Ca-ATPase of human myometrium plasma membranes.

We determined and characterized the Mg2+-dependent, Ca2+-stimulated ATPase (Ca-ATPase) activity in cell plasma membranes from the myometrium of pregnant women, and compared these characteristics to those of the active Ca2+-transport already demonstrated in this tissue. Similarly to the Ca2+-transport system, the Ca2+-ATPase is Mg2+-dependent, stimulated by calmodulin, and inhibited by vanadate. The Km for Ca2+ activation is 0.40 microM, very similar to that found for active calcium transport, i.e. 0.25 microM. Consequently, this Ca2+-ATPase can be responsible for the active calcium transport a

The involvement of calcium carriers and of the vacuole in the glucose-induced calcium signaling and activation of the plasma membrane H(+)-ATPase in Saccharomyces cerevisiae cells.

Previous work from our laboratories demonstrated that the sugar-induced activation of plasma membrane H(+)-ATPase in Saccharomyces cerevisiae is dependent on calcium metabolism with the contribution of calcium influx from external medium. Our results demonstrate that a glucose-induced calcium (GIC) transporter, a new and still unidentified calcium carrier, sensitive to nifedipine and gadolinium and activated by glucose addition, seems to be partially involved in the glucose-induced activation of the plasma membrane H(+)-ATPase. On the other hand, the importance of calcium carriers that can rel

Time-resolved proteolipidomics of the human aorta identifies stage-specific lipid-protein modules across development and aging.

An established vascular network is a prerequisite to ensuring an optimal supply of oxygen and nutrients for sustaining developmental events and systemic function. Herein, we construct a time-resolved proteolipidomic atlas of the aorta across the human life cycle. trans-omics integration reveals that postnatal ganglioside GM3 accumulation is functionally coregulated with calcium homeostasis mediated by plasma membrane calcium-transporting ATPases (PMCAs). We then verify mechanistically in senescence-induced primary vascular smooth muscle cells (VSMCs) that knockdown of GM3 synthase leads to dim

Kinetics of erythrocyte plasma membrane (Ca2+, Mg2+)ATPase in familial benign hypercalcemia.

The pathogenesis of familial benign hypercalcemia (FBH) is unknown. Possible explanations for the disorder include a set-point error in parathyroid gland regulation and intrinsic renal hyperreabsorption of calcium. Thus, FBH may involve an alteration in cellular calcium transport, especially in renal and parathyroid cells. A primary mediator of cellular calcium transport is (Ca2+,Mg2+)ATPase. Therefore, we examined in detail the kinetics of (Ca2+,Mg2+)ATPase activity in erythrocyte plasma membranes from 11 patients with FBH from 7 families, 5 patients with untreated primary hyperparathyroidism

The enzymes of the red blood cell plasma membrane.

Red blood cell plasma membranes contain a number of enzymes: ATPases, anion transport protein, glyceraldehyde 3-phosphate dehydrogenase, protein kinases, adenylate cyclase, acetylcholinesterase. Most of them are tightly bound to the membrane and are present in small amounts. As a result, structural characterization of erythrocyte membrane enzymes has not yet been successful. Functional studies have, however, yielded a great deal of information. ATPases allow active transport of cations (calcium, sodium, potassium). Anion transport protein controls movements of chloride and phosphate ions, and

The plasma membrane calcium ATPase and disease.

The plasma membrane calcium ATPase (PMCA) uses energy to pump calcium (Ca2+) ions out of the cytosol into the extracellular milieu, usually against a strong chemical gradient. This energy expenditure is necessary to maintain a relatively low intracellular net Ca2+ load. Mammals have four genes (ATP2B1-ATP2B4), encoding the proteins PMCA1 through PMCA4. Transcripts from each of these genes are alternatively spliced to generate several variant proteins that are in turn post-translationally modified in a variety of ways. Expressed ubiquitously and with some level of functional redundancy in most

Characterization of calcium efflux by osteoblasts derived from long bone periosteum.

Inside-out vesicles were prepared from the plasma membrane of osteoblasts which had been isolated from the periosteum of 2-3-week-old chicken tibia and cultured for 6-8 days. Calcium uptake was initiated by adding 0.4 mM ATP and detected as a reduction in fluorescence from the reaction medium using the Ca(2+)-specific fluoroprobe, fluo-3. The reaction medium contained ouabain (1 mM) to block Na+, K(+)-ATPase activity and oligomycin (20 micrograms/ml) to block mitochondrial activity. Thapsigargin (5 microM) had little effect, indicating that contributions to Ca2+ uptake by endoplasmic reticulum

The plasma membrane calcium pumps: focus on the role in (neuro)pathology.

