Pathology of polychlorinated biphenyls in rabbits.

Rabbits were administered, orally, 300 mg of polychlorinated biphenyl Aroclor 1221, 1242 or 1254 once a week for 14 weeks. Elevated SGPT and SGOT levels occurred in the male and female 1254- and male 1242-treated rabbits as early as 2 weeks. The female 1242-treated rabbits had increased SGPT and SGOT activities at 4 and 8 weeks, respectively. These serum enzyme activities in the 1221-treated rabbits were similar to the controls during the 14-week period. Serum cholesterol concentrations were moderately elevated in the 1254-treated males throughout the experiment. The BUN, serum proteins, liver

[Polychlorinated biphenyls (PCBs) and reproduction disturbances].

A review of the impact of chlorinated biphenyls (PCBs) on reproduction is presented. PCBs are able to pass the placenta; accumulation in offspring is mainly achieved by breast feeding. Disturbances of fertility and embryotoxicity were detected in animals; teratogenicity without maternal toxicity was only observed in mice. In animal experiments disturbances of fetal and postnatal development were detected as well as in human studies. Neurotoxicity was observed even at low-level exposures in monkeys, which are most comparable to humans because of the development of the same clinical symptoms. Te

Placental markers of human exposure to polychlorinated biphenyls and polychlorinated dibenzofurans.

Our studies have evaluated biochemical changes in placentae from humans exposed to rice oil contaminated with polychlorinated biphenyls (PCBs) and polychlorinated dibenzofurans (PCDFs) in Taiwan. Placentae were obtained from nonsmoking women 4 to 5 years after the exposure had occurred. The exposed individuals ingested approximately 1 to 3 g PCBs and 5 mg PCDFs, and many exhibited symptoms characteristic of PCB poisoning. This disease was termed "Yu-Cheng" in Chinese. Based on data from experimental animal models, we examined a number of parameters in placentae from control and exposed women,

Increase in accumulation of polychlorinated biphenyls in offspring mouse brain via maternal coexposure to methylmercury and polychlorinated biphenyls.

Methylmercury (MeHg) and polychlorinated biphenyls (PCBs) are environmentally persistent neurodevelopmental toxicants. In Japan, the most common source of human exposure is the consumption of contaminated fish and seafood. We investigated the accumulation of MeHg and PCBs in the brains of dams and offspring mice maternally exposed to MeHg and/or PCBs. Pregnant mice (C57BL/6Cr) were assigned to one of four exposure groups: control, MeHg alone (MeHg in diet at 5 mg/kg as Hg), PCB alone (Aroclor1254 by gavage at the dose of 18 mg/kg body weight/3 days) and MeHg+PCB. Levels of MeHg and PCBs were m

Binding of polychlorinated biphenyls/metabolites to hemoglobin.

Polychlorinated biphenyls (PCBs) may form reactive electrophiles that can bind covalently to hemoglobin in vivo. Female C57/BL6 mice, pretreated with phenobarbital and P-naphthoflavone, were injected with either radiolabeled 4-chlorobiphenyl, 3,3',4.4'-tetrachlorobiphenyl, benzo[a]pyrene (B[a]P) or the corresponding non-labeled 4-chlorobiphenyl, 3.3',4.4'-tetrachlorobiphenyl, B[a]P or vehicle. Blood was collected at different time points. At 24 h a greater presence of 4-chlorobiphenyl and 3,3',4.4'-tetrachlorobiphenyl in the plasma, compared with the erythrocytes, was observed. For all three t

Polychlorinated Biphenyls and Nonalcoholic Fatty Liver Disease.

Polychlorinated biphenyls (PCBs) have been associated with abnormal liver enzymes and suspected nonalcoholic fatty liver disease (NAFLD) in cohort studies. NAFLD affects greater than 25% of the global population and may result in liver-related mortality. Both dioxin-like and non-dioxin-like PCBs have been associated with NAFLD, but their effects and mechanisms differ. Dioxin-like PCBs altered the gut:liver axis and microbiome and caused hepatic steatosis by disrupting hepatic lipid metabolism. In contrast, NDL PCBs reduced the liver's protective responses to promote diet-induced NAFLD. Mechani

[Polychlorinated biphenyls indoors: attempt at an assessment].

The use of products containing PCB in buildings resulted in indoor air contaminations. The search for appropriate measures is hampered by several different uncertainties in the regulatory toxicological process. These refer to problems in exposure assessment including measurement approaches, the toxicological evaluation of health risks including the derivation of a tolerable intake and the use of TCDD toxicity equivalency factors and finally the legal consequences of health risk evaluation. Since no consensus on these aspects has been achieved, the regulatory consequences vary considerably from

Polychlorinated biphenyls in residential dust: sources of variability.

We characterized the variability in concentrations of polychlorinated biphenyls (PCBs) measured in residential dust. Vacuum cleaner samples were collected from 289 homes in the California Childhood Leukemia Study during two sampling rounds from 2001 to 2010 and 15 PCBs were measured by high resolution gas chromatography-mass spectrometry. Median concentrations of the most abundant PCBs (i.e., PCBs 28, 52, 101, 105, 118, 138, 153, and 180) ranged from 1.0-5.8 ng per g of dust in the first sampling round and from 0.8-3.4 ng/g in the second sampling round. For each of these eight PCBs, we used a

Environmental stressors and violence: lead and polychlorinated biphenyls.

