Decreased Vascular Bundle 1 affects mitochondrial and plant development in rice.

BACKGROUND: Mitochondria are vital regulators of plant growth and development, constitute the predominant source of ATP, and participate in multiple anabolic and catabolic metabolic pathways. But the mechanism by which dysfunctional mitochondria affect plant growth remains unknown, and more mitochondria-defective mutants need to be identified. RESULTS: A mitochondria-defective mutant decreased vascular bundle 1 (dvb1) was isolated from rice mutant library mutagenized by EMS (ethylmethane sulfonate), which shows dwarfism, narrow leaves, short branches, few vascular bundles, and low fertility.

EDAH increases bleeding sap intensity and grain yield of maize under high-density cultivation on saline-alkali soils in Xinjiang.

High-density maize planting compromises stalk strength and increases susceptibility to lodging. While growth regulators enhance lodging resistance and yield by optimizing stalk mechanical properties, their specific effects on the vascular structure, bleeding sap characteristics, and dry matter accumulation of ear internodes and peduncles under high plant densities remain unclear. Field experiments (2023-2024) were performed to evaluate two maize cultivars (Denghai 618, DH618; MC670) under three planting densities (90,000 plants ha-1 (D1), 135,000 plants ha-1 (D2), and 180,000 plants ha-1 (D3))

Hydraulic function of medullary bundles in a tropical root climber.

PREMISE: Medullary bundles, which are vascular bundles in the pith, have evolved multiple times. However, their functional significance remains poorly understood. Here, we investigated how medullary bundles are involved in the vascular architecture and hydraulic function in a tropical root climber, Adelobotrys scandens (Melastomataceae) to understand the functional significance of this vascular architecture in a wet-adapted root climber species as compared to what is known about medullary bundles in the lineages in hydraulically limited arid/saline environments. METHODS: We used light microsc

Linking phenylpropanoid metabolism, lignin deposition, and plant growth inhibition.

Lignin, a polymer found in the plant secondary cell wall, is a major contributor to biomass' recalcitrance toward saccharification. Because of this negative impact toward the value of lignocellulosic crops, there is a special interest in modifying the content and composition of this important plant biopolymer. For many years this endeavor has been hindered by the plant growth inhibition that is often associated with manipulations to phenylpropanoid metabolism. Although the actual mechanism by which dwarfism arises remains unknown, recent advances in tissue-specific lignin complementation and b

Spatial gene expression analysis in tomato hypocotyls suggests cysteine as key precursor of vascular sulfur accumulation implicated in Verticillium dahliae defense.

Verticillium dahliae is a prominent generator of plant vascular wilting disease and sulfur (S)-enhanced defense (SED) mechanisms contribute to its in-planta elimination. The accumulation of S-containing defense compounds (SDCs) including elemental S (S(0) ) has been described based on the comparison of two near-isogenic tomato (Solanum lycopersicum) lines differing in fungal susceptibility. To better understand the effect of S nutrition on V. dahliae resistance both lines were supplied with low, optimal or supraoptimal sulfate-S. An absolute quantification demonstrated a most effective fungal

The evolution of leaf traces in eupolypod ferns.

The expansion or reduction of structural parts is an aspect of phenotypic variation that leads to diverse form (e.g. digits in vertebrates, segments in arthropods, or petals in angiosperms). The number of petiole vascular bundles (leaf traces) in ferns is one such structure. Integrating traditional anatomy and developmental videography with phylogenetic comparative methods, we examined the evolution and development of fern leaf traces. Across the eupolypods - 80% of extant species richness - and their sister lineages, we observe four conserved vascular developmental units (VDUs) that consisten

Low-affinity cation transporter (OsLCT1) regulates cadmium transport into rice grains.

