Transcriptional slippage in the positive-sense RNA virus family Potyviridae.

The family Potyviridae encompasses ~30% of plant viruses and is responsible for significant economic losses worldwide. Recently, a small overlapping coding sequence, termed pipo, was found to be conserved in the genomes of all potyvirids. PIPO is expressed as part of a frameshift protein, P3N-PIPO, which is essential for virus cell-to-cell movement. However, the frameshift expression mechanism has hitherto remained unknown. Here, we demonstrate that transcriptional slippage, specific to the viral RNA polymerase, results in a population of transcripts with an additional "A" inserted within a hi

Discovery of novel viruses related to the Potyviridae family reveals expanded diversity, a broad host range, and evidence of fungal and oomycete infections.

The family Potyviridae represents the largest and most economically important group of plant-infecting RNA viruses. Despite extensive study of crop-associated members, the full diversity, host range, and evolutionary history of potyvirids remain poorly understood. Here, we conducted a large-scale mining of publicly available RNA-seq datasets to systematically search for novel potyvirid sequences. This approach enabled the identification and assembly of 47 previously undescribed members of the family, distributed across 8 recognized genera and, importantly, putative new groups. Beyond expanding

Virus Acquisition and Retention Periods for Transmission of Triticum Mosaic Virus by the Wheat Curl Mite.

The wheat curl mite (WCM), Aceria tosichella Keifer, is an important pest of winter wheat (Triticum aestivum L.) in the North American Great Plains region because of its ability to transmit three important viruses to wheat. Wheat streak mosaic virus (WSMV; Tritimovirus tritici) has been the most studied of these three viruses, but the rapid spread of Triticum mosaic virus (TriMV; Poacevirus tritici) throughout the region, along with its synergistic relationship with WSMV, has increased the need for a more complete understanding of its transmission capabilities and epidemiology. Studies were co

Molecular insights into Cassava brown streak virus susceptibility and resistance by profiling of the early host response.

Cassava brown streak virus (CBSV) and Ugandan cassava brown streak virus (UCBSV) are responsible for significant cassava yield losses in eastern sub-Saharan Africa. To study the possible mechanisms of plant resistance to CBSVs, we inoculated CBSV-susceptible and CBSV-resistant cassava varieties with a mixed infection of CBSVs using top-cleft grafting. Transcriptome profiling of the two cassava varieties was performed at the earliest time point of full infection (28 days after grafting) in the susceptible scions. The expression of genes encoding proteins in RNA silencing, salicylic acid pathway

Incidence of Wheat Curl Mite-Transmitted Viruses in Major Cereal Crops: Potential Roles of Cover and Forage Cereal Crops in the Continuum of Wheat Streak Mosaic Disease Complex.

Wheat streak mosaic disease (WSMD) is the most important viral disease affecting wheat production in the U.S. Great Plains. WSMD is caused by a single or coinfection of wheat curl mite (WCM)-transmitted wheat streak mosaic virus (WSMV), Triticum mosaic virus (TriMV), and/or High Plains wheat mosaic virus (HPWMoV). Nebraska, one of the U.S. Great Plains states, significantly contributes to the national small grain production and utilizes cereal crops as forage and cover crops. In this study, a statewide sampling was conducted during the 2023 to 2025 growing seasons across Nebraska to examine th

Differential Infection by Barley Yellow Mosaic Disease and Its Impact on Key Agronomic Traits in Barley.

Barley yellow mosaic disease (BYMD), a soilborne viral disease, severely compromises winter barley productivity. This disease is caused by barley yellow mosaic virus (BaYMV) and the barley mild mosaic virus (BaMMV), occurring in either single or mixed infections. To elucidate pathogenesis, 10 barley varieties with differential resistance were systematically analyzed. Virus-specific RT-qPCR revealed distinct accumulation patterns of BaYMV and BaMMV in infected tissues. Crucially, transmission requires temperatures persistently below 10°C for 40 to 54 days, facilitating viral entry into roots fo

CRISPR-Cas13-based amplification-free detection of three quarantine-significant sugarcane viruses.

