Phytomyxea (SAR: Rhizaria: Endomyxa) is a group of obligate biotrophic parasitic protists comprised of two orders: Plasmodiophorida, found in terrestrial or freshwater environments, and Phagomyxida, found in marine environments. While Plasmodiophorida has been extensively studied due to its economic importance as plant pathogens, Phagomyxida remains poorly investigated despite its ecological significance in marine ecosystems. During intensive sampling along the Korean coast from April to December 2023, novel parasitoids infecting dinoflagellates were discovered in seawater collected at 10 coas

Plasmodiophora brassicae is a soil-borne intracellular obligate biotrophic protist and the causal agent of clubroot disease in Brassicaceae. Phylogenetically placed within the Rhizaria supergroup, it exhibits a complex two-stage life cycle leading to the formation of club-shaped galls, driven by phytohormone and effector-mediated host reprogramming. A recently obtained telomere-to-telomere genome assembly of the Canadian pathotype Pb3A revealed a compact genome structure encoding a complex secretome. Despite its agronomic relevance, key aspects of its biology, including the existence of sexual

The plant hormone salicylic acid (SA) plays a critical role in defense against biotrophic pathogens such as Plasmodiophora brassicae, which is an obligate pathogen of crucifer species and the causal agent of clubroot disease of canola (Brassica napus). P. brassicae encodes a protein, predicted to be secreted, with very limited homology to benzoic acid (BA)/SA-methyltransferase, designated PbBSMT. PbBSMT has a SA- and an indole-3-acetic acid-binding domain, which are also present in Arabidopsis thaliana BSMT1 (AtBSMT1) and, like AtBSMT1, has been shown to methylate BA and SA. In support of the

Methylesterases (MES) are involved in hydrolysis of carboxylic esters, which have substantial roles in plant metabolic activities and defense mechanisms. This study aimed to comprehensively investigate Brassica napus BnMESs and characterize their role in response to Plasmodiophora brassicae stress. Forty-four BnMES members were identified and categorized into three groups based on their phylogenetic relationships and structural similarities. Through functional predictions in the promoter regions and analysis of RNA-Seq data, BnMES emerged as pivotal in growth, development, and stress responses

The clubroot pathogen Plasmodiophora brassicae has an effector protein that can methylate the plant defense compound salicylic acid (SA), and the protein sequence is highly conserved in different isolates worldwide. Sequencing the full-length cDNA of P. brassicae Benzoic acid/Salicylic acid Methyltransferase (PbBSMT) from a small collection of clubs indicated the occurrence of a natural variation that differs by 18 amino acids, which was also found within the worldwide data collection for P. brassicae isolates. A search for more possible substrates including defense-inducing compounds revealed

Clubroot, caused by the obligate parasite Plasmodiophora brassicae, is a soilborne disease affecting canola (Brassica napus) and other crucifers. Although planting resistant cultivars remains the primary strategy for managing clubroot, the emergence of resistance-breaking P. brassicae pathotypes continues to threaten canola production. In this context, soil and root microorganisms may play a role in suppressing the disease. This study investigated the impact of P. brassicae infection on the microbial communities of soil, seeds, roots, and the rhizosphere in susceptible and resistant canola lin

Didymella macrostoma P2 was isolated from rapeseed (Brassica napus), and it is an endophyte of rapeseed and an antagonist of three rapeseed pathogens, Botrytis cinerea, Leptosphaeria biglobosa, and Sclerotinia sclerotiorum. However, whether P2 has a suppressive effect on infection of rapeseed by the clubroot pathogen Plasmodiophora brassicae remains unknown. This study was conducted to detect production of antimicrobials by P2 and to determine the efficacy of the antimicrobials and P2 pycnidiospores in suppression of rapeseed clubroot. The results showed that cultural filtrates (CFs) of P2 in

Boron (B) and calcium (Ca) are essential nutrients for plant growth that have also been reported to reduce clubroot disease caused by Plasmodiophora brassicae. This study evaluated the effects of soil-applied B (0, 8 or 16 kg B ha-1) and Ca (0, 500 or 1000 kg Ca ha-1) on clubroot development in Brassica napus lines differing in B sensitivity, and investigated underlying mechanisms. Ca consistently reduced clubroot severity, whereas B had a weaker, inconsistent effect and caused phytotoxicity at higher rates. Ca also alleviated B toxicity and increased shoot biomass under high B conditions. Fou

The role of salicylic acid (SA) and jasmonic acid (JA) signaling in resistance to root pathogens has been poorly documented. We assessed the contribution of SA and JA to basal and partial resistance of Arabidopsis to the biotrophic clubroot agent Plasmodiophora brassicae. SA and JA levels as well as the expression of the SA-responsive genes PR2 and PR5 and the JA-responsive genes ARGAH2 and THI2.1 were monitored in infected roots of the accessions Col-0 (susceptible) and Bur-0 (partially resistant). SA signaling was activated in Bur-0 but not in Col-0. The JA pathway was weakly activated in Bu

The role of primary metabolism during Brassica napus-Plasmodiophora brassicae interaction leading to clubroot resistance has not yet been investigated thoroughly. In this study, we investigated some of the primary metabolites and their derivatives as well as expression of the genes involved in their biosynthesis to decipher this host-pathogen interaction. For this, two sets (clubroot resistant and susceptible) of canola lines were inoculated with P. brassicae pathotype 3A to investigate the endogenous levels of primary metabolites at 7-, 14-, and 21-days after inoculation (DAI). The associated

Brassica oleracea var. capitata (cabbage) is an important vegetable crop in Asian countries such as Korea, China, and Japan. Cabbage production is severely affected by clubroot disease caused by the soil-borne plant pathogen Plasmodiophora brassicae. During clubroot development, methyl salicylate (MeSA) is biosynthesized from salicylic acid (SA) by methyltransferase. In addition, methyl salicylate esterase (MES) plays a major role in the conversion of MeSA back into free SA. The interrelationship between MES and methytransferases during clubroot development has not been fully explored. To begi

The obligate biotrophic pathogen Plasmodiophora brassicae causes clubroot disease in Arabidopsis thaliana, which is characterized by large root galls. Salicylic acid (SA) production is a defence response in plants, and its methyl ester is involved in systemic signalling. Plasmodiophora brassicae seems to suppress plant defence reactions, but information on how this is achieved is scarce. Here, we profile the changes in SA metabolism during Arabidopsis clubroot disease. The accumulation of SA and the emission of methylated SA (methyl salicylate, MeSA) were observed in P. brassicae-infected Arab

Biotrophic pathogens depend on host carbon sources for proliferation. Here, we identified BnaA07.SUC2 in oilseed rape (Brassica napus cv. Westar), which encodes a plasma membrane-localized proton-dependent sucrose transporter. Its expression is markedly induced in roots during the late stages of Plasmodiophora brassicae infection. The Arabidopsis thaliana suc2 mutant exhibited a strong clubroot-resistant phenotype, and functional complementation with BnaA07.SUC2 restored susceptibility to P. brassicae. Overexpression of BnaA07.SUC2 in oilseed rape significantly increased sucrose accumulation a

Salicylic acid (SA) biosynthesis, the expression of SA-related genes and the effect of SA on the Arabidopsis-Plasmodiophora brassicae interaction were examined. Biochemical analyses revealed that, in P. brassicae-infected Arabidopsis, the majority of SA is synthesized from chorismate. Real-time monitored expression of a gene for isochorismate synthase was induced on infection. SA can be modified after accumulation, either by methylation, improving its mobility, or by glycosylation, as one possible reaction for inactivation. Quantitative reverse transcription-polymerase chain reaction (qPCR) co
