Recent insights into air space formation in plant shoots.

An adequate supply of oxygen and carbon dioxide is essential for plant survival. Although plant cell walls are somewhat porous, their hydrated nature hampers gas diffusion. Furthermore, the cuticular wax coating the epidermal layer of aerial tissues strongly inhibits gas exchange. Because plants lack specialized systems that bind and transport gases, gases must be directly delivered to the target cells. This necessitates the establishment of effective gas transport pathways connecting stomata to the target cells. However, our understanding of this process remains fragmented. Recent studies hav

Navigating shoot apical meristem plasticity for plant adaptation.

Plants exhibit striking developmental plasticity, driven by stem cell populations within meristematic tissues such as the shoot apical meristem (SAM). While the genetic networks governing stem cell homeostasis are well-characterized, recent studies reveal that the SAM also functions as an active environmental sensor to modulate growth plasticity. This Tansley review synthesizes advances in how the SAM perceives and integrates key environmental signals - including light, temperature, oxygen, and humidity - to direct adaptive growth. We detail the molecular mechanisms, such as photoreceptor-medi

Droplet-vitrification cryopreservation of shoot tips of the endangered plant Lilium tsingtauense.

This study established an efficient droplet-vitrification cryopreservation protocol for the endangered plant Lilium tsingtauense. Through optimization, the best conditions were determined as follows: shoot tips were precultured in 0.3 M sucrose at 4 °C for 7 days, treated with loading solution for 20 min, dehydrated with PVS2 solution at 0 °C for 90 min, and then rapidly plunged into liquid nitrogen using the aluminum foil strip droplet method. A mean regrowth rate of 67% was achieved. Histological observations showed that cells recovered normal morphology after protective dehydration, and ISS

Fuelling the top - Metabolic control of shoot apical meristem function and plant development.

Plant development relies on a complex network of transcription factors and hormone signalling pathways that guide pattern formation and developmental transitions in the meristems. Yet, how this core network is regulated to enable developmental plasticity remains poorly understood. A key question is how information about nutrient availability influences growth decisions, with nutrients acting not only as substrates for energy production and anabolism but also as regulatory signals. Here, we review recent advances in plant nutrient signalling during development, focussing on how carbon and nitro

Insights into plant-part-specific N2O production in roots and shoots of chicory (Cichorium intybus) using stable isotope labelling.

Nitrous oxide (N2O) contributes substantially to climate change and stratospheric ozone degradation, yet large uncertainties in its global budget point to overlooked sources. Growing evidence suggests plants produce N2O, but the mechanisms remain poorly constrained. For the first time, we resolved organ-specific N2O formation in roots and shoots of aseptically cultivated chicory (Cichorium intybus) under light and dark conditions, combining 15N-labelling (15NO3 -/15NH4 +) with position-specific isotope analysis to identify N2O formation pathways. All cultures produced N2O under dark conditions

Callus mediated shoot organogenesis and regeneration of cytologically stable plants of Ledebouria revoluta: An ethnomedicinal plant with promising antimicrobial potency.

Ledebouria revoluta are important ethnomedicinal plant found in India and South Africa. Micropropagation via indirect shoot organogenesis had been established from three types of explant (i.e. scale leaf, leaf lamina and root) of L. revoluta. Scale leaf was found superior as compared to leaf lamina and root explant with respect to their organogenic callus induction potentiality. Murashige and Skoog (1962) [MS] media supplemented with 3.0 mg L-1 2,4-dichlorophenoxyacetic acid, 0.75 mg L-1 β-naphthoxyacetic acid were best effective for inducing organogenic callus. Melon shoot organization 1, encoding an AGRONAUTE7 protein, plays a crucial role in plant development.

A melon gene MSO1 located on chromosome 10 by map-based cloning strategy, which encodes an ARGONAUTE 7 protein, is responsible for the development of shoot organization. Plant endogenous small RNAs (sRNAs) are involved in various plant developmental processes. In Arabidopsis, sRNAs combined with ARGONAUTE (AGO) proteins are incorporated into the RNA-induced silencing complex (RISC), which functions in RNA silencing or biogenesis of trans-acting siRNAs (ta-siRNAs). However, their roles in melon (Cucumis melo L.) are still unclear. Here, the melon shoot organization 1 (mso1) mutant was identifie

Dissecting the Roles of Cuticular Wax in Plant Resistance to Shoot Dehydration and Low-Temperature Stress in Arabidopsis.

