Pregnancy-specific beta-1 glycoprotein in ectopic pregnancy.

PSBG and hCG were estimated by radioimmunoassays in the serum of 19 consecutive patients undergoing abdominal surgery for suspected ectopic pregnancy. Ectopic pregnancy was verified at operation in 14 cases, and the other five patients had a hemorrhagic cyst of the corpus luteum. Demonstrable PSBG and hCG were found in all patients with ectopic pregnancy. One of them had taken bromocriptine for infertility, and in serial blood samples taken during the luteal phase both PSBG and hCG were identified on day 24 of the fertile cycle. One of the five patients with a hemorrhagic cyst of the corpus lu

Pregnancy specific beta 1-glycoprotein in human intestine.

Pregnancy-specific beta 1-glycoprotein (PSG) transcripts have been identified in a number of placental and non-placental tissues. Using a placental PSG cDNA probe to screen a normal human intestinal cDNA library we have isolated 22 hybridizing clones. These clones could be divided into four groups. Nucleotide sequence analysis showed that one group of clones correspond to functional and another group correspond to non-functional PSG cDNAs. The other two groups are homologous to the nonspecific cross-reacting antigen (NCA) and biliary glycoprotein (BGP), both of which are members of the carcino

Pregnancy-specific beta 1-glycoprotein as a screening test for at-risk pregnancies.

A serum assay was performed on 605 women for pregnancy-specific beta 1-glycoprotein (SP-1) between 31 and 34 weeks' gestation. SP-1 was also estimated for 255 of these women between 15 and 20 weeks' gestation, coincidental with maternal serum alpha-fetoprotein (AFP) screening. A clinical classification of pregnancy outcome was constructed for ech group. SP-1 values at 31 to 34 weeks' gestation in pregnancies that resulted in the birth of a small-for-dates (SFD) infant were statistically different from those in normal pregnancies (p less than 0.001). A value of 100 mg per liter was chosen as a

Pregnancy-specific beta 1 glycoprotein (SP1) in the cerebrospinal fluid.

Pregnancy-specific beta 1 glycoprotein (SP1) was assayed by Particle Counting Assay in the cerebrospinal fluid (CSF) from 26 non-neurological patients, from 190 patients with various neurological disorders and from 84 patients with malignant hemopathies. With a sensitivity limit of 0.5 microgram/l, SP1 was undetectable in normal CSF. High levels were observed in CSF from one pregnant woman with herpetic encephalitis and from another woman with post-puerperal thrombophlebitis as a result of high serum concentrations and leakage of the blood-brain barrier. SP1 was detected at low levels in the C

Production of pregnancy-specific beta 1-glycoprotein by cultured placental cells.

The synthesis of pregnancy-specific beta 1-glycoprotein (PS beta G) was studied in simian virus 40 temperature-sensitive A mutant-transformed human first trimester and term placental cells. At the permissive temperature (33 C, transformed phenotype), they produced low levels of PS beta G. At the restrictive temperature (40 C), the transformed phenotype was lost, and the production of PS beta G was greatly enhanced. The PS beta G produced by these transformed placental cells resembled the purified placental PS beta G by several criteria. Both cell and placental PS beta G bound to Concanavalin A

Trophoblastic disease monitoring: Evaluation of pregnancy-specific beta 1-glycoprotein.

Pregnancy-specific beta 1-glycoprotein (SP1) was evaluated as a potential marker protein for monitoring trophoblastic disease. Four patients with post-molar pregnancy accompanied by spontaneous titer remission and three patients with nonmetastatic trophoblastic disease were found to have regression curves for both human chorionic gonadotropin (hCG) and SP, which closely followed each other. Of three patients with metastatic choriocarcinoma, two were shown to have discordant hCG and SP1 patterns, SP1 in both cases was plateauing or rising while hCG continued to fall. Two other patients are desc

Pregnancy-specific beta 1-glycoprotein in amniotic fluid in high risk late pregnancy.

Pregnancy-specific beta 1-glycoprotein (PSBG) levels in amniotic fluid were studied by radioimmunoassay in 70 normal and 90 high risk pregnancies between 34 and 41 weeks. Most values from high risk singleton pregnancies were within the normal range. Intra-uterine growth retardation (n = 22) was associated with normal amniotic fluid PSBG levels. In twin pregnancies, the amniotic fluid PSBG values were usually above the normal mean, but within the normal range. The average ratio between amniotic fluid and maternal serum PSBG concentrations was 1:16. Amniotic fluid PSBG concentration did not corr

The diagnostic value of maternal pregnancy-specific beta 1-glycoprotein in threatened abortion.

208 serum SP1 measurements have been made in 124 women admitted at the 8th to 20th gestational week for the treatment of threatened abortion. Pregnancy ended in abortion in 85,6 p.c. of the cases with pathological serum levels, while only 16,9 p.c. of the women with normal serum concentration aborted. The results demonstrate the prognostic value of serum SP1 estimations in early pregnancy.

