Pinealectomy in Rats.

The pinealectomy technique consists of the surgical removal of the superficial pineal gland. This procedure allows the ablation of circulating indoles produced by this gland. Withdrawal of systemic melatonin, a pineal hormone, affects animal circadian rhythms and induces several physiological changes that are the subject of many investigations. In this chapter, we describe the pinealectomy protocol adapted to rats. We describe the animal placement on the stereotaxic fixation system, and the procedure for the pineal gland removal and animal recovery from surgery.

Surgical approach for pinealectomy in the calf.

Pinealectomy was performed on 7 calves, and a sham operation was performed on 3 calves. Two pinealectomized calves died immediately. The 5 remaining pinealectomized and 3 sham-operated control calves are presently involved in experimental studies. The surgical technique for exposure of the pineal gland was a rectangular (rostrotentorial) craniectomy between the median line and the horn bud. Removal of portions of the frontal and parietal bones, measuring 6 cm by 4 cm, provided adequate exposure. Positioning the head was important in order to gain the best access to the pineal gland. Modulation of estrous cycle and LH, FSH and melatonin levels by pinealectomy and sham-pinealectomy in female rats.

1. The pineal has been shown to have a role in controlling reproduction of polyestrus mammals (like humans and laboratory rodents). It influences the age of sexual maturation; the timing of the ovulatory cycle; and gonadal steroidogenesis. 2. Here the authors report the early and late effects of pinealectomy (Px) and sham-pinealectomy (SPx) on the estrous cycle periodicity, plasma LH, FSH and urinary 6-sulphatoxymelatonin (6-SMT) excretion in female rats. 3. Female Wistar rats (3-4 months of age) were maintained on 12/12 L/D cycle. Orbital venous plexus blood and urine samples were collected f

Pinealectomy increases thermogenesis and decreases lipogenesis.

The present study was designed to determine the effects of pineal gland‑derived melatonin on obesity by employing a rat pinealectomy (Pnx) model. After 10 weeks of a high‑fat diet, rats received sham or Pnx surgery followed by a normal chow diet for 10 weeks. Reverse transcription‑quantitative PCR, western blotting analysis, immunohistochemistry and ELISA were used to determine the effects of Pnx. Pnx decreased the expression of melatonin receptor (MTNR)1A and MTNR1B, in brown adipose tissues (BAT) and white adipose tissues (WAT). Pnx rats showed increased insulin sensitivity compared with tho

Pinealectomy aggravates acute pancreatitis in the rat.

Melatonin, a pineal indoleamine, protects the pancreas against acute damage; however, the involvement of the pineal gland in the pancreatoprotective action of melatonin is unknown. The primary aim of this study was to determine the effects of pinealectomy on the course of acute caerulein-induced pancreatitis (AP) in rats. AP was induced by a subcutaneous infusion of caerulein (25 μg/kg) into pinealectomized or sham-operated animals. Melatonin (5 or 25 mg/kg) was given via intraperitoneal (ip) injection 30 min prior to the induction of AP. The pancreatic content of the lipid peroxidation produc

Pinealectomy in the chicken: a good model of scoliosis?

The phenomenon of spinal deformity in the pinealectomized chicken has led researchers to postulate a disturbance of melatonin activity as a potential cause of adolescent idiopathic scoliosis (AIS). More recently, structural differences between curves seen in this model and those seen in scoliosis have been highlighted suggesting the deformities observed are not as similar as first thought. We examined melatonin levels, and the radiological and histological characteristics of scoliosis after pinealectomy in chickens. They underwent pinealectomy (P) at 2 days of age, sham surgery (S) or served a

Effect of pinealectomy on cortically kindled rats.

Cortical kindling in pinealectomized rats was studied to observe the effect of pinealectomy on local afterdischarge thresholds and seizure development. Although the local afterdischarge thresholds were not affected by pinealectomy, the kindling process itself was hastened. These results suggest that the anticonvulsive function of the pineal body is due not to a decrease in local afterdischarge thresholds, but to retardation of secondary generalization.

A surgical modification in the technique of rat pinealectomy.

BACKGROUND: Several experimental intents require pineal gland removal. The main challenge of the pinealectomy surgical procedure is the hemorrhage due to the transverse sinus torn. The study aimed to modify the rat pinealectomy surgical procedure to reduce the risk of bleeding and the mortality rate. METHODS: Adult male rats experienced pinealectomy surgery. A mini-drill was used to remove a small skull area in the junction of the lambda and sagittal sutures. The pineal gland was removed using a curved-head hook. Animals experienced intensive post-surgical care. Locomotion, cerebellar motor f

Effect of pinealectomy on circadian rhythm of spermatogenesis.

