Piscirickettsia salmonis, the etiological agent of piscirickettsiosis, represents the main health challenge for Chilean salmon farming and an emerging threat to global salmonid production. Inspection of all P. salmonis genomes available shows that the comEC gene, encoding the DNA uptake channel required for natural transformation (NT) in competent bacteria, is interrupted by various transposable elements, whereas other NT determinants (comEA, comFB, comM, comL/bamD, and dprA) remain intact and highly conserved. Using P. salmonis Psal-103 (EM-genogroup) and Psal-104b (LF-genogroup) as represent

Piscirickettsiosis or salmonid rickettsial septicaemia (SRS) caused by Piscirickettsia salmonis constitutes one of the main problems in farmed salmonid and marine fishes. Since the first reports of the disease, it has been successfully isolated and maintained in eukaryotic cell--culture systems, but these systems are time-consuming, the media are costly, and eliminating heavily contaminated host cell debris is difficult. In this report, we describe a marine-based broth supplemented with L-cysteine, named AUSTRAL-SRS broth, that facilitates superior growth of P. salmonis strains. Strains reache

Research on Piscirickettsia salmonis, a critical pathogen affecting salmon aquaculture, has been hampered by inadequate host models to study the infection process in vivo. Here, we highlight zebrafish as a surrogate host model for in-depth analysis of P. salmonis infection by using fluorescent dyes and protein markers to visualize host-pathogen interactions. Using transgenic zebrafish lines with fluorescent immune cells, we observed a strong increase in neutrophil numbers in the infected larvae, resembling an emergency granulopoiesis response. We observed the presence of P. salmonis in the cau

Caligidosis and Piscirickettsiosis are currently the most important sanitary challenges for the Chilean salmon industry. Caligidosis is caused by the ectoparasite, Caligus rogercresseyi and Piscirickettsiosis is caused by the intracellular bacterium, Piscirickettsia salmonis. Both diseases are highly prevalent and widely distributed in farming areas in Chile. The co-occurrence of the two diseases is frequently reported on salmon farms. However, there is little epidemiological evidence as to whether these two diseases are associated and generate interactive effects. This study was undertaken to

Mortality among hatchery-reared juvenile white seabass Atractoscion nobilis in southern California, USA, was associated with infections by a Piscirickettsia salmonis-like organism (WSPSLO). Infected fish had no consistent external signs other than pale gills, lethargy and impaired swimming behavior. Internally, the kidney and spleen were enlarged, and some fish had livers with multiple pale foci. Smears from infected kidney, liver, and spleen stained with Wright-Giemsa had intracytoplasmic coccoid organisms, often in pairs, that ranged in size from 0.5 to 1.0 microm. Microscopic lesions includ

The composition and structure of lipid A isolated from the lipopolysaccharide (LPS) of Piscirickettsia salmonis were investigated by chemical analyses, gas chromatography/mass spectrometry (GCMS), and electrospray ionization (ESI) combined with the tandem mass spectrometry (MS/MS). Our study revealed moderate compositional and structural heterogeneity of lipid A with respect to the content of phosphate groups and 4-amino-4-deoxy-L-arabinopyranose (Ara4N) residues and with regard to the degree of acylation. It appeared that at least two molecular species were present in lipid A. The major speci

An intracellular bacterium originally isolated from hatchery-reared juvenile white seabass Atractoscion nobilis in southern California, USA, was identified by sequences of the small and large subunit ribosomal (16S and 23S) DNA and the internal transcribed spacer (ITS) as Piscirickettsia salmonis. Considering all rDNA sequences compared, the white seabass isolate (WSB-98) had a 96.3 to 98.7% homology with 4 previously described strains of P. salmonis isolated from salmon in Chile, Norway, and British Columbia, Canada. Experimental infections induced by intraperitoneal injections of juvenile wh

BACKGROUND: Vertebrate hosts limit the availability of iron to microbial pathogens in order to nutritionally starve the invaders. The impact of iron deficiency induced by the iron chelator deferoxamine mesylate (DFO) was investigated in Atlantic salmon SHK-1 cells infected with the facultative intracellular bacterium Piscirickettsia salmonis.
RESULTS: Effects of the DFO treatment and P. salmonis on SHK-1 cells were gaged by assessing cytopathic effects, bacterial load and activity, and gene expression profiles of eight immune biomarkers at 4- and 7-days post infection (dpi) in the control gro

Piscirickettsia salmonis is a facultative intracellular bacterium that generates piscirickettsiosis affecting salmonids in Chile. The bacterium has the adaptability to survive in the marine environment under multiple stressful conditions. In this sense, this work focused on the analysis of a gene battery associated with biofilm formation under different culture conditions and on the adaptability of this biofilm to different media. The results indicated that the strains LF-89, IBM-034 and IBM-040 were strong biofilm producers, evidencing adaptability to the media by increasing the amount of bio

Antibiotic resistance is a major public health concern due to its association with the loss of efficacy of antimicrobial therapies. Horizontal transfer events may play a significant role in the dissemination of resistant bacterial phenotypes, being mobilizable plasmids a well-known mechanism. In this study, we aimed to gain insights into the genetics underlying the development of antibiotic resistance by Piscirickettsia salmonis isolates, a bacterial fish pathogen and causative agent of salmonid piscirickettsiosis, and the main target of antibiotics used in Chilean salmon farming. We provide e

