In modern life petroleum-based plastic has become indispensable due to its frequent use as an easily available and a low cost packaging and moulding material. However, its rapidly growing use is causing aquatic and terrestrial pollution. Under these circumstances, research and development for biodegradable plastic (bioplastics) is inevitable. Polyhydroxybutyrate (PHB), a type of microbial polyester that accumulates as a carbon/energy storage material in various microorganisms can be a good alternative. In this study, 23 cyanobacterial strains (15 heterocystous and 8 non-heterocystous) were scr

The bacterium Paracoccus denitrificans was investigated for polyhydroxybutyrate (PHB) production on eight organic carbon substrates. Aerobic cultivation on crotonate resulted in the most significant PHB accumulation. Additionally, PHB production on butyrate increased with an addition of HCO₃⁻ to the mineral medium. Based on the substrate specificity of the cell fraction and the results of the reverse transcription quantitative real-time polymerase chain reaction (RT-qPCR) for the related gene, it is likely that butyrate and crotonate activations are catalyzed by propionyl-CoA synthetase. Polyhydroxybutyrate (PHB) is fully biodegradable under home-composting conditions, but its low yield and brittleness hinder widespread adoption. Microalgae have gained attention for PHB production due to their photoautotrophic nature, which enables the use of light and carbon dioxide as primary energy and carbon sources. This characteristic offers a low-cost and environmentally sustainable alternative for biopolymer synthesis. This review consolidates recent research on optimizing PHB productivity from microalgae, including strain selection, cultivation methods and growth media conditions. Chl

Polyhydroxybutyrate (PHB) is completely biodegradable which is metabolised by microorganisms in the soil as their sole food source in few years. The level of PHB up to 10.6% of algal dry weight is of great potential of the eco-friendly nature. Botryococcus braunii is mainly used for the production of biodiesel and is also capable of producing biopolymer polyhydroxy butyrate (PHB). In this study, Botryococcus braunii is used which generally produce PHB to around 20% of the dry weight. Three different microalgae were isolated from the fresh water of Kolavoi lake of Tamil Nadu. BACKGROUND: Plastic plays a crucial role in everyday life of human living, nevertheless it represents an undeniable source of land and water pollution. Polyhydroxybutyrate (PHB) is a bio-based and biodegradable polyester, which can be naturally produced by microorganisms capable of converting and accumulating carbon as intracellular granules. Hence, PHB-producing strains stand out as an alternative source to fossil-derived counterparts. However, the extraction strategy affects the recovery efficiency and the quality of PHB. In this study, PHB was produced by a genetically modified Escherichia

The use of antioxidants such as curcumin (Cur) or quercetin (Que) in biomedical and biotechnological applications has been studied owing to their capability to prevent oxidative stress and inhibit free radicals. Using polyhydroxybutyrate (PHB) electrospun fibers is presented as a proper option to encapsulate curcumin and quercetin due to its biocompatibility and biodegradability characteristics. Electrospun fibers were obtained dissolving commercial PHB in chloroform:N,N-dimethylformamide (DMF) (4:1) at 7% m/V, and adding two different concentrations of antioxidant (Cur, and Que) 1%m/m, and 7%

Methylorubrum extorquens DSM13060 is an endosymbiont that lives in the cells of shoot tip meristems. The bacterium is methylotrophic and consumes plant-derived methanol for the production of polyhydroxybutyrate (PHB). The PHB provides protection against oxidative stress for both host and endosymbiont cells through its fragments, methyl-esterified 3-hydroxybutyrate (ME-3HB) oligomers. We evaluated the role of the genes involved in the production of ME-3HB oligomers in the host colonization by the endosymbiont M. extorquens DSM13060 through targeted genetic mutations. The strains with deletions

Polyhydroxybutyrate (PHB) is an attractive biodegradable polymer that can be produced through the microbial fermentation of organic wastes or wastewater. However, its mass production has been restricted by the poor utilization of organic wastes due to the presence of inhibitory substances, slow microbial growth, and high energy input required for feedstock sterilization. Here, Vibrio natriegens, a fast-growing bacterium with a broad substrate spectrum and high tolerance to salt and toxic substances, was genetically engineered to enable efficient PHB production from nonsterilized fermentation o

