Valsa mali negative strand RNA virus 1 (VmNSRV1), a fungal virus infecting Valsa mali and belonging to the Phenuiviridae family, was recently shown to be capable of naturally infecting plants. Its RNA2 encodes a homologue of a plant virus movement protein (MP) that can complement MP-deficient virus vectors and localizes to plasmodesmata, allowing natural cross-kingdom infection. To determine whether this property is a common feature among mycoviruses possessing distant MP homologues or an isolate-specific trait, we investigated Trichoderma gamsii cogu-like virus 1 (TgClV1), a Phenuiviridae mem

The chloroplast, a key organelle for plant immunity, is frequently targeted by viral proteins to suppress host defense. Here, we demonstrate that NSvc4, the movement protein of rice stripe virus (Tenuivirus oryzaclavatae; genus Tenuivirus), functions as a chloroplast-localized virulence effector. We show that NSvc4 enters chloroplasts and directly associates with NbPsbQ, a subunit of the oxygen-evolving complex (OEC) of Photosystem II. This interaction competitively disrupts the binding of NbPsbQ to its native partners NbPsbO and NbPsbP, thereby dampening the accumulation of chloroplast-derive

A number of viral proteins from plant viruses, other than movement proteins, have been shown to traffic intracellularly along actin filaments and to be involved in viral infection. However, there has been no report that a viral capsid protein may traffic within a cell by utilizing the actin/endoplasmic reticulum (ER) network. We used Tomato spotted wilt tospovirus (TSWV) as a model virus to study the cell biological properties of a nucleocapsid (N) protein. We found that TSWV N protein was capable of forming highly motile cytoplasmic inclusions that moved along the ER and actin network. The di

Autophagy is an evolutionarily conserved degradation and recycling pathway that plays essential roles in plant development, stress adaptation, and immunity. Over the past decade, increasing evidence has revealed that autophagy also shapes the outcome of plant-virus interactions. This review summarizes current understanding of the plant autophagy machinery as well as the experimental approaches used to monitor autophagic flux during viral infection. We then explore the diverse roles of autophagy in the plant-virus interface, highlighting its function as an antiviral mechanism that targets viral

The intercellular movement of plant viruses requires both viral and host proteins. Previous studies have demonstrated that the frame-shift protein P3N-PIPO (for the protein encoded by the open reading frame [ORF] containing 5'-terminus of P3 and a +2 frame-shift ORF called Pretty Interesting Potyviridae ORF and embedded in the P3) and CYLINDRICAL INCLUSION (CI) proteins were required for potyvirus cell-to-cell movement. Here, we provide genetic evidence showing that a Tobacco vein banding mosaic virus (TVBMV; genus Potyvirus) mutant carrying a truncated PIPO domain of 58 amino acid residues co

An important aspect of plant-virus interaction is the way viruses dynamically move over long distances and how plant immunity modulates viral systemic movement. Salicylic acid (SA), a well-characterized hormone responsible for immune responses against virus, is activated through different transcription factors including TGA and WRKY. In tobamoviruses, evidence suggests that capsid protein (CP) is required for long-distance movement, although its precise role has not been fully characterized yet. Previously, we showed that the CP of Tobacco Mosaic Virus (TMV)-Cg negatively modulates the SA-medi

Lipid droplets (LDs), which are dedicated to storing neutral lipids (NLs), are dynamic organelles involved in numerous other functions, including membrane remodeling during abiotic stress. While the hijacking of LDs by animal viruses is well documented, their role(s) in plant virus infection remains largely unexplored. Similar to animal viruses, plant potyviruses reroute host proteins, intracellular membranes, and lipids to create an optimized microenvironment for their efficient viral replication compartment (VRC) assembly and movement. We therefore investigated whether potyviruses can also e

Seven isolates of a putative cytorhabdovirus (family Rhabdoviridae, order Mononegavirales) designated as citrus-associated rhabdovirus (CiaRV) were identified in citrus, passion fruit, and paper bush from the same geographical area in China. CiaRV, bean-associated cytorhabdovirus (Brazil), and papaya virus E (Ecuador) should be taxonomically classified in the species Papaya cytorhabdovirus. Due to natural mutations, the glycoprotein (G) and P4 genes were impaired in citrus-infecting isolates of CiaRV, resulting in an atypical rhabdovirus genome organization of 3' leader-N-P-P3-M-L-5' trailer.

The effect of high-pressure treatment (200-600 MPa for 20 min) on the texture of cherry tomatoes and on the key softening enzymes (pectinmethylesterase and polygalacturonase) was investigated. When subjected to high-pressure treatment whole cherry tomatoes showed increasing textural damage with increasing pressures up to 400 MPa. However, treatment at pressures above 400 MPa (500-600 MPa) led to less apparent damage than treatment at 300 and 400 MPa; the tomatoes appearing more like the untreated samples. These visual changes were reflected in the texture (firmness) and amount of cell rupture

Potyvirus rapae (turnip mosaic virus, TuMV) is widely used as a model system in plant-virus interaction studies. The TuMV RNA genome encodes 11 proteins, some of which remain poorly characterized, while the functions of others are well defined. Studying individual proteins in isolation may not recapitulate native expression levels, subcellular localization, and interaction with host factors during virus replication and movement. An alternative approach is to tag individual viral proteins in the context of an infectious clone. Epitope tags may alter protein functions and affect viral replicatio

