Venom proteins from polydnavirus-producing endoparasitoids: their role in host-parasite interactions.

Endoparasitoid wasps have evolved various mechanisms to ensure successful development of their progeny, including co-injection of a cocktail of maternal secretions into the host hemocoel, including venom, calyx fluid, and polydnaviruses. The components of each type of secretion may influence host physiology and development independently or in a synergistic fashion. For example, venom fluid consists of several peptides and proteins that promote expression of polydnavirus genes in addition to other activities, such as inhibition of prophenoloxidase activation, inhibition of hemocytes spreading a

Parasitic castration of Plutella xylostella larvae induced by polydnaviruses and venom of Cotesia vestalis and Diadegma semiclausum.

In the present study, we used gamma-ray to irradiate the female parasitoids to make wasp eggs infertile, resulting in pseudoparasitization, which allowed the analysis of maternal secretions such as polydnaviruses (PDVs) and venom in the absence of larval secretions or teratocytes by the growing parasitoids. We then investigated the spermatogenesis and components of testicular proteins of male Plutella xylostella larvae pseudoparasitized by two endoparasitoids (Cotesia vestalis and Diadegma semiclausum). The results showed that pseudoparasitism by the two endoparasitoids at the early third inst

Interspecific competition between two endoparasitoids Cotesia vestalis (Hymenoptera: Braconidae) and Oomyzus sokolowskii (Hymenoptera: Eulophidae).

Two endoparasitoids, Cotesia vestalis and Oomyzus sokolowskii, parasitize the same host, larvae of Plutella xylostella. These two species have evolved different parasitization strategies. O. sokolowskii expresses a single factor, venom, and exerts virtually no detrimental effects on the development of its host. C. vestalis, on the other hand, injects polydnavirus (PDV) and venom during oviposition, and teratocytes are released into the host's hemolymph after egg hatching. Parasitization suppresses host immune reactions and redirects its developmental program. Because both these species parasit

Per os infectivity factors are essential for bracovirus infection and wasp parasitism.

Many parasitoid wasps in the family Braconidae rely on bracoviruses to parasitize lepidopteran hosts. Bracoviruses evolved from a virus in the family Nudiviridae, while nudiviruses are closely related to baculoviruses. A per os infectivity factor (PIF) complex is essential for oral infection of hosts by baculoviruses. Bracoviruses encode 8 PIF homologs but have no per os infection route because wasps inject virions into hosts. Thus, the mechanism underlying how bracoviruses enter lepidopteran host cells is unknown. Here we used the parasitoid Microplitis mediator, its lepidopteran hosts Mythim

Transgenic Expression of a Viral Cystatin Gene CpBV-CST1 in Tobacco Confers Insect Resistance.

A viral gene, CpBV-CST1, was identified from a polydnavirus Cotesia plutellae bracovirus (CpBV). Its protein product was significantly toxic to lepidopteran insects. This study generated a transgenic tobacco plant expressing CpBV-CST1 Expression of transgene CpBV-CST1 was confirmed in T1 generation (second generation after transgenesis) in both mRNA and protein levels. Young larvae of Spodoptera exigua (Hübner) suffered high mortalities after feeding on transgenic tobacco. All 10 T1 transgenic tobacco plants had no significant variation in speed-to-kill. Systematic analysis of a wasp parasitism arsenal.

Parasitoid wasps are among the most diverse insects on earth with many species causing major mortality in host populations. Parasitoids introduce a variety of factors into hosts to promote parasitism, including symbiotic viruses, venom, teratocytes and wasp larvae. Polydnavirus-carrying wasps use viruses to globally suppress host immunity and prevent rejection of developing parasites. Although prior results provide detailed insights into the genes viruses deliver to hosts, little is known about other products. RNAseq and proteomics were used to characterize the proteins secreted by venom gland

Modulation of antiviral immunity by the ichnovirus HdIV in Spodoptera frugiperda.

Polydnaviruses (PDVs) are obligatory symbionts found in thousands of endoparasitoid species and essential for successful parasitism. The two genera of PDVs, ichnovirus (IV) and bracovirus (BV), use different sets of virulence factors to ensure successful parasitization of the host. Previous studies have shown that PDVs target apoptosis, one of the innate antiviral responses in many host organisms. However, IV and BV have been shown to have opposite effects on this process. BV induces apoptosis in host cells, whereas some IV proteins have been shown to have anti-apoptotic activity. The differen

The calyx fluid of Microplitis rufiventris parasitoid and growth of its host Spodoptera littoralis larvae.

The morphology of the female reproductive system of pupal and adult stages of Microplitis rufiventris and the ultrastructure of the ovaries are described and illustrated. Two morphologically distinct types of particles of nuclear origin, i.e., polydnavirus (PDV) and a virus-like filamentous particle (VLFP) were detected in the ovarian calyx fluid of the female wasp. It is likely that these particles are injected into the host during oviposition. PDV initiated replication in the calyx of mid-aged pupae and in pharate adults and were present throughout adult life. VLFP were only seen in the caly

The virome of HPV-positive tonsil squamous cell carcinoma and neck metastasis.

