Arabinogalactan proteins (AGPs) are a plant-specific family of extracellular proteoglycans characterized by large and complex galactose-rich polysaccharide chains. Functional elucidation of AGPs, however, has been hindered by the high degree of redundancy of AGP genes. To uncover as yet unexplored roles of AGPs in Arabidopsis, a mutant of Hyp O-galactosyltransferase (HPGT), a critical enzyme that catalyzes the common initial step of Hyp-linked arabinogalactan chain biosynthesis, was used. Here we show, using the hpgt1,2,3 triple mutant, that a reduction in functional AGPs leads to a stomatal p

In plants, mounting an effective innate immune strategy against microbial pathogens involves triggering local cell death within infected cells as well as boosting the immunity of the uninfected neighboring and systemically located cells. Although not much is known about this, it is evident that well-coordinated cell-cell signaling is critical in this process to confine infection to local tissue while allowing for the spread of systemic immune signals throughout the whole plant. In support of this notion, direct cell-to-cell communication was recently found to play a crucial role in plant defen

Systemic acquired resistance (SAR) in plants is mediated by the signaling molecules azelaic acid (AzA), glycerol-3-phosphate (G3P), and salicylic acid (SA). Here, we show that AzA and G3P transport occurs via the symplastic route, which is regulated by channels known as plasmodesmata (PD). In contrast, SA moves via the extracytosolic apoplast compartment. We found that PD localizing proteins (PDLP) 1 and 5 were required for SAR even though PD permeability in pdlp1 and 5 mutants was comparable to or higher than wild-type plants, respectively. Furthermore, PDLP function was required in the recip

Plant cell-cell communication plays a crucial role during growth and development. While plasmodesmata (PD) are well known in this communication, their biogenesis and regulation remain poorly understood. In this study, we demonstrate that xyloglucan endotransglucosylases (XETs), enzymes involved in transglycosylation of xyloglucan chains, could impact not only cellular growth but also PD development in the moss Physcomitrium patens (P. patens). Knockout mutant lines of two XET isoforms, named Ppxth5, Ppxth6, and Ppxth5,6dko, showed growth reductions in both protonemal and gametophytic stages. S

Plasmodesmata (PD)-mediated intercellular communication is critical for plant stress adaptation, yet its dynamic regulation by the actin cytoskeleton under abiotic stress remains poorly characterised. Here, we show that rice OsFH14 modulates salt-induced PD permeability to enhance salt tolerance. Through CRISPR-Cas9 mutagenesis and subcellular localisation analyses, we first demonstrate that OsFH14 specifically localises to PD and is strictly required for salt stress-induced increases in PD permeability. Osfh14 mutants exhibit reduced PD permeability and enhanced sensitivity to NaCl treatment.

Viruses are obligate intracellular parasites that rely on their hosts for the cellular machinery necessary for their replication and transmission to new hosts. Viruses are therefore invaluable for illuminating numerous facets of their hosts' biology. Plant viruses take advantage of the cell-to-cell connectivity provided by plasmodesmata for their movement from sites of replication to the vasculature for eventual systemic dissemination in their plant hosts, a strategy distinct from viruses that use animal and fungal hosts. Given their reliance on plasmodesmata for successful infection and sprea

Numerous cell surface receptors and receptor-like proteins (RLPs) undergo activation or deactivation via a transmembrane domain (TMD). A subset of plant RLPs distinctively localizes to the plasma membrane-lined pores called plasmodesmata. Those RLPs include the Arabidopsis thaliana Plasmodesmata-located protein (PDLP) 5, which is well known for its vital function regulating plasmodesmal gating and molecular movement between cells. In this study, we report that the TMD, although not a determining factor for the plasmodesmal targeting, serves essential roles for the PDLP5 function. In addition t

Plasmodesmata (PDs) play vital roles in plant growth and defense by controlling the symplastic transport of important molecules. Here we report that a conserved COBRA-like protein, COBL3, positively regulates callose accumulation and is required for PD-mediated antiviral defense (PMAD) against divergent plant RNA viruses in wheat (Triticum aestivum) and tobacco (Nicotiana benthamiana). The wheat COBL3 protein, TaCOBL3, interacts with the 17K movement protein (MP) of barley yellow dwarf virus-GAV (BYDV-GAV). TaCOBL3 is associated with the plasma membrane and co-localizes with 17K MP at PDs. Gen

The intercellular movement of plant viruses requires both viral and host proteins. Previous studies have demonstrated that the frame-shift protein P3N-PIPO (for the protein encoded by the open reading frame [ORF] containing 5'-terminus of P3 and a +2 frame-shift ORF called Pretty Interesting Potyviridae ORF and embedded in the P3) and CYLINDRICAL INCLUSION (CI) proteins were required for potyvirus cell-to-cell movement. Here, we provide genetic evidence showing that a Tobacco vein banding mosaic virus (TVBMV; genus Potyvirus) mutant carrying a truncated PIPO domain of 58 amino acid residues co

