Caligidosis and Piscirickettsiosis are currently the most important sanitary challenges for the Chilean salmon industry. Caligidosis is caused by the ectoparasite, Caligus rogercresseyi and Piscirickettsiosis is caused by the intracellular bacterium, Piscirickettsia salmonis. Both diseases are highly prevalent and widely distributed in farming areas in Chile. The co-occurrence of the two diseases is frequently reported on salmon farms. However, there is little epidemiological evidence as to whether these two diseases are associated and generate interactive effects. This study was undertaken to

A novel approach has been developed for the simultaneous description of reaction kinetics to describe the formation of polysulfide and sulfate anions from the biological oxidation of hydrogen sulfide (H2S) using a quick, sulfide-dependent respiration test. Next to H2S, thiols are commonly present in sour gas streams. We investigated the inhibition mode and the corresponding inhibition constants of six thiols and the corresponding diorgano polysulfanes on the biological oxidation of H2S. A linear relationship was found between the calculated IC50 values and the lipophilicity of the inhibitors.

Aqueous-soluble hydrocarbons dissolve into the ocean's interior and structure deep-sea microbial populations influenced by natural oil seeps and spills. Among these hydrocarbons, n-pentane, is a seawater-soluble, volatile compound abundant in petroleum products and reservoirs, which partially partitions to the deep-water column following release from the seafloor. In this study, we explore the ecology and niche partitioning of two free-living Cycloclasticus strains recovered from seawater incubations with n-pentane and distinguish them as an open ocean variant and a seep-proximal variant, each

Piscirickettsia salmonis is a well-established intracellular pathogen of farmed Atlantic salmon around the world. The bacterium has also been detected from a number of other hosts including lumpfish (Cyclopterus lumpus), seabass (Dicentrarchus labrax) and rainbow trout (Oncorhynchus mykiss). In February 2025, increased mortality was observed in a population of turbot, Scophthalmus maximus, farmed in Norway. Macroscopic signs of disease included stomach mucosa bleedings. Piscirickettsiosis was diagnosed based on histopathological findings including rickettsia-like inclusions immunostaining posi

BACKGROUND: Nitrate (NO3 -) can accumulate in closed-circuit ecosystems to a toxic level. Adding heterotrophic denitrification process to the water treatment is a strategy to reduce this level. This type of process usually requires the addition of a carbon source. Carbon-to-nitrogen ratio (C/N) is a key parameter known to influence both the function and the activity of microbial communities in bioprocesses. Few studies have examined the influence of C/N on denitrification systems operated under methylotrophic and marine conditions. Here we assessed the influence of C/N (methanol and NO3 -) on

Ferritin is a major iron storage protein essential not only in the infectious process, but also in any circumstance generating oxidative stress. In this study, the cDNA coding sequence of ferritin-H was obtained from the sub-Antarctic Notothenioid fish Eleginops maclovinus through transcriptomic analysis of the head kidney. This sequence contained a 534 bp open reading frame that coded for a 177 amino acid protein with a molecular weight of 20,786.2 Da and a theoretical pI of 5.56. The protein displayed a region of iron putative response elements in the 5'UTR, two putative ferritin iron-bindin

Representative microbial cultures from an oil reservoir and electrochemical techniques including potentiodynamic scan and linear polarization were used to investigate the time dependent corrosion rate associated with control of biogenic sulphide production through addition of nitrite, nitrate and a combination of nitrate-reducing, sulphide-oxidizing bacteria (NR-SOB) and nitrate. The addition of nitrate alone did not prevent the biogenic production of sulphide but the produced sulphide was eventually oxidized and removed from the system. The addition of nitrate and NR-SOB had a similar effect

Salmonid Rickettsial Septicaemia (SRS) is a highly infectious, endemic disease that spreads among saltwater salmonid farms in Chile, making control efforts challenging. Here, we present a model that simulates SRS transmission between saltwater salmonid farms in the Aysén region of Chile, using the state-transition model framework InterSpread Plus (ISP). In ISP, the status (e.g., susceptible or infectious) of farms is individually defined, and the simulation determines the transition of the farms' state over time. Farm characteristics, such as fish species and production size, were incorporated

An intracellular bacterium originally isolated from hatchery-reared juvenile white seabass Atractoscion nobilis in southern California, USA, was identified by sequences of the small and large subunit ribosomal (16S and 23S) DNA and the internal transcribed spacer (ITS) as Piscirickettsia salmonis. Considering all rDNA sequences compared, the white seabass isolate (WSB-98) had a 96.3 to 98.7% homology with 4 previously described strains of P. salmonis isolated from salmon in Chile, Norway, and British Columbia, Canada. Experimental infections induced by intraperitoneal injections of juvenile wh

Phosphoribulokinase (PRK) catalyzes the ATP-dependent phosphorylation of ribulose 5-phosphate (Ru5P) to produce ribulose 1,5-bisphosphate in the Calvin-Benson-Bassham cycle. Of PRK's four classifications, class III consists of PRKs from pseudomonadota and α-cyanobacteria. They are known as octameric enzymes regulated allosterically by NADH. In this study, we characterized the PRK from a hydrogen- and sulfur-oxidizing chemolithoautotroph Hydrogenovibrio marinus MH-110 (HmPRK). Phylogenetic analysis revealed that PRKs from organisms of Piscirickettsiaceae, including H. marinus, formed a clearly

