Cadmium sulfate induces apoptosis in planarians.

With rapid socio-economic development, heavy metal pollution in water has become common and affects both environment and human health. Cadmium (Cd) has been recognized as one of the heavy metals which cause acute or chronic toxic effects if ingested. Although its toxicity is undisputed, the underlying molecular mechanisms in vivo are not fully understood. Planarians, a model organism famous for their regenerative prowess, have long been utilized to study the effects of chemical exposure. In this study, we observed apoptosis with TUNEL assay in planarians induced by cadmium sulfate (CdSO4) in a

Nicotine behavioral pharmacology: clues from planarians.

BACKGROUND: Nicotine is one of the world's most addictive substances and the primary reason that humans inhale tobacco smoke. The pharmacological effects of nicotine can be investigated in planarians, aquatic flatworms that possess an integrated neural network including cephalic ganglia that some consider the earliest 'brain' and spinal cord. Here, we tested the hypothesis that nicotine exposure elicits mammalian-like behaviors in planarians. METHODS: Planarian motility and stereotypy (C-shape hyperkinesias) were quantified following acute nicotine exposure. During repeated nicotine exposure,

Energy drink produces aversive effects in planarians.

Energy drinks pose consumer and environmental risks. One of the few organisms suitable for investigating both risks are planarians, which display mammalian-like behavioral effects during drug exposure and reside in aquatic environments. We investigated effects of Monster Energy® (0.001 - 10%) on planarian behaviors using established assays. For acute exposure, only higher concentrations reduced motility (>1%) and caused stereotypies (>1%). Lower concentrations (0.1-1%) enhanced light avoidance, a measure of defensive responding. In place conditioning experiments conducted with low concentratio

Host-transposon mutualism supports regeneration in planarians.

Transposons make up a significant fraction of eukaryotic genomes. They are commonly viewed as selfish elements that are detrimental to their hosts, and they are prime targets of specialized host defenses that constrain their expansion. Mutualistic interactions, in which elements co-exist and benefit each other, have so far not been found between transposable elements and eukaryote hosts. Here, we present evidence for an active transposon that confers a direct benefit to its planarian host. We find that the Ty3-like giant transposon Burro1 is an ancient element that retains its mobility. Burro1

Quantitative Analysis of Planarian Pigmentation.

The ommochrome and porphyrin body pigments that give freshwater planarians their brown color are produced by specialized dendritic cells located just beneath the epidermis. During embryonic development and regeneration, differentiation of new pigment cells gradually darkens newly formed tissue. Conversely, prolonged light exposure ablates pigment cells through a porphyrin-based mechanism similar to the one that causes light sensitivity in rare human disorders called porphyrias. Here, we describe a novel program using image-processing algorithms to quantify relative pigment levels in live anima

Psilocybin enhances conditioned place preference learning in planarians.

Psilocybin has emerged as a promising therapeutic agent, with clinical benefits attributed to enhanced neuroplasticity rather than acute pharmacological effects. However, the fundamental mechanisms underlying these effects remain poorly understood. Using planarians as a model system, we investigated whether psilocybin enhances associative learning using a conditioned place preference paradigm. Following a single exposure to 5 µM psilocybin, planarians underwent 5 days of conditioning in which light was paired with sucrose. Psilocybin-treated planarians showed a significantly greater shift in l

Surgical Ablation Assay for Studying Eye Regeneration in Planarians.

In the study of adult stem cells and regenerative mechanisms, planarian flatworms are a staple in vivo model system. This is due in large part to their abundant pluripotent stem cell population and ability to regenerate all cell and tissue types after injuries that would be catastrophic for most animals. Recently, planarians have gained popularity as a model for eye regeneration. Their ability to regenerate the entire eye (comprised of two tissue types: pigment cells and photoreceptors) allows for the dissection of the mechanisms regulating visual system regeneration. Eye ablation has several

Nicotine-induced C-shape movements in planarians are reduced by antinociceptive drugs: Implications for pain in planarian paroxysm etiology?

C-shapes are stereotyped movements in planarians that are elicited by diverse stimuli (e.g. acidity, excitatory neurotransmitters, psychostimulants, and pro-convulsants). Muscle contraction and seizure contribute to the expression of C-shape movements, but a causative role for pain is understudied and unclear. Here, using nicotine-induced C-shapes as the endpoint, we tested the efficacy of three classes of antinociceptive compounds - an opioid, NSAID (non-steroidal anti-inflammatory drug), and transient receptor potential ankyrin 1 (TRPA1) channel antagonist. For comparison we also tested effe

Advances in ecotoxicological research using planarians: a comprehensive review.

Planarians, distinguished by their extraordinary regenerative capacity and ecological role as benthic predators, have emerged as a transformative model in ecotoxicology. This review provides a comprehensive synthesis of research on the effects of diverse classes of pollutants, including metals, pesticides, pharmaceuticals, personal care products, nanomaterials, and a dedicated category of "other pollutants," namely biotoxins and physical stressors, on these organisms. We introduce a novel conceptual framework of "regenerative toxicology," which focuses on how contaminants impair an organism's

Xylazine and opioid coadministration enhances stereotyped movements in planarians.

