Bacterial polysaccharides as vaccines--immunity and chemical characterization.

Studies on protective immunity and biochemical characterization of bacterial capsular polysaccharides have led to significant contributions to understanding of the mechanisms of infectious diseases and development of effective vaccines. Immunity to encapsulated bacteria is related to antibody response to polysaccharide (PS) antigen, interactions with T- and B-lymphocytes, and host defense mechanisms. Meningococcal, pneumococcal and Salmonella vi PSs and Haemophilus type b PS-protein conjugate vaccines have been licensed and provided effective immunity for prevention of these bacterial infectio

Bacterial polysaccharide-protein conjugate vaccines.

Following demonstration that chemical conjugation of polysaccharide antigens to proteins could enhance their immunogenicity in the 1920s, interest in this approach to primary prevention of bacterial infections waned with the development and widespread use of antibiotics. Emergence of resistant bacteria rekindled interest in the late 20th century, which saw extremely rapid development and implementation of several vaccines which are already rapidly changing the epidemiology of childhood infections with Haemophilus influenzae type b, Streptococcus pneumoniae and Neisseria meningitidis. Others su

Interaction of Human Peripheral NK Cells with Bacterial Polysaccharides.

NK cells participate in the antimicrobial immune response. NK cell activation by bacterial polysaccharides (PSs) can occur indirectly through signals from other immune cells or through direct influence of PSs on NK cells via interaction with activating receptors. This study demonstrated that peripheral NK cells bind fluorescently labeled PSs from the Gram-negative bacteria Escherichia coli, Shigella flexneri, and Pseudomonas aeruginosa with varying efficiency depending on the PS origin and the NK cell donor. PSs from Escherichia coli strain O19ab induced an increased IFN-γ production by NK cel

Human immune responses to polysaccharide antigens: an analysis of bacterial polysaccharide vaccines in infants.

Mechanisms of human immunity to polysaccharide encapsulated bacteria and the development and testing of the currently available purified polysaccharide bacterial vaccines are reviewed. These vaccines appear to be poorly immunogenic in infants under the age of two years--those at greatest risk for infection. In an effort to understand the poor responses of infants, the human immune response to polysaccharide antigens was characterized in more detail. Using pneumococcal polysaccharide type 3 as an example, it appears that human polysaccharide antibody responses are analogous to the type 2 T cell

Detection and characterization of bacterial polysaccharides in drug-resistant enterococci.

Enterococcus faecium (E. faecium) has emerged as one of today's leading causes of health care-associated infections that is difficult to treat with the available antibiotics. These pathogens produce capsular polysaccharides on the cell surface which play a significant role in adhesion, virulence and evasion. Therefore, we aimed at the identification and characterization of bacterial polysaccharide antigens which are central for the development of vaccine-based prophylactic approaches. The crude cell wall-associated polysaccharides from E. faecium, its mutant and complemented strains were purif

Antibodies to Conserved Surface Polysaccharides Protect Mice Against Bacterial Conjunctivitis.

PURPOSE: Bacterial conjunctivitis is a major problem in ocular health. Little is known about protective immune effectors in the conjunctiva. We evaluated whether opsonic antibody to the conserved surface/capsular polysaccharide poly-N-acetyl glucosamine (PNAG) expressed by Streptococcus pneumoniae and Staphylococcus aureus was protective against bacterial conjunctivitis, as well as an antibody to the Pseudomonas aeruginosa surface polysaccharide alginate. METHODS: Bacteria were injected directly into the conjunctivae of either A/J mice or into conjunctivae of wild type C57Bl/6 mice for compar

Synthesis of bacterial polysaccharides via the Wzx/Wzy-dependent pathway.

The surfaces of bacteria mediate a multitude of functions in the environment and in an infected host, including adhesion to both biotic and abiotic substrata, motility, immune system interaction and (or) activation, biofilm formation, and cell-cell communication, with many of these features directly influenced by cell-surface glycans. In both Gram-negative and Gram-positive bacteria, the majority of cell-surface polysaccharides are produced via the Wzx/Wzy-dependent assembly pathway; these glycans include heteropolymeric O-antigen, enterobacterial common antigen, exopolysaccharide, spore coat,

Prospects for the prevention of bacterial meningitis with polysaccharide vaccines.

MOST SUPPURATIVE INFECTIONS OF THE MENINGES ARE CAUSED BY FIVE BACTERIAL SPECIES: Escherichia coli, Haemophilus influenzae type b, Streptococcus pneumoniae, Neisseria meningitidis, and group B streptococcus. The immune response of adults to pneumococcal capsular polysaccharides has been studied in great detail and their responses to meningococcal and H. influenzae type b capsular polysaccharides are quite similar. Immune responses of adults to E. coli and group B streptococcal antigens are disappointing. The responses of children below the age of 7 years differ both quantitatively and in durat

Bletilla Striata polysaccharides thermosensitive gel for photothermal treatment of bacterial infection.

Skin is the most important defense shield which touched external environment directly. Effectively clearing microbes in infected wound via non-antibiotic therapy is crucial for the promotion of recovery in complex biological environments, and the wound healing is a crucial process after sterilization to avoid superinfection. Herein, a kind of Prussian blue-based photothermal responsive gel, Bletilla striata polysaccharide-mingled, isatin-functionalized Prussian blue gel (PB-ISA/BSP gel) was reported for effective treatment of bacterial infection and wound healing. Capsule polysaccharide mediates bacterial resistance to antimicrobial peptides.

