Prevotella intermedia-Derived Extracellular Vesicles Promote Pulpitis.

Although bacterial infection is the primary etiological factor of pulpitis, the underlying mechanisms remain unclear. Bacterial extracellular vesicles (BEVs) play significant roles in the pathogenesis of infectious diseases independent of bacteria. However, the involvement of BEVs in pulpitis has yet to be explored. In this study, we investigated for the first time the specific mechanisms of action of EVs derived from Prevotella intermedia (Pi) in pulpitis. Our results revealed that Pi-EVs were taken up by macrophages, which amplified their pathogenic effects, thereby inducing inflammation in

Characterization of immunoglobulin G-degrading proteases of Prevotella intermedia and Prevotella nigrescens.

Degradation of immunoglobulins is thought to be an important factor in the causation of periodontal diseases by hindering local host defenses and by providing nutrients to the periodontal microflora. In this study, we characterized the proteolytic activity against human immunoglobulin G (IgG) of 20 strains of Prevotella intermedia and Prevotella nigrescens isolated from periodontal pockets and oral abscesses. IgG degradation was studied by sodium dodecyl sulphate-polyacrylamide gel electrophoresis. All strains degraded IgG within 48 h after growth in trypticase-yeast extract medium (TY) supple

Activation of human B-lymphocytes by Prevotella intermedia.

Black pigmented Gram negative Prevotella intermedia is a potential pathogenic bacterium in periodontal disease. Lipopolysaccharides (LPS) from Gram negative bacteria are the main antigens that stimulate antibody production and cytokine synthesis. Many reports indicate a strong correlation of specific antibodies of the immunoglobulin G (IgG) class with antigens of periopathogenic bacteria in periodontal disease. The aim of this study was to measure lymphocyte proliferation and immunoglobulin (IgA, IgG and IgM) production from B-lymphocytes in response to P. intermedia stimulation. P. intermedia

Current research progress on Prevotella intermedia and associated diseases.

Prevotella intermedia is a Gram-negative anaerobic bacterium that is a common pathogen of periodontitis. Recent studies have revealed that P. intermedia is closely associated with a variety of diseases involving multiple systems. Under the action of its virulence factors such as cysteine protease and adhesins, P. intermedia has the ability to bind and invade various host cells including gingival fibroblasts. It can also copolymerize a variety of pathogenic bacteria, leading to interference with the host's immune inflammatory response and causing various diseases. In this article, we review the

Prevotella intermedia Synergistically Exacerbates Pneumonia Induced by Oral Streptococci.

BACKGROUND: The precise mechanisms of respiratory infection caused by oral anaerobic bacteria remain elusive. Unexpectedly, bacterial microbiota analysis with 16S rRNA revealed "hidden" mixed infections of anaerobic bacteria and commensal oral Streptococcus species in patients with community-acquired pneumonia. The study aimed to elucidate the mechanisms by which Prevotella intermedia exacerbates oral streptococcal pneumonia. METHODS: Oral streptococci were oropharyngeally administered with the culture supernatants of P intermedia (PiSup) in mice to assess survival, microbial burden, inflamma

Infected pericardial effusion caused by Prevotella intermedia: a rare diagnosis.

Prevotella intermedia is a Gram-negative bacterium that thrives in anaerobic environments. It presents challenges in detection through routine laboratory assays, and hitherto, there has been no documented instance of detecting this bacterium in pericardial effusion in China. Metagenomic Next-Generation Sequencing (mNGS) can boost the detection rate of this pathogen and furnish early guidance for clinical management.

Antibacterial and antibiofilm effects of iron chelators against Prevotella intermedia.

Prevotella intermedia, a major periodontopathogen, has been shown to be resistant to many antibiotics. In the present study, we examined the effect of the FDA-approved iron chelators deferoxamine (DFO) and deferasirox (DFRA) against planktonic and biofilm cells of P. intermedia in order to evaluate the possibility of using these iron chelators as alternative control agents against P. intermedia. DFRA showed strong antimicrobial activity (MIC and MBC values of 0.16 mg ml(-1)) against planktonic P. intermedia. At subMICs, DFRA partially inhibited the bacterial growth and considerably prolonged t

Gram-Negative Anaerobic Spondylodiscitis Caused by Prevotella intermedia: A Case Report.

Oral bacteria, such as Prevotella intermedia, can be a rare cause of spondylodiscitis, especially in patients who have recently undergone an esophagogastroduodenoscopy (EGD). Gram-positive bacteria are typically the most common infecting organisms in spondylodiscitis cases. However, if empirical antibiotic treatment fails, it is crucial to perform a biopsy of the infected disc and vertebral body to culture and identify any unusual pathogens, such as anaerobic bacteria. We report the case of a 43-year-old man with L5/S1 spondylodiscitis attributed to P. intermedia. Cauda equina syndrome develop

Characterization of the human immunoglobulin G Fc-binding activity in Prevotella intermedia.

Many pathogenic bacteria possess cell surface receptors which can bind immunoglobulins via the Fc portion. The aim of this study was to characterize the human immunoglobulin G (IgG) Fc-binding activity of Prevotella intermedia, a suspected etiologic agent of adult chronic periodontitis. The Fc-binding activity of P. intermedia on whole cells and on extracellular vesicles was demonstrated. Incubation of P. intermedia cells in the presence of Zwittergent 3-14 allowed complete solubilization of the Fc receptor from the cell surface. This cell envelope extract was thus used to characterize the Fc-