Cuticular waxes are a mixture of hydrophobic very-long-chain fatty acids and their derivatives accumulated in the plant cuticle. Most studies define the role of cuticular wax largely based on reducing nonstomatal water loss. The present study investigated the role of cuticular wax in reducing both low-temperature and dehydration stress in plants using Arabidopsis thaliana mutants and transgenic genotypes altered in the formation of cuticular wax. cer3-6, a known Arabidopsis wax-deficient mutant (with distinct reduction in aldehydes, n-alkanes, secondary n-alcohols, and ketones compared to wild

Responses of the Asian citrus psyllid to volatiles emitted by the flushing shoots of its rutaceous host plants.

Diaphorina citri Kuwayama (Hemiptera: Psyllidae) carries Candidatus liberibacter spp., the putative causal agents of Huanglongbing. D. citri reproduces and develops only on the flushing shoots of its rutaceous host plants. Here we examined whether D. citri is attracted to host plant odors and a mixture of synthetic terpenes. Tests conducted in a vertically oriented Y-tube olfactometer showed that both males and females preferentially entered the Y-tube arm containing the odor from the young shoots of Murraya paniculata (L.) Jack and Citrus limon L. Burm. f. cultivar Eureka. Only males exhibite

The interaction with arbuscular mycorrhizal fungi or Trichoderma harzianum alters the shoot hormonal profile in melon plants.

Arbuscular mycorrhizal fungi (AMF) and Trichoderma harzianum are known to affect plant growth and disease resistance through interaction with phytohormone synthesis or transport in the plant. Cross-talk between these microorganisms and their host plants normally occurs in nature and may affect plant resistance. Simultaneous quantification in the shoots of melon plants revealed significant changes in the levels of several hormones in response to inoculation with T. harzianum and two different AMF (Glomus intraradices and Glomus mosseae). Analysis of zeatin (Ze), indole-3-acetic acid (IAA), 1-am

The Phenolic Compounds in the Young Shoots of Selected Willow Cultivars as a Determinant of the Plants' Attractiveness to Cervids (Cervidae, Mammalia).

This study examined the phenolic acids, flavonoids, and salicylates contents in young, 3-month-old shoots (including the leaves) of willow (Salix spp.). The cultivars were selected based on experiments carried out previously in Poland on fodder and energy willows. It was found, using the HPLC-MS/MS method, that the willow cultivars analyzed from three experimental plots, contained nine different phenolic acids, five salicylates and nine flavonoids, including four flavanols (quercetin, kaempferol, taxifolin and isorhamnetin), two flavanones (prunin, naringenin), two flavones (luteolin, apigenin

Plant-mediated interactions between shoot-feeding aphids and root-feeding nematodes depend on nitrate fertilization.

The plant metabolite composition is modulated by various abiotic and biotic factors including nutrient availability and herbivory. In turn, induced changes in plant quality can affect herbivore performance and mediate indirect interactions between spatially separated herbivores sharing a host. Studies on plant-mediated herbivore interactions have been carried out at single fertilization regimes only, but we hypothesized that nutrient availability modifies these interactions. Therefore, we studied the interactions between two vascular tissue herbivores, the aboveground feeding aphid Brevicoryne

Salt stress-induced Ca2+ waves are associated with rapid, long-distance root-to-shoot signaling in plants.

Their sessile lifestyle means that plants have to be exquisitely sensitive to their environment, integrating many signals to appropriate developmental and physiological responses. Stimuli ranging from wounding and pathogen attack to the distribution of water and nutrients in the soil are frequently presented in a localized manner but responses are often elicited throughout the plant. Such systemic signaling is thought to operate through the redistribution of a host of chemical regulators including peptides, RNAs, ions, metabolites, and hormones. However, there are hints of a much more rapid co

In vitro propagation of Codonopsis pilosula (Franch.) Nannf. using apical shoot segments and phytochemical assessments of the maternal and regenerated plants.

BACKGROUND: Codonopsis pilosula (Franch.) Nannf. is a medicinal plant traditionally used in China, Korea, and Japan to treat many diseases including poor gastrointestinal function, low immunity, gastric ulcers, and chronic gastritis. The increasing therapeutic and preventive use of C. pilosula has subsequently led to depletion of the natural populations of this species thus necessitating propagation of this important medicinal plant. Here, we developed an efficient and effective in vitro propagation protocol for C. pilosula using apical shoot segments. We tested various plant tissue culture me
