Effects of Heavy Metals on the Metabolome of Pinus sylvestris (Pinaceae).

The effects of Cu, Ni, and Cd on the Pinus sylvestris metabolome was studied in experimental conditions by gas chromatography-mass spectrometry (GC-MS). Structural changes in plant metabolite network became detectable on day 6 of exposure to the metals, 3-6 days earlier than visual signs of toxicity developed. Differences at the metabolome level arose earlier in a control group of plants, and specific effects of particular metals on the plant metabolome became distinct on day 9. Both nature and concentration of a metal equally contributed to the plant metabolome clustering. Plant responses (ch

Spatial pattern of allozyme variation in a contact zone of Pinus Ponderosa and P. arizonica (Pinaceae).

The spatial distribution of genotypes for nine polymorphic allozyme loci was examined in a contact zone between Pinus ponderosa var. scopulorum and another tree regarded as either a separate species, Pinus arizonica, or variety, Pinus ponderosa var. arizonica, in southern Arizona. Previous work had identified a steep elevational cline for a key taxonomic trait, number of leaf-needles per fascicle, on the south slope of Mt. Lemmon. The present results indicate that the taxa are not fully interbreeding in this contact zone, because allozyme genotypes are considerably more spatially structured th

Anthelmintic evaluation of triterpenes derived from conifers (family Pinaceae) on the digenean parasite Fasciola hepatica.

Fasciolosis is a devastating food borne parasitic disease of ruminants and is mainly caused by infection with the trematode Fasciola hepatica (liver fluke) in temperate climates. The primary strategy of fasciolosis control relies on a single drug, triclabendazole, although nitroxinil, albendazole, closantel, rafoxanide and clorsulon can also be used to treat chronic infections. Unfortunately, growing evidence indicates that triclabendazole resistant liver fluke populations are pervasive. Therefore, identifying new chemical matter with anthelmintic properties as starting points for drug develop

Dynamic Testing of Lime-Tree (Tilia Europoea) and Pine (Pinaceae) for Wood Model Identification.

The paper presents the results of dynamic testing of two wood species: lime-tree (Tilia europoea) and pine (Pinaceae). The dynamic compressive tests were carried out using the traditional Kolsky method in compression tests. The Kolsky method was modified for testing the specimen in a rigid limiting holder. In the first case, stress-strain diagrams for uniaxial stress state were obtained, while in the second, for uniaxial deformation. To create the load a gas gun was used. According to the results of the experiments, dynamic stress-strain diagrams were obtained. The limiting strength and deform

Clinal differentiation and putative hybridization in a contact zone of Pinus ponderosa and P. arizonica (Pinaceae).

The widely distributed Pinus subsection Ponderosae is a species complex that has a transition zone among taxa in the southwestern United States. In southern Arizona and New Mexico at least two recognized taxa, Pinus ponderosa var. scopulorum and Pinus arizonica or P. ponderosa var. arizonica, are known to coexist in close proximity. In this study, we report the existence of populations where the taxa are sympatric. One of the key characteristics distinguishing taxa is the number of needles per fascicle; P. ponderosa typically has three, P. arizonica has five. We examined the spatial distributi

Expansion of the dehydrin gene family in the Pinaceae is associated with considerable structural diversity and drought-responsive expression.

Temperatures are expected to increase over the next century in all terrestrial biomes and particularly in boreal forests, where drought-induced mortality has been predicted to rise. Genomics research is helping to develop hypotheses regarding the molecular basis of drought tolerance and recent work proposed that the osmo-protecting dehydrin proteins have undergone a clade-specific expansion in the Pinaceae, a major group of conifer trees. The objectives of this study were to identify all of the putative members of the gene family, trace their evolutionary origin, examine their structural diver

Impact of Hemlock Woolly Adelgid (Hemiptera: Adelgidae) Infestation on the Jasmonic Acid-Elicited Defenses of Tsuga canadensis (Pinales: Pinaceae).

Hemlock woolly adelgid is an invasive piercing-sucking insect in eastern North America, which upon infestation of its main host, eastern hemlock ('hemlock'), improves attraction and performance of folivorous insects on hemlock. This increased performance may be mediated by hemlock woolly adelgid feeding causing antagonism between the the jasmonic acid and other hormone pathways. In a common garden experiments using hemlock woolly adelgid infestation and induction with methyl jasmonate (MeJA) and measures of secondary metabolite contents and defense-associated enzyme activities, we explored the

Comparative genome mapping among Picea glauca, P. mariana x P. rubens and P. abies, and correspondence with other Pinaceae.

A composite linkage map was constructed from four individual maps for the conifer Picea glauca (Moench) Voss, from anonymous and gene-specific markfers (714 AFLPs, 38 SSRs, and 53 ESTPs). A total of 12 linkage groups were delineated with an average marker density of 2.7 cM. Macro-synteny and macro-colinearity comparisons with two other composite linkage maps developed for the species complex P. mariana (Mill.) B.S.P. x P. rubens Sarg., and for P. abies (L.) Karst. revealed an identical number of linkage groups and a remarkable conservation of the gene content and gene order of linkage groups o

Geographical Distribution and Long-Term Monitoring of Physokermes hellenicus (Hemiptera: Coccomorpha: Coccidae) on Abies spp. (Pinales: Pinaceae) in Greece.