Crop losses from viral pathogens cost billions of dollars annually, and sugarcane is particularly vulnerable. Sugarcane is the most cultivated crop in the world by volume; therefore, significant yield losses result in large dollar losses for farmers and trade economies. In some cases, viruses contribute to more than half of the yield reduction. Three quarantine-significant pathogens-Sugarcane mosaic virus (ScMV), Sugarcane streak mosaic virus (ScSMV), and Sugarcane yellow leaf virus (ScYLV)- pose major threats to global sugarcane production because of their rapid spread, asymptomatic infection

Comparative analysis of virus-derived small RNAs within cassava (Manihot esculenta Crantz) infected with cassava brown streak viruses.

Infection of plant cells by viral pathogens triggers RNA silencing, an innate antiviral defense mechanism. In response to infection, small RNAs (sRNAs) are produced that associate with Argonaute (AGO)-containing silencing complexes which act to inactivate viral genomes by posttranscriptional gene silencing (PTGS). Deep sequencing was used to compare virus-derived small RNAs (vsRNAs) in cassava genotypes NASE 3, TME 204 and 60444 infected with the positive sense single-stranded RNA (+ssRNA) viruses cassava brown streak virus (CBSV) and Ugandan cassava brown streak virus (UCBSV), the causal agen

Molecular characterization of a proposed novel macluravirus identified in sand ginger (Kaempferia galanga).

A novel macluravirus, designated provisionally as "kaempferia virus A" (KVA), was identified in sand ginger (Kaempferia galanga) by reverse transcription polymerase chain reaction (RT-PCR). The complete genome of this virus is 8,274 nucleotides in length, excluding the poly(A) tail. Its genome organization is typical of members of the genus Macluravirus, with a single large open reading frame (ORF) that encodes a polyprotein of 2,636 amino acids that is predicted to be processed into nine mature proteins. Comparative sequence analysis revealed that KVA shares 55-70% nucleotide sequence identit

A wheat lncRNA-encoded peptide promotes antiviral resistance by targeting viral NIb for autophagic degradation.

Long noncoding RNAs (lncRNAs) are increasingly recognized as sources of functional peptides that regulate plant development and immunity, yet little is known about whether lncRNA-encoded peptides participate in antiviral defense. Through transcriptomic analysis combined with peptide-coding potential prediction, we identified wheat lncRNA19864, which is highly expressed in wheat yellow mosaic virus (WYMV)-resistant cultivars and encodes a 39-amino-acid peptide, TaLEP1. Genetic manipulation of TaLEP1 expression revealed that TaLEP1 acts as a positive regulator of wheat resistance to WYMV. Mechan

Drivers of farmers' intentions to adopt genome-edited cassava resistant to cassava brown streak disease in Uganda.

Genome editing has been increasingly promoted as a promising breeding technology to strengthen the agrifood system. However, its success largely depends on farmers' adoption of the technology, a topic that remains insufficiently addressed in current literature. This study applies the model of goal-directed behavior (MGB) to examine farmers' adoption intention for genome edited (GE) cassava, resistant to cassava brown streak disease, in a hypothetical scenario in a developing country context. Data was collected from a cross-sectional survey of 357 cassava farmers in Northern Uganda and analyzed

A newly discovered 5' translation-regulatory element with a unique three stem-loop structure in sweet potato feathery mottle virus.

Sweet potato feathery mottle virus (SPFMV), a member of the Potyviridae family and Potyvirus genus, severely affects the yield and quality of sweet potato. Internal ribosome entry site (IRES) elements were first identified in animal RNA viruses and have since been identified in plant RNA viruses and eukaryotic mRNAs. Herein, we show that the 5' untranslated region (UTR) of SPFMV possesses IRES activity and contains the complete IRES sequence. Furthermore, the 3'UTR can synergistically enhance the cap-independent translation efficiency of the 5'UTR. Within the 5'UTR of SPFMV, three stem-loop st

Evaluating the Negative-Strand Coding Potential in Plum Pox Virus.