Autotrophic organisms fix dissolved inorganic carbon (DIC: CO2, HCO3-, CO3-2) into biomass, introducing organic carbon into the global carbon cycle. Many lineages of autotrophs developed CO2 concentrating mechanisms (CCMs) to grapple with the catalytic constraints of the carboxylase of the Calvin-Benson-Bassham cycle, ribulose-1,5-bisphosphate carboxylase/oxygenase (RubisCO). Carboxysomes and DIC transporters act together to form CCMs, which facilitate DIC fixation by autotrophic bacteria when DIC is scarce. Autotroph Thiomicrospira pelophila has carboxysomes, with an unusual carbonic anhydras

Piscirickettsia salmonis is the pathogen that most affects the salmon industry in Chile. Large quantities of antibiotics have been used to control it. In search of alternatives, we have developed [Cu(NN1)2]ClO4 where NN1 = 6-((quinolin-2-ylmethylene)amino)-2H-chromen-2-one. The antibacterial capacity of [Cu(NN1)2]ClO4 was determined. Subsequently, the effect of the administration of [Cu(NN1)2]ClO4 on the growth of S. salar, modulation of the immune system and the intestinal microbiota was studied. Finally, the ability to protect against a challenge with P. salmonis was evaluated. The results o

Piscirickettsia salmonis, the etiological agent of piscirickettsiosis, represents the main health challenge for Chilean salmon farming and an emerging threat to global salmonid production. Inspection of all P. salmonis genomes available shows that the comEC gene, encoding the DNA uptake channel required for natural transformation (NT) in competent bacteria, is interrupted by various transposable elements, whereas other NT determinants (comEA, comFB, comM, comL/bamD, and dprA) remain intact and highly conserved. Using P. salmonis Psal-103 (EM-genogroup) and Psal-104b (LF-genogroup) as represent

Vaccine-based approaches provide a sustainable method for controlling pathogens in aquaculture. Our team previously created the IPath® vaccine, a recombinant formulation with metal-chelating properties. This study aimed to evaluate the performance of IPath® in salmon vaccinated with the mandatory vaccines used commercially in Chile, and to assess their effects on the transcriptome profiles of Atlantic salmon challenged with Caligus rogercresseyi and co-infected with Piscirickettsia salmonis. Four experimental groups of Atlantic salmon were immunized: IPath®, BlueGuard® (B) + Alpha Ject LiVac®

Piscirickettsia salmonis is an intracellular bacterium responsible for Salmonid Rickettsial Septicemia (SRS) in Atlantic salmon. This pathogen survives within macrophages, hindering immune clearance. In mammals, CD8+ T lymphocytes represent a crucial component of adaptive immunity, as they specifically recognize and eliminate cells infected by intracellular pathogens. Evidence indicates that an analogous mechanism may also operate in teleost fish. Here, the role of CD8+ T lymphocytes in the immune response against P. salmonis was investigated in Atlantic salmon (Salmo salar). Using autologous

Piscirickettsia salmonis, the causal agent of salmonid rickettsial septicemia (SRS), is the main pathogen affecting farmed salmonids in Chile. Outbreaks of SRS lead to substantial economic losses for producers. Many determinants related to SRS outcomes are still poorly understood. Here, we conducted a retrospective cohort study from 2014 to 2021 to investigate the epidemiology of SRS at the farm level in southern Chile, employing historical monitoring data. Using time series analysis of weekly SRS mortality risk and sea lice (Caligus rogercresseyi ) infestation levels, we found that SRS mortal

Piscirickettsia salmonis is a facultative intracellular bacterium that generates piscirickettsiosis affecting salmonids in Chile. The bacterium has the adaptability to survive in the marine environment under multiple stressful conditions. In this sense, this work focused on the analysis of a gene battery associated with biofilm formation under different culture conditions and on the adaptability of this biofilm to different media. The results indicated that the strains LF-89, IBM-034 and IBM-040 were strong biofilm producers, evidencing adaptability to the media by increasing the amount of bio

Research on Piscirickettsia salmonis, a critical pathogen affecting salmon aquaculture, has been hampered by inadequate host models to study the infection process in vivo. Here, we highlight zebrafish as a surrogate host model for in-depth analysis of P. salmonis infection by using fluorescent dyes and protein markers to visualize host-pathogen interactions. Using transgenic zebrafish lines with fluorescent immune cells, we observed a strong increase in neutrophil numbers in the infected larvae, resembling an emergency granulopoiesis response. We observed the presence of P. salmonis in the cau

Phagocytic cells are pivotal for host homeostasis and infection defense, necessitating metabolic adaptations in glycolysis, the tricarboxylic acid (TCA) cycle, and oxidative phosphorylation (OXPHOS). While mammalian phagocytes shift towards glycolysis and glutaminolysis during polarization, research on fish phagocyte metabolic reprogramming is limited. To address this, the Atlantic salmon phagocytic cell line, SHK-1, serves as a valuable model. Using the Seahorse XFe96 Flux Analyzer, this study compares SHK-1 bioenergetics under glucose-restricted (L-15 medium) and glucose-supplemented (PM) co

BACKGROUND: Vertebrate hosts limit the availability of iron to microbial pathogens in order to nutritionally starve the invaders. The impact of iron deficiency induced by the iron chelator deferoxamine mesylate (DFO) was investigated in Atlantic salmon SHK-1 cells infected with the facultative intracellular bacterium Piscirickettsia salmonis.
RESULTS: Effects of the DFO treatment and P. salmonis on SHK-1 cells were gaged by assessing cytopathic effects, bacterial load and activity, and gene expression profiles of eight immune biomarkers at 4- and 7-days post infection (dpi) in the control gro