The plasma membrane Ca2+ ATPase (PMCA pump) is a member of the superfamily of P-type pumps. It is organized in the plasma membrane with ten transmembrane helices and two main cytosolic loops, one of which contains the catalytic center. It also contains a long C-terminal tail that houses the binding site for calmodulin, the main regulator of the activity of the pump. The pump also contains a number of other regulators, among them acidic phospholipids, kinases, and numerous protein interactors. Separate genes code for 4 basic pump isoforms in mammals, additional isoform complexity being generate

Evidence for a role of plasma membrane calcium pumps in neurodegenerative disease: Recent developments.

Plasma membrane Ca2+ ATPases (PMCAs) are a major system for calcium extrusion from all cells. Different PMCA isoforms and splice variants are involved in the precise temporal and spatial handling of Ca2+ signals and the re-establishment of resting Ca2+ levels in the nervous system. Lack or inappropriate expression of specific PMCAs leads to characteristic neuronal phenotypes, which may be reciprocally exacerbated by genetic predisposition through alleles in other genes that modify PMCA interactions, regulation, and function. PMCA dysfunction is often poorly compensated in neurons and may lead

Effect of acylphosphatase on human erythrocyte membrane Ca2(+)-ATPase.

We studied the effect of human acylphosphatase on the activity of human erythrocyte membrane Ca2(+)-ATPase. Both the acylphosphatase that is contained in hemolysate and the purified enzyme isolated from red blood cells were able to stimulate Ca2(+)-ATPase activity in erythrocyte membranes. Given the same acylphosphatase activity, however, the hemolysate showed higher stimulatory effect than the purified enzyme. Acylphosphatase stimulation was additive to that induced by calmodulin, thus indicating that acylphosphatase acts in a calmodulin-independent manner. Trifluoperazine, a calmodulin antag

Experimental porcine malignant hyperthermia: the activity of certain transporting enzymes and myofibrillar calcium-binding protein content in the muscle fibre.

The activity of enzymes transporting calcium across the membrane of the muscle fibre and sarcoplasmic reticulum was studied. The activity of actomyosin Mg2+-ATPase and the composition of troponin were investigated also. In seven Pietrain pigs halothane alone or with suxamethonium induced the malignant hyperthermia syndrome. In the animals with malignant hyperthermia Ca2+-ATPase was not activated in the plasma membranes of the muscle fibres, when compared with control animals (n = 4). An increase of Ca2+-ATPase of the sarcoplasmic reticulum was observed in animals with malignant hyperthermia; t

Biallelic ATP2B1 variants as a likely cause of a novel neurodevelopmental malformation syndrome with primary hypoparathyroidism.

ATP2B1 encodes plasma membrane calcium-transporting-ATPase1 and plays an essential role in maintaining intracellular calcium homeostasis that regulates diverse signaling pathways. Heterozygous de novo missense and truncating ATP2B1 variants are associated with a neurodevelopmental phenotype of variable expressivity. We describe a proband with distinctive craniofacial gestalt, Pierre-Robin sequence, neurodevelopmental and growth deficit, periventricular heterotopia, brachymesophalangy, cutaneous syndactyly, and persistent hypocalcemia from primary hypoparathyroidism. Chemosensory signalling in human sperm is controlled by Ca2+ influx via CatSper and Ca2+ clearance via plasma membrane Ca2+ ATPases.

BACKGROUND AND PURPOSE: Loss of function of the sperm-specific Ca2+ channel CatSper is a common channelopathy that causes male infertility. CatSper controls the intracellular Ca2+ concentration and, thereby, the motility of human sperm. Activation of CatSper by oviductal ligands evokes a transient Ca2+ increase, which entails changes in the flagellar beat that are required for fertilisation. The CatSper-mediated Ca2+ influx has been studied extensively, whereas the mechanisms underlying Ca2+ clearance and recovery from Ca2+ influx have remained ill-defined. EXPERIMENTAL APPROACH: We examined

Calcium transport in human salivary glands: a proposed model of calcium secretion into saliva.