Recent studies proposed that the negative strands of some single-stranded, positive-sense RNA viruses contain reverse open reading frames (rORFs) that encode functional peptides. Highly conserved rORFs were found in the negative strand of viruses within the Potyviridae family, including turnip mosaic virus (TuMV), where rORF2 was shown to be essential for infection and survival. In this study, we analysed the existence and potential functionality of rORFs in plum pox virus (PPV) using mass spectrometry and mutagenesis. Unbiased mass spectrometry analyses failed to identify any rORF-encoded pep

A novel pathogenicity determinant hijacks maize catalase 1 to enhance viral multiplication and infection.

Pathogens have evolved various strategies to overcome host immunity for successful infection. Maize chlorotic mottle virus (MCMV) can cause lethal necrosis in maize (Zea mays) when it coinfects with a virus in the Potyviridae family. However, the MCMV pathogenicity determinant remains largely unknown. Here we show that the P31 protein of MCMV is important for viral accumulation and essential for symptom development. Ectopic expression of P31 using foxtail mosaic virus or potato virus X induced necrosis in systemically infected maize or Nicotiana benthamiana leaves. Maize catalases (CATs) were

RNAi targeting Myzus persicae (Hemiptera) stylet cuticle proteins reduces ZYMV transmission.

Aphids are one of the most important groups of agricultural pests due to their ability to transmit a wide range of plant viruses. The development of more sustainable and specific methods for plant virus control is an ongoing and active avenue of research. In this study, we demonstrate by feeding dsRNA to aphids via artificial diet assay, the knockdown of two M. persicae (green peach aphid) stylet cuticle proteins (ST-1 and ST-2) implicated in the attachment and retention of the non-persistent, Potyviridae family member, zucchini yellow mosaic virus (ZYMV) (Potyvirus cucurbitaflavitesselati). Whole-genome sequence analysis of zucchini yellow mosaic virus isolated from Cucurbita and Luffa.

Zucchini yellow mosaic virus (ZYMV) is a major pathogen that negatively affects cucurbit crops. To elucidate the molecular characteristics and evolutionary dynamics of ZYMV in China, ZYMV was isolated from Cucurbita and Luffa samples collected from plants exhibiting typical yellow mosaic symptoms in Tai'an, Shandong Province, and its complete genome sequence was determined. Using reverse transcription PCR combined with rapid amplification of cDNA ends, the full-length genome sequences of two isolates obtained from Luffa and Cucurbita (ZYMV-sg, GenBank accession no. High diversity of unknown picorna-like viruses in the sea.

Picorna-like viruses are a loosely defined group of positive-sense single-stranded RNA viruses that are major pathogens of animals, plants and insects. They include viruses that are of enormous economic and public-health concern and are responsible for animal diseases (such as poliomyelitis), plant diseases (such as sharka) and insect diseases (such as sacbrood). Viruses from the six divergent families (the Picornaviridae, Caliciviridae, Comoviridae, Sequiviridae, Dicistroviridae and Potyviridae) that comprise the picorna-like virus superfamily have the following features in common: a genome w

DEVELOPMENTALLY REGULATED PLASMA MEMBRANE PROTEIN of Nicotiana benthamiana contributes to potyvirus movement and transports to plasmodesmata via the early secretory pathway and the actomyosin system.

The intercellular movement of plant viruses requires both viral and host proteins. The Virome of Rhubarb Consists of Diverse Viruses Belonging to at Least Four Families.

Culinary rhubarb (Rheum spp.) comprises herbaceous perennials that are primarily clonally propagated. Like other asexually reproduced crops, rhubarb is vulnerable to virus transmission, but its virome remains poorly characterized. In April 2024, foliar virus-like symptoms including chlorotic and necrotic leaf spots, chlorosis, and stunting were observed on cultivar 'Kerwin' in the rhubarb collection at Cornell AgriTech in Geneva, NY. Symptomatic leaf tissue tested positive for tomato ringspot virus (ToRSV) in double antibody sandwich enzyme-linked immunosorbent assay using specific antibodies.