The scale Physokermes hellenicus (Kozár & Gounari) (Hemiptera: Coccidae) has been recently included in the Greek entomofauna as a beneficial honeydew species. However, there are no adequate data about its geographical distribution and degree of infestation. Therefore, a study was conducted to examine these parameters in fifteen mountains of Greece. Furthermore, the monitoring of P. hellenicus infestation was carried out over a six-year period with regard to natural enemies and honeydew presence at three mountains (i.e., Menalon, Parnis and Tymfristos) that are traditional honeybee foraging are

Use of Pinus sylvestris L. (Pinaceae), Origanum vulgare L. (Lamiaceae), and Thymus vulgaris L. (Lamiaceae) essential oils and their main components to enhance itraconazole activity against azole susceptible/not-susceptible Cryptococcus neoformans strains.

BACKGROUND: Cryptococcal infections, besides being a problem for immunocompromised patients, are occasionally being a problem for immunocompetent patients. In addition, the lower susceptibility of this yeast to azoles is a growing problem in health care. To date, there are very few molecules with any activity towards Cryptococcus neoformans, leading to heightened interest in finding new alternatives or adjuvants to conventional drugs for the treatment of mycosis caused by this yeast. Since the essential oils (EOs) are considered as a potential rich source of bioactive antimicrobial compounds,

Oviposition strategies of conifer seed chalcids in relation to host phenology.

Insects are considered the most important predators of seed cones, the female reproductive structures of conifers, prior to seed dispersal. Slightly more than 100 genera of insects are known to parasitize conifer seed cones. The most diverse (i.e., number of species) of these genera is Megastigmus (Hymenoptera: Torymidae), which comprises many important seed pests of native and exotic conifers. Seed chalcids, Megastigmus spp., lay eggs inside the developing ovules of host conifers and, until recently, oviposition was believed to occur only in fertilized ovules. Ovule development begins just af

Haploid, diploid, and pooled exome capture recapitulate features of biology and paralogy in two non-model tree species.

Despite their suitability for studying evolution, many conifer species have large and repetitive giga-genomes (16-31 Gbp) that create hurdles to producing high coverage SNP data sets that capture diversity from across the entirety of the genome. Due in part to multiple ancient whole genome duplication events, gene family expansion and subsequent evolution within Pinaceae, false diversity from the misalignment of paralog copies creates further challenges in accurately and reproducibly inferring evolutionary history from sequence data. Here, we leverage the cost-saving benefits of pool-seq and e

Differences in the Chloroplast Genome and Its Regulatory Network among Cathaya argyrophylla Populations from Different Locations in China.

Cathaya argyrophylla Chun et Kuang is a severely endangered, tertiary relict plant unique to China whose high physiological sensitivity to the environment, including photosensitivity, is likely closely related to its endangered status; however, the exact mechanism responsible has remained unknown due to the rarity of the plant and the difficulties involved in performing physiological studies on the molecular level. In this study, the chloroplast genomes of six C. argyrophylla populations sampled from different locations in China were characterized and compared. In addition, a gene regulatory n

Phytochemical investigation of the needles of Abies nebrodensis (Lojac.) Mattei.

Abies nebrodensis (Lojac.) Mattei (Pinaceae) is a species living in a very small population only in a confined area of Sicily. In this study, the dichloromethane extract of the leaves was analyzed. Apart from three already known metabolites namely dehydroabietic acid; maltol; and rheosmin, previously detected in other species of Abies, a lanostane derivative was isolated. Its chemical structure was elucidated by means of extensive spectroscopic methods.

Docking in the Abies grandis abietaenol synthase to investigate the mechanism underlying conifer resin acid diterpene synthase evolution.

The pine family (Pinaceae) produces oleoresins in which the diterpenoid resin acids contain tricyclic backbones of either the (iso)pimarane and/or rearranged abietane type. These are produced by closely related (class I) diterpene synthases, whose functional divergence is of significant interest, not least for mechanistic insights into the catalyzed complex carbocation-cascade reactions. The abietaenol synthase from Abies grandis (AgAS) has long served as a model for this enzymatic subfamily and was used here to examine key residues potentially involved in transitions from such ancestral activ

[Storage proteins from seeds of Pinus pinea L].

The Mediterranean stone pine Pinus pinea L. (gymnosperm, Pinaceae) is much appreciated for its seed production, widely used in food preparation in the Mediterranean Basin. Seeds contain 25% proteins on a dry-weight basis. Pinus pinea accumulate globulins as major storage proteins in seeds (75% of total storage proteins), composed of several subunits of 10 to 150 kDa, revealed by SDS-PAGE. The albumin fraction (15%) represents three subunits of 14, 24 and 46 kDa. Glutelins, the least soluble fraction, represents a small proportion (10%). Their constitutive units have frequent PM of 43 kDa. Prol

Cellulase adsorption and an evaluation of enzyme recycle during hydrolysis of steam-exploded softwood residues.

The sugar yield and enzyme adsorption profile obtained during the hydrolysis of SO2-catalyzed steam-exploded Douglas-fir and posttreated steam-exploded Douglas-fir substrates were determined. After hot alkali peroxide posttreatment, the rates and yield of hydrolysis attained from the posttreated Douglas-fir were significantly higher, even at lower enzyme loadings, than those obtained with the corresponding steam-exploded Douglas-fir. The enzymatic adsorption profiles observed during hydrolysis of the two substrates were significantly different. Ultrafiltration was employed to recover enzyme in
