1-D3 ribosomal DNA nanopore amplicon metagenomic sequencing to assess its utility in patients with fungal infections. Eighty-four samples from seventy-three patients were included and categor

Immunology of fungal infections in animals.

The nature of immunity to fungal infection is discussed predominantly for mammals and birds. T-cell-mediated immunity seems essential for recovery both from cutaneous and mucosal infections (Candida, Malassezia and dermatophytes) and from infections of systemic fungal pathogens (Cryptococcus, Blastomyces, Histoplasma, and Coccidioides). Often chronic progressive disease caused by these fungi is associated with a depression or absence of T-cell-mediated immunity to antigens of the infecting fungus. In contrast recovery from disease, or absence of clinical disease after exposure to these fungi,

Apophysomyces variabilis: draft genome sequence and comparison of predictive virulence determinants with other medically important Mucorales.

BACKGROUND: Apophysomyces species are prevalent in tropical countries and A. variabilis is the second most frequent agent causing mucormycosis in India. Among Apophysomyces species, A. elegans, A. trapeziformis and A. variabilis are commonly incriminated in human infections. The genome sequences of A. elegans and A. trapeziformis are available in public database, but not A. variabilis. We, therefore, performed the whole genome sequence of A. variabilis to explore its genomic structure and possible genes determining the virulence of the organism. RESULTS: The whole genome of A. variabilis NCCP
