Selective killing of choriocarcinoma cells in vitro by trichosanthin, a plant protein purified from root tubers of the Chinese medicinal herb Trichosanthes kirilowii.

Trichosanthin is an abortifacient plant protein purified from the Chinese medicinal herb Tian-hua-fen, obtained from root tubers of Trichosanthes kirilowii (Cucurbitaceae). Recently, trichosanthin has also been used in the treatment of trophoblastic tumours, including hydatidiform mole, invasive mole and choriocarcinoma. We have examined the effects of trichosanthin on cultured cells and observed selective cytotoxic effects on choriocarcinoma cells. Mouse melanoma cells were also sensitive to its cytotoxic action. Cytotoxic effects on cultured cells were usually not prominent after 24 hr treat

Systemic Colonization of Potato Plants by Verticillium dahliae Leads to Infection of Tubers and Sprouting Buds.

A green fluorescent protein (GFP)-tagged isolate of Verticillium dahliae was used to study its colonization in potato plants and tubers. Three-week-old potato plants of the highly susceptible cultivar 'Shepody' were inoculated with a conidial suspension of a GFP-tagged isolate of V. dahliae using a wound inoculation method. Colonization was studied using confocal microscopy combined with tissue sections. Conidia germinated and hyphae grew along the root hairs, elongation zones, and root caps between 24 and 96 h postinoculation (HPI). At 7 days postinoculation (DPI), the pathogen advanced to co

Fractionating of Calcium in Tuber and Leaf Tissues Explains the Calcium Deficiency Symptoms in Potato Plant Overexpressing CAX1.

Consistent with reports on other plants we recently reported that a potato transgenic line (AT010901) overexpressing sCAX1 show classic symptoms of calcium deficiency shoot tip injury, leaf curling, leaf margin necrosis and tuber internal defects such as hollow heart and brown spots. The present study was undertaken to quantify calcium in various fraction of leaf and tuber tissues of this transgenic and wild type potato clones to understand the development of these deficiency symptoms at normal calcium nutrition (1mM) and its mitigation at higher calcium nutrition (10mM). Plants were grown in

Ecological Filtering by Tuber Compartments Shapes Stable Core Microbiomes That Underpin Potato Plant Growth Across Environments.

Harnessing plant microbiomes for sustainable agriculture requires understanding not only whether they can boost crop performance, but also how ecological processes govern their assembly, stability, and functional contributions across environments. While we previously showed that seed tuber microbiomes can predict potato vigour using machine learning, it remained unclear how ecological processes shape tuber microbiome stability and functionality across host genotypes, tuber compartments, soil types, and years. Here, we analyzed the national-scale dataset of 240 field-collected potato seedlots,

Auxin synthesis gene tms1 driven by tuber-specific promoter alters hormonal status of transgenic potato plants and their responses to exogenous phytohormones.

Ectopic auxin overproduction in transgenic potato leads to enhanced productivity accompanied with concerted and occasional changes in hormonal status, and causing altered response of transformants to exogenous auxin or cytokinin. Previously, we generated potato transformants expressing Agrobacterium-derived auxin synthesis gene tms1 driven by tuber-specific patatin gene promoter (B33-promoter). Here, we studied the endogenous hormonal status and the response to exogenous phytohormones in tms1 transformants cultured in vitro. Adding indole-3-acetic acid (IAA) or kinetin to culture medium affect

Antioxidant and hepatoprotective activity of tubers of Momordica tuberosa Cogn. against CCl4 induced liver injury in rats.

Hydro alcoholic extract of tubers of M. tuberosa was subjected to preliminary phytochemical screening and evaluated for in vitro and in vivo antioxidant and hepatoprotective activity against CCl4 induced liver damage in rats. Pretreatment with 70% ethanolic extract of M. tuberosa reversed CCl4 induced elevation of levels of serum biomarkers to near normal levels, suggesting that the tubers of M. tuberosa possess hepatoprotective property and this property may be attributed to the antioxidant property of the plant.

Potato Tuberisation Responses to Drought and a Film-Forming Antitranspirant.

