Phytic Acid and Transporters: What Can We Learn from low phytic acid Mutants.

Phytic acid has two main roles in plant tissues: Storage of phosphorus and regulation of different cellular processes. From a nutritional point of view, it is considered an antinutritional compound because, being a cation chelator, its presence reduces mineral bioavailability from the diet. In recent decades, the development of low phytic acid (lpa) mutants has been an important goal for nutritional seed quality improvement, mainly in cereals and legumes. Different lpa mutations affect phytic acid biosynthetic genes. However, other lpa mutations isolated so far, affect genes coding for three c

Phytic acid in health and disease.

Phytic acid (PA), a major phosphorus storage compound of most seeds and cereal grains, contributes about 1 to 7% of their dry weight. It may account for more than 70% of the total kernel phosphorus. PA has the strong ability to chelate multivalent metal ions, especially zinc, calcium, and iron. The binding can result in very insoluble salts that are poorly absorbed from the gastrointestinal tract, which results in poor bioavailability (BV) of minerals. Alternatively, the ability of PA to chelate minerals has been reported to have some protective effects, such as decreasing iron-mediated colon

Phytic acid interactions in food systems.

Phytic acid is present in many plant systems, constituting about 1 to 5% by weight of many cereals and legumes. Concern about its presence in food arises from evidence that it decreases the bioavailability of many essential minerals by interacting with multivalent cations and/or proteins to form complexes that may be insoluble or otherwise unavailable under physiologic conditions. The precise structure of phytic acid and its salts is still a matter of controversy and lack of a good method of analysis is also a problem. It forms fairly stable chelates with almost all multivalent cations which a

Iron, Zinc and Phytic Acid Retention of Biofortified, Low Phytic Acid, and Conventional Bean Varieties When Preparing Common Household Recipes.

Biofortification is an effective method to improve the nutritional content of crops and nutritional intake. Breeding for higher micronutrient mineral content in beans is correlated with an increase in phytic acid, a main inhibitor of mineral absorption in humans. Low phytic acid (lpa) beans have a 90% lower phytic acid content compared to conventional beans. This is the first study to investigate mineral and total phytic acid retention after preparing common household recipes from conventional, biofortified and lpa beans. Mineral retention was determined for two conventional, three biofortifie

Environmental impact of phytic acid in Maize (Zea mays. L) genotypes for the identification of stable inbreds for low phytic acid.

Phytic acid is a ubiquitous compound that chelates the micronutrients in food and hinder their absorption. Hence, breeding for low phytate content for producing stable low phytic acid (lpa) hybrids is essential. Phytic acid content in maize grains has been found to vary across environments and its stable expression has yet to be explored. In a view of this, forty inbreds were screened with two checks viz., CO-6 and CO-H(M)-8 across three locations. Twenty morphological and three quality traits were observed to identify the stable lines for low phytic acid with higher free inorganic phosphorous

Ferrihydrite Transformation Impacted by Coprecipitation of Phytic Acid.

Phytic acid is a common phosphate monoester that is present in soils due to the deposition of plant-derived materials. Thus far, its interaction with dissolved Fe and Fe minerals has not been as extensively investigated as phosphate, although it is expected be highly reactive due to its multiple phosphate functional groups. In this study, the effects of phytic acid on the formation of iron oxyhydroxide was investigated at near neutral pH as a function of the phytic acid/Fe ratio (0.05-0.5) and aging time using zeta potential measurements, X-ray diffraction, Fe K-edge X-ray absorption spectrosc

Hydrolysis of phytic acid and its availability in rabbits.

