Comprehensive screening of antimicrobials to control phytoplasma diseases using an in vitro plant-phytoplasma co-culture system.

Phytoplasmas are plant-pathogenic bacteria that infect many important crops and cause serious economic losses worldwide. However, owing to an inability to culture phytoplasmas, screening of antimicrobials on media is difficult. The only antimicrobials being used to control phytoplasmas are tetracycline-class antibiotics. In this study, we developed an accurate and efficient screening method to evaluate the effects of antimicrobials using an in vitro plant-phytoplasma co-culture system. We tested 40 antimicrobials, in addition to tetracycline, and four of these (doxycycline, chloramphenicol, th

'Candidatus Phytoplasma periplocae' sp. nov. associated with Periploca aphylla witches' broom disease.

Periploca aphylla witches' broom (PapWB) is a phytoplasma-associated disease first reported in south-western Iran in 2010. Recent observations showed that the disease is spreading to new areas. PapWB phytoplasmas, detected in symptomatic P. aphylla plants, exhibited identical nucleotide sequences in the 1.8 kb PCR amplicon encompassing the 16S rRNA gene, the 16S-23S spacer region and the 5'-end of the 23S rRNA gene. When compared with the reference strains of previously described 'Candidatus Phytoplasma' species, the selected representative strain PapWB-S exhibited the highest 16S rRNA gene se

Containment of Phytoplasma-Associated Plant Diseases by Antibiotics and Other Antimicrobial Molecules.

Phytoplasmas are plant-pathogenic bacteria that infect many important crops and environmentally relevant plant species, causing serious economic and environmental losses worldwide. These bacteria, lacking a cell wall, are sensitive to antibiotics such as tetracyclines that affect protein synthesis mechanisms. Phytoplasma cultivation in axenic media has not been achieved for many strains; thus, the screening of antimicrobials must be performed using mainly in vivo materials. Some studies have investigated using in vitro phytoplasma-infected shoots, and several antimicrobials, including tetracyc

First Report of 'Candidatus Phytoplasma mali,' the Causal Agent of Apple Proliferation Disease, in Apple Trees in Finland.

Based on an earlier survey of putative psyllid vectors of apple proliferation (AP), carried out in 2009 and 2010, Cacopsylla picta (Förster) populations infected with 'Candidatus Phytoplasma mali' were detected in at least two commercial apple (Malus domestica Borkh.) orchards in southern Finland (1). To establish the presence of 'Ca. P. mali' in apple trees, a survey was conducted in 17 commercial apple orchards in August 2012. Phytosanitary inspectors tracked the source of the 'Ca. P. mali' by collecting 33 leaf samples from trees showing probable symptoms. Typical symptoms, including elonga

Protective netting as a barrier against the X-disease phytoplasma vector Colladonus reductus (Hemiptera: Cicadellidae) in cherry orchards.

X-disease, caused by "Candidatus Phytoplasma pruni," poses a significant economic threat to the stone fruit industry, causing the removal of vast cherry acreage in Washington State. The most abundant vector in these orchards is the leafhopper Colladonus reductus (Van Duzee). Growers need season-long preventative alternatives to insecticides for a robust integrated pest management program. Thus, this study evaluated the efficacy of protective netting as a barrier to C. reductus. Between 2024 and 2025, 3 field trials were conducted testing 4 types of netting (20% shade, 30% shade, Mesh 25, and M

First Report of Witches'-Broom Disease in a Cannabis spp. in China and Its Association with a Phytoplasma of Elm Yellows Group (16SrV).

Hemp fiber plants (Cannabis spp.) spread naturally in almost every climate zone in China and have a long history of cultivation in the country (1). While hemp stalks provide high-quality fibers for making ropes, clothes, and paper products, hemp seeds are a rich source of edible oil. During the summer of 2004, a disease characterized by witches'-broom symptoms was observed in wild hemp fiber plants growing in suburban Taian, Shandong, China. The diseased plants developed clusters of highly proliferating branches with much shortened internodes and leaves on the affected branches were significan

Potato purple top phytoplasma infection induces autophagy-associated lipid dynamics that support pathogen proliferation.