Salivary calcium plays a vital role in bio-mineralization of dental enamel and exposed dentin. In order to elucidate the yet unknown cellular and molecular mechanisms of calcium secretion in human salivary glands the presence of various relevant plasma membrane transport systems for calcium were investigated. Using an RT-PCR approach, expression of the epithelial calcium channel (CaT-Like), the calcium binding protein (calbindin-2), the endoplasmic reticulum pumps (SERCA-2 and -3), and the plasma membrane calcium ATPases (PMCA-1, -2, and -4), were found in parotid and submandibular glands. Plasma membrane Ca(2+)-ATPase: from a housekeeping function to a versatile signaling role.

Plasma membrane Ca(2+)-ATPases (PMCAs) are high-affinity calcium pumps that contribute to the maintenance of intracellular Ca(2+) homeostasis by exporting Ca(2+) from the cytosol to the extracellular environment. Mammals have four genes encoding the proteins PMCA1 through PMCA4. Each gene transcript is alternatively spliced to generate several variants. Their distribution is tissue- and cell-specific and undergoes regulation during cell development and differentiation. Traditionally, these pumps have been considered to play a housekeeping role in controlling basal Ca(2+) levels, but more recen

Immunolabel imaging of plasma membrane Ca++ pumps (PMCA-pan or PMCA2) implicates the apical cilium on receptor cells as the transduction site for electrosense in two Acipenseriformes fishes.

Specialized receptor cells in ampulla of Lorenzini-like electroreceptors enable Acipenseriformes fishes to detect microvolt signals from aquatic prey. Candidate subcellular sites of electrosensory transduction include each receptor cell's apical face membrane, or its modified primary cilium, or sparse apical microvilli. We aimed to more precisely localize the apical transduction. Given its reported Ca++ dependence, as proxy we immunolabeled electrosensory epithelia for Ca++ ion pumps (PMCA: plasma membrane calcium ATPase), in frozen sections of ampullary organs from rostrum skin of freshwater

Polyamine transport regulation by calcium and calmodulin: role of Ca(2+)-ATPase.

The study was conducted on human leukemia (K 562) cells to characterize the mechanisms implicated in the regulation of the polyamine spermidine (Spd) transport process. The antagonists of calmodulin, trifluoperazine (TFP), W-7 (N-[6-aminohexyl]-5-chloro-1-naphthelenesulfonamide), or mellitin inhibited significantly polyamine Spd uptake in these cells. The translocation of calmodulin towards plasma membrane and a concomitant decrease in its contents in cytosol were directly correlated with the time course increases similar to that of Spd uptake, indicating that calmodulin is recruited towards p

Plasma membrane calcium ATPases and cerebellar pathology: what's the role in the ataxia?

Ca²⁺ signaling is essential for neuronal development, migration, synaptic activity, spine plasticity, neurotransmitter release, membrane excitability, and long-term synaptic plasticity, as well as for the coupling between membrane depolarization and downstream signaling. Traditionally, Plasma Membrane Ca²⁺ ATPases (PMCAs) were considered high-affinity, low-capacity calcium extruders. However, recent evidence reveals that the PMCA-Neuroplastin complex facilitates ultrafast Ca²⁺ clearance at kilohertz frequencies, reshaping our understanding of calcium regulation, in particular in neurons. Plasma membrane Ca2+ ATPase 1 (PMCA1) but not PMCA4 is critical for B-cell development and Ca2+ homeostasis in mice.

The amplitude and duration of Ca2+ signaling is crucial for B-cell development and self-tolerance; however, the mechanisms for terminating Ca2+ signals in B cells have not been determined. In lymphocytes, plasma membrane Ca2+ ATPase (PMCA) isoforms 1 and 4 (PMCA1 and PMCA4, aka ATP2B1 and ATP2B4) are the main candidates for expelling Ca2+ from the cell through the plasma membrane. We report here that Pmca4 (Atp2b4) KO mice had normal B-cell development, while mice with a conditional KO of Pmca1 (Atp2b1) had greatly reduced numbers of B cells, particularly splenic follicular B cells, marginal z

Regulation of PKC mediated signaling by calcium during visceral leishmaniasis.

Calcium is an ubiquitous cellular signaling molecule that controls a variety of cellular processes and is strictly maintained in the cellular compartments by the coordination of various Ca2+ pumps and channels. Two such fundamental calcium pumps are plasma membrane calcium ATPase (PMCA) and Sarco/endoplasmic reticulum calcium ATPase (SERCA) which play a pivotal role in maintaining intracellular calcium homeostasis. This intracellular Ca2+ homeostasis is often disturbed by the protozoan parasite Leishmania donovani, the causative organism of visceral leishmaniasis. In the present study we have
