BACKGROUND: Colour polymorphism in aposematic species is often associated with ecological divergence and predator selection. In amphibians, colouration is achieved through the chromatophore unit, a layered structure of pigment-containing cells (xanthophores, melanophores) and structural cells (iridophores) that interact to produce the final skin colouration. In dendrobatid frogs of the genus Oophaga, striking red and green morphs coexist within and across species, yet the cellular mechanisms underlying this variation remain poorly understood.

Neotropical poison frogs (Dendrobatidae) contain a variety of lipophilic alkaloids in their diffusely distributed cutaneous glands, including a major class of compounds known as pumiliotoxins. Pumiliotoxins are highly toxic and are believed to protect frogs against predators. Their potential activity against ectoparasites, however, has not been investigated. We tested female yellow fever mosquitoes (Aedes aegypti) for responses to 8-hydroxy-8-methyl-6-(2'-methylhexylidene)-1-azabicyclo[4.3.0]nonane, designated pumiliotoxin 251D [PTX (+)-251D], a skin alkaloid present in all genera of dendrobat

Parent-offspring interactions constitute the first contact of many newborns with their environment, priming community assembly of microbes through priority effects and shaping host health and disease. Microbe acquisition during parental care is well studied in humans and agriculturally relevant species but remains poorly understood in other vertebrate groups, such as amphibians. Here, we investigate vertical transmission of skin microbiota in poison frogs (Dendrobatidae), where fathers transport tadpoles piggyback-style from terrestrial clutches to aquatic nurseries. We found that substantial

Amphibians are currently experiencing the most severe declines in biodiversity of any vertebrate, and their requirements for successful reproduction are poorly understood. Here, we show that supplementing the diet of prey items (fruit flies) with carotenoids has strong positive effects on the reproduction of captive strawberry poison frogs (Oophaga pumilio), substantially increasing the number of metamorphs produced by pairs. This improved reproduction most likely arose via increases in the quality of both the fertilized eggs from which tadpoles develop and trophic eggs that are fed to tadpole

Laterality, the preference for using one side of the body over the other, has been observed in various animal species, including frogs and toads. While laterality has been studied widely during locomotion in anurans, its role during other behaviours, such as hunting-related toe-tapping (the quick up and down movement of the middle toe of the hind feet while foraging), remains largely unstudied. This study investigated the toe-tapping rate of the left and right toes (versus both toes simultaneously) in a captive colony of green-and-black poison dart frogs (Dendrobates auratus, Dendrobatidae). Defensive mechanisms, including noxious or toxic substances, are favored by predation-driven natural selection. The acquisition of noxious/toxic substances can be either endogenous, in which the substances are produced by the organism, or exogenous, in which the substances are produced by another organism and are sequestered. Evidence indicates that the defensive skin alkaloids of Neotropical poison frogs (Dendrobatidae) have an exogenous source: a diet of ants and other small alkaloid-containing arthropods, which we term the diet-toxicity hypothesis. A critical prediction of the diet-toxicity

Complex phenotypes typically have a correspondingly multifaceted genetic component. However, the genotype-phenotype association between chemical defense and resistance is often simple: genetic changes in the binding site of a toxin alter how it affects its target. Some toxic organisms, such as poison frogs (Anura: Dendrobatidae), have defensive alkaloids that disrupt the function of ion channels, proteins that are crucial for nerve and muscle activity. Using protein-docking models, we predict that three major classes of poison frog alkaloids (histrionicotoxins, pumiliotoxins, and batrachotoxin

The authors present a case of a patient who used Kambo frog poison for body cleansing that induced severe vomiting and led to esophageal rupture followed by tension pneumothorax and septic shock. Kambo is the waxy substance secreted by the nocturnal giant tree frog Phyllomedusa bicolor. Kambo, which is poisonous, is commonly believed in South America to have cleansing and healing properties. As alternative medicine becomes more common, and as more tourists frequent our hospitals, knowledge of these types of ritual related exposures is important for the practicing emergency physician to be awar

Few ultrastructural studies have been performed on members of the Dendrobatidae, although such investigations can be useful for the understanding of reproductive patterns, as a diagnostic method for males in breeding programs for endangered amphibians and for phylogenetic analysis. The sperm ultrastructure of the Green Poison Frog, Dendrobates auratus, from Panama is described following induced spermiation in living animals. To date only testicular spermatozoa in other dendrobatid frogs have been analysed. Moreover, an electron microscopic preparation method (transmission and scanning electron

Batrachotoxins are neurotoxic steroidal alkaloids first isolated from a Colombian poison-dart frog and later found in certain passerine birds of New Guinea. Neither vertebrate group is thought to produce the toxins de novo, but instead they likely sequester them from dietary sources. Here we describe the presence of high levels of batrachotoxins in a little-studied group of beetles, genus Choresine (family Melyridae). These small beetles and their high toxin concentrations suggest that they might provide a toxin source for the New Guinea birds. Stomach content analyses of Pitohui birds reveale