Oropharyngeal squamous cell carcinoma (OPSCC) represents the most common HPV-related malignancy in the United States with increasing incidence. There is heterogeneity between the behavior and response to treatment of HPV-positive oropharyngeal squamous cell carcinoma that may be linked to the tumor virome. In this prospective study, a pan-pathogen microarray (PathoChip) was used to determine the virome of early stage, p16-positive OPSCC and neck metastasis treated with transoral robotic surgery (TORS) and neck dissection. The virome findings of primary tumors and neck lymph nodes were correlat

Parasitoid Calyx Fluid and Venom Affect Bacterial Communities in Their Lepidopteran Host Labial Salivary Glands.

The influence of gut and gonad bacterial communities on insect physiology, behaviour, and ecology is increasingly recognised. Parasitism by parasitoid wasps alters many physiological processes in their hosts, including gut bacterial communities. However, it remains unclear whether these changes are restricted to the gut or also occur in other tissues and fluids, and the mechanisms underlying such changes are unknown. We hypothesise that host microbiome changes result from the injection of calyx fluid (that contain symbiotic viruses known as polydnaviruses) and venom during parasitoid ovipositi

Symbiotic polydnavirus and venom reveal parasitoid to its hyperparasitoids.

Symbiotic relationships may provide organisms with key innovations that aid in the establishment of new niches. For example, during oviposition, some species of parasitoid wasps, whose larvae develop inside the bodies of other insects, inject polydnaviruses into their hosts. These symbiotic viruses disrupt host immune responses, allowing the parasitoid's progeny to survive. Here we show that symbiotic polydnaviruses also have a downside to the parasitoid's progeny by initiating a multitrophic chain of interactions that reveals the parasitoid larvae to their enemies. These enemies are hyperpara

Parasitization of Manduca sexta larvae by the parasitoid wasp Cotesia congregata induces an impaired host immune response.

During oviposition, the parasitoid wasp Cotesia congregata injects polydnavirus, venom, and parasitoid eggs into larvae of its lepidopteran host, the tobacco hornworm, Manduca sexta. Polydnaviruses (PDVs) suppress the immune system of the host and allow the juvenile parasitoids to develop without being encapsulated by host hemocytes mobilized by the immune system. Previous work identified a gene in the Cotesia rubecula PDV (CrV1) that is responsible for depolymerization of actin in hemocytes of the host Pieris rapae during a narrow temporal window from 4 to 8h post-parasitization. Its expressi

Changes in the haemolymph proteome of Spodoptera littoralis induced by the parasitoid Chelonus inanitus or its polydnavirus and physiological implications.

The egg-larval parasitoid Chelonus inanitus induces in its host Spodoptera littoralis two major developmental effects, namely a precocious onset of metamorphosis followed by a developmental arrest in the prepupal stage. Along with each egg, the wasp injects polydnavirus and venom into the host egg. The polydnavirus has been shown to play a major role in inducing the developmental arrest while the parasitoid larva is instrumental in inducing the precocious onset of metamorphosis. Here we report that experimental dilution of haemolymph of polydnavirus-containing larvae can partially prevent the

A chromosome-scale genome assembly and functional studies implicate the viral RNA polymerase in transcription of most Microplitis demolitor bracovirus late genes.

Microgastroid parasitoids lay eggs in other insects and harbor bracoviruses (BVs), which evolved from a nudivirus. BV genomes are endogenized in the wasp germline and consist of nudivirus-like genes plus proviral segments with virulence genes. Nudivirus genes produce virions in the ovary calyx cells, which wasps use to transfer virulence genes to hosts. Here, we report a chromosome-scale genome assembly for Microplitis demolitor, which harbors Microplitis demolitor bracovirus (MdBV). The assembly identified the locations of all MdBV nudivirus-like genes and proviral segments. Like nudiviruses

Parasitic castration by a viral protein tyrosine phosphatase targeting the host cell cycle checkpoint protein Rad9A.

Parasitic castration is a widespread strategy where parasites hijack host reproductive resources, yet the key molecular mechanisms driving this phenomenon remain poorly understood. Here, we reported that parasitization by the parasitic wasp Cotesia vestalis triggers apoptosis-mediated castration in the larval testes of its lepidopteran host, Plutella xylotella. Such a phenomenon was mediated by CvBV_22-9, a testis-enriched protein tyrosine phosphatase (PTP) encoded by Cotesia vestalis bracovirus (CvBV), a domesticated virus endogenized in the wasp. Similarly, a homolog of CvBV_22-9, encoded by

Cloning, characterization and analysis by RNA interference of various genes of the Chelonus inanitus polydnavirus.

Successful parasitism of some endoparasitic wasps depends on an obligately symbiotic association with polydnaviruses. These unique viruses have a segmented genome consisting of circles of double-stranded (ds) DNA and do not replicate in the parasitized host. They are produced in the wasp's ovary and injected into the host along with the egg. Chelonus inanitus is an egg-larval parasitoid; its polydnavirus (CiV) has been shown to protect the parasitoid larva from the host's immune system and to induce developmental arrest in the prepupal stage. The genome of CiV consists of at least 10-12 segmen

Similar polydnavirus genes of two parasitoids, Cotesia kariyai and Cotesia ruficrus, of the host Pseudaletia separata.

Endoparasitoids can achieve successful development in suitable hosts with the help of polydnavirus (PDV), if it is injected into the host with their eggs at parasitization. It was predicted that different species of endoparasitoids that parasitize the same host successfully use a similar system to avoid the defense reaction from the host. In this study, we identified similar PDVs genes. In two endoparasitoids, Cotesia kariyai (Cky) and Cotesia ruficrus (Crf), which can parasitize the same host species, Pseudaletia separata.
