We evaluated saliva (SAL) specimens for SARS-CoV-2 reverse transcriptase PCR (RT-PCR) testing by comparison of 459 prospectively paired nasopharyngeal (NP) or midturbinate (MT) swabs from 449 individuals with the aim of using saliva for asymptomatic screening. Samples were collected in a drive-through car line for symptomatic individuals (n = 380) and in the emergency department (ED) (n = 69). The percentages of positive and negative agreement of saliva compared to nasopharyngeal swab were 81.1% (95% confidence interval [CI], 65.8% to 90.5%) and 99.8% (95% CI, 98.7% to 100%), respectively. Rapid and accurate identification of the prevalence of schistosomiasis is key for control and eradication of this devastating disease. The current screening standard for intestinal schistosomiasis is the Katz-Kato method, which look for eggs on slides of fecal matter. Although work has been done to estimate prevalence using the number of eggs on a slide, the procedure is much faster if the laboratory only reports the presence or absence of eggs on each slide. To further help reduce screening costs while maintaining accuracy, we propose a pooled method for estimating prevalence. We compare it t

OBJECTIVES: As Chlamydia trachomatis (CT) and Neisseria gonorrhoeae (NG) are the most commonly reported STIs in Belgium and the majority of women infected are asymptomatic, targeted screening of patients in specified risk groups is indicated. To prevent long-term complications and interrupt transmission, extragenital samples should be included. As this comes with a substantial extra cost, analysis of a pooled sample from vaginal and extragenital sites could be a solution. In this study, we evaluated the feasibility of molecular testing for CT and NG in pooled versus single-site samples in a la

BACKGROUND: Pooled testing, in which biological specimens from multiple subjects are combined into a testing pool and tested via a single test, is a common testing method for both surveillance and screening activities. The sensitivity of pooled testing for various pool sizes is an essential input for surveillance and screening optimization, including testing pool design. However, clinical data on test sensitivity values for different pool sizes are limited, and do not provide a functional relationship between test sensitivity and pool size. We develop a novel methodology to accurately compute

OBJECTIVE: To determine the effects of HIV-1 antibody level and test-format characteristics on testing pooled sera.
DESIGN: This study was designed with a laboratory exercise followed by test observations on serosurveillance samples.
METHODS: Sera with low, medium and high (n = 22, 12 and 20, respectively) antibody titers were pooled with HIV-1-negative sera and tested with two enzyme-linked immunosorbent assays (ELISA) and a particle agglutination test. The same kits were used to test single and pooled (batches of five, 10 and 20) samples collected from 3000 blood donors and sex workers. Th

Pooled testing is a procedure commonly used to reduce the cost of screening a large number of individuals for infectious diseases. In its simplest form, pooled testing works by compositing a set of individual specimens (e.g., blood or urine) into a common pool. If the pool tests negative, all individuals within it are diagnosed as negative. If the pool tests positive, retesting is needed to decode the positive individuals from the negative individuals. Traditionally, pooled testing has assumed that each individual has the same probability of being positive. However, this assumption is often un

Universal congenital cytomegalovirus (cCMV) screening in saliva is increasingly recommended. The aim of our study was to correlate the performance of a point-of-care rapid molecular test with CMV real time PCR (CMV RT-PCR) detection, using saliva pool-testing in newborns under a universal screening strategy. Saliva swabs were prospectively collected from newborns < 21 days old and tested by Alethia-LAMP-CMV assay in pools of 5 samples. In positive pools, subjects were tested individually and by saliva and urine CMV RT-PCR. A subset of negative pools were studied with both techniques and viral

Pooled testing represents a resource-conserving diagnostic strategy, and we propose enterovirus-D68 (EV-D68) as an attractive target for pooling (especially locally). To validate an RT-qPCR-based approach, retrospective pools were contrived from EV-D68-positive nasopharyngeal specimens and non-EV-D68-enterovirus-positive specimens. Five/nine-fold pools demonstrated 90.9 %/88.6 % sensitivity versus-unpooled testing (no false-positives), with commensurate results observed prospectively.