Film-forming antitranspirants may help potatoes tolerate moderate drought, but their effects on early tuberisation and tuber size distribution remain unclear. Two pot experiments were conducted in a polytunnel (late summer) and a glasshouse (winter-spring), with moderate drought imposed during tuber initiation and early bulking, alone (DT) or combined with an antitranspirant (di-1-p-menthene; VGDT). Leaf relative water content (RWC), stolon traits, and tuber yield and size distribution were measured. Moderate drought reduced RWC, stolon number, and tuber set, which indicates the sensitivity of

Toxic encephalopathy due to colchicine--Gloriosa superba poisoning.

Gloriosa superba, a flowering plant widespread in South and Southeast Asia, is implicated in many cases of self-poisoning. Colchicine is concentrated in the seeds and tubers and this mediates its toxicity. We describe a 28-year-old woman who developed delayed encephalopathy after eating G superba tubers. MR scan of brain showed bilateral symmetrical T2 basal ganglia hyperintensities in the caudate and lentiform nuclei. The delay in onset of encephalopathy is attributable to a direct-effect colchicine, probably mediated through its effect on microtubular transport.

Impacts of different ozone treatments on the growth and soil bacterial community of Pinellia ternata.

Continuous cropping causes soil microbial imbalance, which severely reduces the yield and quality of medicinal plants. Ozone, a potent oxidant, has the potential to eradicate soilborne diseases in agricultural systems. However, the application of ozone to alleviate continuous cropping problems in the medicinal plant Pinellia ternata has not been reported. This study applied four treatments to P. ternata tubers and continuous cropping soil. The control treatment (CK) was not exposed to ozone and was irrigated with distilled water. The experimental treatments included ozone-fumigated tubers irri

Tissue-specific metal accumulation and tuber metabolic reprogramming in Cyperus esculentus under multiple-metal stress.

Understanding plant responses to co-occurring metal contamination is essential for evaluating their phytoremediation potential, yet the physiological and molecular responses of Cyperus esculentus (CES) to combined Cu, Zn, and Cd exposure remain poorly understood. Here, CES plants were exposed to combined Cu, Zn, and Cd stress at concentrations of 0-10 mg/L under hydroponic conditions, and plant growth, tissue-specific metal accumulation, physiological and biochemical responses, and tuber transcriptomic changes were systematically analyzed. Metal accumulation exhibited clear tissue specificity,

Poly(phenolic) deposition is a prerequisite to poly(aliphatic) deposition for functional potato suberin.

Suberin deposition in specialized cells is a critical aspect of plant protection, providing both water retention and pathogen protection functions. Tissue specific suberization during plant development is mirrored by its wound induction in cells that do not normally suberize and provides a robust experimental system to investigate the process. Suberized cells are characterized by a cell wall-associated lignin-like poly(phenolic) domain, and an ester-linked poly(aliphatic) domain, with the latter often laid down in distinct lamellar bands between the cell wall and plasma membrane. First Report of Potato Tuber Necrotic Ringspot Disease Caused by PVYNTN in Portugal.

During the last 2 years, potato (Solanum tuberosum L.) tubers displaying superficial necrotic arcs and rings were found in central Portugal. These symptoms increased during storage, and diminished tuber quality of ware (fresh-market) potatoes; however, no internal necrosis, which is typical for infections caused by tobacco rattle virus or potato mop top virus, was observed. The symptoms led to the preliminary diagnosis of potato tuber ringspot disease (PTNRD), caused by a tuber necrosis (TN)-inducing isolate of the tobacco veinal necrosis strain group of potato virus Y (PVYN) that was named PV

Scion-based drought stress memory affects potato response to water deficit.

A scion-based stress memory signal, which was derived from drought-primed potato plants, was transmitted to new potato plants generated through vegetative reproduction. This affected potato tuber yield. Drought is one of the most significant threats to agricultural productivity worldwide. The cultivated potato (Solanum tuberosum L.) is a crop species that is sensitive to drought stress. This study investigated the impact of scion-based drought stress memory on tuber yield, physiological parameters, gene expression, and DNA methylation in the vegetative progeny of grafted plants. The tuber prog

Gene expression and phytohormone levels in the asymptomatic and symptomatic phases of infection in potato tubers inoculated with Dickeya solani.

