Phytophthora infestans enters the genomics era.

UNLABELLED: summary Phytophthora infestans, cause of late-blight, is the most devastating disease of potato world-wide. Recent years have seen a dramatic intensification in molecular biological studies of P. infestans, including the development of novel tools for transformation and gene silencing and the resources for genetical, transcriptional and physical mapping of the genome. This review will focus on the increasing efforts to use these resources to discover the genetic bases of pathogenicity, avirulence and host-specificity. TAXONOMY: Phytophthora infestans (Mont.) de Bary-Kingdom Chromi

N-acetyltransferase AAC(3)-I confers gentamicin resistance to Phytophthora palmivora and Phytophthora infestans.

BACKGROUND: Oomycetes are pathogens of mammals, fish, insects and plants, and the potato late blight agent Phytophthora infestans and the oil palm and cocoa infecting pathogen Phytophthora palmivora cause economically impacting diseases on a wide range of crop plants. Increasing genomic and transcriptomic resources and recent advances in oomycete biology demand new strategies for genetic modification of oomycetes. Most oomycete transformation procedures rely on geneticin-based selection of transgenic strains. RESULTS: We established N-acetyltransferase AAC(3)-I as a gentamicin-based selectabl

Aluminum induces cross-resistance of potato to Phytophthora infestans.

The phenomenon of cross-resistance allows plants to acquire resistance to a broad range of stresses after previous exposure to one specific factor. Although this stress-response relationship has been known for decades, the sequence of events that underpin cross-resistance remains unknown. Our experiments revealed that susceptible potato (Solanum tuberosum L. cv. Bintje) undergoing aluminum (Al) stress at the root level showed enhanced defense responses correlated with reduced disease symptoms after leaf inoculation with Phytophthora infestans. The protection capacity of Al to subsequent stress

Transport of amino acids and calcium in the fungus, Phytophthora infestans.

Amino acid transport in the cells of calcium-dependent fungus Phytophthora infestans has been largely studied with respect to phenylalanine. It is defined as an active process in a number of characteristics. The reasons for that are as follows: 1) accumulation of amino acids is inhibited by carbodiimide and ruthenium red, which suggests the participation of an intermediate; 2) the transport is energy-dependent, since it is inhibited by potassium cyanide, sodium azide and 2,4-dinitrophenol. Cold-osmotic shock results in the inhibition of the amino acid and glucose transport 20-80 times without

Combining Different Potato-Associated Pseudomonas Strains for Improved Biocontrol of Phytophthora infestans.

Late blight caused by Phytophthora infestans is considered as the most devastating disease of potato and is a re-emerging problem worldwide. Current late blight control practices rely mostly on synthetic fungicides or copper-based products, but growing awareness of the negative impact of these compounds on the environment has led to the search for alternative control measures. A collection of Pseudomonas strains isolated from both the rhizosphere and the phyllosphere of potato was recently characterized for in vitro protective effects against P. infestans. In the present study, we used a leaf

Biocontrol potential of Bacillus subtilis WL2.3 in mitigating Phytophthora infestans infection in potatoes.

This research aimed to identify native biocontrol bacteria capable of suppressing Phytophthora infestans in potato crops. A total of forty bacterial strains were isolated from wheat (Triticum aestivum), sunflower (Helianthus annuus), potato (Solanum tuberosum), and cotton (Gossypium herbaceum) plants collected in Sibi, Pakistan. Out of these, ten isolates showed antagonistic effects against P. infestans, with inhibition percentages ranging from 27 to 62%. The isolate WL2.3 exhibited the strongest inhibition at 62%. Furthermore, sporangia germination tests, including those using cell-free super

A nonspecific lipid transfer protein, StLTP10, mediates resistance to Phytophthora infestans in potato.

Nonspecific lipidtransfer proteins (nsLTPs), which are small, cysteine-rich proteins, belong to the pathogenesis-related protein family, and several of them act as positive regulators during plant disease resistance. However, the underlying molecular mechanisms of these proteins in plant immune responses are unclear. In this study, a typical nsLTP gene, StLTP10, was identified and functionally analysed in potato. StLTP10 expression was significantly induced by Phytophthora infestans, which causes late blight in potato, and defence-related phytohormones, including abscisic acid (ABA), salicylic

Performance of a tetracycline-responsive transactivator system for regulating transgenes in the oomycete Phytophthora infestans.

The oomycete genus Phytophthora includes many important plant pathogens for which extensive genome data exist, but lacking is an inducible expression system to study contributions of their genes to growth and pathogenicity. Here the adaptation of the reverse tetracycline transactivator (rtTA) system to P. infestans is described. Vectors were developed containing rtTA expressed from an oomycete promoter, and beta-glucuronidase (GUS) controlled by TetR binding sites fused to a minimal oomycete promoter. Transformants were obtained in which GUS was expressed in a dose-dependent manner by the rtTA

Chloroplasts alter their morphology and accumulate at the pathogen interface during infection by Phytophthora infestans.