BACKGROUND: The Alinity m STI assay (Abbott Molecular, Des Plaines, IL, USA) is designed for the qualitative detection of Chlamydia trachomatis (CT), Neisseria gonorrhoeae (NG), Mycoplasma genitalium (MG), and Trichomonas vaginalis (TV) in individual collected samples. Pooled specimens are not recommended due to the high risk of inhibition.
METHODS: We performed a two-part study, including a preliminary validation on 13 well-known positive samples before and after pooling, as well as a retrospective analysis of 5324 pooled specimens collected between 2021 and 2024. All samples were tested for

Tracheal pooling for Mycoplasma hyopneumoniae (M. hyopneumoniae) DNA detection allows for decreased diagnostic cost, one of the main constraints in surveillance programs. The objectives of this study were to estimate the sensitivity of pooled-sample testing for the detection of M. hyopneumoniae in tracheal samples and to develop probability of M. hyopneumoniae detection estimates for tracheal samples pooled by 3, 5, and 10. A total of 48 M. hyopneumoniae PCR-positive field samples were pooled 3-, 5-, and 10-times using field M. hyopneumoniae DNA-negative samples and tested in triplicate. The s

The most effective measure to prevent or stop the spread of infectious diseases is the early identification and isolation of infected individuals through comprehensive screening. At present, in the COVID-19 pandemic, such screening is often limited to isolated regions as determined by local governments. Screening of potentially infectious individuals should be conducted through coordinated national or global unified actions. Our current research focuses on using resources to conduct comprehensive national and regional regular testing with a risk rate based, algorithmic guided, multiple-level,

BACKGROUND: Human papillomavirus (HPV) testing is the preferred method for cervical cancer screening. Noninvasive urinary HPV testing offers an attractive alternative to improve screening coverage among underscreened populations. However, few studies have assessed its performance in detecting cervical HPV via systematic review and meta-analysis.
METHODS: This study systematically searched PubMed, Embase, Web of Science Core Collection, and the Cochrane Library from inception until September 2023. The aim was to assess the performance of urinary HPV testing for detecting cervical HPV against t

BACKGROUND: Virological monitoring (VM) and drug resistance (DR) analysis are crucial for effective HIV management. Due to the high cost of commercial assays, VM and DR analysis is not performed in resource-limited-settings.
OBJECTIVE: The objective of this study is to develop a pooling based algorithm for the combined identification of virologic treatment failure (VTF) by nucleic acid testing (NAT) and DR by sequencing - NAT+DR assay.
STUDY DESIGN: We enrolled 559 participants on first-line therapy and analyzed for VTF. The virologically suppressed participants were followed-up to see the V

PURPOSE: Trachoma is the leading cause of blindness from infection worldwide. Treatment programs require accurate Chlamydia trachomatis infection prevalence rates to guide decision making. The use of clinical examination is by far the most common way to monitor activity, but may yield overestimates of infection prevalence. Laboratory testing on individual specimens such as polymerase chain reaction (PCR) is highly sensitive and specific, but prohibitively expensive. Here we demonstrate simulations of pooled PCR results may estimate infection prevalence of an entire community yielding substanti

INTRODUCTION: Pooled testing is a potentially efficient alternative strategy for COVID-19 testing in congregate settings. We evaluated the utility and cost-savings of pooled testing based on imperfect test performance and potential dilution effect due to pooling and created a practical calculator for online use.
METHODS: We developed a 2-stage pooled testing model accounting for dilution. The model was applied to hypothetical scenarios of 100 specimens collected during a one-week time-horizon cycle for varying levels of COVID-19 prevalence and test sensitivity and specificity, and to 338 skil

Dorfman pooled testing combines individual specimens (e.g., blood samples) into one test; if the pooled sample tests positive for infection, each specimen is tested separately. Under small prevalence levels, this method is known to reduce the expected number of tests required to screen a population, as individual tests only occur when a pooled test detects an infection. When conducting Dorfman testing, studies often recommend implementing positive assortative matching, i.e., pooling together samples with a similar risk of infection, as this tends to minimize the expected number of tests, the e

Pool Testing is a strategy introduced by Robert Dorfman in 1943 to address a common problem in pandemics: the high cost of large-scale testing. This two-phase pool test strategy consists of dividing the initial sample into smaller subsamples of equal size (pools) and conducting a combined test on the individuals within each pool. Given a positive result, all individuals in the pool are individually tested. Otherwise, all individuals in the pool will be considered healthy without further testing. Consequently, for the same sample, the number of tests (testing cost) performed is a random variabl