Dickeya solani is a soft rot bacterium with high virulence. In potato, D. solani, like the other potato-infecting soft rot bacteria, causes rotting and wilting of the stems and rotting of tubers in the field and in storage. Latent, asymptomatic infections of potato tubers are common in harvested tubers, and if the storage conditions are not optimal, the latent infection turns into active rotting. We characterized potato gene expression in artificially inoculated tubers in nonsymptomatic, early infections 1 and 24 hours post-inoculation (hpi) and compared the results to the response in symptoma

Profile of C. Robin Buell.

C. Robin Buell has been a leading figure in plant genomics since the advent of DNA sequencing technology. She helped lead multiple consortia to sequence some of the first crop genomes at the turn of the millennium. She has since used the genomes to tackle questions in fundamental biology, plant evolution, and agriculture. Recently, she applied single-cell technologies to uncover how complex biosynthetic pathways are compartmentalized across different rare plant cell types. Now at the University of Georgia, Buell explores in her Inaugural Article how tubers arose repeatedly in the plant family

Accumulation of As, Cd and selected trace elements in tubers of Scirpus maritimus L. from Doñana marshes (South Spain).

The collapse of a pyrite-mining, tailing dam on 1998 contaminated an area of 4286 ha along the Agrio and Guadiamar river valleys in southern Spain. Over 2700 ha of the Doñana marshes, an important wintering area for wetland European birds, were contaminated. This study reports analyses of the tubers of Scirpus maritimus (an important food for greylag geese, Anser anser) collected in 2000 in the "Entremuros" (spill-affected area) and in nearby unaffected Doñana marshes (control areas). In the spill-affected area mean tuber tissue concentrations of Cd (0.25 mg kg-1) and Zn (61 mg kg-1) were grea

Unraveling the metabolic network and key bioactive compound accumulation during critical developmental stages of Gastrodia elata.

Gastrodia elata Blume, a traditional medicinal plant, the dynamic metabolic changes and the mechanisms underlying the accumulation of medicinal components remain poorly understood during its key developmental stages (tubers before bolting, tubers after bolting, stems). This study utilized untargeted metabolomics to systematically analyze the metabolic composition and differences in tubers before bolting, tubers after bolting, and stems of G. elata f. elata, G. elata f. glauca, and dark red G. elata. We identified a total of 6209 metabolites, encompassing 17 major categories, and of these, 1624

A new Streptomyces scabies-infecting bacteriophage from Egypt with promising biocontrol traits.

Potato common scab caused by Streptomyces scabies is one of the most economically important diseases infecting potato. It reduces the quality of potato tubers, which subsequently decreases the tuber prices and causes significant economic losses for potato growers. Biological control using bacteriophages is a promising strategy for controlling this disease. In this study, a novel bacteriophage with high lytic efficacy against S. scabies was isolated from a potato field at El-Minya, Egypt, and was designated SscP1EGY. The phage has an icosahedral head of 55 nm and a short tail of 7.5 nm, typical

Co-option of ancestral stem regulators drove recurrent evolution of underground storage organs.

Geophytes are plants that produce underground storage organs such as tubers, rhizomes, and bulbs, to facilitate asexual reproduction and withstand a myriad of environmental challenges. While the potato (Solanum tuberosum L.) serves as the primary model for studying tuberization, the genetic mechanisms encoding this trait across diverse angiosperm lineages remains unclear. This study utilized a phylogenomic-transcriptomic approach to compare tuber development across nine tuberizing species with five nontuberizing sister taxa. We identified orthologs of key potato tuberization genes that exhibit

Starch and tuber traits of diploid potato lines B26 and B100 and their hybrids.

Diploid potato breeding enables faster genetic improvement via selection against deleterious alleles in inbred lines, unlike breeding by intercrossing tetraploid varieties. Starch is the major source of calories in potato tubers, but the starch properties of diploid lines have rarely been reported. In this study, we provide a comprehensive characterisation of tuber and starch properties in two diploid lines that are early isolates from the Solynta breeding program, B26 and B100, and their F1 hybrids. B100 produced fewer, but larger tubers compared to B26, and both diploid lines produced tubers