Upon immune activation, chloroplasts switch off photosynthesis, produce antimicrobial compounds and associate with the nucleus through tubular extensions called stromules. Although it is well established that chloroplasts alter their position in response to light, little is known about the dynamics of chloroplast movement in response to pathogen attack. Here, we report that during infection with the Irish potato famine pathogen Phytophthora infestans, chloroplasts accumulate at the pathogen interface, associating with the specialized membrane that engulfs the pathogen haustorium. The chemical

Contribution of Hydrogen Cyanide to the Antagonistic Activity of Pseudomonas Strains Against Phytophthora infestans.

Plants face many biotic and abiotic challenges in nature; one of them is attack by disease-causing microbes. Phytophthora infestans, the causal agent of late blight is one of the most prominent pathogens of the potato responsible for multi-billion-dollar losses every year. We have previously reported that potato-associated Pseudomonas strains inhibited P. infestans at various developmental stages. A comparative genomics approach identified several factors putatively involved in this anti-oomycete activity, among which was the production of hydrogen cyanide (HCN). Here, we report the relative c

Editing miR482b and miR482c Simultaneously by CRISPR/Cas9 Enhanced Tomato Resistance to Phytophthora infestans.

Late blight, caused by Phytophthora infestans, is severely damaging to the global tomato industry. Micro-RNAs (miRNAs) have been widely demonstrated to play vital roles in plant resistance by repressing their target genes. Recently, the clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated protein 9 (Cas9) method has been continuously improved and extensively applied to edit plant genomes. However, editing multiplex miRNAs by CRISPR/Cas9 in tomato has not been studied yet. We knocked out miR482b and miR482c simultaneously in tomato through the multiplex CRISPR/Ca

Copper ions suppress abscisic acid biosynthesis to enhance defence against Phytophthora infestans in potato.

Copper-based antimicrobial compounds are widely and historically used to control plant diseases, such as late blight caused by Phytophthora infestans, which seriously affects the yield and quality of potato. We previously identified that copper ion (Cu2+ ) acts as an extremely sensitive elicitor to induce ethylene (ET)-dependent immunity in Arabidopsis. Here, we found that Cu2+ induces the defence response to P. infestans in potato. Cu2+ suppresses the transcription of the abscisic acid (ABA) biosynthetic genes StABA1 and StNCED1, resulting in decreased ABA content. Treatment with ABA or inhib

Phytophthora infestans RXLR effector PITG20303 targets a potato MKK1 protein to suppress plant immunity.

Pathogens secret a plethora of effectors into the host cell to modulate plant immunity. Analysing the role of effectors in altering the function of their host target proteins will reveal critical components of the plant immune system. Here we show that Phytophthora infestans RXLR effector PITG20303, a virulent variant of AVRblb2 (PITG20300) that escapes recognition by the resistance protein Rpi-blb2, suppresses PAMP-triggered immunity (PTI) and promotes pathogen colonization by targeting and stabilizing a potato MAPK cascade protein, StMKK1. Both PITG20300 and PITG20303 target StMKK1, as confi

Potato foliar infection with Phytophthora infestans drives strong, cultivar-specific shifts in rhizosphere communities.

BACKGROUND: Potato is an important crop worldwide, yet its production is severely threatened by Phytophthora infestans, the causal agent of late blight. Alternatives to the current control strategies are needed, as these rely heavily on environmentally harmful treatments. The recruitment of beneficial microbes by plants upon stress ("cry-for-help" mechanism) may represent an opportunity to find new biocontrol agents but this has not yet been reported for potato. The aim of this study was to analyse whether foliar late blight infection induces shifts in the phyllosphere, rhizosphere and soil ba

Inhibition of Phytophthora infestans chitin synthase via cyclic peptide targeting for sustainable disease control.

Phytophthora spp. are devastating plant pathogens causing significant economic losses in agriculture, but unlike true fungi, oomycetes have cell walls composed predominantly of cellulose and glucans, with little to no detectable chitin. Despite this, one to two putative chitin synthase (Chs) genes are present in Phytophthora spp. suggesting a functional role in its biology. In this study, we characterized the single chitin synthase of P. infestans (PiChs) to better understand its function and explore its potential as a target for novel fungicidal treatments. Unlike typical chitin synthases tha

Structure guided development of phenoxyethanol based N-aryloxyethyl heterocycles for the control of Phytophthora infestans.

BACKGROUND: Late blight caused by the oomycete Phytophthora infestans remains one of the most devastating diseases threatening global potato and tomato production. Overuse of a narrow range of fungicides has sped up resistance, making it urgent to develop new agents that work differently and are safer. RESULTS: In this study, a series of N-aryloxyethyl heterocycles were rationally designed by integrating the aryloxyethyl scaffold of phenoxyethanol with bioactive heterocyclic frameworks. Among the 40 synthesized derivatives, compound 8c exhibited the highest anti-oomycete activity against P. i

The effect of nitrosative stress on histone H3 and H4 acetylation in Phytophthora infestans life cycle.

The oomycete Phytophthora infestans is one of the most destructive phytopathogens globally. It has a proven ability to adapt to changing environments rapidly; however, molecular mechanisms responsible for host invasion and adaptation to new environmental conditions still need to be explored. The study aims to understand the epigenetic mechanisms exploited by P. infestans in response to nitrosative stress conditions created by the (micro)environment and the host plant. To characterize reactive nitrogen species (RNS)-dependent acetylation profiles in avirulent/virulent (avr/vr) P. infestans, a t