Polyhydroxybutyrate (PHB) is a biodegradable bioplastic with potential applications as an alternative to petroleum-based plastics. However, efficient PHB production remains difficult. The main cost of PHB production is attributed to carbon sources; hence, finding inexpensive sources is important. Galactose is a possible substrate for polyhydroxyalkanoate production as it is abundant in marine environments. Marine bacteria that produce PHB from galactose could be an effective resource that can be used for efficient PHB production. In this study, to identify a galactose utilizing PHB producer, w

In this work, polyhydroxy butyrate (PHB) and biochar from pine wood (PWB) are used in a mini-pilot scale biological reactor (11.3 L of geometric volume) for trichloroethylene (TCE) removal (80 mgTCE/day and 6 L/day of flow rate). The PHB-biochar reactor was realized with two sequential reactive areas to simulate a multi-reactive permeable barrier. The PHB acts as an electron donor source in the first "fermentative" area. First, the thermogravimetric (TGA) and differential scanning calorimetry (DSC) analyses were performed. The PHB-powder and pellets have different purity (96% and 93% w/w) and

Biodegradable polyhydroxybutyrate (PHB) films were fabricated using acetic acid as an alternative to common solvents such as chloroform. The PHB films were prepared using a solvent casting process at temperatures ranging from 80 °C to 160 °C. The crystallinity, mechanical properties and surface morphology of the films cast at different temperatures were characterized and compared to PHB films cast using chloroform as a solvent. Results revealed that the properties of the PHB film varied considerably with solvent casting temperature. In general, samples processed with acetic acid at low tempera

There are many published studies presenting ethanol and acetone as PHAs-poor solvents, where these two solvents are shown to dissolve <2% (w/v) of PHAs at low temperatures. In this study, the suitability of ethanol and acetone for the recovery of PHB at different temperatures (from room temperature to near boiling point) in Cupriavidus necator was investigated. Experiments were performed using response surface methodology to examine the effects of different temperatures and heating incubation times on recovery percentage using the two solvents. The highest recovery percentage (92.3%) and produ

Polyhydroxybutyrate (PHB) is a biopolymer produced by various microorganisms for carbon and energy storage and stands out in terms of sustainable bioplastic production. PHB is a biodegradable and sustainable substitute for petrochemical-based plastics frequently used nowadays. Interest in waste-based/byproduct carbon sources has recently increased due to the high production cost of microbial PHB, and byproducts and wastes are being evaluated within the scope of zero-waste projects. In this study, the metabolic changes of Chlorella vulgaris were examined in the existence of different substrates

The valorization of industrial streams containing high concentration of aromatic compounds is of great significance yet remaining a critical challenge. Herein, an innovative strategy was proposed to transition the wastewater treatment from "removal-centered" to "recovery-oriented". Specifically, the newly isolated Cupriavidus sp. CY-1 realized the sustainable synthesis of value-added bioproduct polyhydroxybutyrate (PHB) from benzoate wastewater. For the first time, the upregulation of the ben and dan gene clusters in response to benzoate and aniline was demonstrated by transcriptome profiling,

BACKGROUND: Within lignocellulosic biomass, xylose is the second most abundant sugar after glucose. As a renewable and sustainable substrate, it is gaining attention as a feedstock for microbial bioprocesses. In this study, we demonstrated the co-production of polyhydroxybutyrate (PHB) and violacein from xylose. We initially confirmed the feasibility of co-production through genome-scale metabolic simulations, followed by optimization using a hybrid expression system that combines a conventional tac promoter and synthetic promoter-ribosome-binding site-terminator (semi-endo PRT) elements in du

The branched multiblock copolymer based on fatty acid, methyl methacrylate and PHB was first time synthesized. Autoxidation is the reaction of unsaturated fatty acids with air oxygen leading to macroperoxide initiators. Poly (3-hydroxy butyrate), PHB, is a natural bio polyester accumulated in the bacterium cell of some bacteria. Tri hydroxyl derivatives of PHB were obtained by the modification reactions of PHB with di-ethanol amine. Hydroxyl ends of the modified PHB were capped with the autoxidized unsaturated fatty acid oligomeric peroxides in order to the polyoleic acid, polylinoleic acid an