Phytoplasmas are unculturable, phloem-restricted bacterial pathogens responsible for devastating diseases in crops and ornamentals worldwide. Their mechanism for nutrient acquisition from host plants remains largely unknown. This study demonstrated that infection with potato purple top phytoplasma induced extensive remodeling of lipid metabolism in tomato plants, closely linked to autophagy activation. Western blot and confocal analyses revealed increased ATG8 lipidation and autophagosome formation at endoplasmic reticulum stress sites, alongside the redistribution of lipid droplets toward phy

Phytoplasma-plant interactions: effector-mediated host reprogramming, hormonal crosstalk, metabolic alterations and plant-mediated vector manipulation.

Phytoplasmas are wall-less, phloem-restricted bacterial pathogens that infect over 1,000 plant species, causing substantial losses in agriculture, horticulture, and forestry worldwide. Despite their reduced genomes and limited metabolic autonomy, these obligate parasites colonize diverse hosts through secreted effector proteins that extensively reprogram plant development, metabolism, immune signalling, and vector interactions. Advances in genomics, transcriptomics, proteomics, metabolomics, and functional studies have substantially clarified the molecular basis of phytoplasma pathogenicity an

First Cophylogenetic Reconstruction of Hemipteran Insect-Phytoplasma Associations Reveals Eco-Evolutionary Dynamics.

Vector-borne plant pathogens exhibit a remarkable capacity to colonize distantly related hosts, reflecting long-term ecological and evolutionary interactions among pathogens, plants, and insect vectors. Phytoplasmas, which are obligate bacterial parasites transmitted by hemipteran insects, represent a model system for investigating how host-pathogen associations diversify across ecological and evolutionary timescales. Despite extensive knowledge of phytoplasma diversity and vector relationships, the historical processes shaping these associations remain poorly understood. Here, we investigated

'Candidatus Phytoplasma zeae': community-driven species delineation of the maize bushy stunt phytoplasma, a Dalbulus-transmitted corn pathogen confined to the Americas.

A novel phytoplasma species, 'Candidatus Phytoplasma zeae', is proposed based on ecological distinctiveness, vector specificity, whole-genome comparisons and community consensus. This phytoplasma is associated with maize bushy stunt (MBS) disease in corn (Zea mays) and is transmitted exclusively by Dalbulus maidis and Dalbulus elimatus, two leafhopper species endemic to the Americas that has been reported in Brazil, Colombia, Mexico, Peru and several U.S. states. Here, we sequenced and assembled the genome of MBS phytoplasma (MBSP) strains from Brazil and the USA to describe and propose this n

The ecological dynamics and consequences of phytoplasma infection in white trillium.

BACKGROUND AND AIMS: Ecologists have long sought to link within-population dynamics to patterns of species occurrence and persistence across the range. Theoretical models suggest pathogens have important implications for plant populations and perhaps even limit plant distributions, though more empirical evidence is needed, especially in wild plant populations. METHODS: In the field, we characterized the infection dynamics of phytoplasma, a vectored and sterilizing bacterial pathogen, in wild populations of white trillium. We identified the highest molecular matches for the local bacterial str

Host-driven structuring shapes the landscape of phytoplasma diversity across Indian agroecosystems revealed by multilocus phylogenomics.

Phytoplasmas are phloem-limited, wall-less plant pathogens that impose significant biotic stress on host plants, resulting in substantial yield losses worldwide. However, their genetic diversity, host associations, and population structure in tropical agroecosystems remain poorly understood. This study presents a nationwide multilocus assessment of 'Ca. Phytoplasma' diversity, distribution, and population structure across Indian agroecosystems. A survey (2020-2023) across ten states identified 124 phytoplasma-positive samples from 42 host species and associated leafhopper insect vectors. Multi

Transcriptomics-based analysis using RNA-Seq of the coconut (Cocos nucifera) leaf in response to yellow decline phytoplasma infection.

Invasive phytoplasmas wreak havoc on coconut palms worldwide, leading to high loss of income, food insecurity and extreme poverty of farmers in producing countries. Phytoplasmas as strictly biotrophic insect-transmitted bacterial pathogens instigate distinct changes in developmental processes and defence responses of the infected plants and manipulate plants to their own advantage; however, little is known about the cellular and molecular mechanisms underlying host-phytoplasma interactions. Further, phytoplasma-mediated transcriptional alterations in coconut palm genes have not yet been identi