Territorial males of the pan-Amazonian Dart-poison frog, Epipedobates femoralis, are known to present stereotypic phonotactic responses to the playback of conspecific and synthetic calls. Fixed site attachment and a long calling period within an environment of little temperature change render this terrestrial and diurnal pan-Amazonian frog a rewarding species for field bioacoustics. In experiments at the field station Aratai, French Guiana, we tested whether the prominent frequency modulation of the advertisement-call notes is critical for eliciting phonotactic responses. A non-opioid analgesic, epibatidine (1), isolated from Ecuadoran poison frogs was synthesized in the racemic form starting from a readily available compound 2. A partial Curtius rearrangement product 5 of 2 was converted into 12c by way of the ketone 3 and its condensation product with the pyridine moiety 9c, and catalytic hydrogenation of 12c was specifically conducted in hydrochloric acid-containing 2-propanol for the preferential formation of an exo-product 15b. Conversion of the substituent from 15b to 15a in a single operation using the Vilsmeier reagent, followed by deprotection of the p

Cutaneous granular glands are a shared character of adult amphibians, including caecilians, and are thought to be the source of most biologically active compounds in amphibian skin. Data are available from one or more species in over 100 of nearly 400 genera comprising the three living orders of Amphibia. Many species contain unidentified substances judged to be noxious based on predator aversion or human taste. Additionally, there is a great diversity of known compounds, some highly toxic as well as noxious, which can be tabulated under four broad categories: biogenic amines, peptides, bufodi

The ability to use small molecule alkaloids as defensive chemicals, often acquired via trophic interactions, has evolved in many organisms. Animals with diet-derived defences must balance food choices to maintain their defence reservoirs along with other physiological needs. Poison frogs accumulate skin alkaloids from their arthropod diet, but whether they show selectivity for specific prey remains unexplored. We investigated the role of leaf litter prey availability and dietary selectivity in shaping poison frog chemical defences along a geographic gradient. Specifically, we examined skin alk

Animal acoustic signals play seminal roles in mate attraction and regulation of male spacing, maintenance of pairbonds, localization of hosts by parasites, and feeding behavior. Among vertebrate signals, it is becoming clear that no single stereotyped signal feature reliably elicits species-specific behavior, but rather, that a suite of characters is involved. Within the largely nocturnal clade of anuran amphibians, the dart-poison frog, Epipedobates femoralis, is a diurnal species that physically and vigorously defends its calling territory against conspecific intruders. Here we report that p

Brain microRNA (miRNA) expression can shift in response to the social environment, translating external cues into regulatory action that can influence brain structure, neural activity, and behavior. miRNAs are key drivers of behavioral flexibility, regulating gene networks involved in neuroplasticity. We manipulated social rearing conditions of brilliant-thighed poison frog tadpoles ( Allobates femoralis ) to test whether isolation-linked behavioral changes are associated with altered miRNA expression in the midbrain and eyes, tissues linked by the retinotectal tract, a known connection for tr

The poison frog family Dendrobatidae has radiated into over 300 species throughout the past 30 million years, occupying the majority of neotropical forest habitats. Species in this group have diversified widely in multiple aspects of their biology such as predator avoidance and parental care strategies, reproductive ecology, chemical ecology, and elevational range. This diversification has been marked by dozens of historical dispersal events between the Northern Andes and their adjacent lowland forests (Amazonia, Chocó, Interandean valleys), as well as substantial in situ diversification in mo

Understanding the nutritional requirements of captive animals is necessary for proper animal husbandry, however, the specific dietary requirements for many amphibian species commonly kept in captivity are unknown. Like most vertebrates, frogs cannot synthesize carotenoids and must therefore obtain these essential nutrients through diet. It is unclear if amphibians can cleave provitamin A carotenoids to form vitamin A metabolically within the body, so common practice is to supplement their captive diets with both preformed vitamin A and provitamin A carotenoids. We carried out a feeding experim

Blue poison dart frogs (Dendrobates tinctorius azureus) are commonly maintained in zoological institutions and are becoming popular in the pet trade industry. Sedation or light anesthesia is required for safe and effective handling of this species. In this study, the sedative effects of subcutaneously administered alfaxalone-midazolam-dexmedetomidine (AMD) (20, 40, 5 mg/kg, respectively) and ketamine-midazolam-dexmedetomidine (KMD) (100, 40, 5 mg/kg, respectively) were compared in a prospective, randomized, blinded, crossover study in juvenile blue poison dart frogs (n = 10). The morphology of larvae stages of most amphibians are often completely different than in adults. Tadpole descriptions have historically been based on external characters like morphometrics, color pattern and oral disc structure. Other papers described anatomical details by the use of dissections. The increase in micro-CT scanning technology provides an opportunity to quantify and describe in detail internal characters like skeleton, musculature and organs. To date, no such tadpole descriptions exist for the well-studied Neotropical poison dart frog genus Ranitomeya (Anura: Dendrobatidae). Her