Many environmental risk factors for antisocial and violent behavior have been described. In recent years, this topic has become widely researched in the fields of environmental health, psychology, sociology, and many other disciplines. The results from a myriad of studies have shown that the etiologies of violent and aggressive behavior range from definitive biological environmental stressors like lead or polychlorinated biphenyls to various socio-cultural environmental stressors, such as social, economic, and racial factors. The aim of this paper is (a) to provide an overview of the specific

Advances in polychlorinated biphenyls-induced female reproductive toxicity.

Polychlorinated biphenyls (PCBs) are a class of endocrine-disrupting chemicals (EDCs) widely present in the environment. PCBs have been of concern due to their anti/estrogen-like effects, which make them more toxic to the female reproductive system. However, there is still a lack of systematic reviews on the reproductive toxicity of PCBs in females, so the adverse effects and mechanisms of PCBs on the female reproductive system were summarized in this paper. Our findings showed that PCBs are positively associated with lower pregnancy rate, hormone disruption, miscarriage and various reproducti

Polychlorinated biphenyls and thyroid function: a scoping review.

OBJECTIVE: Numerous evidence indicates the association between polychlorinated biphenyls (PCBs), an endocrine disrupter, with thyroid hormone disruption, contradictory findings also exist. Herein, we tried to address this question by performing a scoping review. CONTENT: The search was performed on PubMed, Scopus, Web of Science, and Google Scholar databases from 2010 onwards. Animal studies on PCBs' effect on thyroid function were searched. The SYRCLE's RoB scale assessed the risk of bias. I2 and Q tests are used for investigating heterogeneity. Environmental fate and global distribution of polychlorinated biphenyls.

In recent decades, regulators, academia, and industry have all paid increasing attention to the crucial task of determining how xenobiotic exposures affect biota populations, communities, or entire ecosystems. For decades, PCBs have been recognized as important and potentially harmful environmental contaminants. The intrinsic properties of PCBs, such as high environmental persistence, resistance to metabolism in organisms, and tendency to accumulate in lipids have contributed to their ubiquity in environmental media and have induced concern for their toxic effects after prolonged exposure. PCB

[Cytogenetic changes in persons exposed to polychlorinated biphenyls].

A cytogenetic study performed in a group of workers exposed to polychlorinated biphenyls comprised analysis of structural chromosomal aberrations in the lymphocytes in the first division in vitro, analysis of micronuclei frequency in binuclear lymphoblasts induced by cytochalasin B and analysis of sister chromatid exchanges. The study included 48 exposed workers and 15 controls. In 16 exposed workers increased incidence of unstable chromosomal aberrations (dicentrics, rings and acentrics) was found, while in 19 no structural chromosomal aberrations could be detected. In 11 workers changes defi

The inducing properties of polychlorinated biphenyls on hepatic monooxygenases.

The polychlorinated biphenyls (PCBs) represent a newly recognized and widely distributed category of environmental pollutants whose biologic impact on animals and man may be both substantial and highly detrimental. The pharmacologic effects of these agents on enzymes in the liver which metabolize drugs and other foreign compounds, such as carcinogens, are powerful and long lasting. PCBs mimic the effects produced on these enzymes by drugs, such as phenobarbital, and carcinogens, such as 3-methylcholanthrene. They are potent inducers of cytochromes P-45O and P-448 and associated enzymic activit

Examining Reproductive Health Outcomes in Females Exposed to Polychlorinated Biphenyl and Polybrominated Biphenyl.

In 1973, accidental contamination of Michigan livestock with polybrominated biphenyls (PBBs) led to the establishment of a registry of exposed individuals that have been followed for > 40 years. Besides being exposed to PBBs, this cohort has also been exposed to polychlorinated biphenyls (PCBs), a structurally similar class of environmental pollutants, at levels similar to average US exposure. In this study, we examined the association between current serum PCB and PBB levels and various female reproductive health outcomes to build upon previous work and inconsistencies. Polychlorinated biphenyls and thyroid status in humans: a review.

Animal studies show that exposure to polychlorinated biphenyls (PCBs) or other persistent organochlorine compounds can disrupt thyroid hormone homeostasis. In some reports dietary exposures to PCBs have also been claimed to affect circulating levels of thyroid hormones and thyrotropin (TSH) in humans. The aim of the present study was to review available epidemiologic studies within this field. A total of 13 studies fulfilled the inclusion criteria for the review. The overall impression is a lack of consistency between studies of reported correlations, neither are there any obvious interstudy d

Mortality of workers exposed to polychlorinated biphenyls--an update.

A retrospective cohort mortality study of workers exposed to polychlorinated biphenyls (PCBs) in two plants manufacturing electrical capacitors was reported in 1981. The study was conducted primarily to examine the risk of cancer mortality associated with exposure to PCBs. Based on animal data, liver cancer was the disease of most interest. Due to the small number of deaths and a relatively short observation period, the study was considered inconclusive. This study has been updated by adding 7 yr of observation. The number of deaths in the study cohort has increased from 163 to 295. Mortality

Effect of polychlorinated biphenyl on viral infections in mice.

Mice that were fed a diet containing 400, 200 or 100 microgram of polychlorinated biphenyls (PCB) per g were significantly more susceptible to Herpes simplex virus than mice that were fed a PCB-free diet. The mortality of mice that were fed a diet containing 400 or 200 microgram of PCB per g and infected with ectromelia virus was higher than that of normal control mice infected with virus. There was no significant difference in inducibility of interferon by polyinosinic acid-polycytoidylic acid between PCB-fed mice and control mice.
