Propitious Profile of Peppery Piperine.

BACKGROUND: Piperine is a key bioactive alkaloid found in plants of piperaceae family. The compound possesses various medicinal and pharmacological activities (cholesterol-lowering, anti-cancer, Alzheimer's disease etc.). Owing to its various target receptors (TRPV1, P-gp, CYP3A4 etc.) and several mechanisms, piperine has been studied as bio-enhancer for other drugs and its role has been evidenced in the literature. When administered with other drugs, it increases the absorption of other drugs, thereby reducing the dose and dose-related toxic potential. There are various mechanisms of piperine

Endocannabinoids in Saliva: Origins, Significance, and Research Directions.

INTRODUCTION: The endocannabinoid system regulates a broad range of psychological and physiological processes throughout the body, and its dysregulation has been implicated in numerous pathological conditions. As a result, components of this system are common targets in diagnostic and therapeutic research. Given the challenges associated with obtaining tissue samples from human participants, current research commonly relies on the quantification of circulating endocannabinoids in blood as a surrogate for tissue-level endocannabinoid activity. However, this approach presents limitations due to

The detection and modulation of piperine in the human oral cavity.

Piperine is an alkaloid that is responsible for the pungency of black pepper and long pepper. This hydrophobic compound causes a spicy sensation when it comes in contact with trigeminal neurons of the oral cavity. Piperine has low solubility in water, which presents difficulties in examining the psychophysical properties of this stimulus by standard aqueous chemosensory tests. This report describes approaches that utilize novel edible film formulations for delivering precise amounts of piperine to the human oral cavity. These films were then used to identify detection thresholds for piperine,

The potential role of Piper guineense (black pepper) in managing geriatric brain aging: a review.

Brain aging is one of the unavoidable aspects of geriatric life. As one ages, changes such as the shrinking of certain parts (particularly the frontal cortex, which is vital to learning and other complex mental activities) of the brain may occur. Consequently, communications between neurons are less effective, and blood flow to the brain could also decrease. Efforts made at the biological level for repair become inadequate, leading to the accumulation of β-amyloid peptide in the brain faster than its probable degradation mechanism, resulting in cognitive malfunction. Subsequent clinical usage

Protocol to produce plant-based hybrid nanovesicles from fresh turmeric and pepper using polyethylene glycol.

In addition to proteins, microRNAs, and lipids, plant-derived exosome-like nanovesicles (ENVs) are also enriched with host plant bioactives. Both curcumin and piperine are water insoluble, lack bioavailability, and are extracted by non-ecofriendly solvents. Herein, we present an eco-friendly protocol for co-isolating both curcumin and piperine in the form of hybrid ENVs. We describe steps for sample pre-processing, combined homogenization of plant materials, filtration, and differential centrifugation. We then detail procedures for polyethylene glycol-based fusion and precipitation of hybrid E

Therapeutic spectrum of piperine for clinical practice: a scoping review.

Translation of traditional knowledge of herbs into a viable product for clinical use is still an uphill task. Piperine, a pungent alkaloid molecule derived from Piper nigrum and Piper longum possesses diverse pharmacological effects. Traditionally, pepper is used for arthritis, bronchitis, gastritis, diarrhea, snake bite, menstrual pain, fever, and bacterial infections, etc. The anti-inflammatory, antioxidant and immunomodulatory actions of piperine are the possible mechanisms behind its therapeutic potential. Various in-silico and experimental studies have shown piperine as a possible promisi

Solid state NMR study of the bioenhancer piperine, supported by GIPAW DFT calculations.

Piperine, an alkaloid found in black pepper fruits, has the properties of promoting the absorption of other substances (e.g. curcumin), therefore it is used in solid forms of dietary supplements as an additive increasing bioavailability. The aim of the study was to analyze piperine in a solid state and then to use it in the analysis of solid dietary supplements with the addition of piperine as an absorption promoter. The 13C CP MAS NMR spectra were recorded using variable contact time and dipolar dephasing experiment. The calculation of theoretical chemical shift values for three polymorphic f

Piperine treating sciatica through regulating inflammation and MiR-520a/P65 pathway.

Although the etiology of sciatica remains uncertain, there is increasing evidence that the disease process of sciatica is associated with the levels of inflammatory factors. Piperine, an alkaloid isolated from Piper nigrum, has previously been demonstrated to inhibit inflammation and analgesic effects. The purpose of this study is to verify the regulatory relationship between miR-520a and p65 and to explore how miR-520a/P65 affects the level of cytokines under the action of piperine, so as to play a therapeutic role in sciatica. Through ELISA experiment, we confirmed that four inflammatory fac

Piperine and its derivatives as a therapeutic alternative against leishmaniasis: A comprehensive review.

Leishmaniasis remains a major global health problem, especially in tropical and subtropical regions, where current treatments are limited by toxicity, cost, and drug resistance. The disease affects more than 98 endemic countries, causing approximately 1.3 million new cases and 30,000 deaths each year. Despite occasional advances, its global burden remains substantial and demands sustained attention. Piperine, a bioactive alkaloid from Piper species, shows promising antileishmanial effects, but its clinical use is limited by poor solubility and bioavailability. This review evaluates the potenti

Co-administration of curcumin with other phytochemicals improves anticancer activity by regulating multiple molecular targets.

Natural plant phytochemicals are effective against different types of diseases, including cancer. Curcumin, a powerful herbal polyphenol, exerts inhibitory effects on cancer cell proliferation, angiogenesis, invasion, and metastasis through interaction with different molecular targets. However, the clinical use of curcumin is limited due to poor solubility in water and metabolism in the liver and intestine. The synergistic effects of curcumin with some phytochemicals such as resveratrol, quercetin, epigallocatechin-3-gallate, and piperine can improve its clinical efficacy in cancer treatment.

Inhibition of the morphological transition of Candida spp. by riparins I-IV.

Candida spp. is an opportunistic pathogen capable of causing superficial to invasive infections. Morphological transition is one of the main virulence factors of this genus and, therefore, is an important variable to be considered in pharmacological interventions. Riparins I, II, III, and IV are alkamide-type alkaloids extracted from the unripe fruit of Aniba riparia, whose remarkable pharmacological properties were previously demonstrated. This work aimed to evaluate in silico and in vitro the inhibitory effects of Riparins on the morphological transition of Candida albicans, Candida tropical
