Proflavine lotion is a commonly used topical antiseptic in the tropics, but its sensitizing potential was never emphasized and many who developed allergic contact dermatitis were never aware of it. In a study of 45 patients, most presented with acute or subacute dermatitis which started on the arms and legs. Concomitant cutaneous sensitivity to other medicaments and lanolin occurred in 66% of the patients.

The effect of purified proflavine and light exposure was assessed in rabbits whose eyes had been infected with one of two strains of herpesvirus. In comparing proflavine-light and placebo-light treatment, 0.1% proflavine administered twice daily for 5 days had a significant effect in suppressing herpetic eye disease, but 0.05% proflavine was less effective. Proflavine allergy is uncommon, occurring in approximately 6% of patients attending contact dermatitis clinics. Proflavine wool is used by many surgeons in the UK as a dressing that can be moulded to conform to the contours of a corrected prominent ear. It may have bacteriostatic properties. We present a case where contact dermatitis in response to proflavine developed after pinnaplasty. This caused diagnostic confusion, a lengthened hospital stay and an unsightly hypertrophic scar.

The prevalence of contact allergy in 2471 patients patch tested in Singapore was 49.2% (571/1160) in women and 49.8% (653/1311) in men (sex difference not significant). The rate appeared to increase with age (43.7% in those less than 20 years, 50.6% between 20 and 49 years, and 63.1% in those greater than 49 years). Mutagenization of measles virus with proflavine produced a temperature-sensitive mutant capable of inducing hydrocephalus following intracranial inoculation of newborn hamsters. Hydrocephalus was not produced by the parental strain or by other measles virus mutants. Thus, mutants can be the causative agents of disease not associated with the parental strain. The results dictate caution in the use and distribution of experimentally induced virus variants.