Accumulation of cadmium (Cd) in rice (Oryza sativa L.) grains poses a potential health problem, especially in Asia. Most Cd in rice grains accumulates through phloem transport, but the molecular mechanism of this transport has not been revealed. In this study, we identified a rice Cd transporter, OsLCT1, involved in Cd transport to the grains. OsLCT1-GFP was localized at the plasma membrane in plant cells, and OsLCT1 showed Cd efflux activity in yeast. In rice plants, strong OsLCT1 expression was observed in leaf blades and nodes during the reproductive stage. In the uppermost node, OsLCT1 tra

Improved mesophyll-bundle sheath connectivity is achieved via different mechanisms in C2 vs C4 Alternanthera.

Connectivity between mesophyll (M) and bundle sheath (BS) cells must improve during the evolution of C4 photosynthesis to facilitate large metabolite fluxes between these cell types, but the trait combinations that enhance M-BS connectivity and the points at which these enhancements occur along the C3 to C4 evolutionary trajectory remain unknown. We investigated shifts in M-BS connectivity that accompanied photosynthetic diversification within the eudicot genus Alternanthera from the agriculturally important Amaranthaceae family. We combined transmission electron microscope and confocal micros

Infrared Microimaging of Sucrose Distribution in Plant Vascular Tissues.

Sucrose is the primary transport sugar in plants, serving as an essential energy source and signaling molecule. Detection, visualization, and quantification of sucrose in various plant tissues are essential for understanding the metabolic and physiological processes that sustain plant life. Traditional metabolite-mapping techniques have struggled to visualize the quantitative distribution of sucrose at sufficient resolution to distinguish vascular bundles from surrounding tissues. Here, we present a Fourier-transform infrared (FTIR) imaging approach that can visualize sucrose in plant tissues

Reporter-based forward genetic screen to identify bundle sheath anatomy mutants in A. thaliana.

The evolution of C4 photosynthesis proceeded stepwise with each small step increasing the fitness of the plant. An important pre-condition for the introduction of a functional C4 cycle is the photosynthetic activation of the C3 bundle sheath by increasing its volume and organelle number. Therefore, to engineer C4 photosynthesis into existing C3 crops, information about genes that control the bundle sheath cell size and organelle content is needed. However, very little information is known about the genes that could be manipulated to create a more C4 -like bundle sheath. To this end, an ethylme

Vascular Bundles Mediate Systemic Reactive Oxygen Signaling during Light Stress.

Systemic signaling and systemic acquired acclimation (SAA) are essential for plant survival during episodes of environmental stress. Recent studies highlighted a key role for reactive oxygen species (ROS) signaling in mediating systemic responses and SAA during light stress in Arabidopsis (Arabidopsis thaliana). These studies further identified the RESPIRATORY BURST OXIDASE HOMOLOG D (RBOHD) protein as a key player in mediating rapid systemic ROS responses. Here, we report that tissue-specific expression of RBOHD in phloem or xylem parenchyma cells of the rbohD mutant restores systemic ROS sig

Biomechanics and anatomical investigation on the nodes and internodes of an arborescent monocotyledon.

PREMISE: Bamboos have extremely long internodes, making it possible to measure tensile properties and anatomical features of discrete nodes and internodes and understand how the plants endure mechanical stress and how the tissues function in commercial products. We compared upper, middle, and lower parts of culms in black bamboo, Phyllostachys nigra, to determine whether the nodes comply with the Niklas spring-like joint model. METHODS: We measured flexural stiffness, modulus of elasticity (E), load at failure (Fmax), modulus of rupture (MOR), and density and fiber cap size of vascular bundle

Structural adaptations of aboveground vegetative organs to cangas: A case study with the Amazonian species Carajasia cangae R.M.Salas, E.L.Cabral & Dessein (Rubiaceae), which grows on ferruginous outcrops.

The Brazilian ironstone outcrops (cangas) impose challenging conditions for establishing species. Adapted species should exhibit different responses to xeric conditions of cangas. To understand these adaptations, our plant model was Carajasia cangae R.M.Salas, E.L.Cabral & Dessein, an endemic and threatened species restricted to the cangas of the Amazon Forest. Herein, we investigate structural adaptations that are important for its conservation, including anatomical characteristics of stems and leaves. To this goal, we evaluated the plant anatomy, including developmental aspects, between plan

Metabolite profiling and histochemical localization of alkaloids in Hippeastrum papilio (Ravena) van Scheepen.