The innate immune system plays a critical role in the defense of areas exposed to microorganisms. There is an increasing body of evidence indicating that antimicrobial peptides and proteins (APs) are one of the most important weapons of this system and that they make up the protective front for the respiratory tract. On the other hand, it is known that pathogenic organisms have developed countermeasures to resist these agents such as reducing the net negative charge of the bacterial membranes. Here we report the characterization of a novel mechanism of resistance to APs that is dependent on th

Role of O-Acetylation in the Immunogenicity of Bacterial Polysaccharide Vaccines.

The incidence of infectious diseases caused by several bacterial pathogens such as Haemophilus influenzae type b, Streptococcus pneumoniae, and Neisseria meningitidis, has been dramatically reduced over the last 25 years through the use of glycoconjugate vaccines. The structures of the bacterial capsular polysaccharide (CPS) antigens, extracted and purified from microbial cultures and obtained with very high purity, show that many of them are decorated by O-acetyl groups. While these groups are often considered important for the structural identity of the polysaccharides, they play a major rol

Modeling and Simulation of Bacterial Outer Membranes with Lipopolysaccharides and Capsular Polysaccharides.

Capsule is one of the common virulence factors in Gram-negative bacteria protecting pathogens from host defenses and consists of long-chain capsular polysaccharides (CPS) anchored in the outer membrane (OM). Elucidating structural properties of CPS is important to understand its biological functions as well as the OM properties. However, the outer leaflet of the OM in current simulation studies is represented exclusively by LPS due to the complexity and diversity of CPS. In this work, representative Escherichia coli CPS, KLPS (a lipid A-linked form) and KPG (a phosphatidylglycerol-linked form)

Suppression of blood group agglutinability of human erythrocytes by certain bacterial polysaccharides.

Erythrocytes coated with bacterial capsular polysaccharides, notably the Vi antigen, were no longer agglutinated by antibodies directed against the various antigens native to the red cell surface. These effects could not be attributed to prevention of antibody uptake even though in some systems the uptake of antibody was diminished. In fact, agglutination by Rh-incomplete antibody was brought back to the original titer only after the sensitized Vi-coated cells had been subjected to ten alternating exposures to globulin and antiglobulin. Hemagglutination by Newcastle, mumps, and influenza virus

Phosphate deprivation restricts bacterial degradation of the marine polysaccharide fucoidan.

Brown algae and diatoms convert carbon dioxide into the polysaccharide fucoidan, which sequesters carbon in the ocean despite the prevalence of marine bacterial fucoidanase genes. Bacteria with fucoidanase genes also have high-affinity phosphate transporters, suggesting that phosphate could impact fucoidan degradation and subsequent carbon sequestration. Here, to test this hypothesis, we assembled a system consisting of a microalga that produces and a bacterium that degrades fucoidan. The fixation of carbon dioxide into fucoidan by the microalga Glossomastix sp. PLY432 occurred independent of

Flocculation of colloidal clay by bacterial polysaccharides: effect of macromolecule charge and structure.

The molecular mechanism of montmorillonite flocculation by bacterial polysaccharides was investigated, with special emphasis on the effect of carboxylic charges in the macromolecules on the mechanisms of interaction with the clay surface. An indirect way to quantify the energy of interaction was used, by comparing the flocculation ability of variously acidic polysaccharides. Data on tensile strength of aggregates in diluted suspension were collected by timed size measurements in the domain 0.1-600 microm, using laser diffraction. The flow behavior of settled aggregates was studied by rheology

Suppressive effect of bacterial polysaccharides on BAFF system is responsible for their poor immunogenicity.

Capsular polysaccharides of encapsulated bacteria are weakly immunogenic T cell-independent type 2 (TI-2) Ags. Recent findings suggest that BAFF system molecules have a critical role in the development of Ab responses against TI-2 Ags. In this study, we investigated the effect of bacterial polysaccharides on B cell responses to BAFF and a proliferation-inducing ligand (APRIL). We determined that B cells exposed to meningococcal type C polysaccharide (MCPS) or group B Streptococcus serotype V (GBS-V) were unresponsive to BAFF- and APRIL-induced Ig secretion. Moreover, MCPS and GBS-V strongly do

Phenylboronic Acid-Modified Copper Nanozymes as Nanoscavengers of Bacterial Polysaccharides Causing Acute Lung Injury.

Acute lung injury (ALI), especially burn-induced cases complicated by secondary infections and hyperinflammation, remains challenging to treat. This study developed 4-mercaptobenzoic acid (MPBA)-modified copper nanozymes (CuMPBA) to simultaneously combat bacterial infections and toxin-triggered immune overactivation. CuMPBA binds bacterial surface polysaccharides via boronate ester bonds, neutralizing lipopolysaccharides (LPS) and lipoteichoic acid (LTA). It demonstrates dual enzymatic activity: peroxidase (POD)-like and glutathione peroxidase (GPx)-like activities for antimicrobial effects. A