Pregnancy-specific beta-1 glycoprotein (SP-1) in breast carcinoma. Pathologic and clinical considerations.

Ectopic production of placental proteins by a variety of nontrophoblastic epithelial tumors is well recognized. Pregnancy-specific beta-1 glycoprotein (SP-1), a recently described placental protein, has been detected both in the serum and tumors of patients with breast carcinoma. To assess the significance of SP-1 in breast carcinoma, we stained 139 cases of primary breast carcinoma for SP-1 using the immunoperoxidase technique. Overall, 55 (40%) of breast cancers were positive for SP-1; focal positivity was also noted in normal breast epithelium and fibrocystic disease. Both intraductal (32%)

Expression of pregnancy specific beta 1-glycoprotein gene in human placenta and hydatiform mole.

Transcriptional studies of the placental protein Pregnancy Specific beta 1-Glycoprotein (SP1 or PS beta G) gene with a cDNA probe in Northern blot analysis showed 15-20 folds mRNA increase in term placenta compared with early placenta and hydatiform mole. This value parallels the SP1 amount translated in wheat germ cell-free system. We conclude that SP1 biosynthesis is regulated at transcriptional level during placental development and a similar mechanism would occur in hydatiform mole which is a hyperplastic trophoblast tumor.

Investigation of Serum Pregnancy-Specific Beta-1-Glycoprotein and Relationship with Fetal Growth Restriction.

OBJECTIVE: The most used definition for fetal growth restriction (FGR) is a fetus whose estimated weight is below the 10th percentile for its gestational age. Pregnancy-specific beta-1-glycoprotein (PSG-1) is an immunomodulator found in maternal serum during pregnancy. This study aimed to determine the serum levels of PSG-1 and clarify the potential role of this molecule in the etiopathogenesis of FGR. METHODS: Eighty women carrying fetuses with FGR and 80 healthy pregnant women were included in the study. Demographic data, laboratory values, and Doppler Ultrasonography (USG) results of all c

Monoclonal antibodies against pregnancy-specific beta 1-glycoprotein (SP1) in immunohistochemistry and radioimmunoassay.

Monoclonal mouse antibodies against pregnancy-specific beta-1-glycoprotein (SP1) have been studied for their suitability in immunoperoxidase staining and radioimmunoassay methodologies. These antibodies were useful in staining normal placentas, hydatidiform moles, invasive moles and choriocarcinomas. They showed good specificity, with minimal background staining, and will thus be superior to conventional polyclonal antisera in immunohistochemistry. However, the presently tested monoclonal anti-SP1 antibodies were found not to be suitable for radioimmunoassay.

Second trimester maternal serum pregnancy specific beta-1 glycoprotein (SP-1) levels in normal and Down syndrome pregnancies.

Maternal serum pregnancy specific beta-1 glycoprotein (SP-1) levels in the second trimester may be predictive of Down syndrome (DS). An enzyme immunoassay was used to measure SP-1 sera from 46 DS pregnancies and 117 normal control women matched for maternal age, gestational age, and length of storage. In the normal control samples, there were slight correlations between the SP-1 concentration and maternal age. The maternal serum SP-1 levels increased with each week of gestation from 15 to 20 weeks. All but one of the DS sera had SP-1 levels greater than the normal median. Using a cutoff of 2.8

Human chorionic gonadotrophin and pregnancy-specific beta-1-glycoprotein in predicting pregnancy outcome and in association with early pregnancy vomiting.

Serum concentrations of human chorionic gonadotrophin (HCG) and pregnancy-specific beta 1-glycoprotein (SP1) were measured in 99 women between the 6th and 10th gestational week, before the onset of pregnancy-threatening complications. The markers failed to predict threatening abortion appearing 9-112 days after sampling (n = 11), regardless of whether the pregnancy continued (n = 2) or ended with abortion (n = 9). Normal HCG and SP1 values were observed in 4 women who subsequently had preterm delivery. 14 women with daily vomiting had higher mean serum concentrations of HCG (p less than 0.01)

Comparison of serum human pregnancy-specific beta-1-glycoprotein 1 levels in pregnant women with or without preeclampsia.

The aim of this study was to investigate the relationship between the maternal serum levels of pregnancy-specific beta-1-glycoprotein 1 (PSG1) and preeclampsia, and to compare levels of PSG1 in pregnancies with preeclampsia and uneventful pregnancies. A case-control study was conducted in a research and training hospital. A total of 40 women with preeclampsia and 42 healthy pregnant women who were gestational age-matched were included. Serum PSG1 levels were measured using enzyme-linked immunosorbent assay. The maternal serum PSG1 levels were significantly lower in patients with preeclampsia c