Hippeastrum papilio (Amaryllidaceae) is a promising new source of galanthamine - an alkaloid used for the cognitive treatment of Alzheimer's disease. The biosynthesis and accumulation of alkaloids are tissue - and organ-specific. In the present study, histochemical localization of alkaloids in H. papilio's plant organs with Dragendorff's reagent, revealed their presence in all studied samples. Alkaloids were observed in vascular bundles, vacuoles, and intracellular spaces, while in other plant tissues and structures depended on the plant organ. The leaf parenchyma and the vascular bundles were

Distribution of Lily symptomless virus in tissues of Lanzhou lily (Lilium davidii var. unicolor) by fluorescence in situ hybridisation.

Lily symptomless virus (LSV) is one of the main viruses that endanger Lilium davidii var. unicolor, causing billions of losses to the lily planting industry every year. The distribution of viruses in plants is uneven and identifying the virus-free tissue regions in lily is crucial for obtaining virus-free resources through in vitro culture. In this study, Lilium davidii var. unicolor carrying LSV was used as the experimental material. RNA fluorescent probes were designed and prepared based on the genomic sequence of LSV. RNA in situ hybridization experiments were conducted on the meristematic

Differential cadmium and zinc distribution in relation to their physiological impact in the leaves of the accumulating Zygophyllum fabago L.

Cadmium and zinc share many similar physiochemical properties, but their compartmentation, complexation and impact on other mineral element distribution in plant tissues may drastically differ. In this study, we address the impact of 10 μm Cd or 50 μm Zn treatments on ion distribution in leaves of a metallicolous population of the non-hyperaccumulating species Zygophyllum fabago at tissue and cell level, and the consequences on the plant response through a combined physiological, proteomic and metabolite approach. Overexpression of TCP5 or Its Dominant Repressor Form, TCP5-SRDX, Causes Male Infertility in Arabidopsis.

TCP transcription factors have long been known to play a crucial role in leaf development, but their significance in reproduction has recently been revealed. TCP5 is a member of class II of the TCP family, which predominantly regulates cell differentiation. This study used overexpression and SRDX fusion to evaluate the role of TCP5 in anther development. TCP5 overexpression resulted in lower fertility, primarily due to anther non-dehiscence. We also observed reduced lignin accumulation in the anther endothecium. In addition, TCP5 overexpression resulted in smaller anthers with fewer pollen sac

Influence of blue light on the leaf morphoanatomy of in vitro Kalanchoe pinnata (Lamarck) Persoon (Crassulaceae).

Kalanchoe pinnata (Lamarck) Persoon (Crassulaceae) (air plant, miracle leaf) is popularly used to treat gastrointestinal disorders and wounds. Recently, the species was tested to treat cutaneous leishmaniasis with successful results. This medicinal activity was associated with the phenolic fraction of the plant. Blue light induces biosynthesis of phenolic compounds and many changes in anatomical characteristics. We studied the effects of supplementary blue light on the leaf morphology of in vitro K. pinnata. Plants cultured under white light (W plants) only and white light plus blue light (WB

Zea mays Drought-Overly Sensitive1/TUBA4 Is Wilty3, and Transcriptome Co-Expression Analysis of Shoot Meristem Mutant Tissues Reveals Wilty2/TUB6:Wi3 Interactions Associated With Stem Vascular Bundle Development.

Plant vasculature is essential for the transport of water, nutrients, and signaling molecules across organs, while also providing critical mechanical support for growth and development. Disruptions in vascular bundle formation can therefore lead to severe physiological and developmental defects. In maize, ethyl methanesulfonate (EMS)-induced dominant nonallelic Wilty mutants exhibit a pronounced wilting phenotype even under well-watered conditions, indicating underlying defects in vascular function. In this study, we characterized the Wi3 mutant, identified as ZmDrought-Overly-Sensitive1/DOS1,