The plant immune system relies on the perception of molecules that signal the presence of a microbe threat. This triggers signal transduction that mediates a range of cellular responses via a collection of molecular machinery including receptors, small molecules, and enzymes. One response to pathogen perception is the restriction of cell-to-cell communication by plasmodesmal closure. We previously found that while chitin and flg22 trigger specialized immune signaling cascades in the plasmodesmal plasma membrane, both execute plasmodesmal closure via callose synthesis at the plasmodesmata. Super-resolution microscopy (SRM) has transformed the study of cellular structures, enabling imaging beyond the diffraction limit. Yet, the need for costly instrumentation has limited its accessibility. Expansion Microscopy microscopy (ExM), pioneered by Ed Boyden's laboratory, offers an alternative by physically enlarging samples embedded within a swellable hydrogel. This simple principle makes nanoscale resolution achievable with conventional fluorescence microscopy. Since its introduction in 2015, ExM has rapidly diversified. Iterative ExM (iExM) increases resolution through repetitive expa

Plant reticulon (RTN) proteins are capable of constricting membranes and are vital for creating and maintaining tubules in the endoplasmic reticulum (ER), making them prime candidates for the formation of the desmotubule in plasmodesmata (PD). RTN3 and RTN6 have previously been detected in an Arabidopsis PD proteome and have been shown to be present in primary PD at cytokinesis. It has been suggested that RTN proteins form protein complexes with proteins in the PD plasma membrane and desmotubule to stabilize the desmotubule constriction and regulate PD aperture. Viral movement proteins (vMPs)

Verticillium wilt, caused by Verticillium dahliae Kleb., is a serious threat to cotton (Gossypium spp.) crop production. To enhance our understanding of the plant's complex defensive mechanism, we examined colonization patterns and interactions between V. dahliae and two cotton species, the resistant G. barbadense and the susceptible G. hirsutum. Microscopic examinations and grafting experiments showed that the progression of infection was restricted within G. barbadense. At all pre- and postinoculation sampling times, levels of salicylic acid (SA) were also higher in that species than in G. h

Phloem loading is a critical step in the transport of photosynthetically produced sugars from leaves to sink tissues. The type of phloem loading-active apoplasmic, active symplasmic, or passive symplasmic-can be inferred from the density and structure of plasmodesmata (PD) along the phloem-loading pathway. This protocol describes a method to predict phloem loading types using transmission electron microscopy (TEM) to quantify PD density. The method follows established standards for sample preparation, imaging, and data analysis, enabling comparative studies across species.

Cassava brown streak virus (CBSV) and Ugandan cassava brown streak virus (UCBSV) are responsible for significant cassava yield losses in eastern sub-Saharan Africa. To study the possible mechanisms of plant resistance to CBSVs, we inoculated CBSV-susceptible and CBSV-resistant cassava varieties with a mixed infection of CBSVs using top-cleft grafting. Transcriptome profiling of the two cassava varieties was performed at the earliest time point of full infection (28 days after grafting) in the susceptible scions. Connectivity between mesophyll (M) and bundle sheath (BS) cells must improve during the evolution of C4 photosynthesis to facilitate large metabolite fluxes between these cell types, but the trait combinations that enhance M-BS connectivity and the points at which these enhancements occur along the C3 to C4 evolutionary trajectory remain unknown. We investigated shifts in M-BS connectivity that accompanied photosynthetic diversification within the eudicot genus Alternanthera from the agriculturally important Amaranthaceae family. We combined transmission electron microscope and confocal micros

Valsa mali negative strand RNA virus 1 (VmNSRV1), a fungal virus infecting Valsa mali and belonging to the Phenuiviridae family, was recently shown to be capable of naturally infecting plants. Its RNA2 encodes a homologue of a plant virus movement protein (MP) that can complement MP-deficient virus vectors and localizes to plasmodesmata, allowing natural cross-kingdom infection. To determine whether this property is a common feature among mycoviruses possessing distant MP homologues or an isolate-specific trait, we investigated Trichoderma gamsii cogu-like virus 1 (TgClV1), a Phenuiviridae mem

Plasmodesmata (PD) are plasma membrane-lined cytoplasmic nanochannels that mediate cell-to-cell communication across the cell wall. A range of proteins are embedded in the PD plasma membrane and endoplasmic reticulum (ER), and function in regulating PD-mediated symplasmic trafficking. However, knowledge of the nature and function of the ER-embedded proteins in the intercellular movement of non-cell-autonomous proteins is limited. Here, we report the functional characterization of two ER luminal proteins, AtBiP1/2, and two ER integral membrane proteins, AtERdj2A/B, which are located within the

Efficient use of plant-derived materials requires enabling technologies for non-disruptive composition analysis. The ability to identify and spatially locate polysaccharides in native plant tissues is difficult but essential. Here, we develop an optical method for cellulose identification using the structure-responsive, heptameric oligothiophene h-FTAA as molecular fluorophore. Spectrophotometric analysis of h-FTAA interacting with closely related glucans revealed an exceptional specificity for β-linked glucans. This optical, non-disruptive method for stereochemical differentiation of glycosid

Systemic acquired resistance (SAR) is a form of broad-spectrum resistance induced in response to local infections that protects uninfected parts against subsequent secondary infections by related or unrelated pathogens. SAR signaling requires two parallel branches, one regulated by salicylic acid (SA), and the other by azelaic acid (AzA) and glycerol-3-phosphate (G3P). AzA and G3P function downstream of the free radicals nitric oxide (NO) and reactive oxygen species (ROS). During SAR, SA, AzA and G3P accumulate in the infected leaves, but only a small portion of these is transported to distal
