A Multifunctionalized Potyvirus-Derived Nanoparticle That Targets and Internalizes into Cancer Cells.

Plant viral nanoparticles (VNPs) are attractive to nanomedicine researchers because of their safety, ease of production, resistance, and straightforward functionalization. In this paper, we developed and successfully purified a VNP derived from turnip mosaic virus (TuMV), a well-known plant pathogen, that exhibits a high affinity for immunoglobulins G (IgG) thanks to its functionalization with the Z domain of staphylococcal Protein A via gene fusion. We selected cetuximab as a model IgG to demonstrate the versatility of this novel TuMV VNP by developing a fluorescent nanoplatform to mark tumor

Immune response to a potyvirus with exposed amino groups available for chemical conjugation.

BACKGROUND: The amino terminus of the tobacco etch virus (TEV) capsid protein is located on the external surface of infectious TEV particles, as proposed by previous studies and an in silico model. The epsilon amino groups on the exposed lysine residues are available for chemical conjugation to any given protein, and can thus act as antigen carriers. The availability of amino groups on the surfaces of TEV particles was determined and the immune response to TEV evaluated. RESULTS: Using a biotin-tagged molecule that reacts specifically with amino groups, we found that the TEV capsid protein ha

Mutagenesis Scanning Uncovers Evolutionary Constraints on Tobacco Etch Potyvirus Membrane-Associated 6K2 Protein.

RNA virus high mutation rate is a double-edged sword. At the one side, most mutations jeopardize proteins functions; at the other side, mutations are needed to fuel adaptation. The relevant question then is the ratio between beneficial and deleterious mutations. To evaluate this ratio, we created a mutant library of the 6K2 gene of tobacco etch potyvirus that contains every possible single-nucleotide substitution. 6K2 protein anchors the virus replication complex to the network of endoplasmic reticulum membranes. The library was inoculated into the natural host Nicotiana tabacum, allowing comp

Binding immunoglobulin 2 functions as a proviral factor for potyvirus infections in Nicotiana benthamiana.

Infection of viruses from the genera Bromovirus, Potyvirus, and Potexvirus in Nicotiana benthamiana induces significant up-regulation of the genes that encode the HSP70 family, including binding immunoglobulin protein 2 (BiP2). Three up-regulated genes were knocked down and infection assays with these knockdown lines demonstrated the importance of the BiP2 gene for potyvirus infection but not for infection by the other tested viruses. Distinct symptoms of cucumber mosaic virus (CMV) and potato virus X (PVX) were observed in the BiP2 knockdown line at 10 days postagroinfiltration. Interestingly

Generation of a Bivalent Recombinant Vaccine Conferring Dual Protection Against Potyvirus and Orthotospovirus in Cucurbits.

Climate warming has facilitated the expansion of insect vectors and plant viral pathogens, leading to increased incidence of viral diseases in crops. Cucurbit crops, including cucumber (Cucumis sativus), melon (Cucumis melo), squash (Cucurbita pepo), and watermelon (Citrullus lanatus), are of major economic importance worldwide, but their production is severely threatened by viral infections. Among the most damaging viruses are zucchini yellow mosaic virus (ZYMV; genus Potyvirus), transmitted by aphids, and melon yellow spot virus (MYSV; genus Orthotospovirus), transmitted by thrips, both of w

A Modified Cas9 Scaffold Allows Extension of the Virus-Induced Gene Editing Technology to the Large Potyvirus Genus.

Plant viruses are recognized as rapid and effective vectors to deliver CRISPR-Cas reaction components into plants, a strategy termed virus-induced gene editing (VIGE). However, VIGE is limited by the host range of the viral vectors. Development of new viral vectors to target a broad range of plant species will potentially enable the delivery of the editing components to new cultivars. Potyviruses (genus Potyvirus) comprise the largest group of plant RNA viruses. The main limitation of potyviral vectors to express a non-coding RNA consists of potential insertion of stop codons that interrupt th

Molecular characterization of passion fruit yellow spot virus, a novel potyvirus infecting passion fruit in La Réunion.