Twenty weaned rabbits were fed ad libitum two granulated feeds containing lucerne meal, barley, oats, wheat bran, oilseed meals and sugarbeet pulp in different proportions. Phytate P in these feeds represented 28.6 and 29.3 % of the total P. Digestibility trials were carried out in rabbits 7 and 10 weeks old. Digestibility of phytate P was 82.1 %, on average. Apparent digestibility of total P was 48.1 and 35.5 % in rabbits aged 7 and 10 weeks, respectively. Concentration of P in the faecal DM of these rabbits averaged 11.9 and 14.7 mg/g. Most of the faecal P was phosphates P (68.1 %). Proporti

Effects of low-phytic acid corn, low-phytic acid soybean meal, and phytase on nutrient digestibility and excretion in growing pigs.

Forty-eight grower pigs were used to evaluate the effects of feeding low phytic acid (LPA) corn, LPA soybean meal, normal corn (NC), normal soybean meal (NSBM), and phytase on nutrient digestibility and excretion. Barrows were blocked by BW (initial BW=45.3+/-1.6 kg) and randomly assigned to 1 of 8 dietary treatments in a 2 x 2 x 2 factorial arrangement (6 pigs/treatment). Pigs were fed twice daily (0700 and 1700 h) at 3 times the ME requirement for maintenance. Phytase was added to the diet at 510 phytase units/kg of feed (where 1 phytase unit is the quantity of enzyme that liberates 1 mumol

Phytic acid protects corneal endothelium from UVA induced ferroptosis.

Corneal endothelial decompensation is a common vision-threatening disease, with a diverse range of pathogenic factors, ultimately necessitating corneal transplantation. Corneal endothelial cells (CECs) act as a barrier between the aqueous humor and stroma, and are non-proliferative in vivo. CECs are susceptible to oxidative stress damage due to their lifelong high metabolic state, which enables them to constantly pump liquid to maintain the dehydrated status of the cornea. Consequently, CECs are subjected to the accumulation of oxidative stress over time, which can result in cell death in the

Phytic acid derived bioactive CaO-P2O5-SiO2 gel-glasses.

The possibility of using phytic acid as a precursor to synthesize CaO-P(2)O(5)-SiO(2) glasses by sol-gel method has been explored and the pseudo ternary phase diagram has been established. It was shown that gel-glasses over a broader range of compositions could be prepared compared to other phosphorus precursors or melt-quenching method. Furthermore, phytic acid was found to assist calcium being incorporated into glass networks. In vitro tests in simulated body fluid (SBF) were performed on the above gel-glasses and it was found that they were bioactive over a much broader compositional range

Effect of phytic acid on colonic epithelial cell proliferation.

Cell proliferation was studied in descending, transverse and ascending colonic crypts of rats fed either 0%, 0.6%, 1.2% or 2.0% phytic acid (PA). Significant effects of phytic acid were seen only in the descending colon. No changes in crypt height were observed. Both the 1.2 and 2.0% phytic acid groups exhibited significantly lower uppermost labelled cells than the control group. Animals given 1.2% and 2.0% phytic acid also showed significantly reduced DNA synthetic cells, as indicated by lower labelling indices.

Regulation on Aggregation Behavior and In Vitro Digestibility of Phytic Acid-Whey Protein Isolate Complexes: Effects of Heating, pH and Phytic Acid Levels.

The impact of heat treatment, pH and phytic acid (PA) concentration on the aggregation behavior and digestibility of whey protein isolate (WPI) was investigated. The experimental results indicated that below the isoelectric point of WPI, heat treatment and elevated PA levels significantly increased turbidity and particle size, leading to the aggregation of WPI molecules. No new chemical bonds were formed and the thermodynamic parameters ΔH < 0, ΔS > 0 and ΔG < 0 suggested that the interaction between PA and WPI was primarily a spontaneous electrostatic interaction driven by enthalpy. After the

Gibberellic acid promoting phytic acid degradation in germinating soybean under calcium lactate treatment.