Expression of recombinant vaccine antigens and monoclonal antibodies using plant viral vectors has developed extensively during the past several years. The approach benefits from high yields of recombinant protein obtained within days after transient delivery of viral vectors to leaves of Nicotiana benthamiana, a tobacco relative. Modified viral genomes of both RNA and DNA viruses have been created. Geminiviruses such as bean yellow dwarf virus (BeYDV) have a small, single stranded DNA genome that replicates in the nucleus of an infected plant cell, using the cellular DNA synthesis apparatus a

Viruses alter host gene expression to create a proviral environment, while the host simultaneously regulates gene expression to restrict viral spread. Owing to RNA viruses' complete reliance on the host translational machinery, it is important to assess translational control during virus infection. Therefore, we used ribosome profiling (Ribo-seq) paired with RNA-seq to observe how red clover necrotic mosaic virus (RCNMV) infection of Arabidopsis plants alters cellular gene expression at the levels of mRNA abundance and translation efficiency. We determined that at 5 days post-inoculation (dpi)

Understanding the innate immune mechanisms of plants is necessary for the breeding of disease-resistant lines. Previously, we identified the antiviral resistance gene JAX1 from Arabidopsis thaliana, which inhibits infection by potexviruses. JAX1 encodes a unique jacalin-type lectin protein. In this study, we analyzed the molecular mechanisms of JAX1-mediated resistance. JAX1 restricted the multiplication of a potexviral replicon lacking movement-associated proteins, suggesting inhibition of viral replication. Therefore, we developed an in vitro potato virus X (PVX) translation/replication syst

IRE1, BI-1, and bZIP60 monitor compatible plant-potexvirus interactions though recognition of the viral TGB3 protein. This study was undertaken to elucidate the roles of three IRE1 isoforms, the bZIP60U and bZIP60S, and BI-1 roles in genetic reprogramming of cells during potexvirus infection. Experiments were performed using Arabidopsis thaliana knockout lines and Plantago asiatica mosaic virus infectious clone tagged with the green fluorescent protein gene (PlAMV-GFP). There were more PlAMV-GFP infection foci in ire1a/b, ire1c, bzip60, and bi-1 knockout than wild-type (WT) plants. Cell-to-cel

A previously undescribed group of related plant viruses has recently been identified within the Benyviridae family. This group is called reclovirids, named after the type species, red clover RNA virus 1 (RCRV1). The single genomic RNA of most reclovirids contains two main open reading frames (ORFs): a large, well-conserved 5′-proximal ORF1 that encodes a viral replicase and a less conserved 5′-distal ORF2 that encodes a polypeptide of unknown function with a C4-type zinc finger motif. In this study, the ORF2 proteins of RCRV1 and a Gymnadenia rhellicani virus, another reclovirid, are found to

Infection of plant cells by RNA viruses leads to the generation of organelle-like subcellular structures that contain the viral replication complex. During Turnip mosaic virus (TuMV) infection of Nicotiana benthamiana, the viral membrane protein 6K2 plays a key role in the release of motile replication vesicles from the host endoplasmic reticulum (ER). Here, we demonstrate that 6K2 contains a GxxxG motif within its predicted transmembrane domain that is vital for TuMV infection. Replacement of the Gly with Val within this motif inhibited virus production, and this was due to a relocation of th

Grapevine fanleaf virus (GFLV) and arabis mosaic virus (ArMV) are nepoviruses responsible for grapevine degeneration. They are specifically transmitted from grapevine to grapevine by two distinct ectoparasitic dagger nematodes of the genus Xiphinema. GFLV and ArMV move from cell to cell as virions through tubules formed into plasmodesmata by the self-assembly of the viral movement protein. Five surface-exposed regions in the coat protein called R1 to R5, which differ between the two viruses, were previously defined and exchanged to test their involvement in virus transmission, leading to the i

Given the dearth of effective antiviral drugs, the exogenous delivery of dsRNA for RNAi against plant viral diseases holds great promise. Here, we present an effective delivery approach of dsRNA utilizing graphene oxide nanosheets (GONs) on mature plant leaves via a spray. Our method achieves rapid and sustained gene knockdown, reducing the level of the target gene to 46% by day 2, and continuously releases dsRNA for at least 6 days. The coupling of GONs with specific fragments of coat protein and replicase gene dsRNA exhibited a superior antiviral effect compared to specific fragments of RNA-

The turnip mosaic virus (TuMV) is an RNA virus that poses a significant threat to crops. The coat protein (CP) plays a crucial role in the viral life cycle, including mediating host infection and cell-to-cell movement, which makes it an ideal target for antiviral drug development. In this study, a series of structurally diverse synthetic indolylureas were screened against TuMV CP using high-throughput methods such as isothermal titration calorimetry (ITC) and microscale thermophoresis (MST). This led to the identification of compound SST21, which exhibits a high affinity for TuMV CP. Further e

The plant sterol biosynthesis pathway is essential for normal plant growth and development, but its role in plant-virus interactions remains poorly understood. In this study, we investigated the regulation of sterol methyltransferase 2 (SMT2), a key enzyme of the plant sterol biosynthesis pathway, during rice stripe virus (RSV) infection. Our findings demonstrate that Nicotiana benthamiana SMT2 (NbSMT2) is transcriptionally upregulated during RSV infection. Together with sterol 4α-methyl oxidase 2 and sterol Δ(7)-reductase, two other enzymes in the sterol biosynthesis pathway, NbSMT2 positivel