Passion fruit (Passiflora edulis) leaves displaying yellow spot symptoms were collected in La Réunion. Whilst viral indexing suggested single infections with potyviruses, high-throughput sequencing revealed both single and mixed infections involving cowpea aphid-borne mosaic virus (CABMV; species Potyvirus vignae) and a previously undescribed potyvirus, for which the name passion fruit yellow spot virus (PaYSV; tentative species Potyvirus passifloraflavamaculae) is proposed. Complete genome sequences (9,626 nucleotides) of two PaYSV isolates were obtained from these samples, using RNA sequenci

Complete genome sequence and molecular characterization of a novel potyvirus (sweet potato virus H) infecting sweet potato.

Sweet potato (Ipomoea batatas [L.] Lam.), an important food crop, is extensively cultivated in more than 100 countries and regions worldwide, particularly in developing countries. In this study, a novel RNA virus, tentatively named "sweet potato virus H" (SPVH), with the proposed species name "Potyvirus hebatatae", was identified in Hainan, China. Its complete genomic sequence was obtained through small-RNA sequencing (sRNA-seq) technology and RT-PCR. The viral genome is 10,901 nt in length (excluding the 3'-terminal poly(A) tail) and encodes a putative polyprotein of 3,498 amino acids. The ge

DEVELOPMENTALLY REGULATED PLASMA MEMBRANE PROTEIN of Nicotiana benthamiana contributes to potyvirus movement and transports to plasmodesmata via the early secretory pathway and the actomyosin system.

The intercellular movement of plant viruses requires both viral and host proteins. Previous studies have demonstrated that the frame-shift protein P3N-PIPO (for the protein encoded by the open reading frame [ORF] containing 5'-terminus of P3 and a +2 frame-shift ORF called Pretty Interesting Potyviridae ORF and embedded in the P3) and CYLINDRICAL INCLUSION (CI) proteins were required for potyvirus cell-to-cell movement. Here, we provide genetic evidence showing that a Tobacco vein banding mosaic virus (TVBMV; genus Potyvirus) mutant carrying a truncated PIPO domain of 58 amino acid residues co

Dahlia Virome Analysis Reveals Multiple Novel Viruses in Mixed Infections.

Dahlia is an economically important ornamental crop in the United States and worldwide. Dahlia is primarily propagated vegetatively through bulbs, which can result in vertical transmission of and constant accumulation of viruses. In this study, mosaic and yellowing symptoms were observed in Dahlia plants in a residential garden in California. Dahlia samples were collected and sequenced using high-throughput sequencing (HTS). HTS analysis was used to explore the Dahlia virome in the symptomatic plants, which revealed the presence of several contigs with homology to known Dahlia-infecting viruse

Characterization and Construction of Full-Length cDNA Infectious Clone of a Novel BCMV Isolate in Pathogroup III.

Bean common mosaic virus (BCMV; Potyvirus phaseovulgaris) is one of the primary viruses that severely impacts the yield and quality of common beans (Phaseolus vulgaris L.) and has a worldwide distribution. Utilizing small RNA sequencing and RT-PCR validation, this study identified widespread co-infection by multiple viruses in field-collected common bean samples, with BCMV being the dominant viral species. A novel isolate, designated DY9, was obtained from these field samples. Pathotype characterization confirmed DY9 as pathotype PG-III, while previous studies reported all other PG-III members

Potyviral genome-linked protein targets NbMYB330 to attenuate hydrogen-peroxide-induced antiviral immunity in plants.

MYB transcription factors play crucial roles in orchestrating plant immunity against various pathogens. However, their regulation during virus infection and how viruses counteract these defenses remain poorly understood. In this study, we demonstrated that turnip mosaic virus (TuMV) viral genome-linked protein (VPg) interacts with Nicotiana benthamiana MYB330 (NbMYB330), an R2R3-MYB transcription factor, in the nucleus. NbMYB330 expression was activated at the early stage of TuMV infection, and NbMYB330 positively regulated host antiviral defense. NbMYB330 directly bound to the promoter region

The first complete genome sequence of Ammi majus latent virus from the new natural host culantro.