The mitotic index of spermatogonia and 24-h dynamics of stages IV, VI, and XIV of spermatogenic cycle are characterized by circadian rhythm. No circadian rhythm was detected for 11 of 14 stages. Pinealectomy led to an increase of the mitotic index of spermatogonia but did not modulate the incidence of spermatogenic cycle stages, and led to disappearance of the circadian rhythm of both the mitotic index and spermatogenic cycle stages.

Rat pinealectomy: a modified direct visual approach.

PURPOSE: To report a new, direct visual approach for rat pinealectomy. METHODS: Eighty adult female rats (Rattus norvegicus albinus EPM-1 strain) were weighted and anesthetized intraperitoneally with 15 mg/kg xylazine and 30 mg/kg ketamine. The animal was fastened to a dissection table, an incision was made in the skin and the subcutaneous tissue, bringing the lambda into view. The skullcap was opened with a dental drill, bringing the cerebral hemispheres and the superior sagittal sinus into view. The pineal gland, located under the venous sinus, was removed in a single piece using tweezers.

Pinealectomy-Induced Neuroinflammation Varies with Age in Rats.

It is widely accepted that chronic inflammation constitutes a significant mechanism that promotes the biological aging process. The pineal gland is regarded as being closely related to the control of the "life clock". The present study aimed to determine the inflammation associated with pinealectomy in the rat hippocampus and to investigate the extent to which age stage impacts the severity of this inflammation. We evaluated the expression of the Akt/NF-kB signaling pathway in neurons and gliosis level in the dorsal hippocampus (dHipp) of rats subjected to sham surgery or pinealectomy at 3, 14

Pinealectomy inhibits antioxidant system in rats with hyperthyroidism.

OBJECTIVE: Thyroid hormones regulate energy metabolism and act on the mitochondria, which is an important source of free radicals in the cell. Reactive oxygen types play a significant role in physiological mechanisms, but in excessive amounts they can cause oxidative damage in molecules. The aim of the present study was to determine levels of lipid peroxidation caused by induced hyperthyroidism in cerebral, hepatic and cardiac tissues of pinealectomized rats. METHODS: Experimental animals used in the study were allocated to three groups as general control group, hyperthyroidism-sham pinealect

An improved technique for visually controlled pinealectomy in the rat.

An improved method for pinealectomizing rats and other small laboratory rodents is described which avoids bleeding and allows excision of the organ under exact visual control. After removing a small piece of the skull dorsal to the confluence of the transverse and dorsal sagittal sinuses, the latter is ligated and cut. The underlying pineal gland is then exposed and may be excised reliably without damage to the gland tissue or to adjacent brain structures. Intra- and post-operative complications are extremely rare.

Inflammatory response modulated by pinealectomy: effect of light.

In view of the known interactions between pineal and adrenal glands and the importance of glucocorticoids on the acute phase of inflammatory process, we investigated the effect of the pineal gland on the paw edema response induced by carrageenin in female Wistar rats, weighing 170-220 g, measured by the pletysmographic method. Adrenalectomy increased the rate of the inflammatory response (149.02 +/- 2.93%, N = 7 vs 113.48 +/- 2.99%, N = 7) whereas pinealectomy and sham-pinealectomy under white light had an inhibitory effect (81.73 +/- 2.16%, N = 15 and 80.21 +/- 1.85%, N = 14 vs 113.48 +/- 2.9

Noradrenergic involvement in pinealectomy induced convulsions in gerbils.

Gerbils treated with various noradrenergic compounds were pinealectomized (PINX) or sham-operated (SHAM) and observed for convulsive activity. Although declines in cortical and hypothalamic norepinephrine content were found after alpha-methylparatyrosine (alpha MPT) or PINX, treatment with alpha MPT did not produce convulsions in SHAM gerbils or in gerbils which were PINX one week earlier. Likewise, alpha MPT did not increase the number or severity of convulsions observed in acutely PINX gerbils. Treatment with propranolol or isoproterenol did not have any effect on convulsive activity of PINX

Influence of pinealectomy on the amygdala kindling development in rats.

A considerable number of studies have demonstrated the anticonvulsant effect of melatonin. The present study examines the influence of pinealectomy on the amygdala kindling development in rats. Animals were divided into three groups: Pinealectomized rats (PNT) Sham rats (SHAM) and Control rats (CTL). The mean number of stimulations and the afterdischarge (AD) duration for each stage of kindling were similar among CTL and SHAM animals. Conversely, PNT animals showed particular characteristics during kindling development. They did not present stage 1 and spent a shorter time in stage 2 in relati