BACKGROUND: Phytic acid as a phosphorus storage vault provides phosphorus for plant development. It is an anti-nutritional factor for humans and some animals. However, its degradation products lower inositol phosphates have positive effects on human health. In this study, the effect of gibberellic acid (GA) on phytic acid degradation under calcium lactate (Ca) existence was investigated. RESULTS: The results showed that Ca + GA treatment promoted the growth status, hormone metabolism and phytic acid degradation in germinating soybean. At the same time, the availability of phosphorus, the acti

Rapid Analysis of Phytic Acid by Paper Spray Mass Spectrometry.

Phytic acid (PA), owing to its strong acidity and multidentate metal-chelating properties, readily forms multiple adduct/complex ions in mass spectrometry and is prone to pronounced matrix effects, resulting in complicated spectra and compromised sensitivity and quantitative robustness, which poses a major challenge for rapid and accurate PA quantification. Herein, we developed a rapid quantitative method for PA based on trimethylsilyldiazomethane (TMSD) methyl-ester derivatization coupled with paper spray mass spectrometry (PS-MS). PA was derivatized with TMSD to yield the methylated product

Phytic acid impairs macrophage inflammatory response in Echinococcus multilocularis infection.

The helminth Echinococcus multilocularis relies on immune evasion strategies to persist within its host. The laminated layer (LL) surrounding the parasite provides physical protection while modulating host immune responses. E. multilocularis' immunomodulatory mechanisms are poorly understood and we explored the role of phytic acid, a known component of E. granulosus sensu lato. We show that phytic acid is also present in E. multilocularis-infected tissue and impairs macrophage inflammation. In vivo, inflammatory macrophages accumulate near the metacestode, yet do not express IL-6, indicating a

Alginate enriched with phytic acid for hydrogels preparation. Therapeutic applications.

In the last decade, numerous innovative strategies have been used to obtain highly efficient synthetic or semi-synthetic biomaterials. Between these innovative biomaterials, hydrogels occupy a distinct place due to their superior biological and physico-chemical characteristics. Alginate is a natural linear polysaccharide with important physico-chemical and biological properties. Recently, we obtained a new hydrogel based on alginate and phytic acid with improved physico-chemical properties. In the present study, the hydrogels previously obtained were tested in terms of their biological propert

Complex Formation of Phytic Acid With Selected Monovalent and Divalent Metals.

The formation of metal complexes with phytic acid is a complex process that depends strongly on the metal-to-ligand molar ratio, pH value and consequent protonation level of the phytate ligand as well as accompanying side reactions, in particular metal hydrolysis and precipitation of the formed coordination compounds. In the present work, the potentiometric titration technique was used in combination with a detailed analysis of the equivalent point dependencies for selected biologically relevant monovalent and divalent cations from the groups of alkaline earths and transition metals, namely: M

Seed Biofortification and Phytic Acid Reduction: A Conflict of Interest for the Plant?

Most of the phosphorus in seeds is accumulated in the form of phytic acid (myo-inositol-1,2,3,4,5,6-hexakisphosphate, InsP₆). This molecule is a strong chelator of cations important for nutrition, such as iron, zinc, magnesium, and calcium. For this reason, InsP₆ is considered an antinutritional factor. In recent years, efforts to biofortify seeds through the generation of low phytic acid (lpa) mutants have been noteworthy. Moreover, genes involved in the biosynthesis and accumulation of this molecule have been isolated and characterized in different species. Beyond its role in phosphorus stor

Peroxidase Sensor Based on Electroactive Ce3+-Phytic Acid-Cu2+@Cu Composite.

Peroxide is a vital oxidant that can quickly kill bacteria, viruses, and fungi in food and ensure food safety. However, excessive intake will stimulate the gastrointestinal tract, cause abdominal pain and vomiting, and its strong oxidation will also destroy the vitamin and protein structure in food. Cerium-based nanomaterials exhibit good biocompatibility and strong chemical stability, and have significant application value in biological detection. Cu nanoparticles were deposited on the surface of a glassy carbon electrode (GCE) by potentiostatic deposition. The Ce3+/PA-Cu2+@Cu/GCE sensor was