A potyvirus (isolate AMLV-CQ) infecting culantro (Eryngium foetidum L.) imported from Vietnam was identified by RT-PCR. The complete genome sequence of AMLV-CQ was determined to be 9,549 nucleotides in length. It contains a large open reading frame encoding a 3,082-amino-acid putative polyprotein, flanked by 5´ and 3´ untranslated regions (UTRs) of 77 and 226 nt, respectively. AMLV-CQ is closely related to five other completely sequenced potyviruses, sharing 68-69% nucleotide and 69-70% amino acid sequence identity. However, the coat protein (CP) gene shares 89% nucleotide and 93% amino acid s

cmv1-Mediated Resistance to CMV in Melon Can Be Overcome by Mixed Infections with Potyviruses.

Resistance to cucumber mosaic virus (CMV) strain LS in melon is controlled by the gene cmv1, which restricts phloem entry. In nature, CMV is commonly found in mixed infections, particularly with potyviruses, where a synergistic effect is frequently produced. We have explored the possibility that this synergism could help CMV-LS to overcome cmv1-mediated resistance. We demonstrate that during mixed infection with a potyvirus, CMV-LS is able to overcome cmv1-controlled resistance and develop a systemic infection and that this ability does not depend on an increased accumulation of CMV-LS in mech

Turnip mosaic virus utilizes the lipid droplet biogenesis machinery to facilitate its propagation in plants.

Lipid droplets (LDs), which are dedicated to storing neutral lipids (NLs), are dynamic organelles involved in numerous other functions, including membrane remodeling during abiotic stress. While the hijacking of LDs by animal viruses is well documented, their role(s) in plant virus infection remains largely unexplored. Similar to animal viruses, plant potyviruses reroute host proteins, intracellular membranes, and lipids to create an optimized microenvironment for their efficient viral replication compartment (VRC) assembly and movement. We therefore investigated whether potyviruses can also e

Targeting ParPMC Susceptibility Hubs in Prunus: eEF1A as a Conserved Mediator of Plum Pox Virus Infection in Nicotiana benthamiana.

Sharka, caused by Potyvirus plumpoxi (plum pox virus, PPV), is the most destructive viral disease affecting Prunus species worldwide. Two MATH domain-containing genes, ParPMC1 and ParPMC2, have been identified in apricot as host susceptibility factors required for PPV infection, but their molecular function remains unclear. Here, we investigated ParPMC functions by identifying their interacting partners and assessing their contribution to viral replication and movement. Yeast two-hybrid screening of PPV-infected apricot tissues revealed 13 ParPMC interactors involved in defence and stress resp

Molecular insights into Cassava brown streak virus susceptibility and resistance by profiling of the early host response.

Cassava brown streak virus (CBSV) and Ugandan cassava brown streak virus (UCBSV) are responsible for significant cassava yield losses in eastern sub-Saharan Africa. To study the possible mechanisms of plant resistance to CBSVs, we inoculated CBSV-susceptible and CBSV-resistant cassava varieties with a mixed infection of CBSVs using top-cleft grafting. Transcriptome profiling of the two cassava varieties was performed at the earliest time point of full infection (28 days after grafting) in the susceptible scions. Turnip Mosaic Virus-Based gRNA Delivery System for Plant Genome Editing.

Plant virus-based gRNA delivery systems offer a rapid alternative to stable transformation for CRISPR-mediated genome editing, but potyvirus-based platforms in Cas9-expressing plants are still underexplored. Mild Strain Cross-Protection in Passion Fruit.

Cross-protection is a phenomenon in which a plant prior infected with a mild viral strain can be protected from subsequent infection by severe strains of the same or closely related viruses. Since its discovery in the 1920s, this phenomenon has become an important strategy for controlling viral diseases in plants. Telosma mosaic virus (TelMV, a species of Potyvirus) is an emerging pathogen that threatens passion fruit production in subtropical and tropical regions. Here, we present a protocol for screening mild TelMV strains and their application in protecting passion fruit from severe TelMV i