Piscirickettsia salmonis is an intracellular bacterium responsible for Salmonid Rickettsial Septicemia (SRS) in Atlantic salmon. This pathogen survives within macrophages, hindering immune clearance. In mammals, CD8+ T lymphocytes represent a crucial component of adaptive immunity, as they specifically recognize and eliminate cells infected by intracellular pathogens. Evidence indicates that an analogous mechanism may also operate in teleost fish. Here, the role of CD8+ T lymphocytes in the immune response against P. salmonis was investigated in Atlantic salmon (Salmo salar). Using autologous

Vaccine-based approaches provide a sustainable method for controlling pathogens in aquaculture. Our team previously created the IPath® vaccine, a recombinant formulation with metal-chelating properties. This study aimed to evaluate the performance of IPath® in salmon vaccinated with the mandatory vaccines used commercially in Chile, and to assess their effects on the transcriptome profiles of Atlantic salmon challenged with Caligus rogercresseyi and co-infected with Piscirickettsia salmonis. Four experimental groups of Atlantic salmon were immunized: IPath®, BlueGuard® (B) + Alpha Ject LiVac®

Piscirickettsia salmonis is the etiological agent of Piscirickettsiosis, a severe disease that affects Atlantic salmon (Salmo salar) farmed in Chile and many other areas (Norway, Scotland, Ireland, Canada and the USA). This study investigated the effects of low-dose P. salmonis infection (1 × 102 CFU/ml) on Atlantic salmon. In this study, we challenged fish with an isolated representative of the EM-90 genogroup via intraperitoneal injection for 42 days. Infected fish displayed decreased haematocrit and haemoglobin levels at day 13 post-infection, indicating erythropenia, haemolysis and haemod

Ferritin is a major iron storage protein essential not only in the infectious process, but also in any circumstance generating oxidative stress. In this study, the cDNA coding sequence of ferritin-H was obtained from the sub-Antarctic Notothenioid fish Eleginops maclovinus through transcriptomic analysis of the head kidney. This sequence contained a 534 bp open reading frame that coded for a 177 amino acid protein with a molecular weight of 20,786.2 Da and a theoretical pI of 5.56. The protein displayed a region of iron putative response elements in the 5'UTR, two putative ferritin iron-bindin

During the winter of 2013 and 2016, several Croatian fish farms experienced mortalities in the fry of European sea bass, Dicentrarchus labrax. Affected fish showed abnormal swimming behaviour and reduced appetite, and death ensued several days after the onset of clinical signs of disease. Necropsy revealed pale liver, empty digestive tract, distended gall bladder, and hyperaemia and congestion of the meninges. Routine bacteriological examination tested negative, and virological examination ruled out nodavirus infection. Histological examination revealed multifocal necrosis and extensive inflam

Piscirickettsiosis is a fish disease caused by the facultative intracellular bacterium Piscirickettsia salmonis. This research aimed to study the immune response of rainbow trout (Oncorhynchus mykiss) during the first seawater farming stage upon infection with P. salmonis. Fish were challenged by immersion with P. salmonis type strain LF-89T (genogroup 1) and the field isolates Psal-103 (LF-89-like genotype or genogroup 3) and Psal-104 (EM-90-like genotype or genogroup 4). A group of fish was treated with sterile AUSTRAL-SRS (Salmon Rickettsial Septicemia) medium. This group and fish from the

Piscirickettsia salmonis is a fastidious intracellular pathogen responsible for high mortality rates in farmed salmonids, with serious economic consequences for the Chilean aquaculture industry. Oxytetracycline and florfenicol are the most frequently used antibiotics against P. salmonis, but routine use could contribute to drug resistance. This study identified differentiated florfenicol susceptibilities in two P. salmonis strains, LF-89 and AUSTRAL-005. The less susceptible isolate, AUSTRAL-005, also showed a high ethidium bromide efflux rate, indicating a higher activity of general efflux pu

The intraperitoneal route is favored for administration of inactivated and attenuated vaccines in Atlantic salmon. Nevertheless, the immune responses in the teleost peritoneal cavity (PerC) are still incompletely defined. In this study, we investigated the B cell responses after intraperitoneal Piscirickettsia salmonis (P. salmonis) challenge of Atlantic salmon, focusing on the local PerC response versus responses in the lymphatic organs: spleen and head kidney. We observed a major increase of leukocytes, total IgM antibody secreting cells (ASC), and P. salmonis-specific ASC in the PerC at 3-

Piscirickettsia salmonis is a well-established intracellular pathogen of farmed Atlantic salmon around the world. The bacterium has also been detected from a number of other hosts including lumpfish (Cyclopterus lumpus), seabass (Dicentrarchus labrax) and rainbow trout (Oncorhynchus mykiss). In February 2025, increased mortality was observed in a population of turbot, Scophthalmus maximus, farmed in Norway. Macroscopic signs of disease included stomach mucosa bleedings. Piscirickettsiosis was diagnosed based on histopathological findings including rickettsia-like inclusions immunostaining posi