Xylazine is a veterinary drug and α2-adrenoceptor agonist that has been increasingly misused as an adulterant in illicit opioids; however, only a few preclinical studies have investigated xylazine's pharmacological profile and impact on opioid-derived behaviors. We investigated the behavioral effects of xylazine alone and in combination with morphine in planarians, which are the simplest living animals having a central nervous system with cephalization. Planarians also express mammalian-like behaviors and neurotransmitters. Our specific experiments investigated the effects of xylazine, morphin

Recording and spectrum analysis of the planarian electroencephalogram.

Many animals produce continuous brainwaves, known as the electroencephalogram (EEG), but it is not known at what point in evolution the EEG developed. Planarians possess the most primitive form of brain, but still exhibit learning and memory behaviors. Here, we observed and characterized the EEG waveform of the planarian. We inserted a monopole electrode into the head of a planarian on a cold stage, and were able to observe the EEG at sub-microvolt amplitudes. The EEG had a continuous waveform, similar to that of evolutionarily advanced animals with more developed brains. Occasional myogenic p

Nociceptin attenuates methamphetamine abstinence-induced withdrawal-like behavior in planarians.

Planarians display a concentration-related reduction in locomotor activity when amphetamine, cocaine, cannabinoid, or benzodiazepine exposure is abruptly discontinued. In the present study, we tested the hypothesis that abrupt discontinuation of methamphetamine would also cause withdrawal-like behavior in planarians and that the withdrawal-like behavior would be prevented by nociceptin, which has been shown to modulate the effects of methamphetamine in mammals. We observed a concentration-related reduction of locomotor behavior when planarians exposed to methamphetamine (0.1-100 microM) were t

Planarian as an Animal Model for Experimental Acute Seizure.

Epilepsy is among the most prevalent neurological disorders characterized by recurring spontaneous seizures. Seizures represent a clinical manifestation of uncontrolled, excessively synchronized neural cell activity. The extent of brain damage from seizures depends on their duration and intensity. Regrettably, there is no effective remedy for epilepsy. The aim of this investigation is to assess whether the planaria worm Dugesia dorotocephala could serve as a model to aid in identifying and developing treatments for epilepsy that can target acute seizures. Currently, various models, such as mar

Total RNA Isolation from Planarian Tissues.

Isolation of high-quality and nondegraded RNA is a prerequisite for many modern molecular biology applications. A variety of RNA extraction products and protocols are available for the standard model organisms, yet species-specific peculiarities of less well studied organisms often require specific protocol adaptations. Here we describe a robust RNA extraction protocol for planarians that is widely used in the community. The protocol combines tissue homogenization in TRIzol with phenol-chloroform extraction and subsequent purification over commercial columns. The purified RNA can be used for m

Ephrin Signaling Patterns Sensory Neurons During Tissue Homeostasis in Planarians.

Eph and ephrin genes encode receptor-ligand pairs that mediate contact-dependent cell signaling and are essential for nervous system development. However, less is known about the role of Ephrin signaling during adult tissue homeostasis and regeneration. Here, we investigated the role of Ephrin signaling in neural patterning in the planarian Schmidtea mediterranea . We discovered that RNAi against the Eph receptor EphR1 led to striking ectopic expression of the mechanosensory neuron markers pkd1L-2 and hmcn-1-L, without obvious disruption of the overall architecture of the central nervous syste

Backward scrunching: planarians escape from noxious stimuli, cannibalism, and predation.

Animals have evolved escape behaviors to survive. Freshwater planarians have many predators, including conspecifics in cannibalistic species such as Dugesia japonica. When agitated, planarians escape via a locomotory behavior called "scrunching", whereby the planarian periodically elongates and contracts its whole body. Current understanding is that planarians move forward when they scrunch. However, when transversely cutting D. japonica, we observed that while head pieces scrunched forward, tail pieces scrunched backward instead. Previous work has shown that forward scrunching is a conserved

Artificial intelligence assisted behavioral profiling of synthetic cannabinoids in planarians.

PURPOSE: Freshwater planarians provide a rapid and scalable biological model for detecting drug-induced neurobehavioral effects. This study evaluated whether automated behavioral profiling in Dugesia dorotocephala could distinguish phytocannabinoid and synthetic cannabinoid exposure based on organism-level motor responses. METHODS: Planarians were acutely exposed to Δ⁹-tetrahydrocannabinol (Δ⁹-THC), AB-PINACA, MA-CHMINACA, A-796,260, or JWH-412 at concentrations of 5–60 µg/mL in artificial spring water containing PEG-400. Annual rhythmicity in the switching of reproductive mode in planarians.

Species of planarians include both asexually reproducing individuals (reproduce through fission and regeneration) and sexually reproducing individuals (hermaphrodites that mate to produce cocoons). While some individuals can switch between the asexual and sexual modes of reproduction. In this study, we examined the reproductive modes and ploidy of Dugesia japonica and Dugesia ryukyuensis from three spring wells in Okinawa (Japan) during two consecutive years. D. japonica are mostly asexual and triploid. In contrast, only 40 % of D. ryukyuensis are asexual and triploid; the remaining are sexual
